Agilent Technologies: Determination of 41 Per- and Polyfluoroalkyl Substances (PFAS) in Cosmetics</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_captiva_emr_pfas_food_ii_cartridge_5994_9111en_agilent_be91861646.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Per- and polyfluoroalkyl substances (PFAS) are synthetic compounds composed of highly stable carbon–fluorine (C–F) bonds, giving them exceptional thermal and chemical stability, leading to poor environmental degradability. PFAS have been used in cosmetics to provide water resistance, oil repellency, or application smoothness. The unintentionally presented PFAS residues in raw materials may remain in finished cosmetic products, and can be absorbed through the skin. Lip products and spray type products can also lead to inhalation or oral exposure to residues.<sup>1,2</sup>&nbsp;</p><p>Agilent Captiva EMR PFAS Food cartridges are designed to provide streamlined and comprehensive matrix removal after QuEChERS extraction for complex sample matrices, demonstrating effective matrix cleanup efficiency and excellent performance for quantitative PFAS analysis.<sup>3,4</sup> In this study, we performed QuEChERS extraction followed by cleanup with Captiva EMR PFAS Food II cartridges for PFAS analysis in five types of cosmetics.</p><h4><strong>Experimental</strong></h4><h5><strong>Equipment and materials&nbsp;</strong></h5><p>The study was performed using an <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=10&amp;search=1290+Infinity+III"><strong>Agilent 1290 Infinity III LC</strong></a> system consisting of an Agilent 1290 Infinity III high-speed pump (G7120A), an Agilent 1290 Infinity III multisampler (G7167B), and an Agilent 1290 Infinity III multicolumn thermostat (G7116B). The LC system was coupled to an <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=10&amp;search=6495D"><strong>Agilent 6495D triple quadrupole LC/MS system</strong></a> (G6495D) equipped with an Agilent Jet Stream Electrospray ion source. Agilent MassHunter Workstation software was used for data acquisition and analysis.&nbsp;</p><p>The 1290 Infinity III LC system was modified using an Agilent InfinityLab PFAS Analysis HPLC conversion kit (part number 5004-0006), including an Agilent InfinityLab PFC delay column, 4.6 × 30 mm (part number 5062-8100). Chromatographic separation was performed using an Agilent <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=10&amp;search=ZORBAX+RRHD+Eclipse+Plus"><strong>ZORBAX RRHD Eclipse Plus</strong></a> C18, 95 Å, 2.1 × 100 mm, 1.8 µm (part number 959758-902).</p><h4><strong>Results and discussion&nbsp;</strong></h4><h5><strong>Chromatography and calibration curve linearity&nbsp;</strong></h5><p>Figure 2 shows the overlaid chromatograms for 41 PFAS calibration standards, including 0.05, 0.1, 0.2, 0.5, 1.0, 2.0, 5.0, and 10.0 ng/mL in ACN. The table on the right shows the peaks identification and corresponding retention time. Across 41 PFAS compounds, R² values ranged from 0.9940 to 0.9998, indicating excellent linearity for all analytes' calibration curves across the calibration range of 0.05 to 10.0 ng/mL.</p><h5><strong>PFAS detection in real cosmetic samples&nbsp;</strong></h5><p>Of the 15 cosmetic products assessed, one eyeshadow sample was detected with a trace level of positive PFAS analytes, including PFHeA, PFHpA, PFNA, and PFOA, at about 0.1 ng/g in the sample. Figure 5 shows the method blanks and positive detection of PFHpA and PFNA in this eyeshadow&nbsp;product.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>A simplified, rapid, and reliable method using QuEChERS extraction followed by EMR mixed-mode passthrough cleanup using the Agilent Captiva EMR PFAS Food II cartridge and LC/MS/MS detection was developed for 41 PFAS analysis in cosmetics.</p><h3><strong>2. Metrohm: brightRC – Advanced brightener analysis with a response curve</strong></h3><p><strong>Next-level brightener determination in nonlinear systems</strong></p><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_v_242_download_83376b2923.pdf"><strong>Full PDF for download</strong></a></li></ul><h4><strong>WHAT IS BRIGHTRC?