These sulfur-, nitrogen-, and oxygen-containing or substituted multi-ringed aromatic analytes can provide further insight into the processes used to generate these fuels and their potential impacts on fuel performance.</p><p>This presentation explores the different types of results that are possible when approaching diverse samples using several statistical methods to highlight and identify significant variation between aviation fuel samples. Multiple aviation fuels from production processes including Fischer-Tropsch reactions and hydroprocessed esters and fatty acids (HEFA) are compared to traditional petroleum-based fuels.<br>&nbsp;</p>

Their coexistence worsens lung function and leads to poor clinical outcomes. To investigate their metabolic interactions, we analyzed the volatile space of P. aeruginosa and A. fumigatus using </strong><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?instrumentationItems=2-3-13-7"><strong>SPME-GC×GC-TOFMS</strong></a><strong> across four sample classes: Media, P. aeruginosa monoculture, A. fumigatus monoculture, and their co-culture. GC×GC-TOFMS provides high-resolution, high-sensitivity separation of complex metabolomic samples, but it also generates high-dimensional data that can be challenging to analyze.&nbsp;</strong></p><p>Therefore, feature selection and chemometric methods are essential to extract meaningful information. Additionally, peak table alignment issues due to retention time shifting across samples can in principle hinder the comparative analysis of multiple samples. Hence, we applied tile-based Fisher-ratio analysis using ChromaTOF Tile software to discover analytes that are statistically significant in concentration differences across samples with replicates while minimizing retention-time shifts. This platform generates a comprehensive hit list that links detected analytes across all samples. In summary, our study integrated tile-based analysis to generate a comprehensive peak table that relates all analytes in all samples in terms of up and down regulation of their concentrations. With further analyte identification, we have characterized analytes specific to each sample class to ultimately learn more about Pseudomonas aeruginosa and Aspergillus fumigatus pathogens and their co-culture.</p><h3><strong>Video Transcription</strong></h3><p>In this study, <span>Wenjing Ma</span> from the <span>University of Washington</span> presents a data-driven approach for analyzing complex GC×GC–MS datasets, with a focus on volatile metabolites produced by clinically relevant microorganisms. This technique provides high acquisition speed and full mass spectral information, allowing detailed chemical characterization. However, it also generates extremely complex datasets. A single chromatogram may contain up to a million data points, and when multiple samples are considered, the data effectively become four-dimensional. Such complexity makes direct manual interpretation impractical.</p><h4><strong>Data Processing Strategy</strong></h4><p>Traditional approaches to data reduction, such as generating peak tables or averaging chromatograms, present significant limitations. Peak alignment can be compromised by retention time shifts, while averaging can reduce resolution through peak broadening. To overcome these issues, the study applies a tile-based feature selection strategy built on statistical analysis.</p><p>The chromatographic space is divided into small regions, referred to as tiles, and each tile is evaluated using a statistical metric that compares variability between sample classes to variability within classes. This approach allows the identification of features that show the greatest differences across experimental conditions. Importantly, the method is extended to two-dimensional chromatographic data and applied across individual mass channels, enabling a highly detailed and systematic analysis.</p><p>The workflow is implemented using <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/348"><strong>Chromatof Tile software</strong></a>, which produces ranked lists of statistically significant features and supports visualization tools such as principal component analysis (PCA) plots for confirming class separation.</p><h4><strong>Results and Interpretation</strong></h4><p>Initial inspection of raw chromatograms reveals that while the media control exhibits relatively low signal intensity, the biological samples show highly complex and overlapping patterns. Differences between monocultures and co-cultures are not readily apparent without advanced data processing.</p><p>Using tile-based analysis, hundreds of statistically significant features were identified. These features provide insight into metabolic behavior. For example, compounds that are abundant in the media but reduced in biological samples are likely being consumed by the organisms. Conversely, compounds that are elevated in specific cultures may represent metabolites produced by one organism or influenced by microbial interaction.</p><p>The reproducibility of the experimental setup is supported by consistent signal patterns across replicates, indicating reliable sample preparation and data acquisition.