<p><strong>Diane Cotter knew cancer was a risk of her husband’s job. Paul had been a firefighter for 27 years, and they had been advised of not only the safety risks, but also the health implications from exposure to diesel exhaust and products of combustion. But when Paul, a 55-year-old in great shape, who ate healthy and took care of himself, received a prostate cancer diagnosis, Diane suspected something other than smoke was the culprit.</strong></p><p>A hunch drew her to consider his gear — the protective clothing that had kept him safe as he rushed into burning buildings, saved lives and put out fires. As it turned out, that gear may have been a cause of his cancer.</p><p>Firefighter gear is complex — it has to be able to withstand both fire and water, and to resist extreme temperatures. The multiple layers — a thermal liner, a moisture barrier and an outer shell — must pass the National Fire Protection Association standards. Research had been done to study the gear and its textiles, but little existed about the chemical coatings on the gear that make it so effective.</p><p>Determined to learn more, Diane began digging. She emailed firefighters, activists, manufacturers. Many tried to shrug her off, but she was persistent. She pored over safety recalls, international regulations. Finally, she received a tip: Ask if the chemicals PFOA or PFOS are present in the gear.</p><p>Her research led her to believe that yes, chemicals known collectively as PFAS must be present in the gear, but manufacturers refused to provide information about the chemical content. And so she began looking for someone who could independently assess the clothing and give her an answer.</p><p>“<i>What we didn't know was how much of these coatings were on the turnout gear and we didn’t have the chemical breakdown,” she says. “And we had no one to tell us what was in the coatings until we found Dr. Peaslee.</i>”</p><figure class="image image_resized" style="width:50%;"><img style="aspect-ratio:687/375;" src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/University_of_Notre_Dame_Matt_Cashore_To_Protect_the_Protectors_Graham_Peaslee_professor_of_physics_e021134945.jpg" alt="University of Notre Dame: Graham Peaslee." srcset="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/thumbnail_University_of_Notre_Dame_Matt_Cashore_To_Protect_the_Protectors_Graham_Peaslee_professor_of_physics_e021134945.jpg 245w," sizes="100vw" width="687" height="375"></figure><p><a target="_blank" rel="noopener noreferrer" href="https://physics.nd.edu/people/faculty/graham-peaslee/"><strong>Graham Peaslee</strong></a> is a professor of <a target="_blank" rel="noopener noreferrer" href="https://physics.nd.edu/"><strong>experimental nuclear physics</strong></a><strong> </strong>at Notre Dame. Peaslee had devoted 20 years of his career to fundamental physics research and teaching, but in 2005 a lake near his home was found to be contaminated with chemicals from flame retardants. He began researching the prevalence of toxic chemicals and was thrust into a merger of science and policy, of chemicals and human health.</p><p>The experience tipped him off that harmful chemicals could be lurking in other unlikely places. He zeroed in on a class of fluorinated compounds, per- and polyfluoroalkyl substances, referred to as PFAS, which have been linked to thyroid disease, hypertension, immunosuppression, low birthweight and kidney and testicular cancers.</p><p>“<i>This is a really persistent class of chemicals,</i>” says Peaslee. “I<i>t gets in the bloodstream; it stays there and can accumulate in the body. There are diseases that correlate with its presence, so we really don’t want this class of chemicals out there.</i>”</p><p>Peaslee explains that these chemicals, also known as PFOA, PFOS or GenX, are particularly dangerous because they don’t break down. They move from commercial products to landfills to soil to groundwater. They can be ingested by livestock, or end up in drinking water, as was the case in Little Hocking, Ohio, a community downriver from a DuPont plant that leaked PFAS into soil and water for more than 50 years. Similar stories come from communities around military bases where firefighting foam laden with the compounds eventually made it into drinking water.</p><p>To identify markers of PFAS, scientists perform a complicated and lengthy chemical method. It can analyze 100 times faster than the chemical method for the same sort of detection,</i>” he says. “<i>In that sense it's a development that nobody else has been able to do, because nobody uses nuclear science to screen for this type of chemical. While it doesn’t replace them, it is considerably faster than the chemical approaches and can be used to screen for the fate and transport of these chemicals in the environment.