&nbsp;</strong></h4><p>brightRC is a response curve calibration approach for the determination of brightener-type additives by CVS or CPVS (Cyclic Pulse Voltammetric Stripping), hence the name «brightRC». This technique is suitable to measure brightener content in copper baths for plating. The method is based on signal normalization using the electrolyte value, which is the response for the virgin makeup solution (VMS) saturated with suppressor, and brightener defined as Q0 in the viva software. All recorded signals are divided by Q0 and the normalized values are plotted against concentration to generate an external calibration curve. This calibration can then be used for routine sample determination without repeated standard additions.&nbsp;</p><p>The sample is simply measured using the same method as the calibration, signals are normalized and evaluated using the mathematically fitted calibration curve. By decoupling calibration from sample measurement, brightRC removes the need for repeated standard additions with each sample measurement, enabling faster and more efficient routine analysis.&nbsp;</p><p>The brightRC calibration approach supports flexible regression models including quadratic and nonlinear regression. This allows reliable quantification of additive systems that do not exhibit a wide linear working range or exhibit nonlinear response characteristics, thereby expanding the range of applicable brightener chemistries.&nbsp;</p><p>One important detail that further differentiates brightRC from the MLAT approach relates to normalization of the signal for calibration and sample analysis. This normalization approach improves measurement robustness as variations in absolute signal intensity naturally occurring over the working day are mathematically compensated.</p><h4><strong>HOW DOES BRIGHTRC WORK IN PRACTICE?&nbsp;</strong></h4><p>The brightRC workflow separates calibration and sample analysis. It can be implemented in different setups (from manual to fully automated) depending on the additive system and concentration range. After recording the electrolyte value, calibration standards are measured to establish the calibration curve, otherwise known as the response curve. Once the calibration is stored, samples are determined with the same CVS or CPVS method, without further standard additions. The sample concentration is automatically calculated from the normalized signal using the calibration curve.&nbsp;</p><p>For sample measurement, viva 4.0 provides two practical approaches:&nbsp;</p><ol><li>«brightRC with solution exchange»&nbsp;</li><li>«brightRC with sample dilution» Both variants follow the same calibration principle and differ only in the sample handling strategy.</li></ol><h4><strong>WHEN TO USE BRIGHTRC?&nbsp;</strong></h4><p>brightRC is ideal for routine brightener control in stable plating baths where high sample throughput is required. It is particularly suitable when matrix conditions are consistent and electrode fouling is limited. The approach is especially advantageous for additive systems exhibiting nonlinear signal behavior. If strong matrix variations, significant breakdown products, or severe electrode fouling are expected and a linear correlation between signal and analyte concentration is guaranteed, standard addition techniques such as MLAT are preferred because they may provide additional robustness.</p><h3><strong>3. Shimadzu / ASMS: PFAS Analysis of Leachate: Evaluating Key Sample Preparation and LC–MS/MS Parameters for Improved Performance</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/WP_255_PFAS_Analysis_of_Leachate_Evaluating_Key_Sample_Preparation_and_LC_MSMS_Parameters_for_Improved_Performance_1114ee264c.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Landfill leachate is a highly complex and variable matrix that poses significant challenges for the analysis of per- and polyfluoroalkyl substances (PFAS). Although U.S. Environmental Protection Agency Method 1633A provides a standardized framework for PFAS quantitation,1 analytical performance remains strongly influenced by sample preparation, chromatographic separation, and mass spectrometric conditions. Landfill leachate samples were extracted using WAX SPE following EPA 1633A and analyzed by LC–MS/MS using a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=1&amp;search=LCMS-8060"><strong>Shimadzu LCMS-8060</strong></a><strong> </strong>under negative ESI conditions. This study evaluates the key factors governing LC–MS/MS sensitivity, signal response, and reproducibility in landfill leachate, including a systematic assessment of SPE, chromatographic behavior, and ionsource conditions across PFAS classes (Figure 1).