</p><h4><strong>Statistical Evaluation</strong></h4><p>To further refine the interpretation, pairwise statistical comparisons were performed between all sample classes. This enables identification of specific differences between monocultures and co-cultures, highlighting which metabolites are associated with individual organisms and which are affected by their interaction.</p><p>The results suggest that certain metabolites are dominant in <i>Pseudomonas aeruginosa</i> monocultures but are altered in co-culture conditions, indicating potential biological competition or suppression mechanisms.</p><h4><strong>Alternative Approach for Limited Data</strong></h4><p>Recognizing that biological replicates are not always available in practice, the study also explores an alternative strategy based on the coefficient of variation (CV). This method allows meaningful analysis even when only a single replicate per condition is available, making it particularly valuable for high-throughput or resource-limited experiments.</p><p>By applying this approach, the researchers were able to identify metabolites showing significant changes across conditions, including compounds uniquely produced or suppressed in specific cultures. One example includes the identification of 1-octen-3-ol, a known fungal metabolite, which serves as validation of the analytical workflow.</p><h4><strong>Discussion</strong></h4><p>A key outcome of this work is the demonstration that the choice of statistical method must be aligned with the experimental design. When sufficient replicates are available, variance-based methods provide robust results. In contrast, when data are limited, alternative metrics such as the coefficient of variation offer a practical solution. Additional approaches, such as fold-change analysis, can further enhance interpretation by focusing on relative differences between conditions.</p><p>The tile-based framework proves particularly effective for handling the complexity of GC×GC–MS data, enabling the extraction of meaningful biological information without compromising resolution.</p><h4><strong>Conclusion</strong></h4><p>This study highlights the power of advanced data processing strategies in modern analytical chemistry. Tile-based statistical analysis enables the systematic exploration of highly complex datasets and provides valuable insights into microbial metabolism and interaction. The work was conducted by researchers from the University of Washington in collaboration with Chevron Technical Center and focuses on creating a comprehensive <strong>Master Peak Table (MPT)</strong> containing all detectable analytes across multiple fuel sample classes. The motivation was that moving from conventional GC to GC×GC dramatically increased chromatographic peak capacity and resolution, but also created a need for more sophisticated data analysis methods capable of handling the much larger number of detected compounds.</p><p>The analytical platform employed <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/37"><strong>GC×GC-TOFMS</strong></a> with a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/1289"><strong>thermal-flow modulator</strong></a> and a TOF mass spectrometer detector. Fuel samples were analyzed using an <strong>Rxi-1 column (30 m × 0.25 mm × 0.25 μm)</strong> in the first dimension and an <strong>Rxi-17MS column (2.0 m × 0.18 mm × 0.20 μm)</strong> in the second dimension. The sample set included multiple petroleum products such as hydrobates, reformates, naphthas, FCC gasoline, and blank samples. Log-scale chromatograms revealed numerous late-eluting trace compounds and significant compositional differences between fuel batches that were not readily visible in conventional chromatographic representations.</p><p>To process these highly complex datasets, the researchers implemented a <strong>tile-based Fisher Ratio (F-ratio) chemometric approach</strong> using <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/348"><strong>ChromaTOF Tile software</strong></a>. The method divides GC×GC chromatograms into small tiles and identifies regions where analyte signals differ significantly between sample classes by comparing between-class and within-class variance. Instead of relying solely on F-ratio ranking, the workflow incorporates statistical testing against blank samples. Three blank chromatograms were used to establish tile-specific confidence intervals and p-test thresholds, allowing the system to determine whether detected signals represent genuine compounds rather than background noise. This strategy enabled the creation of a unified master peak table while minimizing problems caused by retention-time shifts and reducing the need for manual user intervention.</p><p>Application of the method generated <strong>719 statistically significant analyte hits</strong> across the fuel classes and demonstrated excellent reproducibility, with only <strong>6.3% of classifications showing inconsistent behavior among replicates</strong>. Detected compounds included hydrocarbons and aromatic species such as <strong>1-methylnaphthalene, 1,2,4-trimethylbenzene, β-vatirenene, 2-octene, and tetramethylheptadecane</strong>, whose occurrence patterns aligned well with expected fuel chemistry. The resulting master peak table contained both shared and sample-specific compounds and provided a much more comprehensive representation of petroleum composition than conventional workflows. The authors conclude that the approach should be broadly applicable to other complex sample types and could be extended using alternative chemometric metrics beyond Fisher Ratio analysis to address different experimental objectives.