</i>”</p><p>His method got results in minutes, rather than hours, which allowed him to start testing many everyday items. Because PFAS are used in water, flame and oil resistance, they’re frequently used in consumer goods ranging from raincoats to nonstick pans to carpet to fast food wrappers. One of Peaslee’s studies discovered the presence of these chemicals in wrappers at 20 different fast food chains. Companies in the outerwear, furniture and food packaging industries are already making adjustments.</p><p>In the midst of his extensive testing for PFAS on products, Peaslee received a plea for help.</p><p>“<i>I received an email from Diane, which was a heartfelt story about her husband's bout with cancer and that nobody would test her firefighting gear. I felt obliged to go and see if there was anything to her claims,</i>” he says.</p><p>“<i>The firefighters that Diane knew volunteered several sets of used and new gear from around the country, and we started getting dozens of sets of turnout gear delivered to our laboratory. When we looked at the gear, there was a remarkable consistency in how they were fabricated and the levels of fluorine that we were seeing in each of the layers of the gear. It was startling high,</i>” he says. “<i>There were perfluorinated compounds being used on the inside and the outside of this firefighting gear.</i>”</p><p>Digging further into the issue, Peaslee began testing gear that was new, used, washed and unwashed, to help determine the extent of the issue. He wanted to educate firefighters about the potential hazards.</p><p>“<i>Knowledge is empowerment,</i>” he says. “<i>These firefighters, men and women, are being exposed to something that they don’t know about. When they know about it they can mitigate the risks. They can treat the gear with respect and use it when they need it and then they can keep it separate from their lives when they don’t need it. The Cotters also continue to advocate for change for firefighters, now with Peaslee’s findings as backing for their cause.</p><p>Diane says, “<i>What we know is that the chemical coatings have been used for at least 20 years. We never knew how much until Dr. Peaslee discovered, with his own specific testing methods, the amounts that are used. We've been told trace amounts, which, to us, is trying to minimize the issue, when in fact we really should be sounding the alarm on the issue.</i>”</p><p>Now certain that there are toxic chemicals in the gear, Diane believes some manufacturers may have knowingly withheld that information. Peaslee was able to provide undeniable evidence that now needs to be addressed. Because of him, she says, the discussion has begun.</p><blockquote><h4><strong>“Scientists have to learn to communicate when there's an issue like this, and not just do it in the safety of a laboratory.”</strong></h4></blockquote><p>Peaslee is quick to shrug off any praise and insists he’s doing what all scientists should.</p><p>He says, “<i>Scientists have to learn to communicate when there's an issue like this, and not just do it in the safety of a laboratory. But to get out and talk to people about what may in fact impact them.</i>”</p><p>“<i>The reason I came to Notre Dame was that they had a facility that was different from what’s available anywhere else in the world. It has cutting-edge research facilities,</i>” he says. “<i>Their mission states that they are a university that wishes to give back to its community. I identify strongly with that mission.</i>”</p><p>As research and awareness on PFAS has grown, so have efforts to reduce and eliminate their use. European firefighters are already moving away from PFAS in their gear.</p><p>In February, the EPA announced a maximum contaminant level for drinking water and also proposed enforcement tools for parties leaking PFAS into the environment.</p><p>In June, a bipartisan amendment passed that required the military to phase PFAS chemicals out of firefighting foam by 2023 — a response to alarming amounts of PFAS in contaminated drinking water at more than 175 military bases to date.</p><p>The state of Washington was the first to pass legislation to restrict the use of PFAS. In March 2018, the Healthy Food Packaging Act banned PFAS chemicals in paper food packaging. Another bill championed by the Washington State Council of Fire Fighters bans firefighting foams with added PFAS and requires gear containing the chemical to carry a warning. By 2020, the state will designate a list of other consumer products to remove PFAS.</p><p>Meanwhile, several corporations have independently gone PFAS-free. Whole Foods Market pulled takeout packaging that contained PFAS, and Kaiser Permanente, one of the largest health care providers in the U.S., and Google have banned the use of PFAS in finishes and materials it uses for building projects.</p><p>Today, Paul is cancer-free, but Diane isn’t willing to give up the fight to protect other firefighters.