Its popularity is due to the positive impact it has on anode cyclability and reduction of irreversible capacity. Small amounts of moisture in Li-ion batteries can significantly decrease performance and lifetime. Therefore, moisture levels in electrolyte solutions and their additives are controlled for use in manufacturing. Until recent years, accurate moisture quantification in vinylene carbonate samples was difficult due to side reactions. With the commercial development of alcohol-free coulometric Karl Fischer reagents, accurate determination is now much more accessible to end-users.</p><h4><strong>EXPERIMENTAL</strong></h4><p>This analysis was performed using an <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=220&amp;search=852+titrando"><strong>852 Titrando</strong></a> (2.852.0060) instrument, equipped with the electrodes listed in Table 1.</p><h4><strong>CONCLUSION&nbsp;</strong></h4><p>During battery manufacturing, control of all moisture present is important to guarantee adequate performance in the finished product. More easily quantifying the moisture contribution from additives such as vinylene carbonate makes this a much simpler task. The introduction of alcohol-free Karl Fischer reagents accomplishes this effectively, allowing the generation of accurate and repeatable moisture data—even for a difficult sample matrix.</p><h3><strong>3. A particularly powerful approach is non-targeted metabolomics, which aims to analyze not only known metabolites but also unknown compounds without prior assumptions to reveal unexpected chemical differences between samples. Quadrupole time-of-flight mass spectrometry (QTOFMS), with its accurate-mass capability, is well suited for this purpose. However, the large volume of generated data creates challenges for feature identification and data comparison.&nbsp;</p><p>This article presents a case study in which LabSolutions Insight Profiler, a software platform for non-targeted analysis, was used to characterize compounds in beer and beer-like beverages. Peak detection, retention-time alignment, and library searching were performed across multiple datasets. Multivariate analysisrevealed compound profiles that likely reflect the differences in raw materials and manufacturing processes, demonstrating the utility of the approach for beverage characterization.</p><h4><strong>LabSolutions Insight Profiler&nbsp;</strong></h4><p><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=1&amp;search=LabSolutions+Insight+Profiler"><strong>LabSolutions Insight Profiler </strong></a>is software for batch processing of LC-QTOFMS data that enables non-targeted analysis through an intuitive workflow. It readily supports easy spectral feature detection, alignment, statistical processing, filtering, list screening, MS/MS spectra-based library searching, principal component analysis (PCA), displaying volcano or box plots, and more. Fig. 1 shows the data analysis workflow.</p><h4><strong>Analysis Conditions&nbsp;</strong></h4><p><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=1&amp;search=nexera+X3"><strong>Nexera X3 UHPLC</strong></a> and<strong> </strong><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=1&amp;search=9030"><strong>LCMS-9030 systems</strong></a> were used as the analytical instruments. The LC method included in the LC/MS/MS Method Package for Primary Metabolites was used as the method. The data-dependent acquisition (DDA) mode was used to simultaneously acquire precursor m/z and MS/MS data. Table 2 shows the analysis conditions.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>In this study, compounds in various beer and beer-like beverages were analyzed by non-targeted analysis using an LCMS-9030 quadrupole time-of-flight mass spectrometer system. Analyzing compounds characteristic of each sample with LabSolutions Insight Profiler software confirmed that the resulting profiles reflected differences in raw materials and manufacturing methods.&nbsp;</p><p>LabSolutions Insight Profiler enables efficient non-targeted analysis by processing LC-QTOFMS data with an intuitive workflow. This workflow allows for objective evaluation of the taste, quality, and nutritional value of food products. Furthermore, this method is applicable not only to the analysis of compounds in beverages but also to non-targeted analysis of other types of samples.</p><h3><strong>4. Thermo Fisher Scientific / ASMS: High throughput targeted proteomic workflows with Adaptive Retention Time on the </strong><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1959"><strong>Orbitrap Astral Zoom MS</strong></a></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/po_004699_lsms_art_astral_zoom_proteomics_po004699_asms2026_na_en_78767efdc9.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Advances in MS instrumentation have accelerated the identification of biomarkers, but validating and translating these discoveries into assays for clinical use remains challenging and typically requires robust and accurate targeted approaches. While these often rely on predetermined reference target retention time (RT) windows, analytical column degradation or replacement, aging solvents, sample matrix effects, etc. can cause peptide RT shifts between injections and across longer-term analyses. To accommodate this, targeted MS experiments generally schedule wide target RT windows (&gt;1 min) or rely on heavy labeled peptides to trigger acquisition of endogenous peptides. Both greatly limit the number of quantifiable targets per assay. To overcome this, we utilize adaptive RT (aRT) alignment for live correction of RTs together with the speed and sensitivity of the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1959"><strong>Orbitrap Astral Zoom MS</strong></a>. This allows reproducible tracking of &gt;2000 targets with narrow RT windows, high throughputs, such as 300 samples per day (SPD), and hybrid workflows combining targeting, of up to 1000 targets, and discovery.</p><h4><strong>Materials and methods&nbsp;</strong></h4><h5><strong>LC-MS Experiments&nbsp;</strong></h5><p>Experiments were performed using Thermo Scientific Pierce HeLa protein digest resuspended in 0.1% TFA, or individual or pooled human plasma samples. Peptides were separated using a 15 cm Thermo Scientific PepMap analytical columns (150 µm inner diameter) and the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/398"><strong>Thermo Scientific Vanquish Neo UHPLC system</strong></a> at throughputs of 60 or 300 SPD. The liquid chromatography (LC) system was interfaced with the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1959"><strong>Orbitrap Astral Zoom MS.</strong></a> Reference runs using MS1 and/or data-independent-acquisition (DIA) scans were acquired to obtain reference aRT spectra, and to determine target RTs. For aRT alignment, methods consisting of aRT MS1 and/or DIA scans, as well as targeted MS2 scans with RT correction were optimized. For hybrid DIA workflows, we used aRT MS1 scans for reference acquisition and alignment, combined with narrow window DIA and a targeted MS2 experiment. Observed RT shifts were caused by natural column degradation, sample matrix effects or when indicated induced by variation in gradient flow rate and buffer composition.