<p><strong>Electrons can behave very differently depending on how much energy they have. Whether you shoot an electron with high or low energy into a solid body determines which effects can be triggered. Electrons with low energy can be responsible for the development of cancer, for example, but conversely they can also be used to destroy tumors. They are also important in technology, for example for the production of tiny structures in microelectronics.</strong></p><p>These slow electrons, however, are extremely difficult to measure. Knowledge about their behaviour in solid materials is limited, and often scientists can only rely on trial and error. However, TU Wien has now succeeded in obtaining valuable new information about the behaviour of these electrons: Fast electrons are used to generate slow electrons directly in the material. This allows details to be deciphered that were previously inaccessible experimentally. The method has now been presented in the journal ‘Physical Review Letters’.</p><h4><strong>Two types of electrons at the same time</strong></h4><p>“<i>We are interested in what the slow electrons do inside a material, for example inside a crystal or inside a living cell,</i>” says Prof Wolfgang Werner from the Institute of Applied Physics at TU Wien. “<i>To find out, you would actually have to build a mini-laboratory directly in the material to be able to measure directly on site. But that's not possible, of course.</i>"</p><p>You can only measure electrons that come out of the material, but that doesn't tell you where in the material they were released and what has happened to them since then. The team at TU Wien solved this problem with the help of fast electrons that penetrate the material and stimulate various processes there. For example, these fast electrons can disturb the balance between the positive and negative electrical charges of the material, which can then lead to another electron detaching itself from its place, travelling at a relatively low speed and in some cases escaping from the material.</p><p>The crucial step now is to measure these different electrons simultaneously: “<i>On the one hand, we shoot an electron into the material and measure its energy when it leaves again. On the other hand, we also measure which slow electrons come out of the material at the same time.</i>” And by combining this data, it is possible to obtain information that was previously inaccessible.</p><h4><strong>Not a wild cascade, but a series of collisions</strong></h4><p>The amount of energy that the fast electron has lost on its journey through the material provides information on how deeply it has penetrated the material. This in turn provides information about the depth at which the slower electrons were released from their place.</p><p>This data can now be used to calculate to what extent and in what way the slow electrons in the material release their energy. Numerical theories on this can be reliably validated for the first time using the data.</p><p>This led to a surprise: it was previously thought that the release of electrons in the material took place in a cascade: A fast electron enters the material and hits another electron, which is then ripped away from its place, causing two electrons to move. These two electrons would then remove two more electrons from their place, and so on. The new data show that this is not true: instead, the fast electron undergoes a series of collisions, but always retains a large part of its energy and only one comparatively slow electron is detached from its place in each of these interactions.</p><p>“<i>Our new method offers opportunities in very different areas,</i>” says Wolfgang Werner. “<i>We can now finally investigate how the electrons release energy in their interaction with the material. It is precisely this energy that determines whether tumor cells can be destroyed in cancer therapy, for example, or whether the finest details of a semiconductor structure can be correctly formed in electron beam lithography.</i>”</p>

New method unravels the mystery of slow electrons

<p><strong>Researchers from CEITEC Masaryk University in Brno have made significant strides in understanding how plants adapt to genotoxic stress and subsequent mutations, specifically through changes in DNA folding. They discovered that when Arabidopsis plants lack certain auxiliary proteins, called histone chaperones, the plants adapt and adjust the composition of proteins that are necessary for the correct arrangement of DNA in the nucleus. This discovery may have implications for biotechnology and agricultural innovation.</strong></p><p>The nucleus of a plant cell is the control centre where DNA is stored and arranged in a structure known as chromatin. This is achieved by wrapping DNA around proteins called histones, similar to winding a string around spools. Auxiliary proteins, called histone chaperones, ensure that the DNA is properly wrapped by helping the histones fold and transport correctly.</p><p><i>“We discovered that when these chaperones are missing, it leads to errors in histone deposition. Plants adapt to this situation by incorporating alternative variants of histone proteins and modifying their chemical properties, so they mimic the original pattern occurring before the mutation,”</i> says Michal Franek from the research team at CEITEC Masaryk University. Defects in chromatin assembly are usually lethal, but plants are exceptional in this respect, having developed skills for adaptation. The researchers are trying to understand what triggers this adaptation as these changes help to keep the plant’s genetic information stable, ensuring proper growth and development.</p><p><i>“It was already known that errors in histone deposition lead to changes in chromatin properties and impact plant health. Proteomics allowed them to comprehensively analyse histone proteins, while fluorescence microscopy provided detailed visualizations of histones at the level of individual cells. The high-quality infrastructure of CEITEC Core Facilities played a key role in their research.</p><p>Given the sessile nature of plant life, adaptation to adverse conditions is essential and represents a key evolutionary tool. If research is to contribute to the development of more resilient crops, it is important to understand how plants cope with mutations and genotoxic stress and to what extent they are able to withstand environmental challenges.</p>

<p><strong>Michal Holčapek, a researcher in Analytical Chemistry of the Faculty of Chemical Technology of the University of Pardubice has won the Czech Brains Jury’s Extraordinary Award for 2024. With the Czech Brain Award (Česká hlava) being the most prestigious research award in the Czech Republic, Michal Holčapek won the award for his research into lipids and discovery of a method that can diagnose the pancreatic cancer from blood analysis only.</strong></p><p><strong>The 2024 award ceremony was broadcast live by the </strong><a target="_blank" rel="noopener noreferrer" href="https://www.ceskatelevize.cz/porady/10213906625-ceska-hlava/22454416015/"><strong>Czech Television</strong></a><strong>.</strong></p><p>The Czech Brains Award is <strong>the most prestigious award</strong> available to researchers in the Czech Republic. The award has been been presented in several categories since 2002. Since 2007 the Czech Brains Initiative has organized a similar award for secondary-school students called <strong>Little Czech Brain</strong> (Česká hlavička). Last year, <strong>Ing. Barbora Kamenická, Ph.D., a graduate from the University of Pardubice, received the award in the Doctorandus category for technical sciences.</strong></p><p><strong>Professor Michal Holčapek has been researching lipid analysis for a long time with special focus on the detection of some cancer types.</strong> Pancreatic cancer is one of the most aggressive tumours with 80% of the cases being diagnosed too late for the therapy to be effective and thus a 5-year survival rate of less than 10%. For more than 20 years, Professor Holčapek’s team has focused on lipidomic analysis using mass spectroscopy combined with liquid or supercritical fluid chromatography, which enabled them to develop a number of validated analytical methods for detailed description of lipids in biological samples. Thanks to their research, it was possible to develop new screening methods to improve early diagnosis of pancreatic cancer.</p><p>The method is based on lipidomic analysis of blood, which determines the concentrations of more than 150 lipids. Thanks to the data from patients with pancreatic cancer and healthy individuals, a statistical model is created that can determine <strong>whether the patient suffers from the cancer with a certainty of 95%</strong>. The blood test has a number of advantages: it only requires a blood sample and it is reliable to detect even early stages of cancer and thus diagnose the cancer even before the onset of the first symptoms. In addition, similar concentrations of lipids have been observed in other cancer types, showing potential of a universal test for more types of cancer.</p><p><strong>Lipidica, a.s., a spin-off company founded by the University of Pardubice and FONS JK Group, a.s. is conducting the clinical trials</strong> in cooperation with Masaryk Oncological Institute in Brno, Olomouc University Hospital and another 13 hospitals across the Czech Republic. If the method is proved to be accurate and reliable in clinical testing, it can become a basis for national screening programme for those with high risk of pancreatic cancer, which would be unprecedented worldwide.
