Therefore, monitoring is of the utmost importance.&nbsp;</p><p>This application note describes the advantages of using an Agilent 1290 Infinity III Bio Online LC solution with Agilent Online LC Monitoring software. A bioreactor was interfaced with a Flownamics Seg-Flow S3 sampling system for aseptic withdrawal and an Agilent 1290 Infinity III Bio Online LC to make an hourly analysis.&nbsp;</p><p>This is a fully automated approach for taking sterile samples from a bioreactor and preparing or diluting them. In addition, the 1290 Infinity III Bio Online LC solution displays data in near real time, which is a crucial element for effective bioprocess&nbsp;monitoring.</p><h4><strong>Experimental&nbsp;</strong></h4><h5><strong>Equipment&nbsp;</strong></h5><p>The <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=10&amp;search=1290+Infinity+III+Bio"><strong>1290 Infinity III Bio </strong></a>Online LC comprised the following&nbsp;modules:&nbsp;</p><ul><li>Agilent 1290 Infinity III Bio Flexible Pump (G7131A) with an InfinityLab Quick Change Bio inline filter assembly (part&nbsp;number 5067-1607), 2.1 mm id, 0.2 µm pore size&nbsp;</li><li>Agilent 1290 Infinity III Bio Online Sample Manager (G3167B) with Sample Thermostat (option #101) clustered with Agilent 1290 Infinity Valve Drive (G1170A), featuring a reactor valve pod (part number 5067-6680) and Online LC Monitoring Software&nbsp;</li><li>Agilent 1290 Infinity III Multicolumn Thermostat (G7116B) equipped with an Agilent Quick Connect Bio Heat Exchanger Std. (G7116-60071)&nbsp;</li><li>Agilent 1290 Infinity III Diode Array Detector (DAD) (G7117B), equipped with a Bio Max-Light Cartridge Cell (G4212-6008), 10 mm, 1 µL&nbsp;</li><li>Agilent 1260 Infinity High-Performance Degasser (G4225A) is connected between S1 (transport solvent) solvent line and Agilent 1290 Infinity III Bio Online Sample Manager (G3167B)&nbsp;</li></ul><h5><strong>Software&nbsp;</strong></h5><ul><li>Agilent <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=10&amp;search=OpenLab+CDS"><strong>OpenLab CDS</strong></a> version 2.8 or later versions&nbsp;</li><li>Agilent Online LC Monitoring Software version 1.3 or later&nbsp;versions&nbsp;</li><li>Flownamics FlowWeb control&nbsp;software</li></ul><h5><strong>Instrumental configuration&nbsp;</strong></h5><p>The 1290 Infinity III Bio Online LC solution was used with a Seg-Flow interface for automatic sampling and online analysis (see Figure 1). The Seg-Flow interface was used to withdraw samples from a bioreactor through a Flownamics F-Series 310 FISP probe with a ceramic membrane filter (0.2&nbsp;µm pore size) from a 5 L bioreactor. The drawn samples were sent through the built-in sample collection cup on the Seg‑Flow to the LC and the samples were retained into vials. The collected samples were then automatically derivatized by using an injector program and subsequently analyzed. A more detailed description of the instrumental configuration has been published.<sup>3</sup></p><h4><strong>Conclusion&nbsp;</strong></h4><p>For online bioprocess monitoring, a fully automated setup consisting of a bioreactor, a Flownamics sampling system, and an Agilent 1290 Infinity III Bio Online LC solution was successfully established. This setup is capable of taking aseptic samples from a bioreactor, preparing, analyzing, and processing samples without the need for manual interaction. In addition, the added value of the Agilent Online LC Monitoring software was evident in the display method and the ability to track it quickly.</p><h3><strong>2. Shimadzu: Analysis of Oligonucleotide Impurities Using Single Quadrupole Mass Spectrometer</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_00974_en_c50bb67c7a.pdf"><strong>Full PDF for download</strong></a></li></ul><h4><strong>User Benefits</strong></h4><ul><li>Combination use of LCMS-2050 single quadrupole mass spectrometer and LabSolutions Insight Biologics provides comprehensive characterization of oligonucleotides and related impurities.</li><li>Purity calculations for target components can be performed even on poorly separated peaks by utilizing MS spectra simultaneously acquired with UV chromatogram.</li></ul><p>In recent years, oligonucleotide therapeutics have been paid attention as a novel drug discovery modality because of their rapid progress of development. During the synthesis of oligonucleotide therapeutics, impurities that have differing base chain lengths are generated along with the target oligonucleotide. To ensure the safety and efficacy of the drug, comprehensive detection and identification of these impurities are required.&nbsp;</p><p>Reversed phase ion-pair chromatography is often employed as a method to separate the target compound from co-existing impurities. However, it is often difficult to establish analytical conditions that provide compete separation of all impurities that are formed in synthesis process due to their similar structures and properties to the principal compound of active ingredient. Against this drawback, liquid chromatography-mass spectrometry (LC-MS) is commonly used for impurity detection and identification. For analysis of oligonucleotide impurities, quadrupole time-of-flight mass spectrometers are mainly used because of their high mass resolution and ability to perform sequence and structure estimations via MS/MS analysis. However, in quality control process, the use of a single quadrupole mass spectrometer is expected to be advantageous because of providing mass information with easy operation when impurity information is already known.