Agilent Technologies / ASMS: Measurement of Metals in Tissue Samples Using Laser Ablation-ICP-MS and an Organic Matrix-Based Standard</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/labrulez-bucket-strapi-h3hsga3/ps_8890_la_icp_ms_metals_tissue_asms_2025_thp_067_en_agilent_ee9bc7a285.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Laser Ablation-Inductively Coupled Plasma-Mass Spectroscopy (LA-ICP-MS) can provide localization information of elements on solid samples. Therefore, bioimaging techniques using LA-ICP-MS have been developed to investigate the distribution of metals on thin sections.&nbsp;</p><p>Primary biliary cholangitis (PBC) is an autoimmune disorder that primarily targets the interlobular bile ducts. The resulting bile duct damage causes chronic cholestasis in the liver, which can eventually progress to cirrhosis. It is well-established that individuals with PBC exhibit impaired copper (Cu) excretion, leading to Cu accumulation within the liver.&nbsp;</p><p>Pathologists have employed orcein or Victoria blue staining to visualize Cu deposition in pathological specimens. 1 Orcein staining is commonly used to assess the stage of PBC. However, both staining methods measure Cu levels indirectly by detecting Cu-binding proteins, which introduces significant variability in measurement accuracy across different facilities and yields relatively low detection sensitivity.&nbsp;</p><p>The aim of the study was to directly quantify Cu in PBC specimen using LA-ICP-MS, a highly sensitive technique, and compare the results with that obtained from normal-looking tissue in liver. The elemental concentrations were determined from external calibration curves generated using an organic matrix-based standard (Solid scaleTM, FUJIFILM Wako Chemicals).&nbsp;</p><h4><strong>Conclusions</strong></h4><p>This study demonstrates the high performance of the Agilent <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?mainauthorItems=10&amp;search=8900+ICP-QQQ"><strong>8900 ICP-QQQ</strong></a> equipped with an ESL 213 laser ablation system for bioimaging human liver specimens.&nbsp;</p><ul><li>An organic matrix-based standard (Solid scaleTM ) provided quantitative results of metals in human liver specimens.&nbsp;</li><li>Patterns of metal distribution (24Mg, 31P, 44Ca, 56Fe, 63Cu, 95Mo) in human liver showed accumulation of Cu in a PBC specimen and its unique localization pattern along the regenerative nodule.&nbsp;</li></ul><p>These results demonstrate that determining metal concentrations in thin sections using LA-ICP-MS provides valuable insights into the mechanisms of PBC and other metal-related diseases.</p><h3><strong>2. Thermo Fisher Scientific / ASMS: Trace metals analysis in baby food using triple quadrupole inductively coupled plasma mass spectrometry (TQ-ICP-MS)</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/labrulez-bucket-strapi-h3hsga3/PO_296_ASMS_25_metals_baby_food_TQ_ICP_MS_na_en_3c869a51aa.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Many countries have established their own regulations and guidelines to ensure the safety and quality of baby food products. These regulations are generally based on scientific research and risk assessments conducted by health and food safety authorities. Among a range of other contaminants, toxic heavy metals, such as lead, arsenic, cadmium, and mercury, are subject to screening.&nbsp;</p><p>The Baby Food Safety Act of 2021 requires manufacturers and the FDA to take long overdue action by setting maximum levels of inorganic arsenic (10 μg∙kg-1, 15 μg∙kg-1 for cereal), lead (5 μg∙kg-1, 10 μg∙kg-1 for cereal), cadmium (5 μg∙kg-1, 10 μg∙kg-1 for cereal), and mercury (2 μg∙kg-1) allowed in baby food.&nbsp;</p><p>Since January 2022, baby food manufacturers need to meet these maximum levels for toxic elements by testing their final products (because only ingredient testing significantly underestimates toxic heavy metal levels in the final product), and to post the results of their product testing online twice per year. Similarly, as the FDA is working on a “closer to zero plan”, the European Union has established regulations and maximum limits for various contaminants, including toxic elements, in baby food. ICP-MS (inductively coupled plasma mass spectrometry) is the commonly used analytical technique to measure trace elements in various samples, including baby food. The specific trace elements to be checked by ICP-MS in baby food may vary depending on the purpose of the analysis and the specific concerns related to the safety and quality of the product.</p><h4><strong>Materials and methods&nbsp;</strong></h4><h5><strong>Instrumentation&nbsp;</strong></h5><p>A <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/products/1488"><strong>Thermo Scientific iCAP MTX ICP-MS</strong></a> and a Thermo Scientific <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/products/1715"><strong>iSC-65 Autosampler</strong></a> were used for analysis. The ICP-MS was operated using the parameters highlighted in Table 1. To facilitate the robustness to analyze a high number of potentially very different sample types, the instrument was operated using argon gas dilution (AGD), applying a mild (approximately 5 times) dilution to all samples. This ensures the productivity and uptime requirements in a high productivity laboratory environment can be met. To meet the stringent detection limit requirements put in place by regulatory authorities, the iCAP MTX ICP-MS was operated in SQ-KED and TQ-O2 modes to confidently eliminate all polyatomic and isobaric interferences but also deliver the sensitivity needed to reach the requested quantification limits for the critical elements.</p><h4><strong>Results&nbsp;</strong></h4><p>Table 3 summarizes the analytical figures of merit for all analytes including the correlation coefficients, the limits of quantification, and the quantification limits of the method (MLOQ). Whereas the limit of quantification (LOQ) only considers what can be quantified by the instrument, the method quantification limit (MLOQ) also accounts for the complete sample preparation, i.e., the sample weight and the dilution factor. Specifically highlighted in the table are the results obtained for As, Cd, Hg, and Pb, elements which are of specific regulatory concern due to their toxicity and risk for bioaccumulation. As can be seen, the achieved MLOQs are at least a factor two lower compared to the nominal limits specified in EAM 4.7.</p><h4><strong>Conclusions&nbsp;</strong></h4><p>This workflow, combining sample preparation using microwave assisted digestion and analysis using the iCAP MTX ICP-MS, allows accurate measurement of the concentration of toxic elements, dangerous for children, as well as the essential elements, necessary for the good growth and health of children, in various baby food products.&nbsp;</p><ul><li>The automatic and constant 5-times dilution allows robust analysis of different types of baby food products, whether dry, liquid, or fatty, without any compromise on sensitivity to reach the regulated limits.&nbsp;</li><li>The stability of the iCAP MTX ICP-MS allows a large variety of different food samples can be analyzed in the same run, ensuring productivity in an applied testing laboratory.&nbsp;</li><li>All types of interferences (polyatomic as well as isobaric interference, including doubly charged ions) are effectively suppressed with either helium collision gas or by using oxygen as a reactive gas in triple quadrupole mode.&nbsp;</li><li><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/products/1489"><strong>Qtegra ISDS Software</strong></a> with the Reaction Finder functionality, allows simplified handling for the operator by automatically defining the isotopes and the acquisition modes to obtain reliable results.&nbsp;</li><li>The maintenance assistant also ensures alerts to the user when actions need to be carried out by the operator to guarantee an instrument with best performance throughout instrument lifetime.</li></ul>