</p><h3><strong>2. Due to its high cost, ghee is adulterated with vegetable oil, to gain commercial benefits. β-sitosterol, a common phytosterol, found in a variety of natural sources, including vegetable fat or oil from plants (Figure 1). It is used as a marker of vegetable oil adulteration in ghee<sup>1</sup>.</p><p>The primary goal of this study is to assess the vegetable oil adulteration in ghee by analyzing β-Sitosterol as a marker using gas chromatography (GC). This will establish a foundation for the rapid detection of ghee adulteration. In this study, a simple, sensitive, and inexpensive method is developed for extraction of β-Sitosterol from ghee which is then subjected to gas chromatography with flame ionization detection for quantitation. The effects of several important parameters influencing the extraction efficiencies of β-Sitosterol including water-immiscible organic solvent (type and volume), volume of water, sonication time, surfactant (type and concentration), and organic modifier (type and volume) were investigated. This method is ideal for routine analysis in the food industry during the manufacturing, processing, and commercial testing of milk fat samples, or for academic research purposes.</p><h4><strong>&nbsp;Sample Preparation and Analysis Conditions</strong></h4><p>Standard and sample preparations were conducted following the guidelines outlined in the proposed FSSAI method No. 01.097:20222. The analysis was performed using a Shimadzu <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=Nexis+GC-2030"><strong>Nexis GC-2030</strong></a> equipped with a Flame Ionization Detector (FID), as shown in Figure 2. The specific configuration and analytical conditions used during the study are provided in Table 1.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>A gas chromatography–based method using GC-2030 was developed for the estimation of β-Sitosterol in ghee. The method exhibits a detection limit of 2 %, enabling the identification of vegetable oil adulteration in ghee at levels as low as 2 %. At this adulteration level, the signal of β-Sitosterol achieved a signal-to-noise (S/N) greater than 3, confirming the reliability of detection at the established limit. This method provides a reliable and effective approach for the quantification of β-Sitosterol, which serves as a marker for vegetable oil adulteration in ghee.</p><h3><strong>3. Waters Corporation: Multidimensional Characterization of Short Chain Chlorinated Paraffins (SCCPs) with GC-APCI and Cyclic Ion Mobility</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/720009386_3232568115.pdf"><strong>Full PDF for download</strong></a></li></ul><p>This poster presents a multidimensional analytical approach for the characterization of <strong>short-chain chlorinated paraffins (SCCPs)</strong>, a class of persistent environmental pollutants included in the Stockholm Convention due to their environmental persistence and bioaccumulation potential. SCCPs are highly complex mixtures of chlorinated hydrocarbons containing thousands of possible congeners, making their analysis particularly challenging. The study evaluates the use of <strong>gas chromatography coupled with atmospheric pressure chemical ionization, cyclic ion mobility spectrometry, and high-resolution mass spectrometry (GC-APCI-IMS-HRMS)</strong> as a powerful platform for improved SCCP characterization.</p><p>The analytical instrumentation consisted of a modified <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8890"><strong>Agilent 8890 gas chromatograph</strong></a> equipped with an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=7&amp;search=Rxi-5SilMS"><strong>Rxi-5SilMS capillary column</strong></a><strong> (30 m × 0.25 mm × 0.25 μm)</strong> and connected to a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/202"><strong>Waters SELECT SERIES Cyclic IMS</strong></a><strong> high-resolution mass spectrometer</strong> using an <strong>Atmospheric Pressure GC (APGC) ion source</strong>. Unlike conventional electron ionization methods, APCI was operated in negative-ion mode with a chloroform modifier, promoting the formation of characteristic <strong>[M+Cl]⁻ molecular ions</strong>. This significantly reduced fragmentation and provided strong molecular-ion signals, improving both qualitative identification and quantitative analysis of SCCP homologues.</p><p>Using three commercial SCCP technical mixtures (51.5%, 55%, and 63% chlorine content), the researchers demonstrated that GC-APCI-HRMS could selectively detect SCCP homologues according to carbon-chain length and degree of chlorination. Although substantial chromatographic overlap was observed among many homologues, the addition of <strong>ion mobility spectrometry (IMS)</strong> introduced a valuable extra separation dimension. Distinct drift-time versus m/z trendlines were observed for compounds sharing the same number of chlorine atoms, allowing homologues to be grouped and characterized more effectively than by chromatographic retention and mass spectrometry alone. The IMS data also enabled estimation of the relative abundance of individual homologue groups within each technical mixture.