<p><strong>Conclusion:</strong> This application note demonstrated our results from the multi-laboratory collaboration study of EPA draft Method 1621. This method shows that CIC is an excellent tool to measure AOF andscreen for PFAS in wastewater. Along with its ease of use, RFIC provides more accurate results as fluoride is well separated from the water dip, ensuring optimized automated peak integration. The method is sensitive, with MBs of 1 ng/mL and MDLs of 2.5 ng/mL, and accurate, with recoveries of 80–120%. Together, these data highlight the power of CIC in eliminating the sample matrix and measuring only the adsorbable organically bound fluorine content in samples, successfully achieving the goals outlined in EPA draft Method 1621.</p>

<p><strong>Conclusion:</strong> Combustion ion chromatography is a straightforward analytical technique to determine halogens and sulfur in environmental and solid matrices. The ceramic setup is especially suitable for CIC analysis of sample matrices with high amounts of alkali metals and/or alkaline earth metals. With the ceramic setup, the robustness of the analysis and the lifetime of consumables is drastically improved. The ease-of-use is further advanced by the possibility to inject different sample volumes – depending on the analyte concentration in the samples – to guarantee that these fit into the calibration range.</p>
<p>Overall, users of this whole validated procedure profit from easy and standardized handling, the precise determination of the analytes, automatic eluent roduction, calibration, and results calculation, low maintenance, and a single-manufacturer setup.</p>

<p><strong>Abstract:</strong> Acid deamidation can mimic the action of tissue transglutaminase in deamidation of specific peptide glutamine residues, and possibly increase the immunotoxicity of the deamidated gluten food ingredient and convey an increased risk to those with coeliac disease. Peptides with multiple deamidation sites may co-elute under the same chromatographic peak and therefore other methods of site determination are required such as ion mobility mass spectrometry, increasing confidence in profiling these post-translational modifications (PTMs).</p>

<p><strong>Abstract:</strong> In this application note, a quantification method was established for catecholamines (3-MT, DA, E, NE, MN, and NMN) using the Agilent Ultivo liquid chromatography/triple quadrupole mass spectrometer (LC/TQ) system with a newly developed Agilent InfinityLab Poroshell 120 Aq-C18 column. The Poroshell 120 Aq-C18 column shows good performance with baseline separation of these six catecholamine compounds. The validation test results give a wide linearity range of 5 to 500 pg/mL in bovine serum albumin (BSA) matrices with excellent accuracy.</p>