Agilent Technologies / ASMS: Analysis of Allergens in Fragrance Samples Using a Comprehensive GCxGC in Combination with a High-Resolution Mass Spectrometry&nbsp;</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/ps_gc_gc_7250_allergens_asms_2025_mp_353_en_agilent_3459141f12.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Fragrances are highly complex mixtures that combine natural and synthetic components. Analytical separation of these samples on a single GC column results in coelution of isomers and structurally similar compounds which produce similar spectral fragmentation patterns. This significantly complicates identification and quantitative analysis of individual components of the fragrances. Here we demonstrate a power of a comprehensive GCxGC approach combined with high resolution MS for identification and quantitative analysis of 64 allergens in fragrances. With added confidence in compound identification provided by accurate mass, this approach also streamlines a non-targeted analysis of the fragrances to determine their composition.</p><h4><strong>Experimental</strong></h4><p>The samples were separated on an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8890+GC"><strong>8890 GC</strong></a> using a comprehensive GCxGC configuration with a reverse flow modulator (RFM). The non-polar/mid-polar column set was used and comprised of a 20m x 0.1mm x 0.1um DB-1ms column (100% Dimethylpolysiloxane) coupled to a 5m x 0.25mm x 0.15um DB-17ms column (equivalent to (50%-phenyl)- methylpolysiloxane). Optimized instrumental parameters are shown in Table 1.</p><p>The data were acquired using the high-resolution <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=7250+GC%2FQ-TOF"><strong>7250 GC/Q-TOF</strong></a> at data rate of 50 Hz. For compound identification the Unknown Analysis tool of <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=MassHunter+Quantitative+Analysis"><strong>MassHunter Quantitative Analysis</strong></a> software version 12.1 and the GC Image software version 2024 R3 were used. The linear retention indices (RIs) were used to increase confidence in compound identification. Statistical analysis was performed in Mass Profiler Professional (MPP) software version 15.1.</p><h4><strong>Results and Discussion</strong></h4><h5><strong>GCxGC Configuration&nbsp;</strong></h5><p>A GCxGC RFM method was developed to achieve an optimal chromatographic separation of all the allergens while also ensuring adequate sensitivity and carrier gas flow to the MS. GCxGC RFM setup included two detectors: the Q-TOF MS and the FID. The configuration provided an optimal flow to the MS of 1.3 mL/min (11.8% of total flow) maintaining a constant split between FID and MS throughout the entire oven temperature ramp. The configuration is shown in Figure 2.</p><h4><strong>Conclusions</strong></h4><ul><li>Allergens in fragrance samples have been analyzed using comprehensive GCxGC combined with the highresolution GC/Q-TOF and RFM.&nbsp;</li><li>Fragrances with different types of aroma were analyzed using the GCxGC/HRMS combined with statistical analysis to identify the major differentiating compounds that may contribute to the particular type of scent.</li></ul><h3><strong>2. Liège University / MDCW: Coupling of Vac-HS-SPME and GC×GC-qMS for simultaneous 5-HMF quantification and volatile profiling in honey</strong></h3><ul><li>Presentation</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/FL_01_Honey_Eggermont_D_c3a35d2053.pdf"><strong>Full PDF for download</strong></a></li></ul><p>The presentation by D. Eggermont et al. explores a novel analytical approach for assessing honey quality and authenticity. The method focuses on the simultaneous quantification of 5-hydroxymethylfurfural (5-HMF)—a key marker of honey freshness, heat exposure, and compliance with EU regulations—and the untargeted profiling of semi-volatile organic compounds (sVOCs).</p><p>The core of the method is <strong>vacuum-assisted headspace solid-phase microextraction (Vac-HS-SPME)</strong>, which improves extraction efficiency for low and semi-volatiles by reducing gas-phase resistance and accelerating mass transfer. This is followed by <strong>comprehensive two-dimensional gas chromatography coupled with quadrupole mass spectrometry (GC×GC-qMS)</strong>, enabling high-resolution separation and detection of both derivatised 5-HMF and various sVOCs such as benzaldehyde, furfuralcohol, phenylethanol, and menthofuran.</p><p><strong>Instrumentation used in the method includes:</strong></p><ul><li><strong>SPME fiber:</strong> DVB/CAR/PDMS (1 cm)</li><li><strong>Thermal desorption and focusing:</strong> Markes and SepSolve</li><li><strong>GC×GC system: </strong>Shimadzu platform with apolar × polar column set</li><li><strong>Mass spectrometer:</strong> Quadrupole MS operated in SIM mode for targeted 5-HMF quantification and full scan for volatile profiling</li></ul><p>The method was rigorously validated (LoQ 4.7 µg/g, LoD 1.6 µg/g, recovery 98%, precision RSD 21%) and compared against HPLC-UV as the official method, showing comparable results with lower sample amounts and fewer preparation steps. Additionally, the technique aligns with green analytical chemistry principles due to its miniaturization, low solvent use, and energy efficiency. This makes it a promising tool for routine honey analysis and food quality control.</p><h3><strong>3. Shimadzu / ASMS: Advancing PFAS Detection in Drinking Water: GC-MS as a Complementary Technique to LC-MS for Closing PFAS Mass Balance</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/ASMS_25_WP_133_fcfd7e41b2.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Analysis of per- and polyfluoroalkyl substances (PFAS) in the environment is pivotal. There are several standardized PFAS methods, such as EPA 533, 537.1 8327, 1633, OTM-45 and OTM 50. Most of these methods are based on Liquid Chromatography/Mass Spectrometry (LC/MS) techniques. However, LC/MS is not suitable to analyze all PFAS compounds because of the diverse physico-chemical properties of chemicals within the PFAS family. Gas Chromatography/Mass Spectrometry (GC/MS) as a complementary technique can address volatile PFAS compounds that are challenging to analyze by LC/MS. In this study, a Head-Space Solid Phase Microextraction Triple Quadrupole Gas Chromatography/Mass Spectrometry analytical method is used to analyze PFAS in drinking water. This technique has advantages of analyzing volatile PFAS in water with minimum sample preparation procedure.</p><h4><strong>Methods</strong></h4><p>A volatile PFAS analysis method was developed on a Shimadzu <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMS-TQ8040"><strong>GCMS-TQ8040</strong></a> NX with an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=AOC-6000+Plus"><strong>AOC-6000 Plus</strong></a> multifunctional autosampler equipped with a solid phase microextraction (SPME) module (Figure 1).&nbsp;</p><p>Thirteen PFAS target compounds were included in the Multiple Reaction Monitoring (MRM) method. The PFAS chemical classes were perfluoroalkyl iodides (PFIs), (n:2) fluorotelomer iodides (FTIs), (n:2) fluorotelomer acrylates (FTACs), (n:2) fluorotelomer methacrylates (FTMACs), (n:2) fluorotelomer alcohols (FTOHs) and perfluoroalkane sulfonamides (FASAs). Internal standards (IS) FTOHs, FASAs and FTAC mass-labelled compounds were added to each vial prior to extraction. Concentrations of the target compounds were calculated using isotope dilution.&nbsp;</p><p>An internal calibration curve was prepared in 10 mL of reagent water at concentrations of 2000, 1000, 500, 100, 50, 10, 2.5 and 1 ng/L. The IS were spiked at 100 ng/L to each calibrator. Sodium Chloride (NaCl) was added to each vial to achieve a final salinity concentration of 2% NaCl (w/v). These calibrators were vortex for 30 seconds and then placed on the AOC-6000 Plus autosampler rack for analysis.</p><p>The optimized parameters of the HSSPME GC/MS method for the targeted PFAS are listed in table 1. Quantifier and qualifier ions for each PFAS target are listed in table 2. The associated internal standard used for each compound is also listed in table 2.&nbsp;</p><p>A laboratory control sample (LCS) was analyzed to determine the general performance of the method in a clean matrix. Drinking water analyzed in the study were from a private well and an utility with surface water as its source.