&nbsp;</p><h5><strong>Data analysis&nbsp;</strong></h5><p>Reference DIA data was analyzed using Spectronaut. Targets lists were then generated from the search output by selecting varying number of targets evenly distributed across the RT dimension. Target RTs for smaller scale assays were manually validated and exported from Skyline, while target RTs for large scale assays were exported from Spectronaut. For alignment runs, DIA and PRM data were analyzed using Spectronaut and Skyline respectively.</p><h4><strong>Conclusions&nbsp;</strong></h4><ul><li>Adaptive RT alignment on Orbitrap Astral Zoom MS shows robustness against various sources of RT shifts and has high target recovery even for thousands of targets&nbsp;</li><li>aRT enables the use of target acquisition windows &lt;15 s for robust targeting at high throughput&nbsp;</li><li>aRT Hybrid DIA allows targeting of up to 1000 targets with minimal cost to DIA depth</li></ul><h3><strong>5. Waters Corporation / ASMS: BOOSTING SENSITIVITY IN TOP-DOWN PROTEIN STUDIES USING CYCLIC IMS-ENABLED WIDEBAND ENHANCEMENT</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/720009471_51f2e8b546.pdf"><strong>Full PDF for download</strong></a></li></ul><div><div><section><div><div><div><div><div><div><p>The poster evaluates <strong>Wideband Enhancement (WBE)</strong> as a way to improve sensitivity in top-down protein mass spectrometry. In top-down workflows, intact proteins are fragmented directly, but the signal is distributed across multiple charge states, isotope envelopes, and numerous product ions, which can limit MS/MS sensitivity. WBE addresses this problem by synchronizing ion packets leaving the ion mobility region with the time-of-flight pusher, thereby increasing the number of ions that reach the detector within a selected <i>m/z</i> range.</p><p>Experiments were performed using a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/2434"><strong>Waters Cyclic IMS P20 Mass Spectrometer</strong></a> equipped with an <strong>electron capture dissociation cell</strong>. Samples were introduced by direct infusion in positive electrospray ionization mode. Two proteins were studied under denaturing conditions: bovine ubiquitin, approximately 8.5 kDa, and bovine carbonic anhydrase II, approximately 29 kDa. Single-pass and bypass ion mobility measurements were carried out with and without WBE, using both <strong>collision-induced dissociation (CID)</strong> and <strong>electron capture dissociation (ECD)</strong>. Data were acquired in <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/635"><strong>MassLynx</strong></a><strong> v4.2</strong> and processed using <strong>DriftScope</strong> and the <strong>waters_connect UNIFI peptide-mapping workflow</strong>.</p><p>For ubiquitin, WBE substantially increased the intensity of CID- and ECD-derived fragment ions. Improvements of up to tenfold were observed across the c- and z-ion series, while bypass ion mobility mode produced the largest overall sensitivity gain. The results indicate that WBE can enhance fragment detection while retaining the benefits of cyclic ion mobility separation and without sacrificing mass-spectral resolution.</p><p>The larger carbonic anhydrase II protein also showed marked improvements. CID fragment signals increased by up to tenfold for lower-<i>m/z</i> ions and about eightfold for higher-<i>m/z</i> fragments. Because ECD is generally less efficient than CID, the benefit of WBE was especially evident in ECD experiments: sequence coverage increased from <strong>54% without WBE to 70% with WBE</strong> in bypass mode. The authors conclude that combining WBE with complementary CID and ECD fragmentation on the Cyclic IMS P20 enables more sensitive and comprehensive top-down protein characterization.</p></div></div></div></div></div></div></section></div></div>

Impurities may originate from raw materials and excipients, leach from processing equipment, remain as residues from catalysts, or arise through environmental or packaging related contamination. Even at very low concentrations, certain metals pose toxicological risks, making their assessment and control essential for ensuring product safety.&nbsp;</p><p>To address these risks, global regulatory bodies—including the U.S. Food and Drug Administration (FDA), the European Medicines Agency (EMA), and other members of the International Council for Harmonization (ICH)—require manufacturers to perform systematic risk assessments and ongoing monitoring of elemental impurities. The harmonized ICH Q3D guideline provides the framework for this evaluation by defining toxicology based permitted daily exposure (PDE) limits. It also outlines expectations for analytical testing, reporting, and control strategies across a variety of dosage forms.&nbsp;</p><p>To align with these international regulatory standards, the draft Indian Pharmacopeia (IP) General Chapter 5.10 has been developed to harmonize with the corresponding requirements of the European, Japanese, and U.S. Pharmacopeias.6 Harmonization will ensure that India’s regulatory framework reflects globally accepted best practices for assessing and controlling elemental impurities. It will also ensure that pharmaceutical products manufactured for the Indian and global markets meet internationally aligned safety standards. As part of these harmonized expectations, IP General Chapter 5.10 mandates the quantitative determination of specified elemental impurities using validated, high performance analytical technologies such as ICP MS or ICP OES. These techniques provide the sensitivity, selectivity, and sample throughput required to reliably measure trace-level elemental impurities and demonstrate compliance with PDE based limits.&nbsp;</p><p>An <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?mainauthorItems=10&amp;search=7850"><strong>Agilent 7850 ICP-MS</strong></a> was used in this study to meet the trace level elemental impurity requirements defined in IP General Chapter 5.10. Designed to quantify elements at low concentrations, the 7850 achieves detection limits suitable for accurate, confident measurement of impurities at sub ppb levels. Its advanced ORS4 collision cell technology uses helium (He) mode to effectively remove polyatomic interferences across multiple analytes, enabling reliable measurement of secondary or qualifier isotopes. This capability supports analyte confirmation as required by regulatory guidelines, including IP 5.10, ICH Q2(R1), USP &lt;233&gt;, and USP &lt;1225&gt;. The combination of sensitivity and spectral clarity is essential when testing regulated pharmaceutical products, where accuracy, reproducibility, and compliance with pharmacopeial limits are required.&nbsp;</p><p>The 7850 ICP-MS delivers robust performance in real-world pharmaceutical environments, where complex matrices and routine, high throughput testing are the norm. The hightemperature plasma improves matrix tolerance, minimizes matrix-induced interferences, and enhances ionization efficiency.8 This capability provides higher and more consistent sensitivity for poorly ionized elements such as As, Cd, and Hg, as well as platinum group elements (PGEs) including Os, Ir, and Pt. T</p><p>he 7850 ICP-MS system’s high matrix tolerance also enables efficient handling of diverse sample types, including active pharmaceutical ingredients (APIs), excipients, and finished dosage forms, without excessive dilution or instrument downtime. Long term signal stability ensures consistent results throughout long analytical runs, minimizing re-analysis and supporting dependable quality control (QC) testing.