</p><h4><strong>Results&nbsp;</strong></h4><p>Chromatographic performance was evaluated across four C18 columns (A–D) using peak area, height, asymmetry, and HETP. Two columns produced a higher signal and improved peak shape, while the remaining columns showed greater efficiency. Under repeated batches followed by storage in acetonitrile, the Shimadzu GIST column produced a consistent signal (≤15% change), whereas another column showed 20–50% signal loss, particularly for long-chain PFAS and precursors, indicating greater susceptibility to matrix accumulation (Figure 2).</p><h4><strong>Conclusion&nbsp;</strong></h4><ul><li>PFAS analytical performance was dependent on compoundclass-dependent extraction, chromatography, and ionization behavior.&nbsp;</li><li>GIST demonstrated lower signal variability under repeated matrix exposure, with lower signal degradation than alternative C18 phases.</li><li>Neutral PFAS showed poor retention on WAX sorbents, requiring optimization beyond standard EPA 1633A extraction conditions.&nbsp;</li><li>PFCA response was more influenced by gas flow conditions, whereas PFSA response was more sensitive to source temperature conditions.</li></ul><h3><strong>4. Thermo Fisher Scientific / ASMS: Ultra-high throughput proteomics at &gt;500 samples/day using advanced </strong><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/398"><strong>Vanquish Neo UHPLC</strong></a><strong> methods with </strong><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1959"><strong>Orbitrap Astral Zoom MS</strong></a></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/po_004708_lsms_ultra_high_throughput_vanquish_astral_proteomics_po004708_asms2026_na_en_f6499c3eec.pdf"><strong>Full PDF for download</strong></a></li></ul><p>High-throughput mass spectrometry is increasingly critical for large-cohort proteomics, population-scale studies, and screening applications. While established LC–MS platforms routinely support several hundred samples per day, further increases in throughput are often limited by chromatographic stability, robustness, and reproducibility under ultra-short gradient conditions, as well as the acquisition speed of the mass spectrometer. Here, we describe an advanced ultra-high-throughput LC–MS methodology developed on the Thermo Scientific Vanquish Neo UHPLC system and Thermo Scientific Orbitrap Astral Zoom mass spectrometer. The goal of this work was to extend the practical throughput limits of LC–MS while achieving deep proteome coverage and maintaining run-to-run reproducibility.</p><h4><strong>Materials and methods</strong></h4><h5><strong>LC-MS analysis&nbsp;</strong></h5><p>Samples were separated on a Thermo Scientific 150 µm × 70 mm C18 column (prototype, not commercially available yet) operated at 4 µL/min using a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/398"><strong>Vanquish Neo UHPLC system</strong></a> in NanoCap Trap&amp;Elute configuration at room temperature. Prototype methods allowing for reduced LC duty cycles (approx. 1 min of sample pickup/loading and integrated trap-column offline wash) were applied, the standard gradients were adjusted to 1 – 3 min. The Thermo Scientific PepMap Neo 5 µm C18 300 µm × 5 mm cartridge was used as trap column and the separation column connected with a 15 µm ID capillary Thermo Scientific EASY-Spray emitter (ES994) to an EASY-Spray source. The <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1959"><strong>Orbitrap Astral Zoom MS</strong></a> (ICSW AST 2.2) was operated in a dataindependent acquisition (DIA) mode, MS method parameters are given in Table 1. Comparative data for the chemoproteomics samples at lower throughputs (60-300 SPD) were acquired using an EASY-Spray 150 µm × 150 mm C18 column (ES906), as described before.&nbsp;</p><h5><strong>Data analysis&nbsp;</strong></h5><p>The raw data files were processed with Spectronaut® 20.5 software (Biognosys AG), <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1782"><strong>Thermo Scientific Proteome Discoverer </strong></a>3.3 SP1 with <strong>CHIMERYS 5.0 </strong>or DIA-NN Enterprise 2.3.2 (Aptila Biotech GmbH) nodes using default settings as specified in the individual figures. The data was searched against the Homo sapiens SwissProt database (TaxID=9606: 42,252 sequences) and contaminants FASTA file. Peptide numbers correspond to unique stripped peptides</p><h4><strong>Conclusions&nbsp;</strong></h4><ul><li>The advanced Vanquish Neo UHPLC methodology in combination with the Orbitrap Astral Zoom MS delivered highly reproducible, deep and precise proteome profiling across ultra-high-throughput regimes (600 SPD) exceeding current field benchmarks.