</p><p>“<i>My husband leaving the fire department was probably the hardest thing we’ve had to endure. Even harder than the cancer, but we’re blessed because he’s survived it,</i>” she says. “<i>At this point in our lives, we’re fighting for change because it’s that important. We have family and friends, not just here in the United States, but across all the lands, across the planet, in Australia, Europe, Italy, who are asking the same questions. <p><strong>IOCB Prague is celebrating two significant achievements in the field of scientific research. The project seeks to overcome longstanding obstacles in enzyme development. Currently, it is impossible to accurately predict the functions of newly discovered enzymes or to generate enzymes for specific reactions. Dr. Pluskal has been awarded an ERC Consolidator Grant to pursue this project, securing more than 2 million euros for the next five years.</p><p><i>“Progress in this field has been slow and remains highly limited. Enzymes play a key role in biotechnology because they catalyze chemical reactions that are often difficult to achieve through organic synthesis.</p><p>With six active ERC grants and a total of nine awarded in its history, IOCB Prague is among the top Czech institutions in obtaining this prestigious European funding.</p><p>In addition to the ERC grant, Dr. Pluskal has also secured a position in the European Molecular Biology Organization (EMBO). Thermo Fisher Scientific: </strong><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_003158_tea_icp_ms_icap_mtx_halogens_an003158_na_en_ba3b6556c8.pdf"><strong>Analysis of fluorine, chlorine, bromine, and iodine at low levels using triple quadrupole ICP-MS</strong></a></h4><ul><li>Application</li></ul><p><strong>Goal</strong></p><p>This application note will demonstrate an analytical workflow for the determination of the halogens fluorine, chlorine, bromine, and iodine at low levels using triple quadrupole ICP-MS.</p><p><strong>Introduction</strong></p><p>Inductively coupled plasma mass spectrometry (ICP-MS) is widely used as an analytical technique for the trace level quantification of elemental impurities in the range of different matrices due to its specificity, detection capability, and ability to provide isotopic information.</p><p>The detection sensitivity in ICP-MS is mostly determined by the first ionization potential (i.e., the energy required to remove an electron from the valence shell) and mass for a given element. In an argon plasma, complete ionization is typically only achieved for elements with ionization potentials below 6 eV, such as alkaline and alkaline earth elements. Typical transition metals, precious metals, and rare earth elements still ionize at close to 100%. However, semi-metals and in particular non-metals, like silicon, sulfur, or halogens, will only poorly ionize. At the same time, their signals are found in the low mass range (often below m/z 50), where many ions are lost in the transition through the mass spectrometer. In addition, other factors may significantly impact the analysis, for example, isobaric and polyatomic interferences leading to bias on the most abundant isotope for a given element, and hence forcing analysts to often select a less sensitive, but interference-free alternative. Especially polyatomic interferences are abundant in the low mass range and may render the analysis using ICP-MS nearly impossible.</p><p>The analysis of halogens is of importance in a variety of sample matrices pertaining to different industries, such as environmental monitoring, pharmaceuticals, oil and gas, and industrial applications such as Li-ion battery and renewable fuels. Ion chromatography (IC) is a commonly used technique for analysis of halogens, whereas argon plasma-based techniques, like ICP-OES or ICP-MS, have limited applications for this purpose. However, its outstanding characteristics as an element selective detection system have led to the development of applications highly complementary to chromatographic techniques, for example, for the detection of PFAS.</p><p>This note describes the analytical workflows developed with the Thermo Scientific™ iCAP™ MTX ICP-MS to mitigate these analytical challenges associated with the analysis of halogens using ICP-MS.</p><p><strong>Conclusion</strong></p><p>This document highlights how triple quadrupole ICP-MS, such as the iCAP MTX ICP-MS operated in TQ-O2 mode, can be used for accurate and precise quantification of challenging analytes such as chlorine, bromine, and iodine, which can be analyzed directly using optimized instrument conditions. It further highlights the feasibility of the iCAP MTX ICP-MS for indirect but accurate and precise determination of fluorine after complex formation with barium under optimized and controlled conditions. It further summarizes details of iodine (127I) analysis using helium as cell gas in SQ-KED mode. The analytical data and observations made during this study strongly suggest that the iCAP MTX ICP-MS equipped with dedicated mass flow controllers (MFCs) for introduction of pure oxygen and pure helium as cell gases is a powerful tool to accomplish the otherwise challenging analysis of halogens.