&nbsp;</p><p>The 7850 offers streamlined, workflow-based operation with intuitive software and pre-configured method templates that are aligned with pharmacopeial guidelines. Instrument parameters are automatically optimized, simplifying method development, minimizing operator intervention, and ensuring consistent analytical performance.&nbsp;</p><p>This application note outlines an end-to-end analytical workflow designed to help laboratories comply with the elemental impurity requirements of IP General Chapter 5.10 using ICP MS. Twenty four elements were determined in three types of oral drug products, and the method was evaluated against the performance criteria specified in IP 5.10.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>The analytical approach described in draft Indian Pharmacopoeia (IP) General Chapter 5.10 for the determination of elemental impurities in pharmaceutical products was applied in this study using ICP-MS. Three over-the-counter oral tablet drugs were acid digested by microwave, spiked at various J levels, and analyzed for 24 target elements using an Agilent 7850 ICP-MS.&nbsp;</p><p>The 7850 ICP-MS, equipped with a robust plasma system and the ORS4 collision/reaction cell operating in helium mode, effectively minimized polyatomic interferences and enabled accurate measurement of target isotopes and qualifier ions. Its wide dynamic range detector further supported the analysis of all three oral drug products using a single analytical method and standard operating conditions.&nbsp;</p><p>Instrument setup and method development were simplified by selecting General Purpose plasma conditions, applying automated lens tuning, and using the ICH/USP preset method in the Agilent ICP-MS MassHunter software.&nbsp;</p><p>The method met all performance criteria outlined in IP 5.10, including specificity, accuracy, spike recovery, stability, and system suitability for both limit and quantitative procedures. Instrument detection limits were significantly lower than the J values for oral drugs, ensuring reliable quantification of elemental impurities at regulatory control levels.&nbsp;</p><p>Overall, the results confirm that the 7850 ICP-MS provides a reliable, sensitive, and efficient platform for compliance with IP 5.10 requirements for elemental impurity testing in oral drug products. The simplified method development, robust interference removal, and workflow-based operation significantly reduce analytical complexity while supporting consistent regulatory compliance for routine pharmaceutical quality control laboratories.</p><h3><strong>2. Shimadzu: Evaluation of Particle Size in Chocolate —Evaluation of Particle Size Distribution and Coarse Particles that Affect Texture</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_00624_en_ea48de171d.pdf"><strong>Full PDF for download</strong></a></li></ul><h5><strong>User benefits</strong></h5><ul><li>The solid particle size distribution in chocolate can be easily measured using the <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?mainauthorItems=1&amp;search=SALD-2300"><strong>SALD-2300</strong></a>.&nbsp;</li><li>The <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?mainauthorItems=1&amp;search=iSpect+DIA-10"><strong>iSpect DIA-10</strong></a> enables not only the measurement of particle size distribution but also, the number concentration of particles in a specified particle size range.</li></ul><p>Particle size distribution is one of the physical properties that describe the characteristics of materials and products containing particulate matter. In some cases, the difference in characteristics can be described simply by the average particle size. In other cases, the amount of particles above a certain size is important. The values to be noted vary depending on the type of material, product, and its intended use.&nbsp;</p><p>Here, chocolate is taken as an example of a product where particle size control is important. An example is presented of measuring the particle size distribution and the quantity of particles within a specific size range to assess the texture of chocolate.&nbsp;</p><p>When performing chocolate quality control, the particle size distribution is generally evaluated using a laser diffraction-type particle size distribution analyzer. In this article, in addition to using the SALD-2300 Laser Diffraction Particle Size Analyzer, the iSpect DIA-10 Dynamic Particle Image Analysis System is also employed to show the possibility of a slightly different evaluation approach to investigating the relationship between particle size/concentration and texture.</p><h4><strong>Measurement of Particle Size Distribution by </strong><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?mainauthorItems=1&amp;search=SALD-2300"><strong>SALD-2300&nbsp;</strong></a></h4><p>Chocolate is solid at room temperature. Therefore, it must be dissolved (dispersed) in a solvent to measure the particle size distribution of the solid particles contained within it. Samples must also be diluted to a concentration suitable for the particle size distribution measuring analyzer. Isopropanol was selected as the dispersant because it can dissolve oil and fat and disperse the solid particles to measure the particle size distribution of just the solid particles in the chocolate.&nbsp;</p><p>Two commercial milk chocolate samples were used: sample F and sample UF. Taste tests were conducted by several individuals (including the author), and comparisons were made prior to measurement. The taste tests focused on texture differences and the smoothness on the palate and tongue. The results showed that although there were differences among individuals, most perceived sample UF as smoother than sample F, while sample F feltslightly rough.</p><h4><strong>Particle Size and Number Concentration Measurement by </strong><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?mainauthorItems=1&amp;search=iSpect+DIA-10"><strong>iSpect DIA-10&nbsp;</strong></a></h4><p>So far, the particle size distribution of two types of chocolate has been measured using the SALD-2300, and the relationship between the results and the texture has been examined. Next, we considered the measurement results obtained using the iSpect DIA-10.&nbsp;</p><p>The iSpect DIA-10 is different from general image analysis type particle size measurement equipment, and it has the advantage of being able to detect almost all particles within the measurement range. As a result, it is possible to accurately calculate the number concentration in the measurement solution.</p><p>From the SALD-2300 measurement results, it is clear that the sample contains about 20 to 30 percent particles that are below 5 µm, which is the minimum measurement size of iSpect DIA-10. Therefore, it is not possible to capture the number concentration of all particlesin the sample.