&nbsp;</li><li>The workflow enables rapid chemoproteomic screening, as demonstrated with activity-based protein profiling (ABPP) samples, reliably identifying enriched putative target proteins of an antibiotic small molecule probe.</li></ul><h3><strong>5. Waters Corporation / ASMS: Assessing Relative Response of Four European-Regulated PFAS in Human Serum Using Cyclic Ion Mobility MS</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/720009410_c98e7c87ec.pdf"><strong>Full PDF for download</strong></a></li></ul><div><div><section><div><div><div><div><div><div><p>The poster presents an LC-Cyclic-IMS-MS approach for assessing four European-regulated PFAS—PFOS, PFOA, PFNA, and PFHxS—in human serum. The study focuses on environmental exposure monitoring in Ghanaian firefighters and e-waste handlers, using ion mobility mass spectrometry to add separation power and improve identification confidence in complex biological matrices. The work is motivated by increasing regulatory concern around PFAS toxicity, bioaccumulation, and the need for reliable exposure assessment in human biofluids.</p><p>The experimental workflow combined reversed-phase LC with a quadrupole-cyclic ion mobility-time-of-flight MS system (<a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/202"><strong>SELECT SERIES</strong> <strong>Cyclic IMS</strong></a>). Human serum samples were prepared using SPE µElution plates with a polymeric reversed-phase/weak anion exchange sorbent. The LC system was modified with PFAS-free components and an isolator column to reduce background contamination, which is especially important for trace-level PFAS analysis. Identification was supported by retention time, accurate mass, isotopically labelled internal standards, and collision cross section values.</p><p>The results showed detectable levels of the four regulated PFAS in both occupational groups, with a combined detection rate of 83% across human serum samples and NIST reference standards. Firefighter samples generally showed higher concentration ranges than e-waste handler samples, especially for PFOS, PFOA, PFNA, and PFHxS. For example, PFNA reached up to 5.713 ng/mL in firefighter serum, compared with up to 1.513 ng/mL in e-waste handler serum.</p><p>Overall, the poster concludes that LC-Cyclic-IMS-MS provides a highly specific strategy for PFAS analysis in complex biological samples. Ion mobility and CCS values improve confidence in trace-level identification and help distinguish target PFAS from co-eluting matrix background. The authors suggest that this approach can support both targeted PFAS monitoring and broader non-targeted screening of known and emerging PFAS, while also helping future studies connect exposure data with environmental fate and potential toxicological effects.</p></div></div></div></div></div></div></section></div></div>

Increasing evidence of environmental and human-health risks has led to heightened global regulatory pressure, including REACH/POPs restrictions, and strengthened requirements from industry frameworks such as ZDHC, OEKO-TEX®, AFIRM, and Bluesign. These initiatives demand highly sensitive, reliable, and standardized analytical workflows capable of quantifying both volatile and non-volatile PFAS in diverse textile matrices. This study demonstrates end-to-end PFAS solutions aligned with EN 17681-1:2025, comprising heat- and ultrasound-assisted (HUA) extraction followed by GC/TQ and LC/TQ determination of volatile and non-volatile PFAS, respectively. The workflow enables robust quantification of a comprehensive PFAS panel in T-shirts, waterproof shorts, and socks.