News from LabRulezLCMS Library Week 27, 2024

<p><strong>Alexandr Malijevský, now a full professor, joined UCT Prague in 2000 as a doctoral student. He completed his Master&#39;s degree at the Charles University Faculty of Mathematics and Physics , where he studied theoretical physics. Since 2006, he has also worked at the Institute of Chemical Processes Fundamentals  at the Czech Academy of Sciences. He conducted postdoctoral research at Imperial College London for two years  and eventually found his workplace of choice at <a href="https://gcms.labrulez.com/companies/11"><strong>UCT Prague</strong></a>, where he was appointed a full professorship at the end of 2023. The line between his personal life and his research activities is often blurred; in his free time, he likes to play chess.</strong></p>
<h5><strong>What brought you to UCT Prague from Charles University’s Faculty of Mathematics and Physics?</strong></h5>
<p>I have asked myself this question more than once, given that my research fits into physics more than chemistry. My life path started out very straight, and I inclined towards mathematics in primary school. Next came high school, where I focused on mathematics and physics, and then Charles University‘s Department of Mathematics and Physics, where I studied theoretical physics. At Charles, I became one of the many „victims“ of the beauty of probably the most elegant physical theory ever, Einstein‘s theory of general relativity, and was also charmed by the professor who lectured on this topic, Professor Bičák. I did my Master thesis with him on a topic related to the problem of the energy of a gravitational field. This research was undeniably engaging, as were my discussions with Bičák, and I always looked forward to those, even if they sometimes only happened once in a couple of months, depending on the length of his many stays abroad. We would talk about domestic and international politics, the Březin brothers, religion, music, and so on. And if there was any time left, we‘d chat a little about my calculations too. I look back fondly to those times. However, after defending my Master thesis, I wasn‘t sure if I should continue in this direction, because the theory is quite well developed. The idea that I would be able to come up with something truly new was kind of naive. That‘s when my father, who was a professor at the UCT Prague Department of Physical Chemistry, noticed my hesitation. He mentioned to me a modern theory within the statistical mechanics of fluids that seemed very promising in terms of future research possibilities. I read something about it, it piqued my curiosity, so I decided to do my doctorate at UCT Prague‘s Department of Physical Chemistry. It was a bit of a shot in the dark, but it worked out as time went on.</p>
<h5><strong>What was the hardest part of making the transition to UCT Prague?</strong></h5>
<p>I had to get used to a different environment. In short, the mentality here is different, more focused on „performances“ and a bit of PR flair (these things are not interesting to me, even today). More than once, I was criticized for „not showing up much“. Many of my colleagues did not have a problem with this. And then, the level of UCT Prague students was somewhat different, especially where mathematics was concerned, which is totally understandable. I had to adapt. For example, it took me some time to align my research within the field of food technology. After an initial misunderstanding, I understood that I should start each exercise giving colleagues a gentle mathematical introduction, and then it settled down. On the other hand, my non-chemical background and technical ineptitude sometimes showed. For example, during laboratory exercises, which I also had to lead, this was sometimes quite adventurous.</p>
<h5><strong>What was it like starting work in a completely new field?</strong></h5>
<p>At that time, no one in the Czech Republic was dealing with the aforementioned theory, the subject of my doctoral thesis, so I was not assigned a classical supervisor with focused expertise in the field. I had to read a lot at first and gain a bit more knowledge on the topic. But the hardest thing was that, at the beginning, I didn’t know what exactly to aim for, i.e. what specific issue to focus my research on. Trips abroad helped me a lot, and I always tried to absorb as much as possible during these visits. All of this, at the same time, led to the fact that my work lacked a systematic nature for quite a while. I did a lot of work yet without getting the full picture. I only started to have greater consistency in my research efforts over time.</p>
<h5><strong>Are you happy or do you regret the transition, in retrospect?</strong></h5>
<p>A person would need to live two lives, or at least parts of them, in order to be able to evaluate which of the two fundamental decisions was the right one. We don’t have this option, so we can either complain about our life choices or, on the contrary, rejoice about them in the spirit of Nietzsche’s amor fati. I have every reason to be happy. I have both scientific and pedagogical freedom here. I was never assigned to teach a class that would take me beyond my comfort zone. No one is interfering with my research. Before I left for a postdoc at Imperial College London, I was still in touch with Charles University’s Department of Mathematics and Physics, but then I definitely decided to focus on just one field and to fully devote myself to it. By the way, I have visited Charles a few times recently (for example, I gave a popularization lecture there earlier this year), and I can’t say I miss the atmosphere there.</p>
<h5><strong>How would you describe your experience at Imperial College London and what did you take away from your time there?</strong></h5>
<p>It was definitely a great experience. Very positive in many ways, because you get a glimpse of how one of the most famous universities in the world works behind the scenes. For example, the great thing was that you don‘t have to travel anywhere to meet the best scholars in your field (and not only your field!); they „come to you“. But in time you discover that all that glitters is not gold. I had my first strange experience quite early on, after I handed over an almost finished manuscript of an article to my boss (in the first weeks there, I usually spent about fifteen hours a day, including weekends, at the university). I was told I must have co-authors for a manuscript on which I worked entirely alone. With genuine surprise, I asked what the contribution of the co-authors was supposed to have been. The answer surprised me: their contribution was to the writing of the grant project from which I was funded. This is apparently a common practice, but I refused to accept this argument. The article was shelved following this discussion and published only many years later. By the way, I still don‘t understand the practice of arbitrary attribution for articles in general; I would feel awkward and uncomfortable to be attributed to a work that I hadn‘t contributed to. What if someone asks me something from that article?</p>
<p>But to return to the more positive experiences. Over time, I established the cooperation with the Mathematical Physics Group, from whom I learned a lot. This collaboration actually continues to this day.</p>
<h5><strong>What were others differences compared to our academic environment?</strong></h5>
<p>I was used to more directness. I understand academia as being a specific independent environment that should be free-spirited and healthily confrontational. But I discovered there that criticism of opinions, within seminar lectures, for example, could be taken quite offensively and as an indication of a lack of respect. The pursuit of scientific debate was sometimes mistaken for a personal attack. But I would not like to generalize too much; it could have been a completely local experience.</p>
<p>Because I still visit Imperial College regularly, I notice the developments there over the years. And I can say that I’m not always very happy, and I observe the same (delayed) trends here as well. I mean, a kind of activism that jumps on modern political issues and that sometimes stands in stark contrast to the interpretation of an academic environment as I described it above.</p>
<h5><strong>Can you introduce us to your team’s research?</strong></h5>
<p>Very generally: we deal with the description of many-particle systems. We know that matter consists of an enormous number of interacting microparticles whose motion cannot be described deterministically. However, we do not need to know the state of each individual particle to describe a given system, but we are interested in how such a system behaves collectively, i.e. as a whole. For such a description, we use the methods of statistical physics, in the language of probabilities, mean values, fluctuations, correlations, and the like. With the help of appropriate microscopic models, we can predict the macroscopic properties of the system, and this relationship between the micro- and macroworld allows us to interpret the observed phenomena. We then specifically deal with, for example, the description of the phase behaviour of systems in an extremely limited environment, which is dramatically different from our normal experience. Here, theory typically precedes experiments.</p>