&nbsp;</p><p>On the other hand, since no particles exceed the measurement limit of 100 µm and almost all particles in the coarse particle region can be detected, it is possible to calculate the number concentration of particles above a certain size.&nbsp;</p><p>The results of measurements of samples F and UF using the iSpect DIA-10 are presented below. The dispersion procedure is the same as that for the SALD-2300 shown in Table 1, but the final concentration is equivalent to that of 0.1 g of chocolate dispersed in 1000 mL of isopropanol due to a further 50 times dilution during measurement.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>In this article, the particle size distribution of chocolate was measured using two instruments, the SALD-2300 and the iSpect DIA-10, and introduced the relationship between these results and the texture perceived when the chocolate was eaten. By looking at the results of the two measurement methods together, it is believed that a more in-depth analysis is possible.&nbsp;</p><p>In particular, the results of the number concentration of coarse particles measured using the iSpect DIA-10 are considered useful for evaluating texture based on the quantity of coarse particles. This can be expected to be effective in the food field and other fields where the abundance of small quantities of coarse particlesisimportant.</p><h3><strong>3. This high volume of polymer production results in large amounts of plastic ending up in our environment. Recycling is a strong potential solution; however, mechanical recycling often leads to molecular weight degradation via chain scission, limiting performance in secondary applications.&nbsp;</p><p>Upcycling through reactive extrusion provides a pathway to restore or enhance performance by incorporating recycled streams into high-value matrices. Polyamide 12 (PA12) offers superior mechanical and thermal properties but is immiscible with polypropylene due to thermodynamic incompatibility.</p><p>Compatibilization using PP-g-MA enables interfacial stabilization through in situ formation of diblock copolymers via reaction between maleic anhydride groups and PA12 amine end groups. <div><div><section><div><div><div><div><div><div><p><strong>This study introduces a highly sensitive compound-specific isotope analysis method for identifying protein binders in aged paintings. Using direct inlet pyrolysis-GC-combustion-isotope ratio mass spectrometry (DIP-GC-C-IRMS), the method targets δ13C values of pyrolytic diketopiperazines and reduces the required sample size from milligrams to micrograms.</strong></p><p>Reference materials showed that diketopiperazine isotope signatures preserve the distinction between terrestrial and marine collagen sources. Formats and hyperlinks were adapted from the original.</strong></p><div><div><div><p>The complexities and challenges presented by the analysis of protein paint binders illustrate the need for novel methods of identification for these materials, in particular to distinguish between terrestrial and fish-animal glues. An alternative approach with great potential may be found in the neighboring field of archeology, where collagen in bones and teeth has been used as a palaeodietary indicator since the 1970s. Determination of bulk δ<sup>15</sup>N values may suggest the position of an organism in the respective food web, while δ<sup>13</sup>C values offer insight into their diet. (<a href="javascript:;">27−29</a>) Bulk δ<sup>13</sup>C and δ<sup>15</sup>N values of collagen are weighted averages of the δ<sup>13</sup>C and δ<sup>15</sup>N values of their constituent AAs, which reflect both dietary sources and metabolic processes. Serine, glycine, and alanine (Ser, Gly, and Ala) are part of the glucogenic AA group, which is biosynthesized from intermediates of glycolysis. As pyruvate is the end product of glycolysis, considered the catabolic pathway of carbohydrates, the isotopic signature of these AAs should reflect that of dietary carbohydrates. Aspartic acid and glutamic acid (Asp and Glu), asparagine (Asn), and Pro are part of the ketogenic AA group, originating from intermediates of the tricarboxylic acid (TCA) cycle. Due to their TCA cycle precursors, the isotopic signature of these AAs should reflect that of the whole diet, if biosynthesized <i>de novo</i>. Finally, Hyp and hydroxylysine cannot be incorporated into proteins from the diet and are formed after the polypeptides are synthesized, during posttranslational processing of collagen. Hydroxylysine is formed from lysine (Lys), whereas Hyp results from hydroxylation of Pro residues. (<a href="javascript:;">30</a>) Moreover, AAs in the “trophic group“ tend to display a large enrichment in <sup>15</sup>N with increasing trophic level, unlike those belonging to the “source group.“</p></div><div><p>Significant limitations therefore surround the interpretation of bulk stable isotope ratios, which, however, may be overcome by compound-specific isotope analysis (CSIA). Individual AA δ<sup>13</sup>C values have proven invaluable for distinguishing terrestrial from marine diets of our ancestors, (<a href="javascript:;">30−35</a>) while AA δ<sup>15</sup>N values are being increasingly used in archeological trophic position studies. (<a href="javascript:;">36−39</a>) Despite the similarity in target material, CSIA has never been extended to the analysis of animal glue, or any other protein, in paintings. Reasons for this are likely linked to the sample sizes required for standard protocols for GC-combustion-isotope ratio MS (GC-C-IRMS), which, however, as illustrated by recent studies, (<a href="javascript:;">40−42</a>) may be significantly reduced by undertaking direct inlet pyrolysis (DIP)-GC-C-IRMS. Here, this technique is applied to the analysis of proteins of paintings, in particular by targeting, for the first time, the stable carbon isotope composition of DKPs, protein pyrolysates. Having confirmed that DKPs reflect the isotopic signature of their constituent AAs, DKP δ<sup>13</sup>C values may be used to distinguish terrestrial from aquatic animal glue in aged paintings.</p><h4><strong>Materials and Methods</strong></h4><h5><strong>Instrumental Analyses</strong></h5><div><p>For elemental analysis (EA)-IRMS, GC-FID, and GC-C-IRMS methods, please see SI1. All chromatograms were replotted in RStudio. DIP-GC-MS was carried out on an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=7890A"><strong>Agilent Technologies 7890A GC</strong></a> coupled to an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=5975C"><strong>Agilent Technologies 5975C MSD</strong></a> (Agilent Technologies, Santa Clara, CA, USA), a single quadrupole MS. The GC was equipped with a nonpolar fused-silica capillary column (30 m × 0.25 mm i.d. × 0.25 μm film thickness) with a BPX5 stationary phase (5% phenyl polysilphenylene-siloxane, SGE) and a TSP in the multimode inlet (MMI), operated in splitless mode. The initial TSP temperature was 50 <span>°</span>C, set to rise to 450 <span>°</span>C (900 <span>°</span>C min<sup>–1</sup>, 3 min isothermal hold), before cooling to 250 <span>°</span>C (25 <span>°</span>C min<sup>–1</sup>, 40 min isothermal hold). The oven temperature began at 40 <span>°</span>C, was increased to 200 <span>°</span>C (10 <span>°</span>C min<sup>–1</sup>), and then to 310 <span>°</span>C (6 <span>°</span>C min<sup>–1</sup>) with a final hold time of 20 min. He was used as a carrier gas at a constant flow (1.2 mL min<sup>–1</sup>).