ProtiFi S-Trap 96-well Micro Plate for Proteomics Sample PreparationProtiFi S-Trap 96-well Micro Plate processes 96 samples containing 1–100 µg protein per well, combining SDS-compatible cleanup, contaminant removal, and direct in-plate digestion before LC-MS/MS.
Features

ProtiFi S-Trap 96-well Micro Plate for Proteomics Sample Preparation

ProtiFi S-Trap™ 96-well Micro Plate is a 96-well sample-preparation plate for parallel processing of up to 96 protein samples prior to LC-MS/MS proteomics. The Micro format is designed for 1–100 µg of total protein per well and uses S-Trap™ technology for SDS-based protein solubilization, protein trapping, contaminant removal, and enzymatic digestion directly within the plate.

The 96-well format is suitable for high-throughput and automated workflows. Plates can be processed by centrifugation, vacuum, or positive pressure and are compatible with automated platforms including the Tecan A200 Positive Pressure Workstation and Agilent Bravo.

S-Trap™ Technology

S-Trap™ sample processing enables the use of SDS for efficient biological sample lysis and protein solubilization. Proteins are subsequently acidified to pH <1 and exposed to a high concentration of methanol, producing extensive denaturation and eliminating unwanted enzymatic activity such as proteases and phosphatases.

Denatured, undigested proteins are trapped within the pores of the derivatized S-Trap™ matrix through multiple weak-affinity interactions. Contaminants are removed during washing, after which the selected protease is added directly to the plate. The resulting peptides are not retained by the S-Trap™ matrix and can be eluted for downstream LC-MS/MS.

Standard Workflow

  1. Protein solubilization and denaturation in SDS
  2. Reduction
  3. Alkylation
  4. Acidification to pH ≤1
  5. Addition of methanolic Binding/Wash Solution
  6. Sample transfer to the S-Trap™ 96-well Micro Plate
  7. Protein trapping
  8. Washing and contaminant removal
  9. Protease addition
  10. In-plate digestion
  11. Peptide elution
  12. Concentration and preparation for LC-MS/MS