<h5><strong>How is your work useful for society and what real applications does it have?</strong></h5>
<p>I am engaged in basic research, the primary motivation of which is a deeper understanding of natural laws. As for its technological application, that is a job for our engineering colleagues, for whom, it can be said, we are preparing a good terrain. Nevertheless, it could be said that our research does have the potential to be applied in modern nanotechnology.</p>
<p>Concerning my teaching, I see my primary role as motivating enthusiastic students who don’t just want a degree, but who genuinely want to pursue science and who have the capacity for this.</p>
<h5><strong>What classes do you teach?</strong></h5>
<p>I currently teach Mathematical Methods for Physical Chemistry and Advanced Methods of Mathematical Methods for Physical Chemistry, Statistical Thermodynamics, and an Introduction to the Modern Theory of Phase Transitions. These are courses for Master’s and doctoral levels students.</p>
<h5><strong>Which of them are popular among students, and which courses are rather challenging?</strong></h5>
<p>Challenging for me or for students? For me, the beginnings of lectures are the most challenging, regardless of what course it is. You are fully concentrated on solving a problem, but then you have to interrupt it and refocus on something completely different. This eventually engulfs me, so I completely refocus on the lecture and try to do my best. I usually end up with a sweaty shirt and strained vocal cords. They say that I speak quite loudly, but at least no one falls asleep. As for feedback, it depends more on the students than the specific class. In any case, whenever a student mentions your lecture in a favourable manner, it gives you satisfaction.</p>
<h5><strong>What was your primary motivation to become a professor?</strong></h5>
<p>The title of professor is (in addition to DSc.) the highest academic rank. In order to fulfil its prerequisites, you need to conduct long-term pedagogical and scientific work. And you do it because you enjoy it and you see the point of it. All the other benefits of this title, including promotion to a higher pay grade, may be nice, but for me they are not motivating enough. As for social prestige, if you look around and see who is also a full professor (including, for example, candidates for the high political appointments and the circumstances they obtained their titles), you start having some doubts about that prestige. What I am happy about, however, is a certain sense of freedom that comes from knowing that I no longer have to prove to anyone that my work is meaningful and that I am doing it well.</p>
<h5><strong>How about your personal life, what do you like to do in your free time?</strong></h5>
<p>The ideal state is for me is actually when I have no personal life. When the problem I’m working on becomes so engrossing that it wakes me up at night, and in the morning, when I can’t wait to continue solving it. Fortunately, this does not happen very often; it would not even be sustainable in the long term, to state it in modern terminology. Thus all I usually do in my free time is living normally. Or normally…I used to play chess a lot during my studies, now I am returning to it, very slowly and within my severely limited schedule. What fascinates me about chess is its parallel with real life: in both life and in chess, you are looking for (or at least you should be looking for) the optimal moves, regardless of the position you are in, trying to predict the consequences of your moves as far as possible. However, it’s often the case that there is objectively no best move; so you choose the one that is at hand, which causes the complexity on both playing fields. As a theorist, I can responsibly declare: life is an excellent model for chess.</p>

<p><strong>Introduction:</strong> Polychlorinated Biphenyl (PCBs) and the Environment </p>
<p>Polychlorinated Biphenyls (PCBs) are synthesized compounds that belong to the chlorinated hydrocarbon family of compounds. PCBs were used in a variety of industrial applications such as electrical components, plasticizers and pigments/dyes, until they were banned in 1979 by the Toxic Substances Control Act (TSCA)1. PCBs are considered to persistent organic pollutants (POPs) as they do not easily degrade in the environment. PCBs, which are classified as probable human carcinogens, can be found in the air, water, and soil and have been known to bioaccumulate in marine life. Overall, users of this whole validated procedure profit from easy and standardized handling, the precise determination of the analytes, automatic eluent roduction, calibration, and results calculation, low maintenance, and a single-manufacturer setup.</p>
<p><strong>Conclusions:</strong> Two Robust methods for EPA Method 1628 was developed to analyze 209 native and 35 labeled PCB Congeners</p>

<li>Using EPA Method 1628 guidelines and requirements, a robust method was developed with helium carrier gas.</li>
<li>Using MassHunter method translator, a faster method that meets EPA Method 1628 guidelines was developed using Hydrogen carrier gas.</li>
<li>Both methods can detect and quantitate PCB compound down to the single digit ppb levels.</li>

<p><strong>Introduction:</strong> Essential oils are typically analyzed to determine their chemical composition contributing to flavor and quality as well as to evaluate their authenticity and identify adulterants. Essential oils are often complex samples with high number of volatile and semi-volatile components that include many isomers with highly similar spectra and close Retention Indices (RIs) when separated on a single column. Hence, a comprehensive GCxGC approach is preferable to achieve an appropriate chromatographic separation for this sample matrix. In this study we used comprehensive GCxGC coupled to a high-resolution accurate mass GC/MS to identify the essential oil components. Here we also investigate approaches for comparison of the complex GCxGC essential oil data.</p>
<p><strong>Conclusions:</strong></p>
<ul>
<li>Several comprehensive GCxGC configurations and method using the high-resolution GC/Q-TOF and RFM were developed and optimized using a diesel sample.</li>
<li>Essential oils were analyzed using the optimized GCxGC approach, and the workflows for compound identification and statistical analysis were evaluated.</li>

