Goal: To demonstrate the analysis of process water from hydrogen fuel cells for inorganic trace contaminants with high sensitivity and accuracy using triple quadrupole ICP-MS.

Conclusions: The iCAP TQe ICP-MS was employed to analyze 53 elements including fluorine in process water derived from hydrogen fuel cells. Among the analytes, several critical interferences can cause unexpected bias, and these were investigated closely for effective and complete removal by means of selective collision / reaction cell reactions with oxygen. This study also included the development of a method for the analysis of fluorine, generally only possible in an indirect measurement. This analytical method was rigorously tested, and the results obtained clearly demonstrate the fowling analytical advantages:

<li>The iCAP TQe ICP-MS operated in TQ-O2 and He KED modes allowed the analysis of 53 elements in relevant sample matrices over a wide concentration range. For minor elements, the method was tested over 4 orders of magnitude (from 0.01 to 100 µg·L-1), whereas major elements were tested between 1 to 100 µg·L-1.</li>
<li>Using a high throughput analysis approach, with a new autosampler equipped with a Step Ahead feature for minimizing total analysis time, all elements relevant for the development of hydrogen fuel cell technology could be measured accurately with low detection limits.</li>
<li>The use of TQ-O₂ mode provided advanced performance for eliminating the polyatomic interferences of phosphorus, sulfur, silver, and cadmium in the simulated hydrogen fuel cell process water. This covered both polyatomic interferences as well as overlaps created by intense signals from isotopes of elements present in high concentrations.</li>
<li>The iCAP TQe ICP-MS offers the flexibility to choose high resolution for the Q1 and Q3 quadrupoles, enabling complete elimination of polyatomic interference and isobaric interferences. </li>
<li>The feasibility of fluorine analysis was demonstrated using an indirect method after complexation with barium. The iCAP TQe ICP-MS showed accurate and precise determination of fluorine as proven by the spike recovery tests performed.</li>
<li>Excellent stability performance was obtained with the iCAP TQe ICP-MS together with excellent CCV and spike recovery results over a 3-hour run without internal standard correction. This provides the advantage of avoiding potential contamination from addition of the internal standard solution.</li>

Basic inclusion analysis
Non-metallic inclusions dramatically affect steel properties and cause process issues like nozzle clogging. The costs associated with bad steel properties, process issues or customer complaints may amount to several hundred thousand dollars every year. Controlling the formation and the modification of non-metallic inclusions has therefore become essential to the production of modern steels as demanded by today’s markets.
The Thermo Scientific ARL iSpark Plus is a high-end metal analyzer that allows precise and accurate elemental analysis of steel. It also enables ultra-fast analysis of non-metallic inclusions, enabling a steel plant to control steel quality and production process even more effectively, and considerably reduce the costs associated with unwanted inclusions.

Abstract: The Agilent Resolve handheld Raman system is a versatile analyzer that can be deployed on location to identify a broad range of materials. It uses traditional surface analysis mode and through-barrier mode through Agilent&#39;s proprietary spatially offset Raman spectroscopy (SORS) technology. SORS enables samples that are concealed behind barriers such as colored and opaque plastics, paper, and highly pigmented paints and varnishes to be identified. This application note details how the Resolve Raman system can be used to detect concealed endangered species products using SORS. Data are presented that show the effectiveness of Resolve operating in traditional surface mode to distinguish ivory from a plastic substitute. Further analysis of endangered animal products using surface mode enabled the division of animal products into two broad categories: mineral-based and protein-based. Principal components analysis (PCA) was used to further differentiate the protein-based samples, which included pangolin scale, rhino horn, dried seahorse, and totoaba fish bladder, based on their spectral differences.
Conclusion: The Agilent Resolve handheld Raman analyzer includes various spectral libraries that enable the identification of a broad range of chemicals and materials. This study has shown the application of the instrument and the SORS technique for the detection of endangered animal species. Using a combination of conventional surface and through-barrier modes of analysis, Raman spectroscopy and SORS technology can be used to:

CHAMPAIGN, Ill. — Voids, or empty spaces, exist within matter at all scales, from the astronomical to the microscopic. In a new study, researchers used high-powered microscopy and mathematical theory to unveil nanoscale voids in three dimensions. This advancement is poised to improve the performance of many materials used in the home and in the chemical, energy and medical industries — particularly in the area of filtration.

