The core concept builds on GC×GC, where compounds are separated using a primary and secondary column connected via a modulator, significantly increasing separation power compared to one-dimensional GC. The presentation contrasts <strong>thermal modulation</strong> (offering sharper peaks and higher theoretical sensitivity but requiring cryogenic cooling) with <strong>flow modulation</strong>, which avoids cryogens, handles volatile analytes well, and is more flexible, but typically demands higher gas flows that standard MS systems cannot tolerate .</p><p>To address this limitation, the study utilizes the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=24&amp;search=JMS-Q1600GC"><strong>JEOL JMS-Q1600GC</strong></a><strong> UltraQuad quadrupole GC-MS system</strong>, which can accept unusually high carrier gas flows (up to ~20 mL/min), enabling direct coupling with a <strong>SepSolve INSIGHT flow modulator</strong> without splitting the effluent . The analytical setup includes dual GC columns (e.g., Zebron ZB-5MSplus for the first dimension and BPX50 for the second), high secondary flow rates (~25 mL/min), and electron ionization (EI) MS detection. Additional system components include optional FID detection, bleed line control, and ChromeSpace software for data processing . This configuration allows full analyte transfer into the MS, eliminating the need for flow splitting and potentially improving detection efficiency.</p><p>Experimental results demonstrate the applicability of this approach to complex mixtures, including a <strong>pesticide mixture containing over 200 compounds</strong> and real-world samples such as perfumes and fuels. The GC×GC-MS system produced detailed two-dimensional chromatograms with high spectral quality, enabling the correct identification of approximately 120 compounds despite relatively moderate spectral match factors . Comparisons at different flow rates (e.g., 25 vs. 50 mL/min) showed that spectral integrity and compound identification remain robust, indicating that higher flow operation does not significantly compromise MS performance.</p><p>In conclusion, the work demonstrates that <strong>flow-modulated GC×GC-QMS without flow splitting is feasible</strong>, provided that the MS system can tolerate high carrier gas flows. While further optimization is needed (e.g., column selection, inlet pressure, and separation conditions), the approach offers a practical alternative to thermal modulation by eliminating cryogens and simplifying operation. Future work will focus on improving sensitivity, optimizing separations, and exploring alternative carrier gases such as hydrogen to reduce costs and expand applicability</p></div></div></div></div></div></div></section><h3><strong>2. Shimadzu: Detailed Hydrocarbon Analysis by Nexis GC-2030 Using ASTM D6730</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_00863_en_eb633bc741.pdf"><strong>Full PDF for download</strong></a></li></ul><h5><strong>User Benefits</strong></h5><ul><li>Detailed compositional analysis of gasoline in accordance with ASTM D6730 is feasible using the Nexis GC-2030.&nbsp;</li><li>By employing PONAsolution, a PONA/DHA analysis software, laborious identification tasks can be reduced, and quantitation calculations and report generation can be performed automatically.</li></ul><p>ASTM D6730 specifies the analysis of hydrocarbons in gasoline and oxygenates such as ethanol, methyl tert-butyl ether (MTBE), and ethyl tert-butyl ether (ETBE) over a boiling point range up to 225 ̊C. Identification and quantification of individual compounds in gasoline are critically important for refinery process control and product quality assurance. In this application news, we report compositional analysis of gasoline conforming to ASTM D6730 using the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=Nexis+GC-2030"><strong>Nexis GC-2030 gas chromatograph</strong></a>. Gasoline typically comprises over 100 hydrocarbons, making identification tasks highly complex; however, PONAsolution, a PONA/DHA analysissoftware,simplifies this process.</p><h4><strong>PONAsolution</strong></h4><p>PONAsolution Ver.6 provides retention index libraries for each relevant standard (ASTM D6730, D6729, D5134, D6733). Identification of n-alkanes automatically corrects the retention times of the remaining several hundred components, thus streamlining the identification process. When only a specific retention region requires adjustment, the two-point identification function is useful (Fig. 1). In the two-point identification procedure, reference and sample chromatograms are compared while assigning corresponding peaks. First, click on an identified component mark in the reference data (Fig. 1, a), then click the matching sample peak (Fig. 1, b). Repeat this for a second pair of peaks. The retention times of components between these two anchor points are then automatically corrected.</p><h4><strong>Conclusion</strong></h4><p>Compositional analysis of gasoline in accordance with ASTM D6730 was performed. The combination of SH PONA Tuning Column and SH-1 PONA main column enabled separation of compounds with closely spaced retention times. Moreover, use of PONAsolution software greatly simplifies complex identification tasks.</p><h3><strong>3. Thermo Fisher Scientific: Fiber optic probes add flexibility to Raman chemical analysis</strong></h3><ul><li>Technical note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/drx3_flex_tech_note_tn54708_bebb56b870.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Fourier Transform Infrared Spectroscopy (FTIR) and Raman Spectroscopy are non-destructive analytical techniques that rely on the interaction of electromagnetic radiation with molecular vibrations to provide information about the chemical composition and structure of a sample. Despite their similarities, there are some distinct differences between the two techniques. In principle, FTIR is based on the absorption of infrared radiation by molecules and measures changes in dipole moment during molecular vibrations. Raman spectroscopy, on the other hand, is based on the inelastic scattering of monochromatic light and measures changes in polarizability during molecular vibrations.&nbsp;</p><p>When applied to bulk chemical analysis, both FTIR and Raman spectroscopy can be used to analyze solids, liquids, and gases, but the nature of Raman spectroscopy lends some unique sampling advantages. Additionally, Raman often requires little to no sample preparation. Presented herein are application examples that highlight some of the sampling advantages of Raman spectroscopy. All the spectral data in this technical note were collected using a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/2238"><strong>Thermo Scientific™ DXR3 Flex Raman Spectrometer</strong></a> equipped with a fiber optic probe. (Fiber optics are readily available for Raman spectroscopy because of the use of visible and NIR light and do not require the exotic materials associated with FTIR fiber optics.) Preliminary tests for all samples were carried out using both 532 nm and 785 nm excitations, and it was determined that the 785 nm laser set gave an overall better balance between fluorescence and Raman scattering efficiency.</p><h4><strong>Summary&nbsp;</strong></h4><p>In this technical note, the analyses of a variety of samples by Raman spectroscopy are demonstrated. All samples were analyzed without sample preparation.&nbsp;</p><ul><li>Precise and localized analysis at the focal point of the Raman-exciting laser allows sampling through glass bottles, blister packaging and plastic containers, which eliminates possible cross contamination and analyst exposure to hazardous materials. The contributions from the packaging and containers depend on the type of container material, the container thickness, and the materials’ chemical compositional complexity, but these contributions can be readily subtracted out when needed.