</p><h4><strong>Conclusion</strong></h4><p>This study demonstrated the satisfactory performance of a HS-SPME GC/MS/MS method to measure PFAS in drinking water. The PFAS family is vast, comprising thousands of different compounds across various chemical classes. Due to this diversity, multiple analytical instruments are necessary to effectively analyze PFAS. While LC-MS is widely recognized for its ability to analyze many PFAS compounds, it is not always practical for measuring certain PFAS. This has led to a gap in the environmental mass balance, especially when it comes to measuring volatile PFAS compounds. Fortunately, unlike LC-MS, GC-MS is well-suited for analyzing volatile PFAS compounds. GC-MS complements LC-MS-based PFAS methods, providing a more comprehensive analytical solution. By expanding the PFAS target list, GC-MS helps close gaps in the PFAS environmental mass balance. The method used in this study demonstrated quantitative capability in analyzing nanogram per liter PFAS compounds in an LCS and matrix influenced drinking water samples.</p><h3><strong>4. Thermo Fisher Scientific: Method transfer to the EXTREVA ASE Accelerated Solvent Extractor&nbsp;</strong></h3><ul><li>Technical note (White paper)</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/wp_001751_sp_extreva_ase_method_transfer_wp001751_en_c7884e1151/wp-001751-sp-extreva-ase-method-transfer-wp001751-en.pdf"><strong>Full PDF for download</strong></a></li></ul><p>The Thermo Scientific™ <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=3&amp;search=EXTREVA+ASE"><strong>EXTREVA™ ASE™</strong></a> Accelerated Solvent Extractor (Figure 1) is a system based on many proprietary technologies including gas-assisted solvent extraction and parallel accelerated solvent extraction. This fully automated system combines the extraction and evaporation capabilities in one instrument, and it can be conveniently used for extracting and concentrating/evaporating extracts from up to 16 solid and semi-solid samples per batch (i.e., 4 sequences of 4 sample cells in parallel). The EXTREVA ASE system combines elevated temperatures and pressurized gas with liquid solvents to achieve fast and efficient collection of analytes from various matrices. The system can use up to six different extraction solvents (or mixtures of them) and extract up to four cells in parallel. The gas-assisted solvent extraction comprises delivering a mixture of extraction solvents and nitrogen gas to the stainless-steel cell to reach the working pressure of 200 psi (~14 bar). The combined effect of temperature and pressure greatly increases the efficiency of the extraction process, significantly reducing the amount of time and solvent required for extraction when compared to traditional techniques such as Soxhlet. The enrichment/concentration process starts immediately after the completion of the extraction step without any analyst interaction, if no offline cleanup of extract needed. The extracts can be evaporated to dryness or concentrated in 2 mL vials to volumes as low as 0.3 mL, with the final volume controlled by artificial intelligence machine vision (Figure 2). Succinctly, the EXTREVA ASE system automates extraction, in-cell clean up, and evaporation, which frees up the analyst’s time for other, more demanding tasks.</p><p>A schematic diagram of the EXTREVA ASE Accelerated Solvent Extractor is shown in Figure 2. This white paper describes the method transfer from a Thermo Scientific™ Dionex™ ASE 350 Accelerated Solvent Extractor to the EXTREVA ASE system, and provides instructions for new EXTREVA ASE system methods.</p><h4><strong>Which options to choose for a particular application?&nbsp;</strong></h4><p>The EXTREVA ASE system workflows comprise three main options: extraction only, evaporation only, or extraction and evaporation combined (integrated) depending on the option of the instrument.&nbsp;</p><p><strong>1. Extraction only:</strong> No evaporation. After extraction, the analyst may perform an offline cleanup, and, subsequently, perform sample enrichment. If evaporation is performed after cleanup, the analyst needs to manually load cells and evaporation bottles back onto the EXTREVA ASE system as per the evaporation workflow below.&nbsp;</p><p><strong>2. Evaporation only:</strong> Evaporation has two modes of operation: evaporation to dryness or evaporation to a fixed volume. When evaporating to a fixed volume, the minimum and maximum volumes are 0.3 and 1.6 mL, respectively. When evaporating to dryness, automatic end point detection is not available; therefore, the analyst needs to optimize evaporation conditions, such as temperature, vacuum and nitrogen gas flow, in order to complete this process in the minimum amount of time while meeting analysis requirements.&nbsp;</p><p><strong>3. Extraction and evaporation combined (integrated):</strong> Extraction followed by evaporation either to dryness (such as fat extraction) or fixed volume (such as PAH, OCP, and PCB) can be pursued based on the need of a particular application or target analytes.</p>

The growing number of invasive fungal infections creates a ripple effect on the rapid advancement of TDM for antifungal agents. Increasing evidence confirms the critical role of TDM in preventing and treating antifungal infections.1 The RECIPE ClinMass TDM kit system for antimycotics in serum (Munich, Germany) was used on a 6475 LC/TQ for the analysis of eight antifungal agents. This application note demonstrates the evaluation and validation of the RECIPE TDM kit, covering sensitivity, linearity, and robustness across five consecutive days (n =&nbsp;5). Data were processed using the Agilent <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?manufacturerItems=1&amp;search=MassHunter+Qualitative+Analysis+software"><strong>MassHunter Qualitative Analysis software</strong></a>, version B.12, and Agilent <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?manufacturerItems=1&amp;search=MassHunter+Quantitative+Analysis+software"><strong>MassHunter Quantitative Analysis software</strong></a>, version B.12.</p><h4><strong>Results and discussion&nbsp;</strong></h4><p>Figure 2 shows the chromatograms of quantifier MRM transitions for the eight antifungal agents at the lowest calibration level. Six out of eight antifungal agents had good chromatographic separation across the run time. The retention times of two antifungals, posaconazole and hydroxy-itraconazole, were slightly overlapped. Despite this overlap, measurement integrity can still be maintained due to MRM selectivity.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>A fast and sensitive analysis of eight antifungal agents in serum was validated on an Agilent 6475 triple quadrupole LC/MS. By employing a ready-to-use RECIPE ClinMass TDM kit, this method demonstrates accurate and precise quantitation of antifungal agents in biological samples. Additionally, the streamlined sample preparation method allows high throughput analysis of up to 350 samples per day.</p><h3><strong>2. Shimadzu / ASMS: Robustness Evaluation of PFAS Analysis in Soil Using LC-MS/MS</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/ASMS_25_WP_137_e932b391c7.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Per- and polyfluoroalkyl substances (PFAS) are widely used in various fields and industries. However, due to their structural stability and resistance to degradation, they accumulate in the environment and are suspected of being harmful to humans. PFAS in soil can accumulate in the human body via agricultural products, thus it is essential to develop sensitive and robust analytical methods for the detection of PFAS in soil with complex matrices.1 This study shows a robustness evaluation by adding 30 PFAS to a soil matrix and using the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?manufacturerItems=3&amp;search=LCMS-8060RX"><strong>LCMS-8060RX</strong></a> to measure these PFAS in the soil matrix 500 consecutive times.</p><h4><strong>Methods&nbsp;</strong></h4><p>For this analysis, LC-MS/MS equipped with a newly designed ESI source was used, and a delay column was installed between the mixer and the autosampler to inhibit the effects of PFAS contamination from the LC system. Six points calibration curves were built, and each calibration point was measured 3 times. A soil sample was prepared referring to part of the soil preparation procedures published by the National Agricultural and Food Research Organization. 2 Thirty PFAS compounds were spiked to the soil sample at 0.1 μg/L after pretreatment and 500 soil sample analyses were performed for the robustness evaluation. This sample contains more than 90% soil matrix. QC samples were also analyzed after every 20 injections of the soil samples.