Magnification of common filters used in the home shows that, while they look like a solid piece of material with uniform holes, they are actually composed of millions of randomly oriented tiny voids that allow small particles to pass through. In some industrial applications, like water and solvent filtration, paper-thin membranes make up the barriers that separate fluids and particles. 
“The materials science community has been aware of these randomly oriented nanoscale voids within filter membranes for a while,” said Falon Kalutantirige, a University of Illinois Urbana-Champaign graduate student. “The problem was that the complex structure of the membrane as a whole — which looks like nanoscale mountain ranges when magnified — was blocking our view of the void spaces. Because we could not see them, we couldn’t fully understand how they affected filtration properties. We knew that if we could find a way to see them, we could then figure out how they work and ultimately improve filter membrane performance.”
The study, directed by Illinois <a href="https://matse.illinois.edu/">materials science and engineering</a> professor <a href="https://matse.illinois.edu/people/profile/qchen20">Qian Chen</a> and University of Wisconsin-Madison professor <a href="https://directory.engr.wisc.edu/me/Faculty/Li_Ying/">Ying Li</a>, is the first to integrate materials science and a mathematical concept called graph theory to help image and map out the random placement of these voids within filtration materials. The findings are published in the journal Nature Communications.

Building on a <a href="https://news.illinois.edu/view/6367/1945681133">previous study</a> that used laboratory models, the researchers said the new study focuses on far more complex membranes used in industrial applications. 
“The surfaces of the membranes we studied in this work look flat to the naked eye, but when we zoomed in using <a href="https://icpms.labrulez.com/library?instrumentationItems=98&amp;search=transmission+electron+microscopy">transmission electron microscopy</a>, electron tomography and <a href="https://icpms.labrulez.com/library?instrumentationItems=98&amp;search=atomic+force+microscopy">atomic force microscopy</a>, we could observe these voids nestled within these nanoscale mountainous landscapes that we call crumples,” said Kalutantirige, the study’s first author. 
However, the team needed a means to measure and map these features to build a quantitative predictive model and gain a more holistic picture of the membrane surfaces. 
“Mapping and measuring alone will work for materials with a regular or periodic structure, making it mathematically simple to scale up our models and predict how structural properties will influence the material’s performance,” Chen said. “But the irregularity we observed in our study pushed us to use graph theory, which gives us a mathematical way to describe this heterogeneous and messy — but practical — material.” 
Graph theory helped the team finally gain a more holistic understanding of the filter membrane structure, which led them to discover a strong correlation between the unique physical and mechanical properties of random empty space and improved filtration performance. 
“Our method is a very universal technique for describing materials,” Kalutantirige said. “Many things we use in everyday life and science are not made of materials composed of repetitive uniform structures. So, the beauty of the method, I think, is that we can capture the ‘regularness’ of irregular structures.”
The team said that this advancement will improve the effectiveness of many next-generation porous materials, such as polymers used in drug delivery.
“The title of this study hints at the concept of ‘beyond nothingness,’ and by that, we mean that these empty, void spaces are really important when it comes to developing the best filtration membranes,” Chen said. “This work is only possible with our wonderful team of collaborators. <a href="https://chbe.illinois.edu/people/profile/x2su">Xiao Su</a> helped us with the membrane performance testing. <a href="https://chemistry.illinois.edu/emad">Emad Tajkhorshid</a>, <a href="https://chbe.illinois.edu/people/profile/cms">Charles Schroeder</a> and <a href="https://chemistry.illinois.edu/jsmoore">Jeffrey Moore</a> worked with us on the synthesis and analysis of the polymer systems.”