&nbsp;</p><p>This article presents a simulated impurity analysis of synthetic oligonucleotide using the LCMS-2050 high-performance liquid chromatography-mass spectrometer and LabSolutions Insight Biologics.</p><h4><strong>Analytical conditions</strong></h4><p>The employed instrument setup was <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=1&amp;search=Nexera+XS"><strong>Nexera XS inert</strong></a> and <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=1&amp;search=LCMS-2050"><strong>LCMS-2050</strong></a>. HPLC conditions are shown in Table 2, and MS conditions are shown in Table 3. The conditions were set where n-1(5') co-eluted with FLP, while n-3(5') was separated (Fig. 1) to demonstrate impurity analysis performance.</p><h4><strong>Conclusion</strong></h4><p>The simulated sample consisting of mainly FLP and small amounts of n-1(5')/n-3(5') was analyzed using the LCMS-2050 and LabSolutions Insight Biologics software and obtained data were processed appropriately. Detection, identification, and quantitation at low concentrations were accomplished not only for n-3(5') well-separated from FLP but also for n-1(5') co-eluted with FLP. In case that establishing analytical conditions is difficult, Using this instrument setup and software will provide efficient quality confirmation of oligonucleotide therapeutics.</p><h3><strong>3. Thermo Fisher Scientific: Simultaneous Quantitation and Untargeted Discovery (SQUAD) workflow using multiple fragmentation techniques for quantitation and characterization of trace level nitrosamine in biological matrices</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/po_squad_workflow_multiple_fragmentation_techniques_po_en_94a5b27b12.pdf"><strong>Full PDF for download</strong></a></li></ul><p>N-Nitrosamines are a class of carcinogenic chemical compounds that pose threat to humans exogenously through matrices like drinking water, food, cosmetics, and endogenously by nitrosation in gastrointestinal tract, nitrate to nitrite conversion, etc. An accurate, reliable analytical method is critical for detection and quantification of these trace-level, polar and low molecular weight compounds to understand the association between their exposure and related diseases.&nbsp;</p><p>An optimized SQUAD workflow is presented that discovers unknown compounds in the matrix by using high resolution orbitrap and a sensitive linear ion trap detector that quantifies the low intensity nitrosamines in a single run. The workflow uses multiple fragmentation types – HCD, CID and UVPD and MSn to help characterize isomeric structures.</p><h4><strong>Methods</strong></h4><p>Serial dilution of Nitrosamine standards were subjected to Simultaneous Quantitation and Discovery (SQUAD) analysis using <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/744"><strong>Thermo Scientific™ Vanquish™ Horizon UHPLC system</strong></a> and the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/561"><strong>Thermo Scientific™ Orbitrap IQ-X™ Tribrid™ mass spectrometer</strong></a> . Nitrosamine spiked matrices of human pooled urine and human Serum were subjected to SQUAD analysis.</p><h4><strong>Data Analysis&nbsp;</strong></h4><p>Spectra were evaluated for nitrosamines using Thermo Scientific™ FreeStyle™ software and annotated using Thermo Scientific™ <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1757"><strong>Mass Frontier™ 8.0 software</strong></a>. Untargeted data analysis was performed using <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1759"><strong>Compound Discoverer 3.4 software</strong></a>. Calibration curves were generated using Thermo Scientific™ <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/77"><strong>TraceFinder™ 5.2 software</strong></a>.</p><h4><strong>Conclusions&nbsp;</strong></h4><ul><li>Detection limits as low ppb levels ( 0.2 ppb –0.5 ppb) and quantitation limits (0.2 ppb- 5 ppb ) in presence of the matrix were achieved for all seven nitrosamines by applying parallel ion trap analysis&nbsp;</li><li>SQUAD analysis was run on Orbitrap IQ-X Tribrid MS and achieved low ppb level quantitation in ion trap and identified unknown compounds through discovery analysis in Orbitrap.&nbsp;</li><li>Employing nano LC helped achieve lower limit of quantitation (5 ppt)</li></ul><h3><strong>4. Following increased public health awareness during 2019, VEA became a marker of interest for quality control and regulatory surveillance across cannabis supply chains due to its potential impact on lung health. In addition to concentrates and vape liquids, trace residues may be present in infused products and processing intermediates. Therefore, reliable quantification of VEA in these chemically diverse, lipid-rich matrices requires an analytical method that is sensitive, selective, and robust against matrix effects. In February 2024, Missouri’s DCR revised the tolerance limit for VEA in cannabis products from 0.2 ppm to 5 ppm.