</p><h4><strong>Conclusion</strong></h4><p>The <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?manufacturerItems=3&amp;search=LCMS-8060RX"><strong>LCMS-8060RX</strong></a> performed 500 consecutive measurements of 30 PFAS spiked to a soil matrix sample. The results showed good peak area repeatability, good peak shapes, and good recovery. The newly designed ESI source showed excellent robustness, and stable analysis was achieved over a long period, even for samples with complex matrices.</p><h3><strong>3. Its reduced ion transfer overheads result in a higher MS2 acquisition rate of up to 270 scans per second. Additionally, an enhanced spectral processing algorithm was implemented to deconvolute overlapping spectral features in MS2, for better handling spectra with increased spectrum density.&nbsp;</p><p>Using Orbitrap Astral Zoom MS and Spectronaut® 19.5 software, there was a 12-17% increase in IDs at the precursor level, 10-15% at the peptide level (stripped sequence), and 4-9% at the protein groups level. With Proteome Discoverer software, the gain was 12-19% at the DIA precursor level, 12-17% at the peptide level, and 4-7% at the protein group level. The increase in identifications was primarily observed for low-abundant species. Orbitrap Astral Zoom mass spectrometer also yielded, on average, a 15% higher median number of MS2 data points per peak, while maintaining consistently high quantitation precision and accuracy at the MS2 level.</p><h4><strong>Conclusions</strong></h4><p>The novel Orbitrap Astral Zoom mass spectrometer demonstrates improved performance for qualitative and quantitative analysis of complex proteomics samples over a wide range of sample loads spanning four orders of magnitude, from 100 pg to 1µg.</p><h3><strong>4. Waters Corporation / ASMS: Investigating Fire Fighter and E-waste Handlers Exposure to PFAS Using Liquid Chromatography and Cyclic Ion Mobility Mass Spectrometry</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/720008883_21620a6293.pdf"><strong>Full PDF for download</strong></a></li></ul><p>PFAS are persistent environmental pollutants linked to health risks, including cancer. Monitoring their levels in human biofluids provides insight into exposure pathways. However, the analysis of PFAS isomers is technically challenging due to their structural similarity. To overcome this, the study employs <strong>LC coupled with cyclic ion mobility mass spectrometry (LC-cIM-MS)</strong> using the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/202"><strong>Waters SELECT SERIES Cyclic IMS</strong></a>, which adds a new separation dimension for better specificity.</p><h4><strong>Methods</strong></h4><p>Serum samples from Ghanaian firefighters and E-waste workers were prepared using <strong>solid-phase extraction (SPE)</strong> on 96-well µElution plates. A <strong>22-minute reversed-phase LC gradient</strong> was used for separation. The <strong>Cyclic IMS system</strong>, with ion mobility resolution R~65 and 145, enabled non-targeted PFAS analysis, including the generation of <strong>collision cross section (CCS)</strong> values to support compound identification.</p><h4><strong>Results</strong></h4><ul><li>PFAS compounds such as <strong>PFOS, PFHxS, PFHpS, PFNA</strong>, and <strong>6:2 FTS</strong> were identified in real serum samples.</li><li>The <strong>cyclic IMS</strong> allowed for the separation of coeluting isobaric compounds, such as <strong>PFOS isomers</strong> and <strong>cholic acid biomarkers</strong>, minimizing false identifications (Figure 3).</li><li><strong>Figure 4</strong> lists CCS values and other parameters for PFAS standards and identified analytes from a firefighter serum sample.</li><li><strong>Figure 5</strong> shows ion mobility conformeric profiles for <strong>PFNA</strong>, enabling reliable identification even at low signal intensities.</li></ul><h4><strong>Conclusion</strong></h4><p>The LC-cIM-MS method:</p><ul><li>Significantly improves specificity in PFAS identification.</li><li>Enables both <strong>targeted and untargeted detection</strong> of known and unknown PFAS in complex human samples.</li><li>Can be a key part of future occupational and environmental exposure assessments.</li></ul>

While total arsenic has traditionally been measured, its toxicity largely depends on its chemical form with inorganic species Arsenite and Arsenate (As(III) and As(V)) being highly toxic and carcinogenic, and organic forms monosodium methyl arsenate (MMA) and disodium methyl arsenate (DMA) displayed in fig. 1, considered less harmful. Speciation analysis is therefore essential for accurate risk assessment.&nbsp;</p><p>Rice and rice-based products are of particular concern, as rice can efficiently accumulate inorganic arsenic from the environment. In response, the European Union established maximum residue levels (MRLs) for inorganic arsenic in rice under Commission Regulation (EU) 2015/1006, highlighting the need for precise speciation.&nbsp;</p><p>To address this, advanced techniques like liquid chromatography coupled with inductively coupled plasma mass spectrometry (LC-ICPMS) are employed. LC-ICPMS provides high sensitivity and specificity for arsenic species determination, ensuring regulatory compliance and protecting consumer health.&nbsp;</p><p>This study presents the effective use of Shimadzu’s <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?mainauthorItems=1&amp;search=LC-ICPMS-2050+LF"><strong>LC-ICPMS-2050 LF</strong></a> system for arsenic speciation in rice and related products.</p><h4><strong>Methods and Materials</strong></h4><p>All standard stock solutions are prepared as specified in the LC-ICP-MS Method Package for Arsenic Speciation Analysis Type-2 by Shimadzu, Japan. Seven levels of mixed calibration standards are prepared and injected for the quantification of arsenic species. Calibration curves are generated using the external standard calibration method with a 1/l weighted regression. All samples are prepared in duplicate to verify method performance. Additionally, two spiked samples containing only inorganic arsenic standards are included to evaluate the extraction efficiency of the method.&nbsp;</p><p>The Shimadzu <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?mainauthorItems=1&amp;search=ICPMS-2050+LF"><strong>ICPMS-2050 LF</strong></a>, coupled with the Nexera LC series (Fig. 2), manufactured by Shimadzu Corporation, Japan, is used to separate and quantify the different arsenic species present in the samples.</p><h4><strong>Conclusion&nbsp;</strong></h4><ul><li>The extraction procedure employed effectively ensures the recovery of inorganic and organic arsenic species from various sample types, including both raw and processed materials.&nbsp;</li><li>A highly sensitive method based on hyphenated techniques, specifically LC-ICPMS, is developed for the speciation and quantification of arsenic using the Shimadzu ICPMS-2050 LF coupled with a Nexera inert LC system.</li></ul><h3><strong>3. Coal ash may contain lead, mercury, arsenic and many other heavy metals. Fly ash and bottom ash are two subcategories of coal ash, typically separated based on particle size.&nbsp;</p><p>More than half of coal leftovers are fly ash, the most abundant and well-known component. Particles of fly ash, the lightest type of coal ash, “fly” up into a power plant’s exhaust stacks because they are so light. About 99 % of the ash are captured by filters inside the stacks, which draw it in with opposing electrical charges. Fly ash can be recycled. The fine particles are a perfect material for wallboard and concrete because they bond together and solidify, especially when combined with water.&nbsp;</p><p>But fly ash from coal is also a dangerous industrial solid waste. The natural environment and human health are seriously threatened by its excessive worldwide production, inappropriate disposal methods, underutilization, and lack of knowledge about its inherent risks. Therefore, a method that is quick, precise, and accurate, is required to quantify coal fly ash. Many companies use X-ray fluorescence spectrometry (XRF) as a preferred technology for process and quality control since it is a proven analytical approach that can accurately assess the chemical composition of materials with limited sample preparation.</p><h4><strong>Instrumentation&nbsp;</strong></h4><p>The <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/products/1209"><strong>Thermo Scientific™ ARL™ QUANT’X EDXRF Spectrometer</strong></a> is equipped with a Silicon Drift Detector (SDD) and a 50 Watt Rh or Ag target X-ray tube which is air cooled with a maximum excitation v The Thermo Scientific™ ARL QUANT’X™ EDXRF Spectrometer is equipped with the following features: and radiation of heat in our atmosphere; N2 and O2 which are present in much higher concentrations do not have this same characteristic effect. This compact instrument allows for reliable and fast analysis of coal fly ash. Accuracy and repeatability results show that satisfactory results are obtainable for an analysis of loose powder in vacuum. This is a significant advantage for simple sample preparation while providing good accuracy &amp; precision.</p>