</p><h4><strong>Experimental</strong></h4><h5><strong>Instrumentation&nbsp;</strong></h5><p>A dual-platform analytical approach combining GC/TQ and LC/TQ systems was used to quantify over 100 PFAS across 14 chemical classes (Figure 1). More than 30 volatile PFAS were analyzed using an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8890"><strong>Agilent 8890 GC</strong></a> coupled to a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=7010D"><strong>7010D TQ</strong></a> and a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=DB-624"><strong>DB-624 column</strong></a>. MDCW / William &amp; Mary: The Effect Of Using Low-Bleed Columns In The First Dimension For Comprehensive Two-Dimensional Gas Chromatographic Analyses</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/Webb_MDCW_Poster_2026_1_5657991949.pdf"><strong>Full PDF for download</strong></a></li></ul><section><div><div><div><div><div><div><p>In gas chromatography, the presence of column bleed impacts peak data quality. As siloxanebased stationary phase components fragment, they release bleed artifacts into the mass spectrometer (MS), producing interference and decreasing signal-to-noise ratios (S/N) of analytes. <sup>1</sup> When using comprehensive twodimensional gas chromatography (GC×GC), a secondary column with an orthogonal stationary phase is used in providing improved separation between column bleed and analytes. However, it also introduces more stationary phase that can be degraded to produce more bleed artifacts.</p><p>The goal of this research was to define how replacing the primary column in a GC×GC-MS system impacts the resulting S/N and analyte identification quality, in order to clarify how column bleed impacts analytes that appear to be chromatographically resolved from column artifacts in GC×GC output. This will demonstrate the advantage that low-bleed columns can provide for future analyses.</p><h5><strong>Methods</strong></h5><p>Samples analyzed were the Century Mix2 , containing approximately 100 different analytes within different classes (i.e. Century Mix2) and a 52- component Indoor Air Standard. Both were assessed using a 5% phenyl/95% dimethylsiloxane standard column in the first dimension and a (50%-phenyl)-methylpolysiloxane standard column in the second dimension. Samples were then reanalyzed with a low-bleed column replacement in the first dimension.</p><p>Analysis of the samples was performed using the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/37"><strong>Pegasus BT4D GC×GCTOFMS</strong></a> with a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/44"><strong>quad-jet dual-stage cryogenic modulator</strong></a><strong> </strong>(LECO Corporation). <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/41"><strong>ChromaTOF software</strong></a><strong> </strong>(LECO Corporation) was used to analyze the chromatographic data.</p><h4><strong>Conclusions</strong></h4><p>This work demonstrated that samples run with the low-bleed first dimension column showed a reduction in the presence of column bleed components identified and maintained or improved the quantitative signal-to-noise ratios of components compared to the samples run on standard first dimension columns. This showed that that low concentration analytes were more reliably detected in the absence of column bleed, even when contour plots showed physically resolved components. In the future, this can be applied to improve the resolution of complex samples and increase analyte signal.</p><h3><strong>3. Shimadzu: Dimethyl carbonate as a green organic modifier in supercritical fluid extraction of pesticide residue analysis in apple samples</strong></h3><ul><li>Technical note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/c190_e347_869b9e2ade.pdf"><strong>Full PDF for download</strong></a></li></ul><p>According to the Green Analytical Chemistry (GAC) principles[1], a highly promising strategy to minimize the side effects of analytical methods involves replacing potentially toxic solvents and/or obtained by non-renewable sources with more environmentally friendly alternatives. In this frame, dimethyl carbonate (DMC) is an environmentally friendly, non-corrosive, safe handling, and biodegradable chemical, extensively used in several application fields, such as pharmaceuticals, coatings, and lithium-ion battery electrolytes [2]. Additionally, instead of the historical use of toxic and corrosive phosgene as a carbonate source, simple and direct synthesis routes from carbon dioxide (CO2) have been proposed [3]. Thus, also falling within the “carbon capture and utilization” processes, ensuring the reduction of atmospheric CO2 concentration and its conversion into useful chemical products.