&nbsp;</li><li>Water is a weak Raman scatterer. Raman therefore has reduced water sensitivity, making it a preferred technique for aqueous solution analysis over FTIR. As demonstrated in the analysis of an energy drink, the Raman bands from water can be subtracted from the spectrum of the aqueous solution, leading to the identification of the components of the solution.&nbsp;</li><li>Raman spectroscopy is well suited for the analysis of inorganic materials, especially in identifying the crystal phases and polymorphs, as demonstrated by the analyses of minerals and pigments in a painting.&nbsp;</li><li>Fiber optic probes provide flexibility and ease of maneuverability. This is particularly advantageous when analyzing samples that are difficult to access or manipulate, such as large containers, geological samples and paintings</li></ul><p>The DXR3 Flex has multiple sampling options available. A properly configured fiber-optic probe is arguably the most versatile accessory for the analysis of bulk solids and liquids. The advantages, both from those inherent to Raman spectroscopy and those resulting from the use of fiber optic probe, provide opportunities for a wide variety of different applications when using the DXR3 Flex Raman Spectrometer.</p>

Non-destructive, fast analysis is preferred to maintain yield and enable at-line monitoring, avoiding lengthy sample preparation. Recent advancements in benchtop NMR spectrometers (up to 90 MHz) allow quick analysis without deuterated solvents. In this proof-of-concept study we purified a pharmaceutical sample via HPLC and confirmed its identity using benchtop NMR. Fractions (0.7–4.4 mM) were analyzed without preparation, using only 300 µL per sample, ensuring rapid batch control with no product loss. WET parameters were adjusted for each sample to yield optimal suppression for all solvent signals. Pooled HPLC fractions were analysed directly without addition of deuterated solvents or other sample preparation.</p><h4><strong>Conclusion</strong></h4><p>This proof-of-concept study successfully shows that HPLC and benchtop NMR can be used together easily. Samples can be analyzed non-destructively within minutes and without any further preparation due to highly efficient suppression of multiple solvent signals. Compounds can be identified without the need of reference standards. This study shows that NMR can provide a valid compatible alternative detector for HPLC fraction analysis.</p><h3><strong>2. Because CFRP consists of carbon fibers and resin, it shows complex fracture behavior, and brittle fracture progresses from damage as the point of origin. Since fracture proceeds instantaneously, a high-speed video camera with a high recording speed is necessary when observing fracture of CFRP. One method for measuring strain in a high-speed tensile test is Digital Image Correlation (DIC), which visualizes the distribution of strain at the specimen surface by comparing a random pattern on the specimen surface before and after deformation. Thus, a high-speed video camera with a high resolution is also necessary in order to improve the DIC performance.&nbsp;</p><p>Newly-developed <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?mainauthorItems=1&amp;search=HPV-X3"><strong>Hyper Vision HPV-X3 high-speed video camera</strong></a> is extremely well-suited to DIC analysis of CFRP, as its recording speed is 2 timesfaster and itsresolution is 3 times higher than the conventional HPV-X2. In this experiment, a high-speed tensile test of CFRP was conducted using a HITS-TX high-speed tensile testing machine. The condition of the test was observed with the new HPV-X3, and the strain distribution of the CFRP test piece was clarified by a DIC analysis of the recorded images.</p><h4><strong>Test Piece and Test Instruments</strong></h4><p>A CFRP quasi-isotropic laminate was used as the test piece. Aluminum foil was glued to the back side of the test piece, and the timing when the electrical conduction of the aluminum foil was interrupted by fracture of the test piece was used as the trigger for video recording. The HITS-TX was used in the highspeed tensile test. The HPV-X3 was used in fracture observation, and for comparison, the fracture process was also recorded with the conventional HPV-X2 camera. Fig. 1 shows the condition of the test, and Table 1 and Table 2 show the equipment composition and test conditions, respectively.</p><h4><strong>DIC Analysis Results in High-Speed Tensile Test</strong></h4><p>Fig. 3 shows the DIC analysis results in the high-speed tensile test. The color bar was set to display the color purple at around 0 % strain and red at around 2 % strain. Fig. 3 (1) is an image at the start of the test. After the test started, strain in the 45˚ direction concentrated from the lower left to the right central area, as can be seen in Fig. 3 (2). Then, as the test progressed, a condition of increasing strain in the initial area of strain concentration can be seen in Fig. 3 (3) to (9). A crack occurs in Fig. 3 (10), and strain decreases in the crack area. Following this, the test piece fractured completely in Fig. 3 (11) and (12), and a decrease in the strain in the area around the crack could be observed.</p><h4><strong>Conclusion</strong></h4><p>A high-speed tensile test of CFRP was conducted using a HITSTX high-speed tensile testing machine and the newlydeveloped HPV-X3 high-speed video camera, and the strain distribution at the time of test piece fracture was visualized by a DIC analysis. Because the HPV-X3 offers a 3 times higher resolution and a 2 times faster recording speed than the conventional device, it is a suitable high-speed video camera for fracture observation and DIC analysis in impact tests of brittle materials such as CFRP. The measurement system introduced in this Application News article is a useful tool for the development of composite materials.</p><h3><strong>3. Thermo Fisher Scientific: Polymer analysis using femtosecond-laser-ablation depth profiling</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/labrulez-bucket-strapi-h3hsga3/xps_depth_profiling_polymer_analysis_appnote_c731e2e7e0.pdf"><strong>Full PDF for download</strong></a></li></ul><p>X-ray photoelectron spectroscopy (XPS) is an essential polymer characterization technique that can identify not just the elemental composition but also the chemical state of polymer samples. This allows the chemical bonding environment to be deduced and, as a result, the polymer species can be identified. This can all be achieved with minimal sample preparation thanks to the built-in charge-compensation systems found in XPS instruments, which simplify the analysis of insulating samples.&nbsp;</p><p>The introduction of gas cluster ion sources (GCIS), such as the Thermo Scientific™ MAGCIS Dual Mode Ion Source, marked a major step forward in polymer analysis. Unlike monatomic ion beams, GCIS are able to clean surface contamination, and etch away layers of sample material, without damaging the underlying chemistry of organic samples such as polymers. As a result, GCIS has allowed chemical-damage-free analysis to reach micrometers below the sample surface. This depth of analysis can be highly time consuming, however, and total experimental times can range from multiple hours to days.&nbsp;</p><p>The addition of femtosecond-laser-ablation (fs-LA) technology is alleviating this, allowing for XPS spectra to be collected from a depth of tens of µm into a sample in a matter of minutes. This is because the laser ablation occurs on the femtosecond timescale (i.e., nearly instantaneously), effectively eliminating the material removal time of the depth profile. Additionally, unlike other material removal techniques such as atomic ion beam impact (where a collision cascade effect is induced to remove surface material) fs-LA is capable of removing material without chemical damage due to the ultrashort columbic explosion process used to eject the material. The resulting profile is therefore representative of the actual sample chemistry.