Johns Hopkins engineers have created a new optical tool that could improve cancer imaging. Their approach, called SPECTRA, uses tiny nanoprobes that light up when they attach to aggressive cancer cells, helping clinicians distinguish between localized cancers and those that are metastatic and have the potential to spread throughout the body.

Scalable and more cost-effective than current methods, SPECTRA leverages a first-of-its-kind combination: <a href="https://icpms.labrulez.com/library?instrumentationItems=48">Raman spectroscopy</a>—which uses the scattering of laser light to provide detailed information about molecular vibrations—and DNA origami, which involves folding DNA into specific shapes, as in the Japanese art of paper-folding. The team used the folded DNA as a scaffold to create precisely arranged plasmonic nanoparticles, Raman reporters (a molecule that produces a strong signal when analyzed using Raman spectroscopy), and cancer-targeting DNA sequences. These multifunctional nanoprobes were then tested on cancer cells.
The team found that, unlike <a href="https://icpms.labrulez.com/library?instrumentationItems=100&amp;search=+CT">CT</a> or MRI scans that can indicate the presence of a tumor but not the specific molecular signatures that can alert physicians to current or impending metastasis, SPECTRA effectively and consistently bound to metastatic prostate cancer DU145 cells and even differentiated between those and non-metastatic cells.
In addition, the researchers selected a Raman reporter—a signaling molecule—that results in an active and distinct signal in a range that made it stand out clearly against the background of normal tissue, helping clinicians more precisely locate disease.
&quot;It&#39;s a smart design that gives high enhancement to the Raman signal, and it&#39;s uniform,&quot; said team member Swati Tanwar, a postdoctoral fellow in mechanical engineering. &quot;It can distinguish aggressive cancer cells from non-aggressive based on the intensity of the signal. In a tumor, if 10% of the cells are aggressive and 90% are non-aggressive, the 10% will light up and give a very high signal.&quot;
Tanwar said that each strand of DNA in the origami scaffold has a unique sequence and occupies a specific position in the folded origami nanostructure. This meticulous arrangement facilitated the creation of multifunctional SPECTRA nanoprobe.
&quot;Raman spectroscopy is a molecular fingerprinting tool,&quot; said team member Lintong Wu, a PhD student in mechanical engineering. &quot;Molecules can look similar at a distance, but using Raman spectroscopy they show different peaks and signals throughout the entire spectrum.&quot;