&nbsp;</p><p>Firstly introduced in the second edition of the GlaxoSmithKline’s solvent selection guide, DMC has been included among the “few issues” solvents, along with water [4]. Thereafter, it has attracted widespread attention as a valuable chemical and/or solvent, and substantial research efforts have been made in the scientific community. Its potential as a green alternative to conventional solvents in both reversed-phase liquid chromatography [5–7] and supercritical fluid chromatography [8] has been explored. The extraction capabilities of DMC toward different target molecules have been also investigated. In 2022, Alfieri et al. optimized an ultrasoundassisted extraction of two lignans from Linum species [9]. The extraction performances of a panel of green solvents and two natural deep eutectic solvents (in combination with water in a solvent/water ratio of 80:20) were tested versus methanol and chloroform (used for comparison with published studies). The results showed that, in terms of purity and recovery, DMC was the optimal media for the 6-methoxypodophyllotoxin extraction. DMC has been also employed in an in-syringe extraction coupled to solid phase extraction to assess nicotine in environmental water and sportspersons’ urine samples [10]. Extracts enriched in oleanolic acid were obtained by solid-liquid extraction of grape pomace using DMC compared to other tested solvents (ethyl acetate, acetone, n-butanol) [11]. In addition to the highest obtained recovery value, the lowest co-extraction of other matrix components was also achieved.&nbsp;</p><p>Supercritical fluid extraction (SFE) represents a reliable alternative to conventional solvent-based extraction methods. Several advantages are provided when CO2 is employed as supercritical fluid due to its non-toxicity, non-flammability, abundancy, renewability (as a by-product of industrial processes, as fermentation or combustion), recyclability, and cost-effectiveness. Supercritical CO2 has a polarity similar to that of n-hexane; thus, the addition of a co-solvent is necessary for the extraction of polar molecules [12, 13].&nbsp;</p><p>This work aims to expand the use of DMC that, to the best of the author’s knowledge, is here firstly proposed as an organic modifier in SFE. The extraction efficiency and selectivity of DMC toward conventional solvents, such as acetonitrile (ACN) and bio-ethanol (EtOH) obtained from edible renewable feedstocks, has been evaluated for multi-residue analysis of pesticides (64 pesticides, representing diverse chemical classes, polarities, and molecular weights) in apple samples. Considering the sustainable and nontoxic nature of CO2 and DMC, the use of DMC in SFE represents a win-win combination. Thus, improving the overall sustainability and automation of the extraction process closely align with the GAC principles. Furthermore, in this research, always following the GAC principles, a low-pressure gas chromatography-tandem mass spectrometry (LP-GC-QqQMS) method was employed, allowing a greater sample loadability and a higher optimal linear velocity allowed by the vacuum conditions of the system, effectively reducing the analysis time at about 10 minutes [14, 15].</p><h4><strong>2. Materials and methods</strong></h4><h5><strong>2-3. Instrumentation</strong></h5><h5><strong>Low‑pressure gas chromatography triple quadrupole mass spectrometry&nbsp;</strong></h5><p>The LP-GC-QqQMS analyses were performed using a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GC-2010"><strong>GC-2010 Plus system</strong></a> (Shimadzu Corporation, Kyoto, Japan) coupled with a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=8040"><strong>triple quadrupole mass spectrometer (TQ8040</strong></a>, Shimadzu). Injection was performed using an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=AOC-20i"><strong>AOC-20i autosampler</strong></a><strong> </strong>and<strong> </strong>a split/splitless injector (injection temperature 280 °C) equipped with a focus liner (volume: 810 μL). The injection was performed in splitless mode (3 μL) using the high-pressure injection mode function (290 kPa for 0.5 min).&nbsp;</p><p>To avoid that sub-ambient pressure conditions reach the injector, an uncoated column (0.48 m × 0.1 mm ID) was installed before the separation column, a non-polar Equity-5 [poly(5% diphenyl/95% dimethyl siloxane)] of the following dimension: 5 m × 0.53 mm ID × 0.53 μm df. Both the columns were provided by Merck KGaA. The oven was temperature programmed as follows: 40 °C (2 min) to 320 °C raised at 30 °C min−1. Helium was used as carrier gas with an average constant linear velocity of 100 cm s−1 (into the separation column).