</p><h4><strong>Paint layer analysis&nbsp;</strong></h4><p>In traditional ion-beam depth profiling, analytical depths are generally limited to several hundred nanometers, up to a micrometer. This means that it is extremely difficult to analyze the interfaces between multiple micrometer-thick layers in a sample without the use of additional techniques such as ultralow-angle microtome (ULAM) sectioning.¹ The introduction of fs-LA technology, however, allows micrometers of material to be removed rapidly, enabling the characterization of layered samples.&nbsp;</p><p>As an example, a multi-layer paint specimen (provided by the University of Surrey, UK) was depth profiled using the <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/products/2206"><strong>Thermo Scientific™ Hypulse™ Surface Analysis System</strong></a>. The sample consists of an aluminum substrate coated with a 5 µm layer of polyurethane primer followed by a 25 µm PMMA/PVdF topcoat.</p><p>The depth profile was generated using a 1,030 nm laser with an energy of 250 µJ for ablation, along with a 30 µm X-ray spot size for data collection. XPS spectra were collected after each ablation cycle to obtain an atomic concentration plot through both paint layers into the substrate material (Figure 2).&nbsp;</p><p>5 distinct regions throughout the analyzed depth can be seen. The top surface of the sample consists of an adventitious layer of carbon contamination, which is commonly found on samples exposed to air. The second layer is the 25 µm PVdF/PMMA topcoat, and the third layer is the PU primer. Between the primer and the final aluminum substrate layer, a clear increase in adventitious carbon is observed, similar to the signal seen at the top sample surface. This suggests residual carbon contamination was present on the substrate before the paint layers were applied.&nbsp;</p><p>It would take hours to produce the XPS depth profile shown in Figure 2 using traditional ion beam approaches. Even with the use of a monatomic beam with a high energy per atom, the length of the analysis could still be just as long, depending on the sample material, and would sacrifice the chemistry of any organic/polymeric materials. The combination of fs-LA depth profiling and snapshot XPS acquisition, meanwhile, can reduce the experimental time to less than 20 minutes without inducing any chemical damage to the sample surface.</p><h4><strong>Summary&nbsp;</strong></h4><p>One of the most critical aspects of successful polymer XPS depth profiling is the preservation of the sample chemistry during material removal. Additionally, experiment time can be a significant limiting factor, and thicker samples could lead to reduced time for repeat experiments. The femtosecond laser ablation capabilities of the Hypulse Surface Analysis System provide fast, efficient, and chemical-damage-free material removal from a sample surface, enabling analysis multiple micrometers into a sample. This means that hidden sample interfaces, that were largely inaccessible before, can now be analyzed easily and efficiently</p>

<p><strong>This study presents a high-temperature GC-APCI-MS/MS method for detecting large polycyclic aromatic hydrocarbons in pyroplastics and environmental samples. The approach enables sensitive analysis of PAHs with molecular weights from 314–424 Da without extensive cleanup or fractionation.</strong></p><p>Application to pyroplastics from the M/V X-Press Pearl fire revealed substantially elevated levels of large PAHs compared to unburnt plastics. Formats and hyperlinks were adapted from the original.</strong></p><div><p>The measurement and reporting of polycyclic aromatic hydrocarbons (PAHs) are routine practices in environmental monitoring and risk assessment. <a href="javascript:void(0);">(1−3)</a> However, of the thousands of PAHs, only the 16 designated as Priority Pollutants by the U.S. Environmental Protection Agency (EPA16) are typically reported. <a href="javascript:void(0);">(4,5)</a> These span two to six ringed parent PAHs, ranging in molecular weight from 128 to 278 Da. It is well recognized that this limited analyte list drastically simplifies and misses a significant fraction of PAHs. <a href="javascript:void(0);">(4)</a> For example, the concentration of C<sub>1</sub>–C<sub>4</sub> alkylated PAHs can be greater than or equal to the concentration of the EPA16 in crude oils and fuels. <a href="javascript:void(0);">(6)</a> Expanding the analytical window of PAHs has proven valuable for forensic analysis of oil spills (e.g., alkylated and heterocycle-containing PAHs) and relevant for toxicological evaluation(e.g., oxygen-, nitrogen-, and sulfur-containing PAHs). <a href="javascript:void(0);">(3,7)</a> Moreover, the sources of PAHs have distinct compositions of PAHs, <a href="javascript:void(0);">(1,8)</a> which enables the use of PAHs for source apportionment. <a href="javascript:void(0);">(2,7)</a> Including alkylated and sulfur-containing PAHs (those beyond the EPA16) in the investigation of environmental samples and other matrices provides greater discriminating power between potential sources (using extracted ion chromatograms). <a href="javascript:void(0);">(3,7,9)</a> Additionally, monitoring the compositional changes of PAHs in the environment can inform about the fate and environmental processing of the matrix (e.g., oil weathering). <a href="javascript:void(0);">(2)</a> While these efforts have made strides in considering other PAHs, an environmentally relevant fraction of PAHs remains underutilized.</p><p>PAHs with more than six rings have been reported in diverse environmental matrices. <a href="javascript:void(0);">(10,11)</a> These large PAHs (≥24 ringed-carbons <a href="javascript:void(0);">(10)</a>) have been found in combustion-derived particulate matter, <a href="javascript:void(0);">(12−19)</a> urban dust, <a href="javascript:void(0);">(20−23)</a> refinery deposits, <a href="javascript:void(0);">(11)</a> coal tars and pitches, <a href="javascript:void(0);">(24−27)</a> cokes, <a href="javascript:void(0);">(28)</a> soils, <a href="javascript:void(0);">(20,22)</a> sediments, <a href="javascript:void(0);">(20−23)</a> and deep-sea hydrothermal vent bitumen. <a href="javascript:void(0);">(29)</a> Most reports of large PAHs in environmental matrices have been from analyses of U.S. National Institute of Standards and Technology (NIST) standard reference materials (SRMs); <a href="javascript:void(0);">(18,21−23,25,26,30)</a> a few 302 Da isomers have been certified in three SRM. <a href="javascript:void(0);">(31)</a> Nonetheless, isomers ranging from 326 to 374 Da have been reported in SRMs for urban dust (1649a/b), diesel particulate (1650b and 2975), coal tar (1597a), and marine sediment (1941b), displaying qualitative differences between the sources in their large PAH compositions; <a href="javascript:void(0);">(18,20,22,23,27)</a> thus, large PAHs should offer forensic utility.