New technique to diagnose cancer metastasis uses origami nanoprobes

<h4>Abstract</h4>
<ul>
<li>Multiple myeloma is an incurable form of blood cancer. Over time, the cancer cells become resistant to the currently prescribed drugs.</li>
<li>Using the fully automated pharmacoscopy platform, ETH Zurich researchers are testing hundreds of drugs and combinations of drugs on bone marrow biopsies in a high-throughput process.</li>
<li>These tests show which treatment options combined with existing therapeutics would be possible for specific patients.</li>
</ul>
Multiple myeloma is a rare blood cancer caused by the uncontrolled multiplication of abnormal plasma cells. These plasma cells are a special type of white blood cells that play an important role in the immune system by producing essential antibodies in the bone marrow and lymph nodes.
Despite an increasing number of approved drugs and treatment approaches such as immunotherapy becoming available, the disease is still not curable. The average life expectancy of patients after diagnosis is only five years.
One of the main challenges is the cancer’s tendency to return even after treatment. This is because treatment makes the cancer cells more resistant to the drugs used, until eventually, after several rounds of treatment, no effective options remain.
To address this issue, ETH researchers have adapted their screening platform to look for ways out of this problem, and thereby offering new hope for more successful treatment outcomes.
<h4>Biopsies under the microscope</h4>
The researchers are using a high-throughput screening method called pharmacoscopy, developed by Professor Berend Snijder, to test the effectiveness of various treatments on the patient’s cancer cells.
This state-of-the-art method allows for several hundred different treatment combinations to be tested simultaneously outside the body. By analysing the reactions of the cells to each treatment, they can determine which method holds the most promise for each patient.
To do this, the cells from the biopsies are placed in special plates with 384 small wells, each containing a different combination of potential treatment substances. After 24 hours, the cells are stained using different antibodies, and their reactions are evaluated using images generated by automated microscopy. A deep-learning algorithm is then used to identify and classify the cell types. The process is largely automated, allowing for efficient and accurate analysis of the results.
<h4>138 biopsies individually tested</h4>
The researchers used pharmacoscopy to closely examine 138 bone marrow biopsies from 89 myeloma patients with different stages of multiple myeloma, including newly diagnosed and untreated, as well as patients that underwent multiple treatment rounds.
The goal was to observe the behaviour of cancer cells in response to various approved drugs and drug combinations in each biopsy. Based on the cells’ appearance, the researchers could determine the best treatment option for each patient.
Although the Snijder lab had previously used pharmacoscopy in similar studies on other types of blood cancer, such as lymphomas and leukaemias, the platform had to be adapted for this myeloma study.
<h4>Hope for more effective treatments</h4>
The groundbreaking work done by the researchers at ETH and University Hospital Zurich offers hope for more effective treatments for multiple myeloma. By using Pharmacoscopy to test hundreds of treatments, the researchers were able to identify new, more effective treatment options for each patient.
This new personalized medicine approach is transferable to the clinic and can thus help doctors find the best option for their patients at an early stage. &quot;First, however, we will have to validate the method further in clinical trials,&quot; says Snijder.
Now, the Snijder lab wants to develop the platform further to expand its use to solid tumours. Unlike blood cancers, solid tumours must first be dissociated to a certain degree before they can be tested in the 384-well plate format. They are currently adapting the screening platform for brain tumours, among others.

Conclusion
A custom process was developed for mixing carbon dioxide in argon and supplying it to the plasma of the Agilent 8900 ICP-QQQ. The study showed that the use of gaseous carbon for plasma ionization enhancement provided advantages over traditional liquid sources of carbon, such as acetic acid, in terms of cost and performance. The scalability of the gas supply system can be enhanced by increasing the size of the ballast tank, enabling the efficient use with multiple ICP-MS instruments. This optimization would lead to more cost reductions.
The use of CO2 in Ar, supplied from a ballast tank, ensures easy and rapid adjustment of the carbon composition of the plasma with minimal flush out times, addressing a common challenge associated with liquid carbon sources. Carbon was shown to enhance the signal of key elements such as arsenic in the plasma, while maintaining the sensitivity of other elements. Abstract: Ensuring the quality of electrolytes used in lithium-ion batteries (LIBs) is crucial for maintaining the safety, performance, and longevity of these energy storage devices. LIB electrolytes consist of lithium salts dissolved in organic solvents. Any discoloration (yellowness) of these near-clear solutions can indicate contamination or degradation. Industry methods such as ASTM D5386-16 Standard Test Method for Color of Liquids Using Tristimulus Colorimetry can be applied to assess the quality of electrolytes by conducting instrumental color measurements using UV-Vis spectroscopy. Given the low absorbance of colorless-to-near-colorless liquid samples like LIB electrolytes and solvents, a highly accurate and sensitive UV-Vis spectrophotometer is essential.
This study demonstrates an effective quality assessment method for LIB electrolytes and solvents using the Agilent Cary 3500 Flexible UV-Vis spectrophotometer fitted with the optional Agilent Cary Sipper flow cell pump. The method is suitable for production quality control of hazardous samples, ensuring safe sample handling, high throughput, and high quality results for production-critical chemicals.
Conclusion: The Agilent Cary 3500 Flexible UV-Vis spectrophotometer provided accurate and precise color measurements for colorless to near-colorless LIB electrolytes and solvents in accordance with the ASTM D5386 standard method. Advantages of the Cary 3500 Flexible method and findings based on the data set include:

<li>Hazardous samples were introduced directly into a 10 mm path length flow cell using the Cary Sipper pump, ensuring user safety and high sample throughput without compromising data quality.</li>
<li>The superior sensitivity (high signal-to-noise ratio) of the Cary 3500 combined with Agilent Cary WinUV Color application software enabled the reliable monitoring and control of electrolyte properties by analyzing light absorption in the visible spectrum.</li>
<li>Higher Pt-Co color (APHA) values of the used LIB electrolytes compared to fresh samples suggested a deterioration in quality due to the color changes.</li>
<li>Color measurements could be used to improve the quality control of LIB electrolytes and solvents by identifying discoloration—an indication of contamination or degradation that could affect battery performance.</li>
<li>This effective method could be implemented in both manufacturing QC settings and R&amp;D laboratories.</li>
</ul>
QC color measurements using the Cary 3500 Flexible UV-Vis would ensure that only high-quality electrolytes are used in the production of LIBs, enhancing their safety, efficiency, and durability.

In the research article published in ACS Analytical Chemistry journal the researchers from the Department of Physical and Analytical Chemistry, and the Department of Functional Biology (Physiology), University of Oviedo, Spain presented novel analytical strategy for detecting miRNA in human plasma without relying on nucleic acid amplification.
MicroRNAs (miRNAs) are short RNA sequences that play a key role in gene expression and respond to physiological changes, such as those induced by physical exercise. This study presents a novel analytical strategy for detecting miRNA in human plasma without relying on nucleic acid amplification (eg PCR). The method uses a double hybridization process with a magnetic microparticle and a gold nanoparticle for highly selective detection, even at single nucleotide variations. The approach is validated against rt-qPCR and can differentiate between sedentary and active individuals based on miRNA levels. Additionally, this versatile platform can be easily adapted for other miRNA sequences.

MicroRNAs (miRNAs), which are short single-stranded RNA sequences between 18 and 24 nucleotides, are known to play a crucial role in gene expression. Changes in their expression are not only involved in many diseases but also as a response to physiological changes, such as physical exercise. In this work, a new analytical strategy for the sensitive and specific analysis of miRNA sequences in human plasma is presented. The developed strategy does not depend on any nucleic acid amplification process and can be obtained in direct correlation to the number of events obtained by using single-particle ICP–MS measurements. The high selectivity of the assay (up to single nucleotide polymorphisms) can be achieved by a double hybridization process of the target miRNA with a complementary capture oligonucleotide that is conjugated to a magnetic microparticle and simultaneously with a complementary reporter oligonucleotide conjugated to a gold nanoparticle. Thanks to the novel approach followed in this method, the stoichiometry of the oligonucleotide-nanoparticle conjugates does not need to be addressed for the quantification of the target miRNA, which also represents a big advantage over other similar methods. The optimized method is applied to the determination of a miRNA as a biomarker of physical exercise in non-spiked human serum samples, and the results are validated against rt-qPCR. The achieved sensitivity permits the direct differentiation among sedentary and sportive subjects. This general platform can be easily applied to any other sequence by only modifying the capture and reporter oligonucleotides, paving the way for multiple clinically interesting applications.
<h3>Introduction</h3>
Microribonucleic acids (miRNAs) are small noncoding RNA biomolecules with a length between 18 and 24 nucleotides that are involved in different cellular processes, such as gene expression modulation. In fact, the main function of miRNAs consists of their union with mRNAs by base complementarity, either preventing their translation or inducing their degradation, resulting in both cases in a negative post-transcriptional regulation of gene expression. ...In this work, miRNA quantification is based on the specificity of nucleic acid hybridization and the highly sensitive and specific nanoparticle counting capabilities of individual nanoparticles by SP-ICP-MS. The amplification-free workflow that we propose is based on the hybridization of a specific target miRNA sequence (miR-16-5p) with two DNA sequences that are each complementary to half of the target, respectively. One of them is conjugated to a magnetic microparticle (capture probe) that is used for the enrichment and washing of the target sequence. The other half-complementary sequence is conjugated to a gold nanoparticle that is detected by SP-ICP-MS (detection probe). Although the analytical strategy is comparable to this from Xu et al., (34) it is important to note that our methodology is applied to quantify different levels of expression of miRNA sequences as short as 22 nucleotides that are naturally present in human serum samples. Scheme 1 summarizes the main differences between the proposed strategy and other methods for miRNA quantification. Therefore, this methodology represents a step forward in facing challenges like miRNA pre-concentration in a complex matrix with the important role of quantification, which is crucial to extracting conclusions depending on the expression levels.