</p><p>The authors further explored the resolving power of the <strong>cyclic IMS device</strong> by employing multiple ion-mobility passes (up to eight cycles) to investigate individual SCCP congeners. While complete separation of individual congeners within a homologue group was not achieved, higher ion-mobility resolution revealed measurable differences between early- and late-eluting species and provided additional insight into compositional differences among SCCP mixtures. The study concludes that combining <strong>GC-APCI, cyclic ion mobility spectrometry, and high-resolution mass spectrometry</strong> offers a powerful multidimensional workflow for the characterization of complex chlorinated paraffin mixtures and may improve environmental monitoring and future regulatory analysis of these challenging contaminants.</p></div></div></div></div></div></div></section>

These microorganisms frequently coexist in the lungs of cystic fibrosis patients, where their interactions are associated with worsening respiratory function and poorer clinical outcomes. The study aims to identify VOC biomarkers that distinguish microbial monocultures, cocultures, and growth media controls, providing insights into pathogen interactions and potential diagnostic markers.</p><p>To achieve this, the researchers combined solid-phase microextraction (SPME) sampling with comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (<a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/37"><strong>GC×GC-TOFMS</strong></a>). This analytical platform offers high chromatographic resolution, rapid spectral acquisition rates of up to 500 spectra per second, and rich three-dimensional datasets containing chromatographic and mass spectral information. However, each chromatogram can contain more than one million data points, creating significant challenges for data processing and interpretation.</p><p>The core of the work focuses on tile-based feature selection methods implemented through the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/348"><strong>ChromaTOF Tile software</strong></a>. The primary approach, tile-based Fisher Ratio (F-ratio) analysis, identifies analytes that differ statistically between sample classes by comparing between-class and within-class variance. Using this strategy, the authors discovered 576 VOC features capable of differentiating growth media, <i>P. aeruginosa</i> monocultures, <i>A. fumigatus</i> monocultures, and cocultures. Additional pairwise statistical testing was then used to determine which specific classes were responsible for the observed differences. Representative VOCs characteristic of each class were identified, including heptanal for <i>P. aeruginosa</i>, 1-octen-3-ol for <i>A. fumigatus</i>, and several unique compounds associated with coculture conditions.</p><p>The presentation also addresses practical challenges associated with high-throughput experimentation, where replicate measurements may be limited by time, cost, or sample availability. To overcome these constraints, the team developed complementary tile-based coefficient of variation (CV) and fold-change (FC) approaches that can identify important analytes using as little as a single replicate per sample class. The authors conclude that selecting the appropriate feature-selection strategy—F-ratio, CV, or fold-change—depends on the experimental design and available data. Contaminants in recycled PET, including acetaldehyde, benzene, limonene, and bisphenol A (BPA), can migrate from the packaging into the contents, potentially affecting the safety and quality of the products stored inside.<sup>1</sup> Monitoring these contaminants is crucial for ensuring the quality of recycled PET products, as they can affect the physical, chemical, and sensory properties of recycled PET, potentially leading to product defects or health concerns.<sup>1</sup> FSSAI officially permits the use of recycled PET (rPET) in food packaging, subject to strict safety and quality standards. This regulatory change was formalized through amendments notified in March 2025, effective immediately.<sup>2</sup> All recycled plastics used in food contact applications must comply with stringent overall and specific migration limits to ensure no harmful chemical transfer into food. The recycling processes require validated decontamination steps to reduce contaminants to safe levels. Manufacturers must certify their packaging materials under FSSAI guidelines.&nbsp;</p><p>GC/MS plays a critical role in assessing the safety and suitability of recycled plastics for food‑contact applications. The method adopted in this work demonstrates the use of an Agilent 8890 GC system coupled with an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=5977C"><strong>Agilent 5977C GC/MS system</strong></a> for quantification of BPA in PET granules.