</p></div><div><p>The MS was set to acquire in full scan mode (<i>m</i>/<i>z</i> 50–650) at 0.2 scan s<sup>–1</sup>, and was operated at an EI potential of 70 eV with a solvent delay of 6.5 min. The MS transfer line temperature was set to 280 <span>°</span>C, the ion source temperature to 230 <span>°</span>C, and the quadrupole to 150 <span>°</span>C. Data acquisition was undertaken using Agilent Technologies Chemstation, and data processing with Agilent Technologies MassHunter software (version 10.0) and the AMDIS deconvolution software (NIST, version 2.66). Peaks were identified using a combination of mass spectral information, GC retention times (RTs), and comparison to the NIST mass spectral library (Mass Spectral Search Program version 2.0 and NIST 08 MS Library), published data sets, and NG databases. An <i>n</i>-alkane “ladder“ (with a characteristic bell-shaped peak profile) standard, containing C<sub>7</sub>–C<sub>40</sub> saturated alkanes (0.01 mg mL<sup>–1</sup> in <i>n</i>-hexane), was used to monitor instrument performance.</p></div><div><p>DIP-GC-QTOF-MS analysis was carried out on an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=7890B"><strong>Agilent Technologies 7890B GC</strong></a> coupled to an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=7200"><strong>Agilent Technologies 7200 Accurate Mass QTOF MS</strong></a> (Agilent Technologies, Santa Clara, CA, USA). The same method was used, except that the MMI was held at 450 <span>°</span>C for 1 min only. The MS was set with an acquisition rate of 5 spectra s<sup>–1</sup>, an acquisition time of 200 ms spectrum<sup>–1</sup>, and an emission current of 2.1 μA. DIP-GC-C-IRMS analysis was carried out on an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8890"><strong>Agilent Technologies 8890 GC</strong></a> coupled to an <strong>Elementar isoprime precisION IRMS</strong> via an Elementar GC5 combustion interface, using a combustion reactor containing CuO pellets and Ag wool (Elementar Analysensysteme GmbH, Hanau, Germany), maintained at 850 <span>°</span>C. The GC parameters were identical to those for DIP-GC-MS, except that the MMI, after being held at 450 <span>°</span>C for 3 min, was cooled to 300 <span>°</span>C (900 <span>°</span>C min<sup>–1</sup>, isothermal hold for 50 min). Additionally, the oven program was typically shortened by setting a lower maximum temperature (280 <span>°</span>C instead of 310 <span>°</span>C), where it was held for 10 min. He was used as a carrier gas at a constant flow (2 mL min<sup>–1</sup>).</p></div></div></div></div></div></div></section></div></div><h4><strong>Results and Discussion</strong></h4><h5><strong>Pyrolytic Marker Identification from DIP-GC-MS of Protein References</strong></h5><div><p>The thermal degradation of intact proteins has been shown to be a multistep reaction, (<a href="javascript:;">45</a>) where the formation of DKPs, occurring in the first degradation step (<i>T</i><sub>max</sub> = 325 <span>°</span>C), does not require high Py temperatures. (<a href="javascript:;">21</a>) Protein markers may therefore be detected through DIP-GC-MS, as testified by the analysis of a soapstone fragment from Qarmaaluit (18th–19th century CE). (<a href="javascript:;">46</a>) Nonetheless, given that this technique is not widely reported in the literature, modern protein references were analyzed by DIP-GC-MS, and pyrolytic profiles obtained were compared to published Py-GC-MS data. Studies have been undertaken with (<a href="javascript:;">22, 47, 48</a>) and without (<a href="javascript:;">22, 26</a>) TMAH, hence, both methods were carried out here. Results were scrutinized, in particular, to identify pyrolytic markers suitable for DIP-GC-C-IRMS analysis. Egg yolk, which presents a similar AA composition as egg white, was not examined, as its high fat content was deemed to interfere with the analysis.</p></div><div><p>In general, DIP-GC-MS analyses involving derivatization with TMAH, in particular for animal glues, featured many more compounds compared to the total ion chromatograms (TICs) obtained without derivatization (see <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/ancham/article-supplement/5211080/pdf/ac5c07901_si_001/">Figure S1</a> for G1, aged gelatin spread). In this case, GC-MS TICs exhibited better baseline separation with the most abundant peaks corresponding to the targeted DKPs. Further analysis of glue samples was therefore carried out without derivatization, as good baseline resolution is central to reliable compound-specific stable isotope determinations. (<a href="javascript:;">43</a>) Analysis of rabbit skin glues (GRS1 and GRS2) and isinglass glues (GI1 and GI2) yielded GC-MS TICs identical to each other. In accordance with the literature, (<a href="javascript:;">21, 22, 49</a>) as illustrated in <a href="javascript:;">Figure 1</a>a, they are dominated by peaks corresponding to diketodipyrrole (peak 8) and cyclo(Pro-Gly) (peak 10). Because of its relatively high polarity, or overloading, the latter produces a broad (fronting) peak. (<a href="javascript:;">15</a>) The author reported AA profiles consistent with the presence of animal glues, which, for this purpose, would be expected to be of terrestrial origin. However, given the low proportion of Pro and Hyp in NG6368, White discussed a possible aquatic origin for this glue. Analysis was repeated with the aforementioned DIP techniques, revealing very similar profiles to those of the reference animal glues (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/ancham/article-supplement/5211080/pdf/ac5c07901_si_001/">Figure S6</a>). The protein binder in the ground layers of both paintings exhibited similar DKP stable carbon isotope signatures. In particular, δ<sup>13</sup>C<sub>Pro-Gly</sub> values were determined to be −18.4‰ (±0.1) and −17.6‰ (±0.1) for NG599 and NG6368, respectively. The δ<sup>13</sup>C<sub>Pro-Hyp</sub> values were found to equal −19.8‰ (±0.4) and −18.6‰ (±0.001), respectively.</p></div><div><p>It should be noted that the SD observed for these determinations is lower than what was reported for some of the reference binders discussed above. These errors are in line with those observed for standard GC-C-IRMS methods. As these paint fragments were in the form of very fine powder, typical of paint samples, these results support that sample inhomogeneities might be primarily responsible for wide SDs, rather than the DIP-GC-C-IRMS method itself. In the case of marine animal glues, more enriched δ<sup>13</sup>C<sub>DKP</sub> values would be expected than those observed here, especially for cyclo(Pro-Gly). Instead, the determined δ<sup>13</sup>C<sub>DKP</sub> values suggest a terrestrial origin for the glues in both paintings. Indeed, by plotting the values as described above, as shown in <a href="javascript:;">Figure 7</a>, both paintings fall extremely close to the reference rabbit skin glues. Hence, for NG599, these results are in agreement with the previous analysis by White (1984). Given the challenges currently faced by this field to differentiate fish glues from other animal glues, this innovative technique might enable new information to be gathered on artist techniques and the history of use of these materials. It is important to note that the method can be applied to many other fields of research. In addition to painting analysis, this minimally invasive protocol is pertinent to any discipline that involves the analysis of degraded proteins, heterogeneous and complex matrices, or where access to samples is restricted due to ethical implications or lack of material. This includes environmental and forensic sciences, paleo- and zooarcheology, paleoecology, and anthropology.