Agilent Technologies / ASMS: Optimizing Hydrogen Carrier Gas on GC/MS Using the HydroInert EI Source and 5Q Columns&nbsp;</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/ps_gc_gc_5q_columns_ei_source_asms_2025_mp_358_en_agilent_c5cf1876f0.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Hydrogen is widely recognized for its benefits in gas chromatography (GC), including cost-effective generation in the lab, rapid chromatography, and higher sample throughput. However, its reactivity can lead to hydrogenation and dechlorination reactions in the mass spectrometer electron ionization (EI) source, complicating its application in many scenarios. To address these issues, Agilent has developed a specially designed extractor source for the Inert Plus GC/MSD and triple quadrupole GC/MS systems, enhancing performance with hydrogen carrier gas. The Agilent HydroInert source maintains mass spectral fidelity and allows users to utilize existing helium-based mass spectral libraries.</p><p>This study investigates how the 5Q series of columns impact the performance and durability of the HydroInert source tune. The study involved cycling through 10,000 oven temperature ramps without any injections to assess the effects of column bleed from the 5Q phase. Furthermore, the study examined how the combination of sample matrix and column bleed from the 5Q phase affects source longevity.</p><h4><strong>Experimental</strong></h4><h5><strong>HydroInert Functionality with 5Q Columns (no Injection)&nbsp;</strong></h5><p>An experiment was designed to investigate the effects of ultra low bleed columns (<a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?instrumentationItems=19&amp;manufacturerItems=1&amp;search=HP-5Q"><strong>HP-5Q</strong></a>) on the functionality of the HydroInert source over time. A method simulating a large oven temperature ramp cycle (Table 1) over 12 minutes was created. The syringe was removed from the ALS tower to simulate injections, but to isolate the source of siloxanes to column bleed from the HP-5Q only. 10,000 injections were executed, and a system tune was performed every 100 injection to monitor source parameter shifts.</p><p>The Agilent <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=1&amp;search=7000E"><strong>7000E</strong></a> was run in SCAN mode, scanning from 50-450 m/z during each run. Air leaks were detected and immediately remedied when found to ensure the experiment was free from oxides. Liner and septum were not changed during this experiment.</p><h4><strong>Results and Discussion</strong></h4><h5><strong>HydroInert Functionality with 5Q Columns (no Injection)&nbsp;</strong></h5><p>Results of the effects of 10,000 oven temperature cycles on the HydroInert source tune parameters are shown in Figure 2. Repeller and ion body voltages are set automatically by the tune algorithm to maximize the transference of ion on the 7000E. Throughout the 10,000 injections the ion body voltage remained stable. The EM voltage gain across 10,000 injections was as expected. The source was in use for 120,000 minutes during this study and data was being collected throughout the duration of the study in SCAN mode. During this study, three leaks were detected at the MS transfer line and were corrected as soon as they were detected. The leaks are reflected in the spike in O2 percentage. A new ion body will typically have a voltage of 16-22 V on a HydroInert source. When an ion body begins to lose functionality the ion body voltage will begin to rise to compensate for the loss. Eventually the ion body voltage tops out at 29-30 V and the tune begins to show lower abundance of tune ions, until the source fails to tune. In the current study the ion body voltage was set by the tune algorithm between 19-24 V. This is a stable and normal operating range for the ion body. It is important to remember that a single elevated ion body tune is not cause for concern, there needs to be a trend of elevation over time. The overall purpose of this first study was to examine what baseline changes would occur to the HydroInert tune parameters when using a 5Q column in leak free conditions. Water and oxygen are known to speed the liberation of column bleed, which in turn reduces the functionality of the HydroInert source. This study shows that under leak free conditions, with a 5Q column, the HydroInert source can function optimally for an extended period. This study adds to a growing body of evidence that has demonstrated one of the main factors in HydroInert longevity is leak state of the system.</p><h4><strong>Conclusions</strong></h4><p>Agilent HydroInert source longevity is dependent on application and method conditions. However, by using the best practices highlighted above, the longevity of HydroInert parts can be prolonged. The HydroInert source cannot be mechanically or chemically cleaned, thus when source parts show reduced functionality, replacement of the recommended parts is required to restore functionality. Replacement intervals will be method and application dependent.</p><h3><strong>2. Liège University / MDCW: MOSH&amp;MOAH in food ingredients and additives and the advantages of using LC/GC×GC(-FID/TOFMS) for their analysis&nbsp;</strong></h3><ul><li>Presentation</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/FL_02_2025_MDCW_AG_MOSH_MOAH_additives_a05df36b39.pdf"><strong>Full PDF for download</strong></a></li></ul><p>This presentation by Aleksandra Gorska and colleagues (Gembloux Agro-Bio Tech) addresses the analysis of MOSH (mineral oil saturated hydrocarbons) and MOAH (mineral oil aromatic hydrocarbons) in food ingredients and additives, particularly those derived from oils and fats. These contaminants are widespread in the food chain due to packaging, lubricants, or storage conditions. While MOSH can accumulate in human tissues, MOAH—especially those with three or more aromatic rings—pose genotoxic risks. The European Food Safety Authority (EFSA) has recently emphasized a lack of data on such impurities in food additives.</p><p>To fill this gap, the researchers developed and applied an advanced analytical workflow based on <strong>HPLC-GC×GC-FID/TOFMS</strong>, replacing the traditional HPLC-GC-FID approach. This multidimensional technique combines high-resolution separation with both flame ionization detection (FID) and time-of-flight mass spectrometry (TOFMS), allowing detailed structural characterization, efficient removal of interfering compounds, and the ability to separate MOAH by the number of aromatic rings.</p><p>Key benefits of the HPLC-GC×GC-FID/TOFMS system include clearer chromatograms with structured elution patterns, improved purification and separation of complex MOAH profiles, and identification of contamination markers such as DIPN (indicating migration from recycled packaging). Additionally, the system enables <strong>simultaneous quantification of MOAH and polycyclic aromatic hydrocarbons (PAHs)</strong>, which have distinct analytical requirements and toxicological profiles.</p><p>The study concludes that, despite its complexity, the HPLC-GC×GC-FID/TOFMS technique provides substantial analytical advantages and aligns with EFSA’s recommendations. It offers a powerful tool for ensuring food safety in products containing oil- or fat-derived additives, enabling more accurate risk assessments and source tracking of contamination.</p><h3><strong>3. Shimadzu / ASMS: Highly sensitive and efficient analysis of aroma components using trapped Headspace GC-MS&nbsp;</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/ASMS_2025_MP_352_97902375e7.pdf"><strong>Full PDF for download</strong></a></li></ul><p>The aroma composition of foods has been the subject of considerable research in recent years. It is well known that the presence of volatile compounds and their composition determine the specific aroma of foods and the flavor of the resulting products. Static headspace technique enables high sensitivity analysis of volatile compounds, normally extracting 1 mL of headspace. But in situations where it is necessary to evaluate the total chemical profile, it is known that components in small quantities also affect the overall aroma. Thus, the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=3&amp;search=HS-20+NX"><strong>HS-20 NX</strong></a> was used running in "Trap mode", where by using trapped headspace sampling, it is possible to extract much more volume and analyze, not only the majority components, but all the compounds with high sensitivity.