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>This application note presents key strategies for the analysis of BPA in PET granules using GC/MS. The calibration used here ranged from 10 to 500 ng/mL, and the LOQ of the method was demonstrated at 500 ng/g for a PET sample. Repeatable results were found for six successive replicates of matrix-based standards at 100 ng/mL. Satisfactory recoveries were obtained for the tested matrix at 500 ng/g spiked concentration levels. Thus, this study demonstrates the applicability of this method for routine analysis of PET samples for identification and quantification of BPA.</p><h3><strong>2. The pathogenic behaviors of P. aeruginosa are controlled by a cell-to-cell communication system called quorum sensing (QS). Many genes activated by QS include those responsible for forming biofilms and producing virulence factors. Interestingly, several virulence factors produced by P. aeruginosa are regulated by an interaction between a receptor, RhlR and a hydrolase, PqsE. Thus, the PqsE-RhlR interaction has become a target for antibiotic development and further characterization. Comprehensive two-dimensional gas chromatography paired with time-of-flight mass spectrometry (GC×GC-TOFMS) was chosen for metabolomic analysis as it is known for its high sensitivity and improved peak capacity. With GC×GC-TOFMS, the volatile organic compound (VOC), acetophenone, was discovered and utilized to measure dose-dependent inhibition of PqsE’s enzymatic activity in vivo by the small molecule inhibitor, Vorinostat. The purpose of E &amp; L studies is to identify and evaluate possible toxicological risks. In this study, the regulatory references aim to identify traces of potential chemical substances. These substances may be harmful to patients due to their toxicity or may impact the activity of the drug product. Hence, to ensure the safety and efficacy of the drug throughout itsshelf-life, E &amp; L studies play an important role.&nbsp;</p><h5><strong>What Are Extractables and Leachables?&nbsp;</strong></h5><p>Extractables are organic and inorganic chemical entities that are released from a pharmaceutical packaging/delivery system, packaging component, or packaging material of construction into an extraction solvent under laboratory conditions[1].&nbsp;</p><p>Leachables are foreign organic and inorganic chemical entities that are present in a packaged drug product because they have leached into the packaged drug product from a packaging/delivery system, packaging component, or packaging material of construction under normal conditions of storage and use or during accelerated drug product stability studies[1].</p><h4><strong>Methods of Analysis</strong></h4><p>In this method, fifteen standards were analyzed by <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMSTQ8050+NX"><strong>GCMSTQ8050 NX</strong></a> with <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=HS-20+NX"><strong>HS-20 NX</strong></a> (Trap) autosampler. For the method suitability, several important parameters like system suitability, linearity and precision were performed.&nbsp;</p><h4><strong>Conclusion</strong>&nbsp;</h4><p>The chromatographic profile of solvent reflux experiment shows higher number of extractables than aqueous incubation. Furthermore, in aqueous incubation, the content of extractables is well below the AET level. Since, placebo of the ophthalmic drug product is completely aqueous, CCP used in the analysis is totally safe and can be used for the storage of Penicillin V.&nbsp;</p><p>Shimadzu’s GCMS-TQ provides high scan speed of 20000 amu/sec with ASSP technology enabling simultaneous SCAN/MRM analysis which is crucial in E&amp;L study.&nbsp;</p><p>Generally, if the drug product falls under parenteral and oral drug product (PODP), evaluation threshold will be at higher side due to having wider SCT. However, AET for Penicillin V was at too low to evaluate with liquid injection and static headspace technique. This has been proved with the trap headspace technology.&nbsp;</p><p>To analyze the complete extractable profile with the sample screening is only possible with dynamic headspace as most of the extractables were not detected in the injection acquired with static headspace.</p><h3><strong>4. VOCs from decomposing remains have been studied before,1 however there is a gap that remains in the knowledge on VOCs that evolve from submerged decomposing remains. 2 In this study, submerged animal tissue was tested to see how the VOCs can be utilized in forensic settings.&nbsp;</p><p>This study used data obtained last summer using comprehensive two-dimensional gas chromatography – time-of-flight mass spectrometry (GC×GC-TOFMS). Pigs (Sus scrofa domesticus) are often used as analogs in decomposition studies due to similarities with humans. 3 In this study, pork belly samples were submerged in mason jars filled with water and stored at different temperatures. The GC×GC-TOFMS instrument was used to collect odor from above water samples for a period of twelve days.</p><p>GC×GC is often used to analyze complex odor profiles. When analyzing samples with GC×GC, there is a lack of standardization to process the data acquired from time trials. There is a need to discover effective ways to track VOCs for chemical identification and longitudinal analysis.</p><p>The goal of this research was to compare different software approaches to analyze GC×GC longitudinal data to achieve accurate analyte identification, representation of data over time, and effective class characterization.