</p><p>Recent advances in soft ionization within an atmospheric pressure chemical ionization source for gas chromatography–mass spectrometry (GC-APCI) are well-suited for the analysis of large PAHs. Two contributing factors for this are the tolerance for high carrier gas flow rates by GC-APCI and the sensitivity of charge exchange APCI for aromatic species. <a href="javascript:void(0);">(48)</a> Increasing the carrier gas flow rate reduces the elution temperature of analytes, making many higher mass analytes accessible using a conventional GC and column, although poor thermal stability may still pose challenges for some analytes. <a href="javascript:void(0);">(49)</a> Classic vacuum-source GC/MS systems using electron ionization (EI) restrict the upper carrier gas flow allowed from the GC, and therefore, cannot fully utilize high GC carrier gas flows. The sensitivity of GC-APCI for components of petroleum, such as PAHs, has been previously reported. <a href="javascript:void(0);">(48)</a> Furthermore, unlike LC-APCI, which commonly exhibits protonation of analytes in positive ion mode, <a href="javascript:void(0);">(50)</a> GC-APCI+ can operate with dry source conditions. These conditions result in nitrogen-mediated charge exchange ionization and ions of the form M+<strong><sup>•</sup></strong>, the same form of molecular ion created using EI. As a result, because the charge site location on an ion dictates which product ions will be stable, the same multiple reaction monitoring (MRM) transitions can be used for both GC-APCI MS/MS and EI MS/MS. <a href="javascript:void(0);">(51)</a></p><p>Herein, a GC-APCI method was developed for the detection of large PAHs. Due to the limited availability of pure standards, previously characterized NIST SRMs were used in method development as sources of large PAHs. The values of the method and detection of large PAHs were demonstrated by analyzing environmental plastic samples collected after the M/V <i>X-Press Pearl</i> ship fire and plastic spill.</p><h4><strong>Materials and Methods</strong></h4><h5><strong>GC-APCI</strong></h5><p>The solvent extracts and dilutions were analyzed by GC-APCI on a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/554"><strong>Xevo TQ Absolute</strong></a> (Waters Corporation) tandem quadrupole mass spectrometer (TQ-MS/MS). The atmospheric pressure gas chromatography (APGC) ionization source was operated in charge transfer mode using dry N<sub>2</sub> as the reagent gas. Dry N<sub>2</sub> reagent gas creates molecular ions of the form M+<strong><sup>•</sup></strong> with no significant adduct formation for large PAHs. The high reagent gas flow rate of 350 mL/min into the ionization chamber, combined with low energies applied to the transfer optics from the atmospheric pressure region to the first quadrupole, leads to a high molecular ion survival rate. Furthermore, the use of MS/MS with quadrupoles operating at unit mass resolution prevents potential effects on response caused by adduct formation. Acquisitions were performed in positive ion, MRM mode. Two MRM transitions were used for each of the 16 precursor masses, ranging from 314 to 424 Da (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acsomega.5c11703/suppl_file/ao5c11703_si_001.pdf">Table S1</a>). No optimization of source conditions specific to the analysis of large PAHs was required. The list of 16 specific precursor masses targeted in this range was developed using multiple sources reporting large PAHs in NIST SRMs and samples from deep-sea hydrothermal vents. <a href="javascript:void(0);">(18,29,30)</a> The two transitions represented constant neutral losses of 2 and 4 Da at high collision energies of 60–100 eV, using N<sub>2</sub> as the collision gas.</p><p>For 1,3,5-TPB analysis, the APGC ionization source was operated in charge transfer mode using N<sub>2</sub> as the collision gas. Acquisitions were performed in positive ion, MRM mode, yielding six MRM transitions for analysis (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acsomega.5c11703/suppl_file/ao5c11703_si_001.pdf">Table S2</a>). Similar to the large PAHs, this analyte also shows neutral losses of 2 and 4 Da, reflecting behavior like that of the more condensed structures.</p><p>The <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8890+GC"><strong>8890 GC (Agilent Technologies)</strong></a> was configured with N<sub>2</sub> as the carrier gas and an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=7&amp;search=Rxi-5HT"><strong>Rxi-5HT column (Restek)</strong></a> of 15 m in length, 0.25 mm inner diameter, and 0.10 μm film thickness. The split/splitless (SSL) injection port was operated at a 10:1 split ratio with a temperature of 380 °C. A 1 μL injection volume was used for all analyses. The temperature program was 30.8 min; it started by holding at 40 °C for 0.5 min, then ramped to 160 °C at 14 °C/min, followed by a ramp to 395 °C at 22 °C/min and a hold at the temperature for 11 min. The flow rate of the N<sub>2</sub> carrier gas was initially 0.60 mL/min, then ramped at 0.015 mL/min<sup>2</sup> to 0.90 mL/min, followed by a ramp at 0.150 mL/min<sup>2</sup> to 3.0 mL/min. To adapt the system for high-temperature work, the SSL was configured with a high-temperature septum (400 °C maximum), a 100% graphite liner O-ring, and a straight 4 mm inner diameter, wool-packed liner (450 °C maximum). Additional instrument and method details are included in the <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acsomega.5c11703/suppl_file/ao5c11703_si_001.pdf">Supporting Information</a>.</p><h4><strong>Results</strong></h4><h5><strong>Analysis of Large PAHs in Pyroplastic Samples by GC-APCI</strong></h5><p>Macroplastic and microplastic debris from the M/V <i>X-Press Pearl</i> ship fire and plastic spill were investigated for the presence and distribution of large PAHs. The field samples included white, unburnt polyethylene nurdles and two types of pyroplastic, termed burnt plastic and combustion remnant (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acsomega.5c11703#fig1">Figure 1</a>), both of which were polyethylene. <a href="javascript:void(0);">(43)</a> The types of pyroplastics were previously operationally defined based on the size and shape of the pieces. <a href="javascript:void(0);">(43)</a></p><p>Investigation of the 302 Da isomers revealed profiles similar to those of the SRMs; however, unique differences were observed (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acsomega.5c11703/suppl_file/ao5c11703_si_001.pdf">Figure S2</a>). Eighteen to twenty-one peaks were detected across the plastic samples. Compared with the SRMs, the pyroplastics showed decreases in the relative abundance of dibenzo[<i>a</i>,<i>e</i>]fluoranthene and increases in dibenzo[<i>a</i>,<i>h</i>]pyrene (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acsomega.5c11703/suppl_file/ao5c11703_si_001.pdf">Figure S2</a>). This trend was not the case for the white nurdle sample. Starting at a retention time of 16.80 min, two peaks were unique to the pyroplastic samples (not present in the SRMs and especially minor in the white nurdles). These peaks followed that for dibenzo[<i>a</i>,<i>h</i>]pyrene, which had previously been reported as the last detected 302 Da isomer compound in the SRMs, <a href="javascript:void(0);">(21)</a> supporting their novelty as marker compounds of pyroplastics.</p><p>The plastic samples displayed similarities and differences in the presence and distribution of large PAHs (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acsomega.5c11703#fig3">Figure 3</a>). The white nurdles, burnt plastic, and combustion remnant all contained high-intensity peaks at 20.50 min for 398 Da and 19.55 min for 374 Da. Peaks between 20.00 and 20.25 min of 398 Da and the 19.35 min peak of 374 Da were reduced in intensity for the pyroplastics compared to the white nurdles (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acsomega.5c11703/suppl_file/ao5c11703_si_001.pdf">Figure S3</a>). These levels included the individual PAHs that were common to the pyroplastics. This trend mirrored that observed for smaller PAHs and the solvent-extractable content of the sample types. <a href="javascript:void(0);">(39,43,46)</a> There were also slight differences in the signatures of the large PAHs found in the pyroplastics. The 398 Da peak at 20.24 min, along with the 374 Da peak at 19.31 min and the 350 Da peak at 18.61 min, were more prominent in the combustion remnant compared to the burnt plastic (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acsomega.5c11703/suppl_file/ao5c11703_si_001.pdf">Figure S3</a>).