</p></div><h4><strong>Conclusions</strong></h4><div><p>Aged proteins encountered in painted cultural heritage have usually undergone extreme structural changes from the freshly applied material, often rendering unambiguous protein source determination unattainable. In particular, the distinction between terrestrial and aquatic animal glues is typically not possible without proteomics approaches. Nonetheless, this paper demonstrates that, with minimal sample sizes, DIP-GC-C-IRMS permits determination of δ<sup>13</sup>C values of DKPs arising from pyrolysis of aged proteinaceous material, providing the first record of the isotopic signature of these compounds. In this study, δ<sup>13</sup>C values of DKPs were shown to reflect the δ<sup>13</sup>C values of the parent AAs and, therefore, to permit unambiguous differentiation between terrestrial and marine animal glues, analogous to the use of CSIA of archeological collagen for palaeodietary reconstruction.</p></div><div><p>Remarkably, DIP-GC-C-IRMS, requiring a considerably smaller sample (in the microgram range) than typical GC-C-IRMS methods (in the milligram range), renders CSIA accessible for the investigation of proteinaceous material in painted cultural heritage for the first time. This widens the application of CSIA to precious, unique, and fragile objects in cultural heritage and beyond. Moreover, the rapidity of the analysis places DIP-GC-C-IRMS as a useful screening method to identify materials of interest for further proteomic analyses.</p></div><div><p>First, the isotopic imprint of modern references for rabbit skin glues, isinglass glues, casein, chicken, and duck eggs was confirmed through established EA-IRMS and GC-C-IRMS methods. As expected, bulk and individual AA δ<sup>13</sup>C and δ<sup>15</sup>N values for terrestrial animal glues exhibited a higher depletion in <sup>13</sup>C and <sup>15</sup>N than the marine glues. The δ<sup>13</sup>C values of DKPs arising from DIP-GC-C-IRMS of the same materials were found to retain this relationship. Furthermore, the ground layers of two 16th-century Italian paintings from the National Gallery collection, known to contain animal glue, yielded δ<sup>13</sup>C<sub>DKP</sub> values consistent with the terrestrial variety, confirming that the isotopic signature of these compounds is retained through aging and degradation.</p></div>

While many imaging approaches can visualize elemental distributions, quantitative determination is limited by preparation artifacts, matrix dependent signal variability, and the lack of validated analytical workflows. (<a href="javascript:;">1−7</a>) These limitations restrict mechanistic interpretation of elemental behavior in biological models and hinder comparison across studies.</p></div><div><p>Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) offers a powerful approach for spatially resolved elemental analysis, combining high sensitivity, broad elemental coverage, and micrometre-scale resolution. It has been widely applied to map metals, nanoparticles, and metal-based therapeutics in biological systems, including tumor spheroids, tissues, and brain models. (<a href="javascript:;">8−12</a>) Despite its sensitivity and multiplexing capability, applications in complex cellular models have largely remained qualitative or relatively quantitative. (<a href="javascript:;">13−15</a>) Quantitative implementation is hindered by methodological factors: sample preparation can redistribute elements, and matrix effects complicate signal interpretation, limiting reproducibility.</p><p>Here, we present a validated workflow for quantitative LA-ICP-MS imaging in heterogeneous 3D tumor spheroid models. We systematically evaluate fixation, embedding, sectioning, drying, calibration, and endogenous normalization to identify conditions that preserve spatial fidelity and analytical sensitivity. We show that endogenous phosphorus provides a robust internal normalizer, consistent with previous studies, (<a href="javascript:;">34−36</a>) and demonstrate that quantitative elemental maps can be integrated with biological context through correlation with immunohistochemistry on consecutive sections. Together, this work establishes an integrated and transferable framework providing a practical platform for quantitative elemental imaging and cross-modal spatial analysis in complex 3D biological systems, with applications spanning drug delivery, nanomedicine, toxicology, and metallomics.</p><h4><strong>Experimental Section</strong></h4><h5><strong>LA-ICP-MS Instrumentation and Acquisition</strong></h5><div><p>Elemental imaging was performed using a 193 nm ArF* excimer laser ablation system (Iridia, Teledyne Photon Machines) equipped with a cobalt long-pulse ablation cell and coupled via an Aerosol Rapid Introduction System (ARIS) to a triple-quadrupole ICP-MS (<a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/products/1488"><strong>iCAP MTX</strong></a>, Thermo Fisher Scientific). Instrument control and data acquisition were conducted using <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/products/1489"><strong>Qtegra software</strong></a> (Thermo Fisher Scientific).</p></div><div><p>Instrument performance was optimized using NIST SRM 612 glass to minimize laser-induced elemental fractionation (238U<sup>+</sup>/232Th<sup>+</sup>), maintain oxide formation below 1% (232Th16O<sup>+</sup>/232Th<sup>+</sup>), and maximize sensitivity of 59Co<sup>+</sup>, 115In<sup>+</sup>, and 238U<sup>+</sup>.</p></div><div><p>Spheroid sections mounted on glass slides were analyzed in fixed dosage raster mode with a 10 μm square laser spot, defining the spatial resolution. Imaging was performed with a laser energy density of 0.6 J cm<sup>–2</sup>, a repetition rate of 500 Hz (10 shots per pixel), and a helium carrier gas flow of 0.4 L min<sup>–1</sup>. Elemental detection was performed using Dynamic Reaction Cell (DRC) mode with oxygen as the reaction gas (0.16 mL min<sup>–1</sup>) to reduce polyatomic interferences on <sup>31</sup>P and <sup>32</sup>S. Two isotopes were monitored per acquisition (<sup>11</sup>B and <sup>31</sup>P, or <sup>11</sup>B and <sup>32</sup>S), with dwell times of 14.3 ms (<sup>11</sup>B) and 1 ms (<sup>31</sup>P or <sup>32</sup>S), a quadrupole total switching time of 4.7 ms, and a total acquisition time of 20 ms per pixel. A section thickness of 30 μm ensured that the imaging depth matched the physical section thickness. Slides were arranged within a four-position sample holder such that a boron-spiked fish gelatin calibration standard was analyzed within the same run to enable signal calibration.