</p><h4><strong>Methods</strong></h4><p>For the first four days, 5 mL of water was taken and run on the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/37"><strong>GC×GC-TOFMS</strong></a>. Thereafter, the samples were sampled and run every three days until day twelve. Data were analyzed initially using <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/41"><strong>ChromaTOF</strong></a> (LECO Corporation). The samples were then analyzed using <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/348"><strong>ChromaTOF Tile </strong></a>and <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/1873"><strong>ChromaTOF Sync 2D</strong></a> to compare how they can be used in a forensic setting.</p><p>Peak tables were curated in Tile and Sync 2D. Then, ChromaTOF was used as a guide to confirm which peaks were identified correctly. If the compound was correctly identified, the peak would be accepted or kept on the table. If not correctly identified, the peak would either be rejected or deleted. Once the tables were complete from the different software approaches, they were exported to Excel for further comparison.</p><p>In excel, the relevant hits that were identified as decomposition VOCs were compared across the software approaches. The compounds that were most commonly found were compared to existing literature on decomposition VOCs. From there, two tables were built; where one table was compounds confirmed with literature and the other table was compounds not confirmed with literature.</p><p>Comparison between software approaches included looking at different criteria to see which method was the most effective and efficient. Due to this diversity of feedstocks, continuous quality control is necessary as the quality of the raw material has an enormousimpact on the quality of the biodiesel.&nbsp;</p><p>The EN 14103:2020 describes the determination of total ester content and linolenic acid methyl ester content in biodiesel by means of gas chromatography with flame ionization detection (GC-FID). The total ester content covers a range of fatty acid methyl esters (FAME) from C6 to C24, and linolenic acid methyl ester is investigated separately as interfering reactive compound that might influence long-term stability of the biodiesel. According to the standard, the total ester content must be greater than 90 % (m/m) and the linolenic acid methyl ester content shall be between 1 and 15 % (m/m)1.</p><p>This study shows the analysis of biodiesel originating from rapeseed,soybean, and palm oil using the Nexis GC-2060.</p><h4><strong>Multi-Mode Injection Unit as Alternative&nbsp;</strong></h4><p>Shimadzu’s multi-mode injection unit (MMI) offers reliable splitsplitless mode functionality over a broad range of carbon numbers. It allows analysis of biodiesel samples with high repeatability and provides chromatographic results comparable to those measured with SPL as dedicated split-splitless injection unit. The use of MMI significantly shortens maintenance downtime from tens of minutes for a classical split-splitless injection unit to just a few minutes. The study addresses a key limitation of conventional LC-MS/MS methods, which are highly effective for many PFAS but less suitable for more volatile species such as fluorotelomer alcohols (FTOHs), fluorotelomer acrylates (FTAcr), and fluorotelomer acetates (FTOAc). By incorporating GC-MS/MS analysis, laboratories can obtain a more complete characterization of PFAS contamination across environmental, biological, and food-contact materials.</p><p>The analytical workflow utilized an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8890"><strong>Agilent 8890 gas chromatograph</strong></a> equipped with an autosampler and coupled to a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/554"><strong>Waters Xevo TQ Absolute tandem mass spectrometer</strong></a> operated with APGC ionization in positive-ion mode. Separation was performed on an Rtx-200 capillary column, while data acquisition and quantification were managed using <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1544"><strong>waters_connect software</strong></a>. Unlike traditional electron ionization (EI), APGC generated intact protonated molecular ions, significantly improving sensitivity and selectivity for GC-amenable PFAS. A method covering 35 PFAS compounds was developed, achieving lower limits of quantification (LLOQs) of 0.025–0.1 ng/mL for most analytes.</p><p>Several sample types were evaluated, including wastewater, surface water, soil, fish tissue, shrimp, and food-packaging materials. Environmental and biological samples were prepared according to EPA Method 1633A and had previously been analyzed by LC-MS/MS, allowing the GC-MS/MS method to serve as a complementary technique. Food-packaging samples were extracted with methanol containing ammonium hydroxide and analyzed for targeted PFAS. While no target PFAS were detected in the tested packaging materials, multiple GC-amenable PFAS were found in environmental samples. Fluorotelomer alcohols were the most frequently detected compounds, with wastewater and ski-wax-impacted soils showing the highest concentrations.</p><p>The results demonstrate that APGC-MS/MS provides a sensitive and robust solution for the analysis of volatile PFAS classes that may be missed by LC-MS/MS alone. The method delivered accurate quantification, effective chromatographic separation, and reliable recovery in spiked samples. The authors conclude that combining APGC-MS/MS with established LC-MS/MS workflows enables a more comprehensive assessment of PFAS contamination, particularly for emerging fluorotelomer compounds and other GC-amenable PFAS that are increasingly relevant in environmental monitoring and food-packaging investigations.</p></div></div></div></div></div></div></section></div></div></div></div></div></div></section>