</p><p>Similar to the differences in large PAH signatures between the SRMs, the plastic samples had different signatures. Such qualitative interpretation of extracted ion chromatograms is forensically accepted for distinguishing sources. <a href="javascript:void(0);">(3)</a> This feature can potentially be utilized to differentiate between plastic types (e.g., virgin, weathered, and partially combusted) as well as to identify sources of the large PAHs and the combustion temperature.</p><h4><strong>Conclusions</strong></h4><p>GC-APCI-MS/MS proved a useful technique for detecting large PAHs in diverse environmental matrices, including extracts from pyroplastic field samples. Despite a lack of widely available analytical standards for large PAHs, NIST SRMs provided ample material for method development for their detection. Nonetheless, with the increasing research on microplastics in the environment and their relevance to human health, an environmental SRM with certified values for large PAHs is needed, and pyroplastics may provide such a source of material. Large PAHs and 1,3,5-TPB were detected in pyroplastic field samples and hold promise as chemical markers for pyroplastics, complementing other means for their detection in environmental samples (e.g., appearance and physical properties). Due to the accessibility of GC-APCI, the detection of large PAHs can be integrated into workflows for evaluating environmental samples, including those for microplastics.</p></div>

Gas Chromatography-Atmospheric Pressure Chemical Ionization (GC-APCI) Expands the Analytical Window for Detection of Large PAHs (≥24 Ringed-Carbons) in Pyroplastics and Other Environmental Matrices

<p><strong>This study investigates the pathogenesis of neurosyphilis through cerebrospinal fluid proteomics using LC–MS/MS and ELISA validation. Comparative analysis identified 234 differentially expressed proteins, with major alterations linked to lysosomal dysfunction, axon guidance, and neurodegeneration pathways.</strong></p><p>Key lysosomal proteins, including PSAP and DNASE2, together with the axonal protein EPHA4, were significantly downregulated in neurosyphilis patients. Formats and hyperlinks were adapted from the original.</strong></p><div><p>Neurosyphilis (NS) is a group of clinical syndromes characterized by neuroinflammation caused by <i>Treponema pallidum</i> (<i>T. pallidum</i>), which occurs in all stages of syphilis. <a href="javascript:void(0);">(1,2)</a> NS is classified based on clinical presentation as either asymptomatic (ANS) or symptomatic (SNS), and based on disease stage as either early (occurring 1–2 years after primary infection) or late. <a href="javascript:void(0);">(1)</a> <i>T. pallidum</i> can cause local nonspecific and central nervous system (CNS) inflammation, leading to possible degenerative changes. <a href="javascript:void(0);">(3)</a> The immune response of CNS in neurosyphilis is comparable to that observed in other neuroinflammatory diseases. Consequently, the presence of neurological symptoms may not reliably indicate active CNS inflammation in syphilis patients. <a href="javascript:void(0);">(3)</a> A cerebrospinal fluid (CSF) proteomic study identified three biomarkers (SEMA7A, SERPINA3, and ITIH4) that contribute to NS and have potential implications for diagnosis and therapeutic strategies. <a href="javascript:void(0);">(4)</a> Through integrated proteomics and single-cell transcriptomics analyses, Toll-like/NF-κB signaling was identified as the key pathway involved in CNS damage caused by <i>T. pallidum</i>. <a href="javascript:void(0);">(5)</a> Despite an increasing understanding of the biomarkers and possible neuroinflammatory damage mechanisms of neurosyphilis, the mechanisms of neurodegeneration caused by <i>T. pallidum</i> infection remain elusive. <a href="javascript:void(0);">(6)</a></p><p>Patients with neurosyphilis exhibit stage-specific profiles of soluble factors in their CSF, which are linked to both immune status and the extent of neuronal damage. <a href="javascript:void(0);">(7)</a> In this study, we analyzed differentially expressed proteins (DEPs) in the CSF proteomes from 28 individuals, including neurosyphilis (NS), suspected neurosyphilis (spNS) and non-neurosyphilis (nNS) individuals, by LC–MS/MS and analyzed a public data set of the neurosyphilis CSF proteome. Validation was then performed by ELISA in an independent group comprising 110 samples. The results revealed that neuroinflammation by <i>T. pallidum</i> infection results in dysregulation of the key proteins in lysosomal and axonal pathway, which suggesting a potential contribution to neurodegeneration in neurosyphilis.</p><h4><strong>Materials and Methods</strong></h4><h5><strong>LC–MS/MS</strong></h5><p>The tryptic peptides were dissolved in 0.1% formic acid (solvent A) and loaded onto a custom-made reversed-phase column (15 cm × 75 μm). Using an <strong>EASY-nLC 1000 UPLC system</strong>, a gradient was run at 400 nL/min as follows: 6% to 23% solvent B (0.1% formic acid in 98% acetonitrile) over 26 min, 23% to 35% in 8 min, up to 80% in 3 min, and held at 80% for 3 min. Peptides were ionized via an NSI source at 2.0 kV and analyzed online by a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/559"><strong>Q Exactive Plus mass spectrometer</strong></a><strong>.</strong> Full scans (<i>m</i>/<i>z</i> 350–1800) were acquired in the Orbitrap at 70,000 resolution. The top precursors were selected for MS/MS with an NCE of 28, and fragments were detected at 17,500 resolution. A data-dependent method cycled between one MS scan and 20 MS/MS scans with a 15.0 s dynamic exclusion. AGC was set at 5E4. MS/MS data were processed with MaxQuant (v.1.5.2.8) using Trypsin/P as the enzyme (up to 2 missed cleavages). Precursor mass tolerance was 20 ppm (first search) and 5 ppm (main search); fragment mass tolerance was 0.02 Da. Fixed modification: carbamidomethyl (C); variable modification: oxidation (M). The FDR threshold was &lt;1%, and the minimum peptide score was &gt;40.</p><h5><strong>ELISA</strong></h5><p>CSF levels of host proteins were quantified using commercial ELISA kits according to the manufacturers’ protocols. Proteins analyzed included PSAP (OKEH07124, Aviva, USA), CTSL (abx150963, Abbexa, UK), NPC2 (A-QEK07857, Biogradetech, USA), and DNASE2 (abx386953, Abbexa, UK). Briefly, 100 μL of CSF was added to precoated plates and incubated at 37 °C for 2 h. After washing, 100 μL of detection antibody was added to each well and incubated at 37 °C for another 2 h. Following a wash, 100 μL of streptavidin-HRP working solution was added and incubated at room temperature for 20 min in the dark. The reaction was stopped with 50 μL of stop solution, and the optical density was measured at 450 nm.