&nbsp;</p><p>The system used to achieve the highest sensitivity is the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=3&amp;search=HS-20+NX"><strong>HS-20 NX</strong></a> coupled to the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=3&amp;search=GCMS-QP2020+NX"><strong>GCMS-QP2020 NX</strong></a> single quadrupole mass spectrometer.</p><h4><strong>Results and Discussion</strong></h4><p>After the incubation time, the acquisitions were done by loop mode and trap mode with 3- and 5-times sample concentration. In this way, it is possible to analyze, via trap, the presence of compounds at trace levels, which cannot be observed when analyzed in loop mode. 18 compounds could be identified in the sample when analyzed in loop mode, whereas in trap mode, 35 were identified.</p><p>Some components such as benzene acetaldehyde, gammacaprolactone, methyl butyrate, o-guaiacol, that have a decisive influence on the overall aroma and flavor were detected only in the trap mode, due to their low concentrations in the sample. Even concentrating the sample 5-times in the trap, it was possible to detect these compounds that require greater sensitivity along with compounds present in high concentrations in the sample, such as butanoic acid, acetic acid, and others, without losing analysis efficiency.</p><h4><strong>Conclusion</strong></h4><p>The improvement in analysis sensitivity when using trap mode can be observed when comparing the area for some components with a major impact on the overall aroma but present in small quantities. When concentrating the sample 3-times in the trap, the peak intensities increased from 351.17 to 1401.08%. And it was still possible to have a significant increase of another 168%, on average, by concentrating the sample 5-times. The peak intensities increased from 521.48 to 2517.86% compared to loop mode analysis.&nbsp;<br>These analyses demonstrate that trapped headspace can be used to determine an overall aroma profile in a single analysis and with a single trapping material. The trapped headspace sampler enables ultra sensitivity analysis, identifying compounds at low concentrations which were undetectable with conventional headspace samplers.</p><h3><strong>4. In this technical note, the new SmartStatus instrument tracking capabilities are highlighted with examples of how it can enable analytical laboratories to keep instruments producing consistent results.</p><h4><strong>SmartStatus software overview&nbsp;</strong></h4><p>SmartStatus intelligent software, available within Thermo Scientific™ <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/76"><strong>Chromeleon™ Chromatography Data System (CDS)</strong></a>, allows monitoring of the entire analytical system including Thermo Scientific™ autosamplers, GC, GC detectors (FID and TCD), and mass spectrometers (MS). Hundreds of key instrument parameters are monitored by SmartStatus software, which provides real-time data and insights informing the user to potential problems before they arise to ensure maximum instrument uptime. This allows users to make informed decisions on which parts of the analytical system require attention and when. By performing preventative maintenance using data-driven guidance, optimal instrument performance is sustained day in and day out. Figure 1 shows the SmartStatus software overview page for a TSQ 9610 triple quadrupole MS coupled to a TRACE 1610 GC and AI/AS 1610 liquid autosampler. This overview page allows a user to quickly determine if the system is ready to run or requires attention.&nbsp;</p><p>For more information on instrument status, there is a detailed view for each part of the system. This next level view shows how many injections have been performed for the various GC consumables, the lifetime of the MS detector and filament, and when the last tune was performed. Figure 2 shows an example of the GC and MS parameters and how they are monitored. Each parameter is represented as a counter gauge. As the usage increases, the gauge changes color to quickly notify the user of any maintenance that is required or will be needed soon. This allows users to plan maintenance in advance to fit it around their schedule and workload. SmartStatus instrument tracking can be monitored directly in Chromeleon CDS, and consumables usage can also be monitored directly on the TRACE 1610 GC touch screen through the dedicated instrument health icon, as shown in Figure 3.</p><p>The ISQ 7610 GC-MS and the TSQ 9610 GC-MS/MS detector lifetime is monitored using a predictive algorithm. This assesses the voltages on the detector and predicts the lifetime remaining on the system so that preventative maintenance can be planned. As the ISQ 7610 GC-MS and TSQ 9610 GC-MS/MS are equipped with the XLXR™ electron multiplier detector as standard, the lifetime of the detector is several years.1 By monitoring the detector intelligently, users can avoid unnecessary downtime. Figure 4 shows monitoring of the detector within SmartStatus software.</p>

Agilent Technologies / ASMS: Faster Than the Speed of Life: A Rapid Screening Method for Select PFAS Compounds in Serum by RapidFire-MS</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/ps_rapidfire_ms_pfas_serum_asms_2025_wp_243_en_agilent_c8b56f4dd5.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Per- and polyfluoroalkyl substances (PFAS) are a broad class of compounds with a high degree of fluorination used in a variety of industries and consumer products over the past seventy years. Because of their widespread use, as well as their persistence and poorly understood effects on the human body, laboratories have started to monitor their presence in biological fluids, including serum. Biological samples are commonly prepared with protein precipitation. Additional cleanup steps such as solid phase extraction (SPE) with either C18 or weak anion exchange (WAX) sorbent can be used for increased sensitivity and cleanup. Alternatively, a pass-through cleanup using Captiva EMR-Lipid removes proteins and phospholipids through filtration, which significantly reduces sample complexity.</p><h4><strong>Experimental</strong></h4><p>Purchased serum was aliquoted into two sets of samples, one for protein precipitation (ppt) only and one for a protein precipitation plus Captiva EMR-Lipid based cleanup. For each preparation, 100 µL serum was aliquoted into a tube and spiked with 25 native and extracted isotopically labeled (EIS) compounds (EPA 533 list). The samples were vortexed for 1 minute and then crashed with 1% formic acid in cold ACN. They were vortexed for 3 minutes and then spun for 10 minutes to complete the crash. Samples undergoing the Captiva-EMR Lipid cleanup were then loaded onto a pre-rinsed cartridge and allowed to filter through (Figure 1). All samples were then spiked with the additional non-extracted isotopically labelled (NIS) compounds, vortexed, and transferred to a plate for analysis.</p><p>The RF/TQ system consisted of a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?manufacturerItems=1&amp;search=RapidFire+400"><strong>RapidFire 400</strong></a> front end with a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?manufacturerItems=1&amp;search=6495D+triple+quadrupole"><strong>6495D triple quadrupole mass spectrometer</strong></a> for detection. Samples were introduced into the mass spectrometer using a RapidFire cartridge for additional cleanup, and the total cycle time was about 12.5 seconds, with analytes detected in negative polarity. Specific method conditions are detailed in Tables 1 (RF) and 2 (MS).</p><h4><strong>Results and Discussion</strong></h4><h5><strong>Early Development Work&nbsp;</strong></h5><p>Feasibility studies for detection of PFAS compounds in serum using the RapidFire started by looking for PFAS background in the instrument and in all steps of the workflow. Early testing demonstrated no detectable background from any stage of the process.