MDCW / University of Washington: Implementing Tile-Based Fisher Ratio Analysis of GC×GC-TOFMS Data to Obtain a Master Peak Table of All Detected Analyte Compounds in Many Petroleum-Based Samples</strong></h3><ul><li>Presentation</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/O_9_Halvorsen_136808e314.pdf"><strong>Full PDF for download</strong></a></li></ul><section><div><div><div><div><div><div><p>The presentation describes the development of a chemometric workflow for advanced fuel sample analysis using GC×GC-TOFMS data. The work was carried out by the University of Washington in collaboration with Chevron Technical Center, with the goal of improving the characterization of complex petroleum-based samples. Moving from conventional GC to comprehensive two-dimensional GC increased peak capacity and chromatographic resolution, but also created the need for more advanced methods to generate reliable peak tables across many chemically diverse fuel samples.</p><p>The key method presented is a tile-based Fisher Ratio approach implemented through <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/348"><strong>ChromaTOF Tile</strong></a><strong>.</strong> This method identifies analytes that show statistically significant concentration differences between sample classes. Instead of relying only on peak detection or manual alignment, the workflow uses chromatographic “tiles” to reduce the impact of retention-time shifting and to capture both major and trace-level components. The study included multiple fuel classes, including hydrobate, reformate, naphtha, FCC gasoline, and blanks.</p><p>Because Fisher Ratio values alone do not directly indicate whether a compound is truly detectable, the authors added a blank-based p-testing strategy. Three blank chromatograms were used to estimate background signal distributions for each tile, and confidence intervals were then applied to determine whether each detected hit was present in individual sample classes. This produced a master peak table covering 719 hits across nine fuel sample classes. The results showed good reproducibility, with only 6.3% of classifications being heterogeneous across replicates.</p><p>Overall, the presentation concludes that the tile-based workflow can generate comprehensive and reproducible peak tables with reduced user intervention, while preserving the ability to recover trace analytes in complex petroleum samples. The approach should be broadly applicable to other complex sample types and could also be adapted using chemometric metrics other than Fisher Ratio, depending on the experimental design and analytical goals.</p><h3><strong>3. Shimadzu / ASMS: Is Your Next Sip Safe? A Simple, Rapid Method for Measuring volatile PFAS in Juices.</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/TP_143_Is_Your_Next_Sip_Safe_A_Simple_Rapid_Method_for_Measuring_volatile_PFAS_in_Juices_a6ed9414c0.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Per- and polyfluoroalkyl substances (PFAS) pollution is a growing global concern due to links with adverse health effects, many of which remain poorly understood. 1 Accurate detection and quantification are therefore essential. While PFAS analysis has traditionally focused on water, attention is rapidly expanding to food and beverages, where testing remains limited despite high stakes and increasing regulations. 2 A recent PFAS lawsuit against a juice manufacturer underscored the heightened public scrutiny of PFAS in juices, placing food safety and brand reputation at risk. 3 This study presents a Headspace Solid-Phase Microextraction– Triple Quadrupole Gas Chromatography/Mass Spectrometry (HS-SPME GC/MS/MS) method for PFAS quantitation in juices. GC/MS enables analysis of volatile PFAS unsuitable for LC/MS, while HS-SPME offers rapid analysis with minimal sample preparation in complex matrices.</p><h4><strong>Methods</strong></h4><p>A volatile PFAS analysis method was developed on a <strong>Shimadzu </strong><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMS-TQ8040+NX"><strong>GCMS-TQ8040 NX</strong></a> with an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=AOC-6000+Plus"><strong>AOC-6000 Plus multifunctional autosampler</strong></a> equipped with a solid phase microextraction (SPME) module (Figure 1).</p><h4><strong>Results&nbsp;</strong></h4><p>The system was deemed free of contaminants and inferences. None of the target PFAS in the method blank were found in quantifiable concentration. In the study, the calibration curve for each target included at least seven calibrators. Calibration curve results showed a good linear fit for all compounds with coefficient of determination (R2 ) ≥ 0.993. The linear range and R2 of each PFAS target are shown in Table 3.