</p><h4><strong>Results</strong></h4><h5><strong>Downregulation of Key Lysosomal Proteins in CSF from Patients With Neurosyphilis</strong></h5><p>By analyzing the DEPs in NS vs nNS, we observed a significant enrichment of downregulated proteins in the lysosome pathway (<i>P</i> = 0.007) (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.jproteome.5c00689#fig2">Figure 2</a>A), with 9 lysosomal proteins showing significant downregulation (<i>P</i><sub>adj</sub> &lt; 0.05) in the NS group (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acs.jproteome.5c00689/suppl_file/pr5c00689_si_001.zip">Table S4</a>). To validate this finding, we examined the public CSF proteomics data set of neurosyphilis (PXD033034) and identified 18 proteins in the lysosomal pathway that were significantly downregulated in patients with NS (<i>P</i><sub>adj</sub> &lt; 0.05) (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acs.jproteome.5c00689/suppl_file/pr5c00689_si_001.zip">Table S4</a>). PPI network analysis determined the top 10 hub genes within the lysosome pathway (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.jproteome.5c00689#fig2">Figure 2</a>B). A Venn diagram revealed 3 proteins (PSAP, CTSL, and NPC2) shared across our current data, the public data set (PXD033034) and the top 10 hub genes (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.jproteome.5c00689#fig2">Figure 2</a>C). The mass spectrometry results indicated that the levels of PSAP (<i>P</i><sub>adj</sub> = 0.043), CTSL (<i>P</i><sub>adj</sub> = 0.028) and NPC2 (<i>P</i><sub>adj</sub> = 0.013) were significantly lower in the NS group compared to the nNS group (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.jproteome.5c00689#fig2">Figure 2</a>D). The PXD033034 data set revealed consistent changes in the PSAP, CTSL, and NPC2 levels in the NS group (<i>P</i><sub>adj</sub> &lt; 0.05) (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acs.jproteome.5c00689/suppl_file/pr5c00689_si_001.zip">Figure S1 and Table S4</a>). In addition, associations between the lysosomal protein DNASE2 and NS were observed in both the present study (<i>P</i><sub>adj</sub> = 0.003) (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.jproteome.5c00689#fig2">Figure 2</a>D) and the PXD033034 data set (<i>P</i><sub>adj</sub> &lt; 0.05) (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acs.jproteome.5c00689/suppl_file/pr5c00689_si_001.zip">Figure S1 and Table S4</a>), with subgroup analysis revealing a significantly lower expression level of DNASE2 in the SNS group compared to the ANS group (<i>P</i> = 0.035) (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.jproteome.5c00689#fig2">Figure 2</a>D and <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acs.jproteome.5c00689/suppl_file/pr5c00689_si_001.zip">Table S3</a>). Further validation by ELISA (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acs.jproteome.5c00689/suppl_file/pr5c00689_si_001.zip">Table S5</a>) in an independent group (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.jproteome.5c00689#tbl1">Table 1</a>) confirmed significantly downregulated CSF levels of PSAP (<i>P</i> &lt; 0.0001), CTSL (<i>P</i> &lt; 0.0001), NPC2 (<i>P</i> = 0.0024) and DNASE2 (<i>P</i> &lt; 0.0001) in patients with NS (n = 21) than in nNS individuals (n = 56) (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.jproteome.5c00689#fig2">Figure 2</a>E) (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acs.jproteome.5c00689/suppl_file/pr5c00689_si_001.zip">Table S6</a>), as well as decreased in the SNS subgroup (<i>P</i> &lt; 0.05) (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acs.jproteome.5c00689/suppl_file/pr5c00689_si_001.zip">Figure S2 and Table S6</a>). Receiver operating characteristic (ROC) curve analysis supported PSAP (AUC = 0.91, 95% CI: 0.83–0.99) and DNASE2 (AUC = 0.95, 95% CI: 0.91–0.99) for distinguishing NS from nNS and as potential biomarkers for assessing neurodegeneration in NS (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.jproteome.5c00689#fig2">Figure 2</a>F). Our proteomic data revealed a total of 24 downregulated (<i>P</i><sub>adj</sub> &lt; 0.05) axonal proteins and 1 upregulated (<i>P</i><sub>adj</sub> &lt; 0.05) axonal protein in the CSF of patients with NS (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acs.jproteome.5c00689/suppl_file/pr5c00689_si_001.zip">Table S9</a>). Furthermore, the PXD033034 data set revealed 55 downregulated (<i>P</i><sub>adj</sub> &lt; 0.05) and 4 upregulated (<i>P</i><sub>adj</sub> &lt; 0.05) axonal proteins in NS (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acs.jproteome.5c00689/suppl_file/pr5c00689_si_001.zip">Table S9</a>). The top 10 hub genes involved in the biological processes of axon development and axonogenesis were identified by PPI network analysis (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.jproteome.5c00689#fig4">Figure 4</a>A). Considering the overlap of our current data, the PXD033034 data set and the top 10 hub genes (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.jproteome.5c00689#fig4">Figure 4</a>B), one key protein, EPHA4, was identified across 3 data sets, with significant downregulation in our current data (<i>P<sub>adj</sub></i> = 0.011) and in the PXD033034 data set (<i>P<sub>adj</sub></i> = 0.028) (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.jproteome.5c00689#fig4">Figure 4</a>C). Correlation analysis further revealed a positive correlation between the levels of PSAP and EPHA4 (<i>r</i> = i 0.83, <i>P</i> &lt; 0.05) (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.jproteome.5c00689#fig4">Figure 4</a>D). Reduced levels of PSAP and EPHA4 in CSF have been implicated in neurodegenerative diseases, including Alzheimer’s disease (AD) and Parkinson’s disease (PD). <a href="javascript:void(0);">(11,12)</a> Our findings suggest that coordinated dysregulation of lysosomal and axonal proteins may contribute to neurodegeneration in NS.</p><p><img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/J_Proteome_Res_2026_25_2_713_722_Figure_4_Key_axonal_protein_downregulated_in_CSF_from_patients_with_neurosyphilis_66f5533e94.jpg" alt="J. First, the use of ELISA to validate a limited number of proteins has certain limitations, such as the inherent cross-reactivity of ELISA. In the future, we can optimize the validation experiments by adding techniques such as Western blotting for verification. Second, this study uses a cross-sectional sampling method for research, which limits our ability to infer disease progression and treatment responses. In the future, we will conduct follow-up on the subjects, continue collecting cerebrospinal fluid samples after treatment, and observe whether the protein dysregulation is reversible after treatment. For spNS patients, follow-up should also be conducted to determine if they have transformed into NS, which will be very valuable for validating the predictive utility of our biomarkers. Finally, although multiple discovery and validation cohort studies have been conducted, we currently lack direct mechanistic evidence. Future work will investigate whether exposing neural cell coculture models to outer-membrane antigens of <i>T. pallidum</i> reproduces the lysosome and axon alterations observed in CSF, clarifying their role in neural damage of neurosyphilis and potential biomarkers for diagnosis and treatment of neurodegeneration in neurosyphilis.</p></div>

Cerebrospinal Fluid Proteome Reveals Dysregulation of Lysosomal and Axonal Proteins in Neurosyphilis

<p><strong>This study examines the combustion behavior of B4C/Bi2O3 pyrotechnic delay compositions as sustainable alternatives to conventional materials. Thermodynamic modeling and experimental testing were used to evaluate the influence of compaction pressure, particle size, and granule size on burning performance.</strong></p><p>Results showed that increased compaction reduced burning rates by lowering porosity and shifting combustion from convective to conductive mechanisms. These compositions are essential in timing energetic events and are used extensively in both civilian and military applications. <a href="javascript:void(0);">(1)</a> This broad usage leads to diverse motivations for studying such materials. In civilian contexts, such as mining, the focus is typically on safety and cost-effectiveness. <a href="javascript:void(0);">(2)</a> For military purposes, reliability and precision are key factors, often with stringent performance requirements. <a href="javascript:void(0);">(3)</a></p><p>Pyrotechnic delay compositions are classified based on their combustion behavior into gassy or gasless types. In confined, sealed and obturated environments, gasless compositions are favored, to avoid gas buildup. <a href="javascript:void(0);">(1,4)</a> However, in unconfined or vented systems, gassy compositions are not problematic. For example, in fireworks, where combustion gases freely dissipate, <a href="javascript:void(0);">(1)</a> or in vented housings like hand grenades or signaling devices, gassy