</p><p>RapidFire method development tested a variety of cartridges for optimal binding of the PFAS compounds, as well as determining the best state times and solvent conditions. For the preliminary work, the C18 cartridge appeared to have the best results, while the optimized elution solvent was 5 mM ammonium acetate in methanol.</p><p>Early experiments were performed to understand the effects of dilution and sample prep on the overall sensitivity of the method. When only a protein precipitation preparation was employed, dilution had a significant positive effect on analyte sensitivity, as shown in Figure 2. However, when the sample prep routine utilized protein precipitation in combination with a Captiva EMR-Lipid cleanup, additional dilution was not necessary and in fact decreased overall sensitivity, as shown in Figure 3. This is attributed to the removal of extraneous matrix using the Captiva EMR-Lipid device, which has a unique selectivity for unbranched alkane chains, capturing phospholipids while allowing PFAS compounds to pass through the sorbent bed unretained.</p><h5><strong>Custom Cartridge Work&nbsp;</strong></h5><p>This preliminary work was done using the C18 cartridge, but the cartridge had poor retention of the short-chain PFAS compounds (C5 and below). To address this issue, a cartridge was custom-packed with mixed mode material that included C18 and anion exchange resins. Further testing and method optimization for this specific packing material yielded better results across the board, but especially for the short-chain PFAS compounds. Sensitivity for most analytes was at or below 0.1 ppb, as summarized in Table 3, with a calibration range of 0.01- 10 ppb tested for all analytes.</p><h4><strong>Conclusions</strong></h4><ul><li>LOQs for most PFAS analytes of interest in serum were 0.1 ppb or below</li><li>Samples were injected every 12 seconds, allowing for high throughput analysis</li><li>Custom mixed mode cartridge had best overall performance&nbsp;</li><li>There was no background PFAS contribution from the RapidFire system</li></ul><h3><strong>2. Shimadzu / ASMS: Benchtop Matrix Assisted Laser Desorption Ionization Mass Spectrometry of PFAS for preliminary screening of environmental samples</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/ASMS_2025_MP_588_4c415fced0.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants which negatively impact human health and the environment.&nbsp;<br>High concentrations of PFAS in test samples can cause contamination of sensitive LC-MS/MS instruments requiring extensive decontamination procedures, resulting in significant downtime and costly delays in sample processing.&nbsp;</p><p>We present the development of a simple, cost-effective method for the rapid screening of PFAS samples using an entry-level linear benchtop MALDI-TOF mass spectrometer to identify samples containing high levels before they enter the analytical workflow.</p><h4><strong>Methods</strong></h4><p>PFOA and PFOS standards were purchased from Merck, UK. Stock solutions (1 ppm prepared in acetonitrile) were diluted in water to the required concentrations.&nbsp;</p><p>Samples were tested with a variety of MALDI matrices (CHCA, DHB, DHAP, DAN, NRM and TMGN), with norharmane (NRM) and 1,8-bis(tetramethylguanidino)-naphthalene (TMGN) identified for further investigation. Diluted samples were mixed with NRM (1 mg/mL) or TMGN (5 mg/mL) in 70:30 Acetonitrile:Water. Blank samples of UHP water were prepared to ensure no contamination was present. 1 µL of the mixture was spotted onto a FlexiMassSR48 slide and the samples were analysed using a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?manufacturerItems=3&amp;search=MALDI-8030"><strong>MALDI-8030 MALDI-TOF mass spectrometer</strong></a> (Shimadzu). Analysis conditions are shown in Fig 1. Peaks at m/z 314.2 and m/z 499.2 were monitored for PFOA and PFOS, respectively. 13C8PFOS (m/z 507) was used as an internal standard for PFOS samples with a final sample concentration of 6.25 ng/mL (see Fig. 2). MALDI-MSI was used to assess spot homogeneity and limits of detection were established.</p><p>Subsequently, PFHxA and PFBS standards were purchased from Merck, UK and analysed using the same method</p><h4><strong>Conclusion</strong></h4><ul><li>We have shown, in principle, that it is possible to use MALDITOF mass spectrometry to implement a simple pre-screening method for aqueous samples to prevent widespread contamination in a PFAS monitoring laboratory. Using just a 200 µL sample and minimal preparation, a full slide of samples can be acquired in less than 15 minutes&nbsp;</li><li>Spot imaging during method development highlighted the differences in homogeneity of PFOS and PFOA spots suggesting that multiple internal standards would be necessary to pursue semi-quantitative analysis.&nbsp;</li><li>Whole slide imaging can provide quick results for multiple samples across a range of PFAS concentrations in a single run with easy visual identification of potentially problematic samples.&nbsp;</li><li>As a linear MALDI-TOF method, these results do not confirm the presence of PFAS but may be used as an indicative result to modify further testing protocols. Confirmatory LC-MS/MS analysis is required to determine the presence and concentration of any PFAS.</li></ul><h3><strong>3. Thermo Fisher Scientific / ASMS: Enhanced resolution with multi-pass mode on the Orbitrap Astral Zoom mass spectrometer&nbsp;</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/PO_319_ms_ASMS_25_Astral_Zoom_TMT_po319_na_en_a16d57e473.pdf"><strong>Full PDF for download</strong></a></li></ul><p>The introduction of Thermo Scientific TMT 32-plex, with reporter ions clustered into quadruplets spaced by 3 mDa, demands a resolving power of approximately 90 k at low m/z, which challenges state-of-the-art time-of-flight mass analyzers. The Thermo Scientific Orbitrap Astral mass spectrometer [1] typically achieves around 60 k resolution at m/z = 138, falling to 50 k under high space charge influence.&nbsp;</p><p>Here we present an enhanced resolution multi-pass method implemented in the new Thermo Scientific <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1959"><strong>Orbitrap Astral Zoom mass spectrometer</strong></a> [2], significantly improving the resolving power at low m/z enabling TMT 32-plex on the Thermo Scientific Astral analyzer. The ions are repeatedly guided through a switchable prism deflector to extend the ion path while the principal aberrations are compensated, thereby more than doubling the resolution.</p><h4><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1959"><strong>The Orbitrap Astral Zoom Mass Spectrometer</strong></a></h4><p>The Orbitrap Astral Zoom mass spectrometer incorporates a range of novel technologies for higher speed, sensitivity, dynamic range and resolving power. A faster ion filter and quadrupole switching in combination with improved ion transfer times in the ion-routing multipole (IRM) and ion processor allow DIA methods (Thermo Scientific Pierce FlexMix calibration solution, 2 Th isolation window, 350-980 m/z range, Thermo Scientific Orbitrap MS and Astral MS/MS) with scan rates faster than 270 Hz (DDA methods &gt; 180 Hz). A duty cycle of up to 80 % at 270 Hz is made possible by a new accumulation stage in the bent trap (linear to 106 ions) which increases the effective ion injection into the IRM by up to 40 % (additional ~0.75 ms on top of 2 ms IT in a DIA method). Furthermore, an additional mode of the high-dynamic range detector of the Astral analyzer for low-input sample applications like single-cell proteomics boosts the single ion detection efficiency by about 10 % and thereby the sensitivity. In combination with a new peak deconvolution algorithm for enhanced spectral processing, this increases the depth of analysis. In addition, modified IRM hardware allows for faster stepped collision energy experiments for deeper spectral coverage (2 CE: 80 Hz (OT Astral), 180 Hz (Orbitrap Astral Zoom MS), 3 CE: 60 Hz, 150 Hz). A modified AGC filling in the Orbitrap by a new enhanced dynamic range mode (EDR) provides additional spectral complexity and sensitivity. Here, multiplexed ion injections with different accumulation times are used for different mass ranges taking the variation in signal intensity into account. In addition, the Astral analyzer incorporates a new multi-pass mode which enhances the resolving power for small masses by more than a factor of two, enabling TMT 32-plex measurement in the Astral analyzer.