<h4><strong>4. Shimadzu: <a href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/MP_205_GC_3_Deferiprone_ASMS_GC_poster_ba23d98e92.pdf">Ultra-sensitive dynamic headspace GC-MS/MS method for trace level quantitation of Nitrosamines in Deferiprone API</a></strong></h4>

<p><strong>Introduction:</strong> Regulatory bodies related to pharmaceutical industry have extensively investigated the presence of genotoxic impurities, called Nitrosamines (NSA), in many drugs. Deferiprone is an iron chelator used to treat patients with transfusional iron overload caused by thalassemia syndromes. This drug is admired in high dose with long term treatment. Hence it is imperative to make Deferiprone drug available with safe levels of NSA. </p>
<p>Pharmaceutical drug product which contains Deferiprone, is supposed to consume for a longer time. Maximum daily intake is of 99 mg/kg/day. Considering worst case scenario, calculated Maximum Daily Dose (MDD) will be 9000 mg/day when weight of the patient as 90 kg. Hence, it is imperative to determine NSA with Limit of Quantitation (LOQ) &lt; 0.1 ppb. Limits were calculated as per MDD of 9000 mg / day and reported in Table-1 with established LOQ’s for each NSA impurity. Developing method for determining total NSA at such low level in Active Pharmaceutical Ingredient (API) &amp; Finish Dosage Form (FDF) creates challenges in pharmaceutical industry.</p>
<p>This poster describes a validated dynamic headspace GC-MS/MS procedure for quantification of various NSAs like N-nitroso-dimethylamine (NDMA), N-nitroso-diethylamine (NDEA), N-nitroso-ethylisopropylamine (NEIPA), N-nitroso-diisopropylamine (NDIPA), N-nitroso-dipropylamine (NDPA), and Nnitroso-dibutylamine (NDBA) in Deferiprone Active Pharmaceutical Ingredient (API). </p>
<p><strong>Conclusion:</strong></p>
<ul>
<li>Dynamic headspace mode, seamlessly meet the current regulatory limits, delivering high sensitivity compared to static headspace as well as liquid injection mode for nitrosamines analysis.</li>
<li>Shimadzu GCMS-TQ8050 NX has features like a highly efficient shielded detector and superior noise reduction technology enhances sensitivity and enables quantitation of nitrosamines even at trace levels.</li>

<p><strong>Introduction:</strong> The purpose of Extractable &amp; Leachable (E&amp;L) studies in the regulatory references, is to identify the traces of potential chemical substances. Because these substances may be harmful to patient due to their toxicity, or the activity of drug product may impact because of their presence. Hence, to ensure the safety and efficacy of the drug till its shelf-life period, E&amp;L study plays an important role. Extractables are organic or inorganic entities which are released from container closure system (CCS) into an extraction medium under laboratory condition. Whereas, leachable are the foreign organic or inorganic chemical substances which are present in the packaged drug product during normal as well as accelerated stability conditions.</p>
<p>Samples of CCS will be used for identification of extractables. However, packaged drug product sample will be used for the evaluation of leachable. Ophthalmic drug product packaging material was used for this study (Figure 1) and the analysis was carried out on Shimadzu triple quadrupole system GCMS-TQ8040 NX equipment (Figure 2).</p>
<p><strong>Conclusion:</strong></p>
<ul>
<li>The migration of extractables from the CCS includes a wide range of compounds. Highly sensitive and robust techniques is required for detection at trace level. Also, Data need to be acquired in both SCAN and SIM/MRM modes to detect known and unknown extractables simultaneously without affecting sensitivity.</li>
<li>Shimadzu’s GCMS TQ-8040 NX triple quadrupole ultra fast mass spectrometer offers a SCAN speed of 20,000 amu/sec, 800 MRM transitions per second along with Advanced Scan Speed Protocol (ASSP) function, facilitates detection and identification extractables even at trace levels.</li>

<li><strong>Purpose:</strong> To demonstrate high sensitivity and robustness of a cost-effective and sustainable Helium saving injector for the analysis of polychlorinated dibenzo-pdioxins/furans (PCDD/PCDF).</li>
<li><strong>Methods:</strong> Sample analysis was carried out on the Thermo Scientific TSQ 9610 GC-MS/MS equipped with the new Thermo Scientific iConnect HeSaver-H2Safer split/splitless (SSL) injector. A sample volume of 1.5 μL was injected for chromatographic separation using a TG-Dioxin (60 m × 0.25 mm, 0.25 μm) capillary GC column with previously described conditions.1The GC-MS/MS system was equipped with an advanced electron ionization (AEI) source to increase sensitivity for analyte detection. Quantification was performed using isotopic dilution in Thermo Scientific Chromeleon Chromatography Data System software using the Dioxin Analyzer workflow.</li>
<li><strong>Results:</strong> The He saver demonstrated excellent robustness, precision, and sensitivity for PCDD/F analysis in soil extracts (analyzed with no prior clean-up) at fg levels while consuming 8 times less helium compared to the standard SSL injector.</li>