<h3>Experimental Section</h3>
<h4>Instrumentation</h4>
The measurements were performed by using the <a href="https://icpms.labrulez.com/library?instrumentationItems=83-90&amp;mainauthorItems=3&amp;search=+iCAP+TQ">iCAP TQ triple quadrupole ICP–MS system from Thermo Fisher Scientific</a>. All measurements, including single particle ICP–MS measurements, were done using the standard configuration for sample introduction, including the MicroMist nebulizer at a sample flow rate of 0.4 mL min–1 and a cyclonic spray chamber because a high transport efficiency was not crucial for this application. The sample introduction system was operated in combination with an ASX-560 autosampler from Teledyne Cetac. The ICP–MS was tuned daily to maximize sensitivity while keeping the formation of oxides and doubly charged species below 3 and 5%, respectively.
For spectrophotometric measurements of nucleic acid concentrations, a NanoDrop spectrophotometer (Thermo Fisher Scientific) was used. A transmission electron microscope MET JEOL-JEM-2100F was used to obtain images of the nanoparticles. A scanning electron microscope MEB JEOL-6610LV with EDX microanalysis was used for the images and elemental analysis of microparticles. <h3>Results and Discussion</h3>
<h4>Characterization of Naked and Conjugated Gold Nanoparticles</h4>
The gold nanoparticles used as metal labels for the detection probe were characterized both before and after their conjugation with the DNA oligo by SP-ICP-MS, transmission electron microscopy (TEM), and DLS. The results of SP-ICP-MS, shown in Table 1 and Figure 1, reveal that the obtained mean diameters were 23.3 ± 3.3 nm, which was slightly larger than the value provided by the manufacturer (22 nm). When a Student-t test was performed to compare the mean diameters of the conjugated and original nanoparticles, no statistical differences between them or compared to the value given by the manufacturer was observed at a 95% confidence level. Under these conditions, the size limit of detection was calculated as 18 nm, well below the size of the nanoparticles. Both sets of particles showed a narrow distribution, as can be seen in Figure 1, although the nanoparticles showed a more monodisperse distribution after the conjugation (Figure 1B), probably due to an increased stability in solution when coated with negatively charged oligonucleotides...

<h3>Conclusions</h3>
In this study, we introduce a novel amplification-free miRNA analysis method using magnetic microparticles for capture and preconcentration and gold nanoparticles as labels that amplify the response for ICP–MS detection. Eliminating the need for nucleic acid sequence amplification, our approach streamlines the analysis process while reducing potential sources of error that are common in conventional techniques. Moreover, it circumvents the often challenging considerations of oligonucleotide-nanoparticle conjugation stoichiometry. Importantly, this methodology also demonstrates remarkable resilience to batch-to-batch variations in conjugation processes, which are accounted for in daily calibration. The high selectivity of the assay, thanks to the complementarity interactions between probes and analytes, has been demonstrated, achieving the possibility of clearly discriminating a 2-base mismatch. Even more, the response to single nucleotide polymorphisms is seriously affected, and these kinds of important sequence variations can also be also discriminated. The developed method, which has been validated against the gold-standard RT-qPCR, can be easily modified to detect any miRNA sequence by changing only the detection and capture probes. This allows for high versatility with minimal optimization for the detection of any miRNA sequence.