</p><h5><strong>ICP-MS Sample Preparation</strong></h5><p>For bulk quantification, spheroids were processed in parallel to LA-ICP-MS samples. Three spheroids per condition were collected into low-binding microcentrifuge tubes following incubation with BPA (4h, 37 <span>°</span>C). Samples were washed once with ice-cold PBS and pelleted by centrifugation (1500 rpm, 5 min). The supernatant was removed and spheroids were digested in 100 μL of 70% HNO<sub>3</sub> (trace-metal grade, Sigma-Aldrich) at 60 <span>°</span>C overnight. Following complete digestion, samples were diluted in 2% HNO<sub>3</sub> spiked with beryllium as an internal standard and analyzed using <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?mainauthorItems=10&amp;search=7900"><strong>7900 ICP-MS</strong></a> (Agilent). Boron concentrations were quantified against external calibration standards and expressed as total boron mass per spheroid.</p><h4><strong>Results</strong></h4><h5><strong>Overview of the LA-ICP-MS Workflow</strong></h5><p>The overall experimental pipeline is summarized in <a href="javascript:;">Figure 1</a>, outlining the sequential steps from spheroid preparation to quantitative image generation. Representative data sets illustrate the multimodal workflow, combining normalized boron distributions with brightfield imaging, propidium iodide (PI) staining, and IHC for GLUT1 and LAT1 (<a href="javascript:;">Figure 5</a>a). Across HT29 and HCT116 spheroids, mean variation in staining intensity between adjacent sections was 10.5 ± 6.7% and 9.5 ± 4.7% for LAT1, and 6.6 ± 0.9% and 11.4 ± 2.1% for GLUT1, respectively (<a href="javascript:;">Figure 5</a>b). Overall variability across all spheroids ranged from 0.9–23.7% (<i>n</i> = 3 spheroids per cell line).</p></div><div><p>Coefficients of variation were 7.4 ± 2.3% and 6.8 ± 2.7% for LAT1, and 4.9 ± 1.3% and 8.3 ± 1.9% for GLUT1 in HT29 and HCT116 spheroids’ consecutive sections, respectively, indicating that spatial protein distributions were largely preserved between consecutive sections at this scale.</p></div><div><p>Consistent with this observation, Pearson correlation analysis demonstrated strong spatial agreement between adjacent sections for most proteins analyzed. GLUT1 expression showed high correlation in both HT29, (<i>r</i> = 0.9678–0.9924) and HCT116 spheroids (<i>r</i> = 0.8274–0.9836), while LAT1 expression was highly correlated in HCT116 (<i>r</i> = 0.9796–09963) but more variable in HT29 (<i>r</i> = 0.5582–0.8190) (<a href="javascript:;">Figure 5</a>c). All correlations were statistically significant (<i>p</i> &lt; 0.05). Section-to-section variability must be quantified before assuming that a consecutive section faithfully represents the biology of the elementally mapped section; where this condition is met, consecutive-section correlation provides a reliable means of relating elemental and protein distributions.</p></div><div><p>Having established this framework, the known relationship between LAT1 expression and boronophenylalanine (BPA) uptake was used as a biological test case. BPA is transported into cells via the large neutral amino acid transporter LAT1, providing a mechanistic link between protein expression and intracellular boron accumulation.</p></div><div><p>Region-of-interest (ROI) analysis demonstrated that LAT-1 enriched regions exhibited higher boron signal compared to LAT1-low regions in HCT116 spheroids (<i>p</i> = 0.0331, paired two-tailed <i>t</i> test; <i>n</i> = 3, 3 spheroids per replicate). Radial analysis further revealed a positive correlation between LAT1 expression and boron distribution (<i>r</i> = 0.5214, <i>p</i> = 0.0221, <a href="javascript:;">Figure 5</a>d), supporting spatial coupling between transporter expression and uptake.</p></div><div><p>Orthogonal validation by flow cytometry confirmed that LAT1-positive cells exhibited significantly higher BPA levels than LAT1-negative cells in both cell lines (36% and 39% increase in HT29 and HCT116, respectively; <i>p</i> &lt; 0.05, <i>n</i> = 3, 25,000 events per replicate).</p></div><div><p>Together, these results demonstrate that combined LA-ICP-MS and consecutive section IHC workflow enables biologically meaningful spatial correlation between elemental distributions and tested proteins expression in 3D tumor models.</p></div><h4><strong>Conclusion</strong></h4><div><p>This study establishes a validated workflow for quantitative LA-ICP-MS imaging in heterogeneous three-dimensional spheroid models by systematically addressing key methodological determinants of accuracy, including sample preparation, calibration, endogenous normalization, and cross-modal validation. Optimized preparation (2% CMC cryo-embedding with freeze-drying) reduced analyte loss by ∼84% relative to OCT, while matrix-matched gelatin standards enabled linear calibration (<i>r</i><sup>2</sup> &gt; 0.99) and conversion of signal intensity to absolute concentration. Under these conditions, the workflow achieved 10 μm spatial resolution with ng g<sup>–1</sup>-level sensitivity (LOD 7.39 ± 2.24 ng g<sup>–1</sup>) and reproducible quantification across independent runs (CV &lt; 20% across most of the spheroid radius).</p></div><div><p>Quantitative accuracy was confirmed by strong agreement with bulk ICP-MS (<i>r</i><sup>2</sup> = 0.9965; &lt; 15% deviation above ∼3 ng), demonstrating that reconstructed LA-ICP-MS measurements provide reliable estimates of total uptake. Integration with protein expression using consecutive-section analysis was feasible when section-to-section variability was characterized (typically ∼5–12% variation; CV &lt; 10%), enabling biologically interpretable correlations between elemental distributions and molecular markers in multicellular systems.</p></div><div><p>While volumetric reconstruction requires simplifying assumptions (e.g., radial symmetry), these do not affect the spatial correlation analyses used to interpret elemental heterogeneity. By integrating preparation optimization, validated normalization, quantitative calibration and cross-modal benchmarking within a single framework, this work moves LA-ICP-MS from relative signal mapping toward reproducible, quantitatively interpretable imaging in complex 3D biological models, providing a practical platform for mechanistic studies of drug distribution and microenvironmental heterogeneity.</p></div></div></div></div></div>