</p><h4><strong>3. Shimadzu: </strong><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_00724_en_1_98d698e2ba.pdf"><strong>Observation of Joined Metal and Rubber Parts Using a Microfocus X-Ray CT System</strong></a></h4><ul><li>Application</li></ul><p><strong>User Benefits</strong></p><ul><li>The progression of rubber deterioration, where rubber is in contact with metal, can be evaluated non-destructively.</li><li>Even locations with no visible abnormalities can be inspected non-destructively to help improve or maintain quality.</li></ul><p><strong>Introduction</strong></p><p>Because metal and rubber materials have different properties, using them together can achieve high strength, vibration-damping effects, lighter weight, tight seals, and various other characteristics that cannot be achieved with only one or the other. Therefore, mechanical parts with rubber joined to metal<br>are widely used in automotive, industrial machinery, and other fields. However, joining parts with dissimilar properties can cause defects from heat, vibration, or other factors present during manufacturing processes or product use. In particular, rubber can potentially deteriorate at metal contact surfaces due to high metal temperatures without showing any symptoms significant enough for the deterioration to be discovered. Consequently, non-destructive inspections are required to evaluate deterioration and defects before they appear.</p><p>X-ray CT systems offer one type of non-destructive inspection. They can visually show the internal status of joined metal-rubber parts in order to assess the position and shape of internal defects without damaging the part. This article describes using an inspeXio SMX-225CT FPD HR Plus microfocus X-ray CT system (Fig. 1) to observe rollers made with rubber joined to metal.</p><p><strong>Conclusion</strong></p><p>his article describes using an inspeXio SMX-225CT FPD HR Plus microfocus X-ray CT system to scan rollers made of rubber bonded to metal.</p><p>The system enables non-destructive discovery of rubber deterioration and debonding, regardless of whether or not the exterior appears normal. The progression of rubber deterioration can be observed by detailed observations of cross-sectional images or by comparing the volumes of deteriorated portions. Color-coding the deteriorated or debonded areas can &nbsp;provide a 3D understanding of the position and shape of those areas. Thus, X-ray CT imaging provides a useful way to inspect the quality of parts made with rubber bonded to metal.</p><h4><strong>4. Anton Paar: Anton Paar’s Multiwave 5000 Helps Colombian Industry Grow: Inside Confia’s Food Analysis</strong></h4><ul><li>Other</li></ul><p><strong>Confia, an analysis service laboratory, plays a significant role in supporting the growth of Colombian industry. Central to its operations is the use of Anton Paar’s Multiwave 5000, a key piece of equipment that enhances efficiency and precision.</strong></p><p>This technology is particularly crucial in the food industry, where Confia focuses much of its project work. Multiwave 5000 facilitates microwave-assisted acid digestion, ensuring fast and efficient sample preparation across as many as 60 food matrices per month. This process is essential for accurately determining a range of elements, including sodium, magnesium, calcium, and various heavy metals in a subsequent analysis.</p><h5><strong>Q: Can you introduce yourself and describe your role at Confia?</strong></h5><p>A: I’m David Moyano, a chemist with 17 years of experience in fields like food, pharmaceuticals, and environmental. However, its effectiveness is limited by sample preparation challenges for solid samples like soils and microplastics. Traditional methods include sample preparation, such as digestion, which is time-consuming and involves reagents, like acids, contributing to measurement uncertainty and higher carbon footprints. Slurry sampling allows direct analysis of suspensions, offering a more efficient alternative. However, maintaining suspension stability is challenging, requiring robust autosampler systems to streamline the process and enhance analytical performance.