<li>FluoroMatch can be used to rapidly annotate PFAS in an automatic fashion, determine unknowns and expand annotation using an interactive visualizer. Formula prediction provides a significant benefit by providing evidence for annotation for molecules which are considered tentative using other lines of evidence, or for which the signal is too low to get quality MS/MS.</li>
<li>The FluoroMatch formula prediction algorithm is highly sophisticated and has several new or optimized approaches for formula prediction of fluorine containing compounds. With the additional use of homologous series assigning the predicted formula with the most common motifs for all members of a series, the resulting annotations are highly confident (0% false positives in this case for the four series with confident assignments).</li>

<p><strong>Introduction:</strong></p>
<p>Post-translation modifications (PTMs) on monoclonal antibodies (mAb) play an important role in the safety, efficacy, and binding of a therapeutic to its target [1]. Common examples of PTMs that seek to be identified include glycosylation and phosphorylation. One challenge in the identification of these PTMs is that dissociation techniques such as collision-induced dissociation (CID) can break apart these fragile modifications. Here, we describe the use of an electron-based dissociation technique (ExD) that supplies low-energy electrons for the fragmentation of tryptic digests that contain glycosylation at asparagine consensus sites (NXS/T). The fragmentation of trastuzumab tryptic digest on a 6545XT AdvanceBio LC/Q-TOF using CID is compared to illustrate that CID alone will fragment these labile PTMs.</p>
<p><strong>Conclusions:</strong> The Agilent 6545XT AdvanceBio LC/Q-TOF with ExD cell enables glycopeptide analysis with excellent sequence coverage assessed using MassHunter BioConfirm 12.1</p>

<p><strong>Introduction:</strong> Amino acids are organic compounds that combine to form proteins. They are typically known as the building blocks of protein. Quantitative analysis of amino acids from biological fluids is of great importance in the clinical diagnosis of inborn errors of amino acid metabolism. The assessment of amino acids in body fluids is needed for the diagnosis and treatment of metabolic diseases. We have here performed a high-throughput method for analysis of amino acids in plasma samples. The method combines classical protein hydrolysis and derivatization with fast separation by UHPLC and detection by a single quadrupole mass spectrometer</p>
<p><strong>Conclusion:</strong></p>
<ul>
<li>Liquid chromatography triple quadrupole mass spectrometry (LCMS/MS) analysis of amino acids is becoming the method of choice by more and more laboratories because of its speed, sensitivity, and increased specificity. Although LC-MS/MS instruments are more selective and sensitive than LC-MS single quadrupole instruments, LC-MS can also be used in the quantitative analysis of amino acids when the necessary conditions are provided.</li>
<li>The required sensitivity and chromatographic separation were achieved with the single quadrupole LC-MS.</li>
<li>The method can quantitatively analyze 37 Amino acids in biological fluids such as plasma in less than 20 minutes.</li>

<p><strong>Introduction:</strong></p>
<ul>
<li>MALDI mass spectrometry imaging (MSI) is a versatile technique capable of characterising lipid, polysaccharide, metabolite, pharmaceutical, protein and peptide distributions within a wide variety of samples. Recently, MSI has been combined with commercially available antibody probes containing photocleavable mass-tags (PC-MTs) which target specific proteins in what has been termed MALDIimmunohistochemistry (MALDI-IHC) MSI.</li>
<li>In this study, we show imaging of FFPE human tonsil sections on an entry level benchtop MALDI-8020 using both a multiplex of 6 antibodies and a cancer marker screening panel consisting of a multiplex of 20 antibodies at a stage step size of 30μm, demonstrating a reliable, lowcost approach with good quality results.</li>
<li>Analysis of a follicular hyperplasia tissue sample showed visible differences from normal human tonsil tissue relating to structural features and cell distributions (Fig. 1)</li>
</ul>
<p><strong>Conclusion:</strong></p>
<ul>
<li>We have successfully mapped up to 20 biomarkers with a stage step size of 30µm using a low cost, linear benchtop MALDI-TOF mass spectrometer in conjunction with the Miralys probe kit.</li>
<li>The spectra were well-resolved allowing for clear and specific imaging in both normal and clinically significant samples.</li>
<li>The Shimadzu MALDI-8020 and MALDI-8030 MALDI-TOF mass spectrometers are robust systems with class leading sensitivity and resolution capable of a wide variety of applications including, as demonstrated here, MALDI imaging. These systems can provide an<br/>invaluable contribution to any research laboratory.</li>

<li><strong>Purpose:</strong> Confident characterization of PROTAC drug metabolites for transformation site localization and soft-spot analysis.</li>
<li><strong>Methods:</strong> Drug metabolites were analyzed using MSn fragmentation data acquisition on a Thermo Scientific Orbitrap Ascend Biopharma Tribrid mass spectrometer and data interpretation in Thermo Scientific Compound Discoverer 3.3 SP3 software.</li>
<li><strong>Results:</strong> PROTAC drug metabolism was found to be extensive for the investigated compounds. Among the detected metabolites, several isomeric compounds were found where MSn fragmentation data could be used to reduce ambiguity in the transformation site localization, especially for metabolites with higher molecular weights.</li>
</ul>
<p><strong>Conclusions:</strong></p>
<ul>
<li>The utility of flexible MS data acquisition methods for metabolite identification could be demonstrated with two representative PROTAC compounds, MZ1 and dBET1, which exhibit extensive metabolism in vitro.</li>
<li>MSn fragmentation data helped to enhance the precision of transformation site localization, particularly for isomeric metabolites and those with high molecular weights.</li>
</ul>