<h4>Description and use:</h4>
These reference materials are aqueous calibration solutions of individual elements and ions with certified/assigned concentration value in the range from 1 mg/l to 10 g/l or 1mg/kg to 10g/kg (m/m) and aqueous calibration solutions of combinations of elements and/or ions with certified/assigned concentrations at various levels relevant to particular analytical requirements. They can be used by analytical techniques analysing aqueous solutions such as atomic spectrometry (<a href="https://icpms.labrulez.com/library?instrumentationItems=85">AAS</a>, AFS, <a href="https://icpms.labrulez.com/library?instrumentationItems=84">ICP-OES</a>, <a href="https://icpms.labrulez.com/library?instrumentationItems=83">ICP-MS</a>), molecular absorption spectrometry, ion chromatography and some selected electroanalytical methods.
<h4>Starting primary substances and other applied materials:</h4>
Metals or compounds (in mass calculated from the content of the certified component and a target volume of the batch) are used as starting materials for the production of these RM. High purity metals with a declared assay of minimum 99.95 % are preferred and compounds of a defined and constant stoichiometry and declared assay and/or impurities content are used when a suitable metal is not available [1, 2]. The supplier of primary substances must comply with ISO 17034 [3], namely to employ a certified Quality Management System. Each supply of the primary substance must be accompanied with complete documentation, including an identification of sources and treatment, assay and impurity contents. As required by this Guide, the supplier’s data are verified by the CRM producer and by other subcontracted accredited laboratories (e.g. trace metal impurities by spectroscopic methods such as ICP-AES, ICP-MS, AAS, the assay by primary gravimetric or volumetric methods).
Acids (prepared by sub-boiling distillation), other ultrapure chemicals (ammonium hydroxide, hydrogen peroxide, etc.) and deionised water with a resistivity 18 MΩ.cm are further applied in the (C)RM preparation. Their impurity contents are regularly monitored. They are generally negligible (~ 1 μg/l) as compared with the concentration of the certified/assigned constituent.
<h4>Weighing, volume and temperature measurement:</h4>
A class 1 electromechanical analytical balances with a valid verification is used to weigh the primary substances. A performance of this balance is regularly controlled by the Czech Metrology Institute. Mettler weights class F1 (5 g and 100 g) are used for an internal calibration control. Two calibrated mercury or alcoholic thermometers 0 – 50 °C (with a scale division by 0.1 °C), regularly calibrated by authorized institutions are used for the temperature measurement.
All volumetric vessels used (flasks, pipettes, burettes) are of Accuracy Class 1, calibrated by their producers and delivered with a relevant certificate. The calibration of all labware is regularly checked.
<h4>Coding and packaging:</h4>
(C)RMs are delivered in amber HDPE bottles or amber glass bottles whenever an interaction of the analyte with HDPE may be suspected (e.g. mercury, precious metals). Prior to filling, the bottles and caps are leached by HCl (5% v/v) for a longer period, washed repeatedly with deionised water and thoroughly dried. The bottle filled with (C)RM is hermetically closed with a polypropylene screw cap and tightened with parafilm immediately after filling and sealed into reclosable aluminium bag or PE foil. The bottle is labelled with a resistant plastic label containing all data as required by (C)RM standards, i.e. (C)RM and batch codes, volume of the packed unit, type of the matrix, producer identification and contact data, further data and warning symbols when required by specific regulations [4, 5].
<h4>Homogeneity and stability:</h4>
The (C)RM are considered homogeneous due to their physico-chemical character (diluted solutions) and thus a detailed homogeneity testing is not required. This fact is supported by a long-term experience of the producer and other recognized producers of similar materials. In compliance with a recommendation of ISO 17034, homogeneity testing is performed (as prescribed in ISO Guide 35) at randomly chosen solutions and evaluated by ANOVA. 
An assumption of stability and the consequent expiration period is based on both over 20 year experience with the preparation of similar RMs and a test of possible instability factors. These factors are of general (e.g. water evaporation by regular weighing of the filled bottle) and specific character (e.g. adsorption and desorption of the particular analyte on the walls of the bottle, precipitation, etc.).
Along with these tests the content of the certified/assigned constituent in the bottle is compared (at least twice during the validity period) with that of a newly prepared batch and/or a corresponding (C)RM form other producers. For these comparisons, both primary analytical methods (gravimetry, titrimetry) and some instrumental methods (ICP-OES, AAS) working in the regime of maximum precision (the application of internal standard, bracketing technique etc.) are used. For selected analytes, an accelerated isochronous stability test (in the temperature range of -20 °C to +40 °C) is realized. Based on these tests, transport conditions for a particular (C)RM are defined.
<h4>Certification, certified and assigned value and their uncertainties:</h4>
The concentration value of CRM/RM are certified/assigned on the basis of gravimetric preparation (refering to ISO Guide 35), i.e. on weighing and volume measurement [6]. They are calculated from an assay of the certified constituent in the primary substance, declared by its supplier and duly verified by the CRM producer. The certified/assigned concentration (m/v) and its uncertainty are expressed in g/l (alternatively in mg/kg) at a temperature of 20 ± 0.1 °C.
The uncertainty of the certified/assigned values is estimated in compliance with ISO and EURACHEM methodologies [7, 8]. It combines the calculated standard uncertainties of the individual CRM/RM preparation steps with an expert estimate of the standard uncertainty of the assay of the certified constituent in the primary substance, usually a dominant contribution. The expanded combined uncertainty calculated using coverage factor k = 2 is expressed as a two-sided half-interval by one significant figure, with the certified value given by the same number of digits.
The particular certified values and uncertainties, mostly at a level of 0.2% (rel.), are specified in the Certificate together with the expiry terms and other relevant information.