</span></p><h5><span lang="EN"><strong>Results</strong></span></h5><p><span lang="EN">We present a novel autosampler extension for slurry sample introduction into GF-AAS. This system ensures suspension stability with a stirring device and closed vessels to prevent evaporation and contamination, incorporating a cooling unit to reduce solvent and analyte losses. It installs and removes in minutes without additional connections. Validation with cadmium analysis in BAM-U110 (Soil) and BAM-H010 (ABS) showed high reliability. For BAM-U110 (Soil), we achieved recovery rates of 94 %&nbsp;±&nbsp;13 % in water suspension. The recovery rate for BAM-H010 (ABS) was 104 %&nbsp;±&nbsp;11 % in acetonitrile suspension. Materials and methods</strong></h3><h4><strong>2.1. Materials, reagents and standards</strong></h4><p>For the calibration, a single element cadmium standard (<i>ρ</i>(<i>Cd)&nbsp;=</i>&nbsp;1000&nbsp;mg&nbsp;L<sup>−1</sup>, Certipur, Merck KGaA, Germany) was used. For the dilution steps, type I reagent grade water obtained from a Milli-Q-System was used (&gt;18.2&nbsp;MΩ&nbsp;cm; Merck Millipore, Germany). To stabilize the ionic standard, double sub-boiled nitric acid (<i>ω</i>(HNO<sub>3</sub>)&nbsp;=&nbsp;65 %) was used. As modifier for the determination of Cd via high resolution-continuum source-graphite furnace atomic absorption spectrometry (HR–CS–GFAAS), palladium, and magnesium ICP-MS standard solutions were used (<i>ρ</i>(Mg)&nbsp;=&nbsp;10000&nbsp;mg&nbsp;L<sup>−1</sup>; <i>ρ</i>(Pd)&nbsp;=&nbsp;10000&nbsp;mg&nbsp;L<sup>−1</sup>, Certipur, Merck KGaA, Germany). To characterize the newly developed autosampler system, two different reference materials were used: BAM-H010 (acrylonitrile butadiene styrene (ABS), granulate, Federal Institute for Materials Research and Testing (BAM), Germany) and BAM-U110 (contaminated soil, BAM). To obtain small plastic particles for the ABS material, two steps (500&nbsp;μm and 150&nbsp;μm sieves) were carried out with a centrifugal mill (ZM 200, Retsch GmbH, Germany) under liquid nitrogen. As sample vials, HPLC vials with PTFE/silicon septa were used. The organic solvents used (methanol (MeOH, LC-MS grade, 99.95 %), ethanol (EtOH, for analysis, 99.9 %), acetonitrile (ACN, LC-MS grade, 99.95 %)) were obtained from Th. Geyer (CHEMSOLUTE, Th. Geyer GmbH &amp; Co. KG., Germany).</p><h4><strong>2.2. For this purpose, four brushless motors (Type 0703B 15000&nbsp;KV, Racerstar Electronic Technology Co., Ltd., China) are utilized as magnetic stirrers by gluing neodymium magnets (diameter 3&nbsp;mm, height 1&nbsp;mm) on top of them. The modified stirrer units are placed under the positions where the sample is taken by the injector of the autosampler. To protect stirrer units from possible spills without reducing the magnetic field, the motors are covered by a thin glass-reinforced epoxy laminate layer (FR-4). An ATtiny85-S20U microcontroller (Microchip Technology Corp., USA) is used to define the timings and control the brushless motors. To avoid contamination nickel wire pieces (99.2 %, Evek GmbH, Germany) of different diameters (1&nbsp;mm and 2&nbsp;mm) and lengths (4 mm–9&nbsp;mm) are used as stirring bars. For easy usage, the autosampler is designed to hold standard 1.5&nbsp;mL HPLC vials. To keep these vials stable and avoid the lifting of the vessels the sample turning table was designed in three layers – PVC/silicon/PVC. The two PVC layers hold the vials in position while the silicon layers prevent them from being lifted after the sample uptake of the needle. As a further modification, a hypodermic needle was installed onto the autosampler to allow the septum of the used HPLC vials to be penetrated and keep the low dead volume within the system. Further, by cooling the aluminum autosampler and using closed HPLC vials the possible loss of solvents is drastically reduced and the time for measurements can be extended without the manual closing of vials with <i>e.g.</i>, parafilm as reported by Metzger et al. [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0003267024012613#bib13"><span>13</span></a>] As a chiller, a Cool-Care 8–16 unit (Van Der Heijden Labortechnik GmbH, Germany, temperature range 5&nbsp;°C–25&nbsp;°C) was used. The implementation of a photoelectric sensor enables precise initiation of the motors, thereby minimizing idle rotation time to prolong motor lifespan and prevent overheating. The sensor is connected to an Arduino NANO (ATMEGA328p) as a control unit between the motor controller and the sensor. The system is designed in a way that it can be changed within a few minutes back to the standard AAS autosampler. This allows a fast, easy-to-use, and reproducible way to analyze slurry samples. The developed autosampler can also be used without the stirring, to avoid the evaporation of solvents and thus, increase the accuracy of the utilized analysis method. In addition, the incorporated fluidic system can be also used for heating, <i>e.g.</i>, trigger reaction before the sample is taken. This wide range of possibilities in a highly automated fashion makes this autosampler perfectly suited for routine measurements.