<p><strong>Conclusions:</strong> The performance of a novel high throughput (HT) DIA workflow for label-free quantitation was demonstrated by combining an Orbitrap Exploris 240 mass spectrometer with fast capillary flow chromatography on a high throughput µPAC Neo HPLC column and Vanquish Neo UHPLC system. <p><strong>INTRODUCTION:</strong></p>
<ul>
<li>In recent years, the health benefits of mushrooms—including anti-  inflammatory effects1 and improved cognitive function2—have led to huge  growth in the number of food and supplement products containing so-  called &quot;functional&quot; mushrooms.</li>
<li>There is a wide variety of coffee products currently on the market that are blended with extracts from an assortment of different mushroom species.</li>
<li>In this study, we characterized mushroom coffee products containing up to  three mushroom species: chaga, cordyceps, and lion&#39;s mane. The chemical  fingerprints were compared with that of traditional coffee.</li>
</ul>
<p><strong>Conclusion:</strong></p>
<ul>
<li>A series of oligosaccharides were observed in mushroom coffees and  cordyceps capsules, but not chaga or lion’s mane capsules, nor in  traditional coffee. These oligosaccharides may contribute to enhanced  health benefits of mushroom coffee.</li>
<li>Trehalose disaccharide displayed variability among the mushroom  coffees, but was relatively consistent across the cordyceps, chaga, and  lion’s mane capsules.</li>
<li>Taken together, these observations suggest that the extraction process  (and/or the parts of the mushrooms extracted) governs the  oligosaccharide content more than the specific mushroom species  included in a product, though further testing is needed.</li>

<h4><strong>8. Waters Corporation: <a href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/2024_asms_sarah_characellides_828ec17fb4.pdf">Ion Mobility Separation Of Marine Natural Products: Characellides from the Deep-sea Sponge Characella pachastrelloides</a></strong></h4>

<p><strong>Introduction:</strong> <em>Characella pachastrelloides</em> was first described in 1876 as part of a collection of deep-  sea sponges.1 Deep-sea sponges are a source of unique natural products partly due to  the extreme environment they inhabit. This study looks at a group of glycolipopeptides  known as characellides that were isolated from <em>C. pachastrelloides</em> and found to be  bioactive metabolites.2,3 While HRMS can be used for some structural characterization,  full elucidation of natural product structures remains challenging. The ‘gold-standard’  method for structural characterization of natural product compounds is nuclear  magnetic resonance (NMR). However, certain compounds can be difficult to isolate or  at concentrations too low for quality NMR analysis. This study investigates if ion  mobility (IMS) can be another tool for structural assignments of these compounds.</p>
<p><strong>Conclusions:</strong></p>
<ul>
<li>The structurally similar characellide isomers A and B were able to be separated in ion mobility space by utilizing multiple passes in the Cyclic IMS device</li>
<li>The added separation power of IMS led to the discovery of two new compounds with the same molecular formula as Characellides C and D.</li>
<li>Additional related compounds were identified with molecular networking and their CCS values were calculated to give insights into the structural diversity of this unique set of compounds.</li>

<p><strong>Introduction:</strong> Introduced in Agilent ICP-MS MassHunter version 4.6, IntelliQuant for ICP-MS provides an automated semiquantitative analysis for every sample. Calibration and background correction are automatically made with no user intervention,  and data is presented in an easily understood periodic table heat map format and a comprehensive data table.</p>
<p>IntelliQuant is possible because a QuickScan can be performed when measuring each sample. The optional two second QuickScan acquires a complete mass spectrum of the entire measurable mass range, facilitating semiquantitative and Total Matrix Solids (TMS) calculations of every sample.</p>
<p>TMS levels are calculated from the IntelliQuant data. Similar to Total Dissolved Solids (TDS), TMS is the sum of the key matrix elements in each sample. Included in the TMS calculation are the elements typically considered to contribute to matrix effects, such as ionization suppression and matrix deposition on interface cones and lenses. Gas elements, including elements in acids such as HCl and HNO₃ , do not cause appreciable matrix effects in ICP-MS, so they are excluded from the calculation.</p>
<h4><strong>2. <p><strong>Introduction:</strong> Synthetic oligonucleotides are short chains (oligomers) of deoxyribonucleic acid (DNA) or ribonucleic acid (RNA) molecules. They are used in applications such as polymerase chain reaction (PCR) tests, DNA sequencing and novel therapeutic products.</p>
<p>The synthesis of oligonucleotides involves a four-step cyclic process followed by multi-step downstream purification. This manufacturing process as well as environment, packaging, and container closure systems can be potential sources of contamination leading to inorganic impurities formation.</p>
<p>To control and monitor product safety, permitted daily exposure (PDE) limits of 24 elements are defined in ICH guideline Q3D(R2)1 and USP chapter &lt;232&gt;2 . ICH Q2(R1)3 and USP&lt;233&gt; 4 describe analytical measurement requirements and procedures for the measurement of those elemental impurities. Although oligonucleotide products have not been included in those regulatory guidelines yet, the industry common practice is to follow those guidelines.</p>
<p>In this study, an analytical workflow (Figure1) was developed in accordance with the ICH/USP guidelines for the analysis of elemental impurities in an oligonucleotide sample using an Agilent 7850 ICP-MS. Data was acquired to evaluate the accuracy, specificity, reproducibility, and ruggedness of the method.</p>
<p><strong>Conclusions:</strong> The study has demonstrated the suitability of the Agilent 7850 ICP-MS for the identification and quantification of 24 elements in a synthetic oligonucleotide sample in compliance with existing USP and ICH guidelines. Instrument and method performance requirements for stability, detectability, precision, specificity, ruggedness, accuracy, and spike recovery were all achieved per the criteria given in USP &lt;232&gt;/&lt;233&gt; and ICH Q3D(R2)/Q2(R1).</p>
<p>As the development and use of oligonucleotides in therapeutic applications continue to advance, the elemental impurities analysis of oligonucleotides using 7850 ICP-MS can safeguard the product quality and safety.</p>