The certified/assigned analyte concentration and its uncertainty is further experimentally verified by means of primary and instrumental analytical methods [9]. However, the results received are not used in the calculation of the certified/assigned value and serve only for the confirmation that this value is true.
<h4>Traceability and quality assurance:</h4>
Direct traceability to SI unit for the weight (1 kg) is secured by using a primary substance within comprehensively verified content of the analyte constituent and by gravimetric method of preparation. This traceability is for each produced batch of (C)RM further confirmed by analytical data acquired from the determination of certified analyte with validated primary and instrumental analytical methods. Only internationally accepted references (SRM NIST, TraceCERT etc.) are used.
The producer operates a certified quality management system ISO 9001 [10] and is accredited according to ISO 17034 [12].
Control analytical laboratory of the producer is accredited according to ISO/IEC 17025 [11].
<h4>Storing and instructions for use:</h4>
This (C)RM must be stored in the original closed bottle (in an upright position) between 5 °C and 30 °C, away from any strong light sources (e.g. sunshine, UV lamp). The producer guarantees declared life time (shelf life) and expiration time (period of validity) provided (C)RM is properly stored and professionally handled. The temperature of the bottle with (C)RM must be 20 ± 0.5 °C before every use. After use, the bottle must be immediately tightly capped. It is recommended to tighten the screw cap with parafilm Do not pipette from the bottle. Do not return removed aliquots to bottle. The user should differentiate between the life time (shelf life) of the particular (C)RM (usually 5 years from the date of production) and expiration time (never more than 24 months from the first opening of the bottle). Both values are presented in the Certificate and Identification sheet. It is necessary to indicate on the Certificate/Identification sheet the date of the first opening of the bottle or aluminium bag and the expiration time, which depends on the date of the first opening. It is necessary to indicate the expiration time on the label of the bottle too.
It is not recommended to use the standard solution when the bottle contains less than 10 % of the solution. Therefore, in case of opaque bottle, it is important to indicate the amount of the solution used, e.g. on the label.
Transpiration through amber HDPE bottles (losses of the solution mass resulting in a gradual increase in the analyte mass fraction) was not observed. It is the producer’s experience that the loss of vapour phase occurs solely around the screw cap of the bottle which is not airtight. Therefore, it is recommended to cover the screw cap and the neck of the bottle with parafilm layer after each opening and put the bottle into the reclosable aluminium bag, if it is delivered in.Safety regulations for transportation, storage, and use of this (C)RM are specified in the respective Safety Data Sheet.
