Features

The Thermo Scientific Orbitrap Elite Hybrid Mass Spectrometer with ETD is a high-performance hybrid MS system combining a high-field Orbitrap™ mass analyzer with the Velos Pro dual-pressure linear ion trap, delivering exceptional resolution, sensitivity, and versatility for advanced research applications.

Key features

  • High-field Orbitrap analyzer for ultra-high resolution and mass accuracy
  • Resolving power >240,000 FWHM at m/z 400
  • Parallel MS and MSⁿ acquisition for increased analytical throughput
  • Multiple fragmentation techniques: CID, HCD, and ETD (Electron Transfer Dissociation)
  • Velos Pro ion trap (Generation II) with neutral blocking technology for improved robustness
  • Advanced signal processing and low-noise detection electronics

Ion sources and options

  • H-ESI II heated electrospray source (required), flow rates from 1 µL/min to <2,000 µL/min
  • NanoSpray Flex ion source for static and dynamic nanospray (50 nL/min–2 µL/min)
  • APCI/APPI compatibility for flow rates from 50 µL/min to 2 mL/min
  • ETD reagent ion source with dedicated nitrogen supply

Performance characteristics

  • Mass range: m/z 50–2,000 (standard), up to m/z 4,000
  • Resolution:
    • 60,000 FWHM at m/z 400 (4 Hz scan rate)
    • Maximum >240,000 FWHM at m/z 400
  • Mass accuracy:
    • <3 ppm RMS (external calibration)
    • <1 ppm RMS (internal calibration)
  • Dynamic range: >5,000 within a single scan
  • MSⁿ capability: up to MS¹⁰
  • ETD performance: >15% fragmentation efficiency (Angiotensin I)

Vacuum and hardware design

  • Differentially pumped vacuum system with split-flow turbomolecular pumps
  • Final operating pressure <2 × 10⁻¹⁰ Torr
  • Gas-filled C-Trap, axial-field HCD collision cell
  • Active temperature control of the Orbitrap analyzer (Peltier element)

Software and data system

  • Thermo Scientific Xcalibur instrument control and data processing
  • Data-dependent acquisition with intelligent decision tree logic
  • Compatible with Proteome Discoverer, ProSightPC, SIEVE, MetWorks, Mass Frontier, and others

Typical applications

  • Proteomics and PTM analysis
  • Metabolomics and lipidomics
  • Top-down and bottom-up protein characterization
  • High-resolution qualitative and quantitative workflows

A fault was detected - insufficient rotation of tmp in the etd part of the system (8%) and an error in the embedded PC in the FT part, probably a defect in the power supply and mainboard.