&nbsp;</p><p>Electron ionization was performed at 70 eV. The interface and ion source temperatures were 320 and 280 ˚C, respectively. The collision gas was argon at a pressure of 200 kPa. The targeted analyses were carried out in the multiple reaction monitoring (MRM) acquisition mode (acquisition frequency: 3.3 Hz). To ensure univocal identification, two different MRM transitions (quantifier and qualifier) were monitored for each compound (Table S1). The SCAN analysis was performed with an acquisition frequency of 3.3 Hz and a mass range of 50–600 m/z. The GCMS Solution v.4.45 software (Shimadzu) was used for data collection and processing.</p><h4><strong>4. Conclusions&nbsp;</strong></h4><p>Under the optimized extraction conditions, the use of DMC as a co-solvent in SFE demonstrated promising performance in the analysis of pesticide residues in apple samples. The method achieved an average extraction yield of 85% across the 64 analyzed pesticides, spanning a broad range of chemical classes, polarities, and molecular weights. Notably, DMC provided cleaner extracts with significantly lower amounts of co-extracted matrix components compared to conventional solvents like ACN and EtOH. Despite the recovery for certain analytes being below 70%, the results remained within acceptable precision limits according to SANTE guidelines. Overall, DMC not only fulfills the analytical requirements for pesticide residue determination but also offers a greener, more sustainable alternative that supports enhanced instrument maintenance and aligns with the principles of GAC.</p><h3><strong>4. The LC/TQ method was developed with a target limit of detection (LOD) at least 20-fold lower than the specification limit to ensure adequate sensitivity. Stevia’s derivatives are significantly sweeter than table sugar. These compounds are estimated to be 50 to 300 times sweeter in comparison. This sweet taste coupled with the fact that the human body cannot metabolize steviol glycosides (i.e., they contribute zero calories to one’s diet), makes them a very attractive sugar replacement.&nbsp;</p><p>The moisture content of stevia materials is a crucial factor when discussing their shelf life and stability. Measuring the moisture content of a stevia derivative, rebaudioside M, using an oven system coupled with volumetric Karl Fischer (KF) titration provided a repeatable approach for quantifying the moisture content of this material.</p><h4><strong>Experimental</strong></h4><p>The analysis was carried out with a Double Pt ring electrode for volumetry (6.00346.100). Testing was executed on the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=220&amp;search=874+Oven+Sample+Processor"><strong>874 Oven Sample Processor</strong></a> (2.874.0120) coupled with an <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=220&amp;search=852+Titrando"><strong>852 Titrando</strong></a> (2.852.0060). Approximately 0.2 g of sample was weighed into an 874 Oven Sample Processor 6 mL vial and placed onto the rack. The oven was set to a temperature of 100 °C, and the evaporated moisture was carried to the volumetric titration vessel by nitrogen gas flow at 70 mL/min. The moisture from the sample was titrated with Hydranal Comp 5 reagent.</p><h4><strong>Conclusion</strong></h4><p>Natural low-calorie sweeteners like stevia derivatives are growing in popularity as people choose healthier alternatives to sugar. Working to develop standardized, specific methodology can help support sweetener industry development, effective quality assessment, and the evaluation of rebaudioside M and other steviol glycosides’ stability.&nbsp;</p><p>This Application Note provides a foundation for the analysis of stevia powder by Karl Fischer titration. An oven system paired with volumetric Karl Fischer analysis provides an accurate, repeatable method to evaluate stevia powder and its derivatives. This method’s compatibility with the 874 Oven Sample Processor allows increased sample throughput through automation, providing operators even more time to complete other important tasks in parallel.</p><h3><strong>3. These drugs are widely prescribed for the treatment of hypertension, angina and heart failure<sup>1</sup> . However, beta blockers may also be misused by athletes resulting in their inclusion on the World Anti-Doping Agency (WADA) prohibited list<sup>2</sup> . Owing to both their therapeutic importance and potential misuse, sensitive and selective analytical methods are required for reliable detection and quantification. Liquid chromatography tandem mass spectrometry (LC-MS/MS) offers high sensitivity and specificity for such analyses.