</p><h4><strong>2.3. Instrumental analysis</strong></h4><p>The determination of cadmium was performed on an Analytik Jena contrAA 800 HR–CS–GFAAS system (Analytik Jena GmbH, Germany). Pyrolytically coated graphite tubes with PIN platform (Analytik Jena) were used. The temperature program applied as well as the injection sequence were adapted from Borges et al. [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0003267024012613#bib14"><span>14</span></a>] Accordingly, the utilized injection loop consisted of 10&nbsp;μL of ultra-pure water, 25&nbsp;μL sample, and 5&nbsp;μL of the modifier mixture (<i>ρ</i>(Pd)&nbsp;=&nbsp;1.0&nbsp;g&nbsp;L<sup>−1</sup>, <i>ρ</i>(Mg)&nbsp;=&nbsp;0.6&nbsp;g&nbsp;L<sup>−1</sup>). Sample cooling unit - temperature profile characterization</strong></h4><p>For the characterization of the temperature profile of the sample cooling unit (see <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0003267024012613#fig1"><span>Fig. 1</span></a> b), an infrared camera (VarioCam HR, InfraTec GmbH Infrarotsensorik und Messtechnik, Germany) with a 30&nbsp;mm lens was used. This has a focal plane array (FPA) with an uncooled microbolometer detector consisting of 640 x 480 pixels. The camera covers a spectral range from 7.5&nbsp;μm to 14&nbsp;μm. Measurements were taken in the calibrated temperature range from −40&nbsp;°C to +120&nbsp;°C. The noise equivalent temperature difference (NETD) is 50&nbsp;mK at 30&nbsp;°C. The measurement uncertainty is ±1.5&nbsp;K or ±2 % for temperatures above 100&nbsp;°C. The data was recorded at 2&nbsp;Hz and analyzed using the IRBIS Professional 3.1.80 thermography software.</p><p>The thermograms were recorded with a lateral geometric resolution of 0.5&nbsp;mm at 65&nbsp;cm between the IR camera and the measurement object.</p><p>For the measurement of the reflective aluminum surface, emissivity stickers from Testo SE &amp; Co. KGaA with a defined emissivity (<i>ɛ</i>) of 0.95 were used to measure the aluminum reflective surface as reference points.</p><h3><strong>4. Conclusion and outlook</strong></h3><p>The presented newly developed autosampler extension for GF-AAS is a promising tool for streamlining arduous and error-prone sample preparation, particularly the digestion of solid samples. This extension offers a cost-effective alternative for the precise, accurate, economical, and rapid determination of metal contents in solid samples. The system's flexibility and adaptability are evident within the two presented matrices, as it can be easily tailored to different matrices by simply changing the solvent. Furthermore, the system is designed to facilitate temperature control of samples and employs closed vessels, which are readily available from various vendors, enabling easy use of organic solvents. This adaptation of the AAS enables access to emerging markets, such as the investigation of electronic scrap, which is challenging to address using conventional techniques due to its complex composition of glass, polymers, and various other matrix components that are difficult to digest. Additionally, this setup could enable further extensions in the analysis of organic fluorine in the form of adsorbable organic fluorine (AOF) using activated carbon suspensions in the future This capability enables comparison and complementary investigations in the increasingly concerning field of emerging pollutants, specifically per- and polyfluorinated substances (PFAS).</p>

Two Postdoctoral Positions in Lipidomics and Glycomics

<p><strong>Customers often want to know the amount of “dead volume” in a connection. However, most people who ask about dead volume actually want to know how much internal volume exists within the connection. What exactly is the matter with these terms?</strong></p><p>The dead volume is that portion of the void volume that is out of the flow path.</p><p>Swept Volume is that portion of the void volume in a connection directly in the fluid pathway.</p><p>Void Volume is simply another way of describe the total internal volume. It is defined as any space within connection into which fluid can flow.</p><p><strong>So a simple equation applies: Void Volume = Dead Volume + Swept Volume</strong></p><p>And why are we so interested in dead volume? Especially in capillary connections, it can cause some very undesirable chromatographic effects, including:</p><ul style="list-style-type:disc;"><li><strong>Analysis Delay</strong></li><li><strong>Broadened Peaks</strong></li><li><strong>Poor Resolution</strong></li><li><strong>Sample Carry-Over</strong></li><li><strong>Split Peaks</strong></li><li><strong>Gas Collection&nbsp;</strong></li></ul><p>Therefore, in addition to keeping the volume inside the connection to a minimum, all dead volume should be removed from the connection if possible. Finally, in capillary connections, the preferred relationship could be expressed as follows:</p><p><strong>Void Volume = Swept Volume (and should be REALLY small!)</strong></p><p>By matching the tube ID as closely as possible to the diameter of the thru-holes in your equipment, you will ensure that most of the void volume will be swept volume. This diameter adjustment also helps to reduce the amount of turbulence that the fluid experiences as it passes through the connection.</p>