<p><strong>WASHINGTON-Today, May 9, the President’s Task Force on Environmental Health Risks and Safety Risks to Children is publishing the <a href="https://ptfcehs.niehs.nih.gov/features/featured-activity/task-force-reports-progress-federal-lead-action-plan">Progress Report on the Federal Lead Action Plan</a>, a comprehensive update on the government’s progress since 2018 toward reducing childhood lead exposures. The U.S. Department of Housing and Urban Development (HUD), the U.S. Environmental Protection Agency (EPA), and the U.S. Department of Health and Human Services (HHS), as co-leading members of the Task Force’s Lead Exposures Subcommittee, are leading aggressive actions to combat lead exposure.</strong></p>

<p><em>&quot;Protecting the health of vulnerable populations, especially children and families with limited resources, is paramount. Our Task Force&#39;s progress in implementing the Action Plan reflects the Biden-Harris administration&#39;s shared commitment to investing resources in lead safety programs,&quot;</em> <strong>said HUD Acting Secretary Adrianne Todman</strong>. <em>&quot;The individual programs to implement Justice40 and additional administration initiatives are complemented by the many interagency activities described in the progress report.&quot;</em></p>
<p>The <a href="https://ptfcehs.niehs.nih.gov/sites/niehs-ptfceh/files/resources/lead_action_plan_508.pdf"><strong>2018 Federal Lead Action Plan</strong></a> was released with a clear vision: to reduce childhood exposure to lead and its harmful effects. Since then, the federal government has been working to implement strategies outlined in the plan, and leveraging partnerships with states, Tribes, local communities, business, and caregivers to achieve this shared goal.</p>
<p>The progress report summarizes the significant strides made toward reducing lead exposure and improving children’s health through landmark initiatives including:</p>
<ul>
<li>Reducing lead in drinking water, land, air, food, housing, and consumer products,</li>
<li>Improving childhood lead poisoning testing to improve children’s health outcomes,</li>
<li>Enhancing lead hazard communication with partners and the public with streamlined messaging,</li>
<li>Supporting critical research that informs efforts to reduce lead exposures and health risks, and much more.</li>
</ul>
<p>The President’s Task Force is the focal point for the federal government to scope, plan, and act together for the betterment of children’s environmental health and safety. The Task Force engages <a href="https://ptfcehs.niehs.nih.gov/about"><strong>multiple government departments, agencies, and other federal partners</strong></a> to coordinate efforts to address the array of environmental and social stressors that threaten the health of children, with particular focus on areas including lead exposures, asthma disparities, chemical exposures, climate change, emergencies, and disasters. These efforts have complemented the <a href="https://www.whitehouse.gov/briefing-room/statements-releases/2021/12/16/fact-sheet-the-biden-harris-lead-pipe-and-paint-action-plan/"><strong>Biden-Harris Administration’s Lead Pipe and Paint Action Plan</strong></a>, which laid out over 15 new commitments from more than 10 federal agencies to make sure that the federal government marshals every resource and every tool it can to make rapid progress towards ensuring a lead-free future. These efforts have also complemented the President’s <a href="https://www.whitehouse.gov/environmentaljustice/justice40/"><strong>Justice40 Initiative</strong></a>, which set a goal that 40 percent of the overall benefits of certain federal investments flow to disadvantaged communities that are marginalized by underinvestment and overburdened by pollution.</p>

Biden-Harris Administration Reports Significant Progress Toward Protecting Children from Lead Poisoning

<p><strong>Physicists have been hoping for this moment for a long time: for many years, scientists all around the world have been searching for a very specific state of thorium atomic nuclei that promises revolutionary technological applications. It could be used, for example, to build an nuclear clock that could measure time more precisely than the best atomic clocks available today. It could also be used to answer completely new fundamental questions in physics - for example, the question of whether the constants of nature are actually constant or whether they change in space and time.</strong></p>