delays are acceptable. <a href="javascript:void(0);">(5,6)</a></p><p>In gassy compositions, the burning rate is influenced by the loading (or consolidation) pressure. Hot gases generated by the chemical reaction penetrate gas channels (voids) in the material, igniting unreacted material through a process known as convective heating. As loading pressure increases, these voids are gradually reduced, which diminishes convection and subsequently lowers the burning rate. In contrast, for gasless compositions, burning is driven by conductive heating, where heat is transferred through direct particle-to-particle contact, leading to the opposite effect. <a href="javascript:void(0);">(7−9)</a></p><p>Historically, up until World War II, black powder served as the foundation for most delay elements. Nowadays, it continues to be utilized for this function and others, but on smaller scales. <a href="javascript:void(0);">(1)</a> Some more recent and traditional formulations for pyrotechnic compositions include fuels like silicon, boron, tungsten, zinc, iron, zirconium, and manganese. Examples of these are Si/Pb3O<sub>4</sub>, <a href="javascript:void(0);">(8,10)</a> B/BaCrO<sub>4</sub> <a href="javascript:void(0);">(8)</a>, W/BaCrO<sub>4</sub>/KClO<sub>4</sub>, <a href="javascript:void(0);">(5,6,11)</a> Zn/PbO<sub>2</sub>, <a href="javascript:void(0);">(12)</a> Fe/BaO<sub>2</sub>, <a href="javascript:void(0);">(13)</a> Zr/KClO<sub>4</sub>, <a href="javascript:void(0);">(8)</a> and Mn/PbCrO<sub>4</sub>/BaCrO<sub>4</sub>. <a href="javascript:void(0);">(14)</a></p><p>Currently, researchers are actively developing several new formulations for pyrotechnic delays. One of the main motivations for this is the need to transform these compositions into “greener” ones. <a href="javascript:void(0);">(1,2)</a> This means that compositions that could cause problems for the environment or human health should be avoided. To achieve this sustainability, the identification and replacement of known or potentially harmful, toxic, and carcinogenic compounds is vital. <a href="javascript:void(0);">(10)</a> Heavy metals, persistent organic pollutants, toxic gases, volatile organic compounds and known causers of human health problems are examples of such substances to be removed. <a href="javascript:void(0);">(2,5,15)</a></p><p>Specifically, there are several attempts to replace lead, barium, chromates, and perchlorates. <a href="javascript:void(0);">(15)</a> The reason is that most of the established delay formulations used today contain these substances. <a href="javascript:void(0);">(2,5)</a> Lead, barium and chromium (contained in chromates) are heavy metals and are known to cause serious health problems in human, specially to nervous, cardiovascular and respiratory systems. <a href="javascript:void(0);">(5,16)</a> Perchlorates are well-documented substances and are used in various pyrotechnic applications. However, they are widely recognized as contributors to thyroid issues. <a href="javascript:void(0);">(5)</a></p><p>From the above, it is clear that modern strategies in pyrotechnics are increasingly focused on developing new materials that prioritize both efficiency and sustainability. This shift imposes numerous restrictions, meaning that formulations must be designed with precise planning and thorough investigation to meet both performance and environmental standards.</p><p>Boron could be considered the ideal fuel for pyrotechnic reactions due to its high energy content. However, it presents combustion issues for its limited reactivity due to boron oxide layer and boron agglomerates formation upon heating. <a href="javascript:void(0);">(17)</a> The boron oxide layer acts as a barrier, hindering oxygen (O) diffusion and consequently limiting the oxidation of boron. <a href="javascript:void(0);">(18)</a></p><p>Boron carbide (B<sub>4</sub>C) is a natural candidate for combustion processes due to its lower ignition point, which is attributed to its higher reactivity. This reactivity stems from the diffusion of carbon (C) particles, which oxidize into gaseous products. These gaseous products create diffusion channels that penetrate the boron oxide layer, breaking down the layer and agglomerates, thereby further improving the reaction. <a href="javascript:void(0);">(18,19)</a> B<sub>4</sub>C presents better combustion properties, with better burning stability and reliable self-propagation, resulting in better combustion efficiency. <a href="javascript:void(0);">(17)</a></p><p>Bismuth trioxide (Bi<sub>2</sub>O<sub>3</sub>) is an attractive oxidizer for pyrotechnic delay compositions due to several advantageous properties. Its high oxygen content and efficient oxygen release facilitate sustained combustion, while its favorable thermal properties, including a relatively low melting and decomposition temperature, ensure that oxygen is available at precisely the right moment to support a controlled reaction. <a href="javascript:void(0);">(20)</a> Additionally, Bi<sub>2</sub>O<sub>3</sub> high density allows for compact formulations, promoting consistent and reliable burn rates. Also, it has a low heat of formation (approximately −582 kJ/mol), which is crucial to minimize the energy required to initiate its decomposition. This enhances the overall reaction efficiency, allowing Bi<sub>2</sub>O<sub>3</sub> to rapidly release oxygen without consuming excessive energy, thus ensuring sustainable and stable combustion. <a href="javascript:void(0);">(8)</a></p><p>In addition to their technical advantages, Bi<sub>2</sub>O<sub>3</sub> and B<sub>4</sub>C offer a significant environmental benefit: they are considered more eco-friendly compared to other materials commonly used in pyrotechnic compositions. Bi<sub>2</sub>O<sub>3</sub> serves as a less toxic alternative to traditional oxidizers like lead-based compounds, reducing the risk of environmental contamination and exposure to harmful heavy metals. <a href="javascript:void(0);">(1,8,16)</a> Similarly, B<sub>4</sub>C contributes to cleaner combustion, producing fewer pollutants and minimizing the environmental impact. <a href="javascript:void(0);">(5)</a> This makes both materials not only effective but also greener choices for sustainable pyrotechnic applications.</p><h4><strong>2. Experimental Section</strong></h4><h5><strong>2.8. Material Characterization</strong></h5><p>Particle and granule size distribution were measured with a <strong>Malvern Mastersizer 3000</strong> (Malvern Panalytical). Scanning Electron Microscopy (SEM) was carried out with a <strong>TESCAN MIRA</strong> (TESCAN) to examine morphology and confirm particle size. A 40 cm<sup>3</sup> combustion chamber was used to analyze pressure profile, quantify solid residues, and collect combustion gases for further analysis. Fourier Transform Infrared Spectroscopy (FTIR) was performed using a <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/products/2392"><strong>Nicolet iS50 FT-IR</strong></a> (Thermo Scientific) and standardized nitrogen (N<sub>2</sub>) background to identify gas-phase products. Thermal stability and water content assessments were conducted using <strong>TGA</strong> (NETZSCH TG 209 F3 Tarsus) and <strong>DSC</strong> (NETZSCH DSC 200 F3 Maia). Energy Dispersive X-ray Spectroscopy (EDS) was performed using a <strong>Bruker Quantax 200</strong> (Bruker) for semiquantitative chemical composition analysis of the combustion residues. X-ray diffraction (XRD) analyses were performed using a Rigaku MiniFlex diffractometer for complementary phase identification, while surface chemical states were characterized by X-ray photoelectron spectroscopy (XPS) using a Scienta Omicron ESCA2SR system.