</p><h4><strong>Conclusions&nbsp;</strong></h4><p>The TMT HR Mode advances the capabilities of the Astral analyzer. Fast electronic switching of the relay prism opens an ion trap and guides the ions after three times their normal flight distance to the detector. This greatly boosts the resolving power, especially for small m/z ratios, thereby enabling baseline-resolved quadruplets of reporter ions in a TMT 32-plex sample. First results show a peak resolution well above 100 k while maintaining high mass accuracy and shot-to-shot stability. The TMT HR Mode is utilized within a novel acquisition scheme delivering greater throughput for protein identification and quantitation for multiplexing up to 32 samples. Achieving a throughput of over 440 samples per day, the proteome coverage included 4,299 quantified protein groups and 22,146 peptides.</p><h3><strong>4. Waters Corporation / ASMS: Assessing the Impact of Part-per-Billion Mass Accuracy Upon Forensic Toxicology Data-Independent Analysis Screening&nbsp;</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/720008882_68ab66ac31.pdf"><strong>Full PDF for download</strong></a></li></ul><p>This poster presents a forensic toxicology screening approach using <strong>ultra-high mass accuracy</strong> to improve specificity and efficiency in the analysis of complex biological matrices, such as urine. The need for rapid, accurate, and cost-effective screening is critical in forensic laboratories. The study evaluates the impact of <strong>parts-per-billion (ppb) mass accuracy</strong> using the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1148"><strong>Waters Xevo MRT Mass Spectrometer</strong></a><strong> </strong>compared to previous DIA methods.</p><p>The method involved analyzing a commercial standard and ten anonymized urine samples, diluted and analyzed via the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/199"><strong>ACQUITY UPLC I-Class FTN PLUS System</strong></a> coupled with the <strong>Xevo MRT MS</strong>. Data acquisition was performed in <strong>MSE mode</strong> (data-independent acquisition) under positive electrospray ionization (ESI+) using a 15-minute gradient. Data were processed in waters_connect software, leveraging a comprehensive spectral library with over 2000 compounds.</p><p>Results demonstrated a <strong>root mean square (RMS) mass error of just 571 ppb</strong> for precursor ions, with 94% of compounds detected within 1 ppm, and 96% of fragment ions within the same tolerance. A key example illustrated how reducing fragment ion tolerance from 2 mDa to 0.2 mDa eliminated a false positive (Anabasine), correctly identifying Nicotine as the true analyte, showing how ppb-level accuracy increases identification confidence and minimizes misidentifications.</p><p>The study concludes that ppb mass accuracy significantly reduces false positives, lowers review workloads, and facilitates independent analysis by less experienced personnel. Additionally, it supports cost savings by minimizing the need for confirmatory testing, potentially reducing both financial and environmental burdens in forensic laboratories.</p>

<h3><strong>1. What exactly is a “source” in mass spectrometry?</strong></h3><p>Whether you’re running LC-MS, imaging-MS, or native-MS, the ion source is where molecules leave the chromatograph, capillary, or sample stage and become charged gas-phase particles that the mass spectrometer can detect. The source you choose governs the kinds of compounds you’ll see, the flow rates you can use, and the robustness you can expect from run to run.</p><p><strong>Sources fall into two broad families:</strong></p><ul><li><strong>Vacuum ionization: </strong>Electron ionization (EI), fast-atom bombardment (FAB), and traditional MALDI create ions inside the instrument’s evacuated housing.</li><li><strong>Atmospheric-pressure ionization (API): </strong>Electrospray (ESI), atmospheric-pressure chemical ionization (APCI), atmospheric-pressure photo-ionization (APPI), and today’s atmospheric-pressure MALDI (AP-MALDI) generate ions in open air and then guide them through an interface into vacuum.</li></ul><blockquote><p>Learn more about Ionization methods here: <a target="_blank" rel="noopener noreferrer" href="https://www.thermofisher.com/content/lifetech/global/en/home/industrial/mass-spectrometry/mass-spectrometry-learning-center/mass-spectrometry-technology-overview/ionization-source-technology-overview.html"><strong>Ionization Source Technology Overview | Thermo Fisher Scientific – US</strong></a></p></blockquote><h3><strong>2. Why electrospray changed the game</strong></h3><p>In 1988 John B. Fenn demonstrated that a fine, high-voltage spray could coax intact proteins into the gas phase (see figure). The work earned a share of the 2002 Nobel Prize in Chemistry. Because electrospray produces multiply charged ions, large biomolecules fit within the m/z window of modern analyzers. Putting ESI sources to work on a Thermo Scientific platform</strong></h3><p>If you have ever run a proteomics experiment on Thermo Scientific mass spectrometry solution, you have already handled at least two ESI sources:</p><ul><li><strong>Thermo Scientific OptaMax NG ion source</strong> or <strong>Thermo Scientific OptaMax Plus ion source</strong> with the Thermo Scientific<sup> </sup>Heated Electrospray Ionization (HESI-II) Probe – easy installation into the MS face, this robust family of sources, paired with adaptable HESI-II probes, supports a wide range of flow rates from 1 – 2,000 µL/min, providing flexibility for various applications. Most proteomics laboratories use it for calibrations or quick QC injections: direct-infuse a Pierce FlexMix solution through the HESI probe and the instrument can calibrate to sub-ppm accuracy</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/2464"><strong>Thermo Scientific EASY-spray source</strong></a> or <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/2463"><strong>Thermo Scientific NanoSpray Flex ion source</strong></a> – once calibration is complete, the OptaMax ion source comes off and a low-flow source goes on. Thermo Scientific<sup> </sup>EASY-Spray HPLC columns combine LC column, liquid junction, and emitter in a single finger-tight connection, eliminiating dead volume and manual x-axis alignment. The NanoSpray Flex offers emitter and geometry control, providing flexibility for method development when custom configurations are needed.</li></ul><p>In a routine bottom-up proteomics run, you might equilibrate an EASY-Spray column on the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/398"><strong>Thermo Scientific Vanquish Neo UHPLC</strong></a>, calibrate the mass spectrometer using the OptaMax NG ion source fitted with HESI-II probe, then swap to an EASY-spray source loaded with your nanoLC column, and finally acquire a 30-minute DIA gradient on an <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1959"><strong>Orbitrap Astral Zoom mass spectrometer</strong></a> - never touching the mass spectrometer itself, only exchanging the front-end source.</p><h3><strong>4. Alternate source spotlight: Native MS with static nano-spray</strong></h3><p>Electrospray isn’t limited to peptide gradients. In native MS the goal is to perform mass analysis on intact, non-denatured molecules- virus capsids, ribosomal subunits, even 10 MDa gene-therapy assemblies- without the salts and organic solvents required by LC. Here the workhorse is static nano-spray: load &lt;5 uL of volatile, salt-stripped buffer into a pre-conductive tip, let capillary action deliver a &lt; 50 nL/min plume straight into the MS inlet, and- because the analyte never touches a column- fragile complexes stay together long enough for high-resolution mass spectrometry analysis. The Nanospray Flex ion source with the Offline Nano Electrospray kit makes difficult-to-analyze native applications compatible with state of the art mass spectrometry. For example, it is often coupled with the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/560"><strong>Thermo Scientific Q Exactive UHMR mass spectrometer</strong></a> and the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/748"><strong>Thermo Scientific Orbitrap Ascend Structural Biology Tribrid mass spectrometer</strong></a>.<br><br>Static nano-spray highlights how choosing the right ion source- and the right accessories- opens new analytical spaces.</p><h3><strong>5.