</p><h4><strong>Method</strong></h4><p>Nadolol, metoprolol, propranolol, atenolol &amp; carvedilol were purchased from Sigma Aldrich and Thermo Fisher Scientific. Standards were prepared in 1 mg/mL in LCMS grade methanol. <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=1&amp;search=Nexera+40"><strong>Nexera 40-series HPLC system</strong></a> connected to a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=1&amp;search=LCMS-8045RX"><strong>LCMS-8045RX</strong></a> was used for data acquisition. The mixture of standards was further diluted in LCMS grade methanol (Sigma Aldrich) and Sigmatrix urine diluent to create the standards for the calibration curves. A Shim-pack Velox Biphenyl column was used for the chromatographic separation. All analytes were ionized with electrospray ionization (ESI) in positive ion mode.</p><h4><strong>Conclusion&nbsp;</strong></h4><ul><li>Superior sensitivity in complex matrices: Limit of Detection (at accuracy ±20% and S/N &gt; 3) of 0.010 ng/mL for all the analytes evaluated in this study.</li><li>Excellent linearity and accuracy: Linear responses (with R2 &gt; 0.99) observed across the evaluated concentration range, and consistent accuracy within ±20% for all 17 calibrants included in this study.&nbsp;</li><li>Optimized throughput: Automated interface and MRM optimization via LabSolutions Connect MRM ensured maximum signal-to-noise determination for this study, while the high-organic flush at the end of each run, maintained column integrity</li></ul><h3><strong>4. A custom firefly luciferase (Fluc) mRNA construct containing a Cap-1 structure, 1,919 nucleotides, and a Poly(A) tail was used as the model sample. Importantly, all three CQAs were assessed using a <strong>single LC-MS assay</strong>, supported by specialized waters_connect software tools designed to streamline nucleic acid characterization from sequence entry through final data processing.</p><p>The analytical platform consisted of a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/191"><strong>Waters ACQUITY Premier Binary UPLC System</strong></a><strong> with TUV detector coupled to a </strong><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1148"><strong>Xevo MRT Q-Tof Mass Spectrometer</strong></a>, which uses multi-reflectron time-of-flight technology. Separations were performed on an ACQUITY Premier OST 1.7 µm, 300 Å, 2.1 × 150 mm column, while data-independent acquisition was carried out in MSE mode. Data processing used the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1544"><strong>waters_connect</strong></a><strong> Informatics Platform together with SYNTHETIC Library 2.0, MAP Sequence 2.0, and INTACT Mass 1.9</strong> applications. For sequence mapping, the mRNA was independently digested using four endonucleases—RNase T1, RapiZyme MC1, RapiZyme Cusativin, and human RNase4—providing complementary cleavage patterns.</p><p>The workflow produced detailed information on both capping and Poly(A) tail characteristics. The Fluc mRNA showed <strong>100% capping efficiency</strong>, with 62.7% carrying an m7GpppAm Cap-1 structure and 37.3% an m7GpppGm Cap-1 structure; no uncapped digestion product or Cap-related impurities were detected. Poly(A) tail analysis identified ten major species ranging from <strong>98-mer to 107-mer</strong>, resulting in an average Poly(A) tail length of <strong>102.3 nucleotides</strong>. The Xevo MRT Q-Tof provided high-resolution measurements—approximately 100,000 resolution in the examples shown—and very good mass accuracy for the detected Poly(A) species.</p><p>For sequence integrity, combining the four complementary enzymatic digestions achieved <strong>93.7% unambiguous mRNA sequence coverage</strong>, demonstrating the value of a multi-enzyme approach for comprehensive sequence confirmation. MAP Sequence 2.0 also automatically distinguished structural isomers using elevated-energy fragment-ion information, helping resolve isobaric and isomeric digestion products with less manual interpretation. Overall, the poster demonstrates that high-resolution MSE acquisition on the <strong>Xevo MRT Q-Tof combined with automated waters_connect informatics</strong> can provide an integrated and efficient approach for comprehensive characterization of key mRNA quality attributes.</p></div></div></div></div></div></div></section></div></div>