</p><h4><strong>3. Results and Discussion</strong></h4><h5><strong>3.7. FTIR Analysis of Combustion Gases</strong></h5><p><a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acsomega.5c10964#fig10">Figure 10</a> compares the FTIR spectra of combustion chamber gases from stoichiometric (blue) and fuel-rich (red) tests, highlighting CO and CO<sub>2</sub> as the main species, consistent with Horiuchi. <a href="javascript:void(0);">(18)</a> Normalizing their absorbance to concentration using standard references yielded CO<sub>2</sub>/CO ratios of 0.67 (stoichiometric) and 0.34 (fuel-rich). However, a quantitative analysis is approximate and inherently imprecise, as potential contamination during gas collection and measurement could affect the results, given the challenge of maintaining a completely controlled internal atmosphere. A small fraction of hydrocarbon species (e.g., CH<sub>4</sub>, C<sub>2</sub>H<sub>4</sub>, and C<sub>2</sub>H<sub>2</sub>) was also identified in both samples.</p><p><img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/ACS_Omega_2026_11_3_4668_4683_Figure_10_Comparative_FTIR_spectra_of_combustion_gas_compositions_in_a_combustion_chamber_ac7b7daf84.jpg" alt="ACS Omega 2026, 11, 3, 4668–4683: Figure 10. Notably, the identified gaseous products are relatively simple and exhibit low toxicity compared to halogenated or metal-containing species commonly found in traditional pyrotechnic formulations, supporting the potential of this system as a more environmentally friendly alternative.</p><p>Since the reaction involves both condensed-phase reactants and products, characterizing the solid residues is essential to evaluate the extent of reaction and gain insight into the actual progression of the reaction pathway. For this, SEM imaging and EDS analysis were employed to investigate the morphology and elemental composition of the combustion residues.</p><h5><strong>3.8. SEM and EDS of Burning Residues</strong></h5><p><a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acsomega.5c10964#fig11">Figure 11</a> presents the SEM analysis of solid residues from the combustion of B4. <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acsomega.5c10964/suppl_file/ao5c10964_si_001.pdf">Figure S5</a> exhibits images of the other samples. <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acsomega.5c10964#fig11">Figure 11</a>a shows the top view of the burned delay element, revealing the distribution of residues across the surface, while <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acsomega.5c10964#fig11">Figure 11</a>b provides a magnified view of a region rich in bright spherical structures. These spheres are likely bismuth that transitioned through a liquid or gaseous phase during the reaction and solidified upon cooling. This phenomenon of spherical formation due to phase transition is well-documented in the literature. <a href="javascript:void(0);">(33)</a></p><p><img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/ACS_Omega_2026_11_3_4668_4683_Figure_11_SEM_analysis_of_B4_after_burning_8fc7a8f520.jpg" alt="ACS Omega 2026, 11, 3, 4668–4683: Figure 11. Surface Chemistry of Combustion Residues (XPS)</strong></h5><p>Survey spectrum scans identified the main photoelectron lines corresponding to carbon, oxygen, boron, and bismuth. High-resolution scans for the O 1s, C 1s, B 1s, and Bi 4f regions of B4 are presented in <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acsomega.5c10964#fig19">Figure 19</a>, allowing detailed peak deconvolution and chemical assignment.</p><p><img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/ACS_Omega_2026_11_3_4668_4683_Figure_19_XPS_high_resolution_spectra_of_O_1s_C_1s_B_1s_and_Bi_4f_regions_for_B4_5c6064d013.jpg" alt="ACS Omega 2026, 11, 3, 4668–4683: Figure 19. XPS high-resolution spectra of O 1s, C 1s, B 1s, and Bi 4f regions for B4." srcset="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/thumbnail_ACS_Omega_2026_11_3_4668_4683_Figure_19_XPS_high_resolution_spectra_of_O_1s_C_1s_B_1s_and_Bi_4f_regions_for_B4_5c6064d013.jpg 214w," sizes="100vw" width="2049" height="1498"></p><p>The analysis confirmed B<sub>2</sub>O<sub>3</sub> as a major surface product and indicated the presence of unreacted B<sub>4</sub>C in the B4 sample through B–C bonds. sp<sup>2</sup> carbon was also detected in B4, suggesting possible graphite formation, as predicted in thermodynamic calculations. A2 showed more Bi<sup>3+</sup> than B4, supporting that the stoichiometric composition had more residual Bi<sub>2</sub>O<sub>3</sub>, while the fuel-rich B4 sample promoted greater reduction to Bi<sup>0</sup>. The presence of Bi<sup>3+</sup> on the surface observed may result from postcombustion reoxidation of Bi<sup>0</sup>. These results reinforce the proposed reaction pathway and the role of fuel excess in enhancing reaction completeness. In this analysis, the B 1s spectrum further confirms the absence of BN by the lack of B–N peaks, consistent with the EDS results, which also revealed no detectable nitrogen signal.</p><p>All experimental findings contribute to the fundamental understanding of boron carbide and bismuth oxide reactions under both basic and applied conditions, particularly in delay compositions. These results provide new insights into the reaction pathway and the underlying combustion mechanism.</p><h4><strong>4. Conclusions</strong></h4><p>Stoichiometric formulations of B<sub>4</sub>C/Bi<sub>2</sub>O<sub>3</sub>, which ignited when not pressed, failed to burn after being lightly compacted; a fuel-rich composition is necessary to ensure combustion. The fuel-rich compositions ignited readily and maintained combustion without quenching. Water facilitated the granulation process and improved the final composition. With increasing loading pressure, burning rates decrease due to reduced convective heat transfer, leading the combustion process to transition toward a conductive regime. Particle size also impacted combustion, particularly with larger Bi<sub>2</sub>O<sub>3</sub> particles, which influenced the combustion profile up to a certain threshold. Additionally, the granule size of the final composition was found to alter the burning behavior, with larger granules promoting faster combustion at lower pressures.</p><p>Thermodynamic calculations provided an initial insight into the reaction process. The findings show that the reaction progresses through distinct stages: starting in a solid–solid preignition phase, transitioning to a solid–liquid phase as components melt, and culminating in a gas phase marked by significant product volatilization, illustrating a complete combustion evolution.</p><p>FTIR analysis confirmed that the main gaseous products are CO and CO<sub>2</sub>, indicating reduced environmental toxicity. Elemental and phase analyses confirmed that the reaction followed the expected pathway, with Bi and B<sub>2</sub>O<sub>3</sub> as the predominant products. No Bi<sub>2</sub>O<sub>3</sub> was detected by XRD, indicating that the oxidizer was fully consumed in the bulk. Morphological analysis of the products suggests that, upon cooling, bismuth solidifies into small metallic spheres, while surface analysis indicates that B<sub>2</sub>O<sub>3</sub> remains predominantly in the outer layer. Altogether, the results are consistent and complementary, reinforcing the reliability and scientific rigor of the study.</p><p>All these observations accentuate the necessity of conducting experiments under application-specific conditions. These findings not only enhance our understanding of the combustion dynamics of low toxicity B<sub>4</sub>C/Bi<sub>2</sub>O<sub>3</sub> mixtures but also underscore their potential for practical applications requiring precise delay times and environmental sustainability. The successful use of water as a green solvent further highlights the viability of developing sustainable pyrotechnic compositions. This research advances the field by providing valuable insights into optimizing performance while minimizing environmental impact through careful material selection and process control.</p></div>