</p><h4><strong>Conclusion</strong></h4><p>Overall, these results demonstrate the robustness and reliability of the method for PFAS analysis in two carton apple juices (Brands X and Y), one plastic bottled apple juice (Brand X), and a berry blend carton juice (Brand X). Given the diversity of matrices evaluated, the method may also be suitable for other beverages, including sports drinks and vitamin-enhanced waters</p><h3><strong>4. Orbitrap technology provides high resolution and mass accuracy, enabling confident identification of coeluting and isobaric compounds while reducing false results. Furthermore, the FS-HRAM acquisition enables simultaneous quantitation and screening within a single analysis with the capability of retrospective data analysis, allowing laboratories to reprocess datasets for additional or emerging compounds without re-injecting samples. The <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/2419"><strong>Orbitrap Exploris GC S mass spectrometer</strong></a><strong> </strong>features the Thermo Scientific NeverVent Advanced Electron Ionization Source (NVAEI) which offers an improved design providing enhanced sensitivity and robustness, allowing for increased instrument uptime and maximized sample throughput for rapid return on investment (ROI).</p><h4><strong>Materials and methods</strong></h4><h5><strong>Data analysis&nbsp;</strong></h5><p>Data was acquired, processed and reported using the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/76"><strong>Thermo Scientific Chromeleon Chromatography Data System (CDS) software</strong></a>, which provides integrated instrument control, automated workflows, and an intuitive interface for data analysis, customizable reporting, and compliant data management in accordance with FDA 21 CFR Part 11 and European Commission (EU) Annex 11. Its advanced reprocessing capability streamlines sequence setup, data review, and reporting offering the possibility to easily flag and check compliance with the current regulations.</p><p>Results Compliant analyte identification The use of HRAM-GCMS reduces the matrix interferences ensuring a confident and selective identification of analytes (Figure 1). All target compounds satisfied the SANTE criteria at the default MRL of 10 µg/kg and more than 90% were confirmed at concentrations of 1 µg/kg following the SANTE/11312/2021v2026 guidance criteria of:&nbsp;</p><ul><li>two ions were detected for each pesticide with mass accuracy ≤ 5 ppm with precursor and product ions in the extracted ion chromatograms fully overlapped,&nbsp;</li><li>due to the added value of accurate mass measurement, matching ion ratios is not necessary, however, an ion ratio (IR) deviation within ± 30% (relative) of the average of calibration standards from the same sequence was considered to provide additional support for identification</li><li>retention times (RT) within a tolerance of ± 0.1 minutes</li></ul><p>Sensitivity performance (IDL) and analytical linearity Instrument sensitivity was evaluated using calculated IDLs derived from replicate injections of low-level, matrix-matched standards. The Orbitrap Exploris GC S demonstrated detection limits at levels at or below 0.5 µg/kg for most of the investigated pesticides (Figure 2). Linear dynamic range was evaluated using matrix-matched calibration curves ranging 0.5–200 µg/kg, with each calibration level injected in triplicate. All target analytes demonstrated excellent linearity, with coefficients of determination (R²) &gt; 0.993, response factor (%RSD) values below 20% across the calibration range, and ion ratios (IRs) within ±30% of the average calibration standard values. System suitability for quantitative analysis was confirmed by calculated concentrations within ±20% of expected values and recoveries ranging from 96% to 104%. An example of calibration curve for Aldrin is shown in Figure 3.</p><h4><strong>Conclusions&nbsp;</strong></h4><p>The results obtained in these experiments demonstrate that the Orbitrap Exploris GC S mass spectrometer equipped with NeverVent AEI ion source represents a reliable tool for analysis of pesticides in complex matrices.&nbsp;</p><ul><li>The Orbitrap Exploris GC S offers a simple approach for the multiresidue analysis of pesticides allowing for full-scan acquisition combined with high mass resolving power&nbsp;</li><li>The NeverVent AEI ion source is designed for ultra-trace concentration detection, making it ideal for challenging applications ensuring detection limits at levels at or below 0.5 µg/kg for most of the investigated pesticides&nbsp;</li><li>Confident identification of compounds can be easily achieved through i). high spectral quality, ii). high spectral fidelity, ensuring consistent spectrum independently from concentration, and iii). consistent mass accuracy irrespectively of analyte concentration and time&nbsp;</li><li>System suitability for quantitative analysis was demonstrated with R2 &gt;0.993, RF %RSD across the entire calibration curve &lt; 20, and IRs within ± 30% of the average of calibration standards, calculated amounts within 20% the expected values, and percentage recovery (%) ranging from 96 to 104.</li></ul></div></div></div></div></div></div></section></div></div></div></div></div></div></section>