<p>Now this hope has come true: the long-sought thorium transition has been found, its energy is now known exactly. For the first time, it has been possible to use a laser to transfer an atomic nucleus into a state of higher energy and then precisely track its return to its original state. This makes it possible to combine two areas of physics that previously had little to do with each other: classical quantum physics and nuclear physics. A crucial prerequisite for this success was the development of special thorium-containing crystals. A research team led by Prof. Thorsten Schumm from TU Wien (Vienna) has now published this success together with a team from the National Metrology Institute Braunschweig (PTB) in the journal &quot;Physical Review Letters&quot;.</p>

<h4><strong>Switching quantum states</strong></h4>
<p>Manipulating atoms or molecules with lasers is commonplace today: if the wavelength of the laser is chosen exactly right, atoms or molecules can be switched from one state to another. In this way, the energies of atoms or molecules can be measured very precisely. Many precision measurement techniques are based on this, such as today&#39;s atomic clocks, but also chemical analysis methods. Lasers are also often used in quantum computers to store information in atoms or molecules.</p>
<p>For a long time, however, it seemed impossible to apply these techniques to atomic nuclei. <em>&quot;Atomic nuclei can also switch between different quantum states. However, it usually takes much more energy to change an atomic nucleus from one state to another – at least a thousand times the energy of electrons in an atom or a molecule,&quot;</em> says Thorsten Schumm. <em>&quot;This is why normally atomic nuclei cannot be manipulated with lasers. The energy of the photons is simply not enough.&quot;</em></p>
<p>This is unfortunate, because atomic nuclei are actually the perfect quantum objects for precision measurements: They are much smaller than atoms and molecules and are therefore much less susceptible to external disturbances, such as electromagnetic fields. In principle, they would therefore allow measurements with unprecedented accuracy.</p>

<h4><strong>The needle in the haystack</strong></h4>
<p>Since the 1970s, there has been speculation that there might be a special atomic nucleus which, unlike other nuclei, could perhaps be manipulated with a laser, namely thorium-229. This nucleus has two very closely adjacent energy states – so closely adjacent that a laser should in principle be sufficient to change the state of the atomic nucleus.</p>
<p>For a long time, however, there was only indirect evidence of the existence of this transition. <em>&quot;The problem is that you have to know the energy of the transition extremely precisely in order to be able to induce the transition with a laser beam,&quot;</em> says Thorsten Schumm. <em>&quot;Knowing the energy of this transition to within one electron volt is of little use, if you have to hit the right energy with a precision of one millionth of an electron volt in order to detect the transition.”</em> It is like looking for a needle in a haystack – or trying to find a small treasure chest buried on a kilometer-long island.</p>
<h4><strong>The thorium crystal trick</strong></h4>
<p>Some research groups have tried to study thorium nuclei by holding them individually in place in electromagnetic traps. However, Thorsten Schumm and his team chose a completely different technique. <em>&quot;We developed crystals in which large numbers of thorium atoms are incorporated,&quot;</em> explains Fabian Schaden, who developed the crystals in Vienna and measured them together with the PTB team. <em>&quot;Although this is technically quite complex, it has the advantage that we can not only study individual thorium nuclei in this way but can hit approximately ten to the power of seventeen thorium nuclei simultaneously with the laser – about a million times more than there are stars in our galaxy.&quot;</em> The large number of thorium nuclei amplifies the effect, shortens the required measurement time and increases the probability of actually finding the energy transition.</p>
<p>On November 21, 2023, the team was finally successful: the correct energy of the thorium transition was hit exactly, the thorium nuclei delivered a clear signal for the first time. The laser beam had actually switched their state. After careful examination and evaluation of the data, the result has now been published.</p>
<p><em>&quot;For us, this is a dream coming true,&quot;</em> says Thorsten Schumm. Since 2009, Schumm had focused his research entirely on the search for the thorium transition. <h4><strong>The dream of the atomic nucleus clock</strong></h4>
<p>This marks the start of a new exciting era of research: now that the team knows how to excite the thorium state, this technology can be used for precision measurements. <em>&quot;From the very beginning, building an atomic clock was an important long-term goal,&quot;</em> says Thorsten Schumm. <em>&quot;Similar to how a pendulum clock uses the swinging of the pendulum as a timer, the oscillation of the light that excites the thorium transition could be used as a timer for a new type of clock that would be significantly more accurate than the best atomic clocks available today.&quot;</em></p>
<p>But it is not just time that could be measured much more precisely in this way than before. For example, the Earth&#39;s gravitational field could be analyzed so precisely that it could provide indications of mineral resources or earthquakes. The measurement method could also be used to get to the bottom of fundamental mysteries of physics: Are the constants of nature really constant? Or can tiny changes perhaps be measured over time? <em>&quot;Our measuring method is just the beginning,&quot;</em> says Thorsten Schumm. <em>&quot;We cannot yet predict what results we will achieve with it. It will certainly be very exciting.&quot;</em></p>
