Features

ProtiFi S-Trap Turbo 96-well Mini Plate for Proteomics Sample Preparation

ProtiFi S-Trap™ Turbo™ 96-well Mini Plate is a high-throughput 96-well sample-preparation plate designed for rapid proteomic workflows prior to LC-MS/MS. It processes 10–250 µg of total protein per well while retaining the key benefits of S-Trap™ technology, including robust SDS-based protein solubilization, contaminant removal, and enzymatic digestion directly within the matrix. Unlike the standard S-Trap™ format, Turbo™ is optimized for low-volume elution, eliminating the need to concentrate peptides prior to LC-MS/MS.

S-Trap™ Turbo™ uses a polymeric material with more than 100× greater surface capture density than the original design. The increased binding efficiency allows less capture matrix to be used, resulting in smaller elution volumes and a single low-volume elution step. Total processing time, including digestion, is reduced from more than five hours with the standard format to approximately 2.5 hours.

S-Trap™ Turbo™ Technology

S-Trap™ Turbo™ advances the original S-Trap™ design by using a polymeric capture material with substantially increased binding-site density. This allows proteins to be efficiently trapped using less matrix while retaining SDS-compatible cleanup and direct digestion.

The optimized design enables:

  • efficient capture with less matrix,
  • reduced elution volume,
  • replacement of multiple elution steps with a single low-volume elution,
  • elimination of lyophilization or SpeedVac concentration before LC-MS/MS,
  • fewer sample-handling steps,
  • reduced potential peptide loss,
  • increased sample-processing throughput.

According to ProtiFi, the shortened workflow provides analytical performance comparable to or better than the standard S-Trap™ format.

Standard Workflow

  1. Protein solubilization and denaturation with SDS
  2. Optional DNA shearing or nuclease treatment
  3. Reduction with TCEP
  4. Alkylation with MMTS
  5. Acidification to pH ≤1
  6. Addition of methanolic Binding/Wash Solution
  7. Transfer to the S-Trap™ Turbo™ 96-well Mini Plate
  8. Protein trapping
  9. Washing and contaminant removal
  10. Protease addition
  11. Digestion for 2 h at 47 °C
  12. Single low-volume elution
  13. Peptides are ready for downstream LC-MS/MS analysis.