However, untargeted metabolomics and downstream data analysis workflows are vulnerable to signal contamination by non-biological variance associated with sample collection and handling and instrumental drift, which can obscure meaningful biological patterns.&nbsp;</p><h4><strong>Objective/Aims</strong></h4><p>The goal of this study is to identify and quantify non-biological variance in untargeted volatile metabolomics arising from sample collection procedures and instrumental drift.</p><h4><strong>Methods</strong></h4><p>Breath samples were collected and transferred onto Thermal Desorption Tubes (TDTs) in technical duplicates. TDTs were stored at 4˚C and analyzed using two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (<a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/37"><strong>GC×GC– TOFMS</strong></a>).</p><h4><strong>Conclusions</strong></h4><p>Instrument drift and other non-biological factors can strongly affect metabolomics data. Consistent, timedependent patterns of dissimilarity highlight the need to identify and control these sources of variance to ensure reliable biomarker discovery.</p><h3><strong>2. Shimadzu: Analysis of Volatile PFAS in Textiles Using </strong><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMS-TQ8040+RX"><strong>GCMS-TQ8040 RX</strong></a><strong> with MMI</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_01257_en_beb993613a.pdf"><strong>Full PDF for download</strong></a></li></ul><h4><strong>User benefits</strong></h4><ul><li>Using the GCMS-TQ8040 RX and the Multi-Mode Injection Unit (MMI), 12 volatile PFAS including compounds listed in EN 17681-2,<br>can be analyzed at the ppb level without preconcentration.</li><li>The spike recovery test shows good percentage recoveries of all compounds (80–120 %) that are at or below the EU Persistent<br>Organic Pollutants (POPs) threshold limit (25 ppb)</li></ul><p>Per- and polyfluoroalkyl substances (PFAS) are used in textile products because of their useful properties, such as heat resistance and water repellency. However, PFAS are also extremely stable substances that resist degradation and accumulate in the environment, which has led to concerns about their effects on humans. Since eliminating sources of PFAS is essential to prevent them from accumulating in the environment and in living organisms, many countries are passing laws that restrict the production, distribution, and use of PFAS in products. For example, the EU POPs Regulation sets a permitted threshold of up to 0.025 mg/kg (25 ppb) for certain PFAS (including PFOA, PFOS, and PFHxS) in substances, mixtures, or articles1) . EN 17681 defines standard test methods for PFAS analysis in textiles2),3) .&nbsp;</p><p>In a previous <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_01108_en_e04fe25939.pdf"><strong>Application News, No. 01-01108</strong></a>, an example of analyzing volatile and nonvolatile PFAS in textiles using LC/MS/MS and GC/MS/MS based on this standard was introduced. This study shows an example of trace analysis of volatile PFAS (Table1) in textiles using the newly developed GCMS-TQ RX series equipped with the Multi-Mode Injection Unit (MMI).</p><h4><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMS-TQ8040+RX"><strong>GCMS-TQ8040 RX&nbsp;</strong></a></h4><p>Delivering highly sensitive multi-component simultaneous analysis, the system improves operational efficiency through Analytical Intelligence , while also providing performance as a single GC-MS. The GCMS-TQ RX Series is a versatile triple quadrupole GC-MS that demonstrates high performance across a wide range of application fields. By combining it with the newly developed Multi-Mode Injection Unit (MMI), multiple injection methods are supported and analytical capability is further expanded.</p><h4><strong>Multi-Mode Injection Unit (MMI)&nbsp;</strong></h4><p>The Multi-Mode Injection Unit (MMI) supports a wide range of injection modes in a single unit, including split/splitless (SPL), programmable temperature vaporization (PTV), large-volume injection (LVI), direct injection, and thermal desorption/extraction (TD/TE). Dedicated software with an intuitive user interface enables easy method creation and seamless method transfer, simplifying analytical workflows. In this Application News, use of the MMI enables a threefold increase the injection volume compared with conventional injection methods, while maintaining peak shape.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>Using the GCMS-TQ8040 RX and the MMI, volatile PFAS in textiles were analyzed. The results showed that all compounds measured in this study could be analyzed at 25 ppb or below. Furthermore, in the spike recovery test at the EU POPs regulatory limit of 25 ppb, recoveries were within 80 to 120 % for all compounds, showing good results. By increasing the injection volume with the MMI, trace levels of volatile PFAS can be measured even with the standard GCMS-TQ8040 RX model.</p><h3><strong>3. While this is a key technique in the analysis of PFAS, there are classes of volatile and semi-volatile PFAS that do not chromatograph well using LC and require the use of gas chromatography (GC) to be successfully analyzed. Classes of PFAS that favor GC analysis include, but are not limited to, fluorotelomer alcohols (FTOH), fluorotelomer acrylates (FTAcr), fluorotelomer methacrylates (FTMAC), and fluorotelomer acetates (FTOAc). Additionally, there are classes that can be analyzed using both LC and GC techniques, including sulfonamides (PFSA) and sulfonamidoethanols (PFSE).&nbsp;</p><p>All of these compounds are considered precursor compounds that terminally transform into the most widely studied PFAS, perfluoroalkyl carboxylates (PFCA) and perfluoroalkyl sulfonates (PFSA). Not only are they precursors in the synthesis of PFCA and PFSA end products, they are also intentionally used in products such as food packaging and textiles to impart grease and water proofing qualities.&nbsp;</p><p>These precursor compounds are also discharged into the environment through similar routes as their end products (landfills, manufacturing waste, firefighting foams, etc), but also through volatilization, transport and deposition through the atmosphere either in the gas phase or bound to particulates. Like most PFAS, not much is known about the human toxicity of these particular PFAS, although some animal and in vitro studies do point to similar health effects as more commonly studied PFCAs and PFSAs, including liver and kidney toxicity, endocrine disruption and negative immune effects.1-4 Furthermore, their ability to transform in the environment to PFCAs and PFSAs, which are known to have toxic health implications, make them of concern.&nbsp;</p><p>GC-MS/MS analysis for the volatile and semi-volatile GC amenable PFAS provides an additional tool in the workflow of PFAS sample analysis, increasing the comprehensiveness of sample characterization. In this application note, the development of an atmospheric pressure gas chromatography (APGC) coupled to tandem quadrupole mass spectrometry (MS/MS) method will be presented. This approach could integrate seamlessly with existing LC-MS/MS workflows, allowing labs to extend compound coverage without requiring changes to established sample preparation protocols. Performance for all compounds was found to be optimal with the use of a siltek deactivated inlet liner and a 30 m x 0.25 mm ID, 0.50 µm Rtx-200 column. This configuration allows for the injection of samples pre-extracted for LC-MS/MS analysis that are in mainly methanol and water, allowing labs to generate additional insights without increasing sample prep burden. Method LLOQs ranged from 0.005 – 5.0 ng/mL, with most compounds being below 0.10 ng/mL, delivering robust performance across compound classes, indicating the method can be used for trace level applications. Overall, APGC-MS/MS adds a complementary capability to LC-MS/MS methods, allowing labs to comprehensively assess PFAS contamination for better understanding and informed regulatory or product safety decisions. Further application notes will demonstrate the use of this method in environmental and food packaging samples, highlighting how labs can gain more complete PFAS insights while leveraging existing workflows.</p></div></div></div></div></div></div></section>