<sup>1&nbsp;</sup></p><p>Liquid chromatography coupled with tandem quadrupole mass spectrometry (LC-MS/MS) provides the sensitivity and specificity required to distinguish VEA from endogenous cannabinoids, terpenes, and other lipophilic excipients, while also offering the sensitivity to support low-ppm and sub-ppm action limits. Unlike UVbased methods, where co-eluting chromophores may interfere, LC-MS/MS enables targeted multiple reaction monitoring (MRM) for confident identification and accurate quantitation in complex cannabis matrices.&nbsp;</p><p>This application note describes the validation of an LC-MS/MS method for the quantitation of VEA in two inhalable products, cannabis flower and cannabis concentrates. The method was developed and validated using the Waters ACQUITY UPLC H-Class System coupled with a Xevo TQ-S micro Triple Quadrupole Mass Spectrometer. Chromatographic separation was achieved with a reversed-phase ACQUITY UPLC BEH C18 Column. A streamlined sample preparation approach was applied for cannabis flowers and concentrates, which minimizes handling time and increases sample throughput.&nbsp;</p><p>The validation study was conducted at GPA. Method performance was evaluated for calibration range, linearity, LOD, LOQ, accuracy, precision, recovery, repeatability, and robustness. Matrix-spiked recoveries across flower and concentrate samples were within the 70–130% acceptance range, and QC samples met all performance criteria, confirming method reliability. Robustness testing across analysts and matrices further validated the method’s reproducibility. Overall, this approach provides a streamlined, sensitive, and regulatory-compliant workflow for laboratories, enabling accurate monitoring of VEA to support quality control, consumer safety, and regulatory compliance in cannabis-derived products</p>

SISs are health care facilities where staff are trained to provide health education, support and connections to services as well as intervening in serious adverse events such as drug overdoses. They provide a safe, hygienic and non-judgement environment in which individuals may consume illicit drugs without fear of arrest.&nbsp;</p><p><strong>The benefits of SIS are to:&nbsp;</strong></p><p>(1) prevent an overdose from becoming fatal through quick intervention, (2) reduce the frequency of public injection and publicly discarded syringes, (3) increase access to health services including substance use treatment, and (4) change behaviors associated with acquiring HIV and hepatitis.&nbsp;</p><p><strong>The benefits of drug checking services within an SIS are:&nbsp;</strong></p><p>(1) it provides the consumer information about the contents of their drugs prior to consumption allowing them take steps to reduce their risk of overdose, (2) it identifies when a highly toxic substance may be in the local drug supply so advisories can be issued to local service agencies and first responders, (3) it encourages use of supervised injection services and the benefits mentioned above, and (4) it detects novel substances or drug combinations that may impact the consumer.&nbsp;</p><p>Initially approved by the Canadian federal government in 2017, the nation now has four official SISs in operation in Ottawa and four sites approved in Toronto. While most SISs are prepared to respond quickly to overdoses, a better practice is to screen drugs for harmful impurities prior to injection. Specifically, Raman can be used in QC to distinguish between types of chocolate, detect adulteration, measure crystallization and texture, and monitor the manufacturing process.&nbsp;</p><p>This Application Note outlines techniques for effectively collecting Raman spectra from various chocolates, providing a foundation for quality assessment and adulteration detection.</p><h4><strong>METHOD&nbsp;</strong></h4><p>Raman data collection with an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=220&amp;search=i-Raman+NxG+1064"><strong>i-Raman NxG 1064 </strong></a>laboratory Raman system (Figure 2) was optimized by adjusting integration time and laser power (Table 3) to determine the best conditions to maximize signal strength and minimize risk of sample melting.</p><p>All chocolate samples were analyzed by placing a piece of chocolate on the stage with a Raman probe securely locked above the sample (Figure 3). The optimal working distance was determined by adjusting the probe’s z-axis position while continuously monitoring the intensity of the Raman signal. Once the optimal focal distance is found, a distance regulator helps the operator position the probe on the sample to ensure consistent and reliable measurement.</p><h4><strong>CONCLUSION</strong></h4><p>This study highlights the capability of Raman spectroscopy for rapid, nondestructive measurements of chocolate quality indicators. PLS models demonstrate high predictive accuracy for both cocoarelated materials and sugar content. Increasing the number of samples and testing a broader range of chocolates would further improve the robustness and accuracy of the model. Overall, Raman spectroscopy, combined with chemometric modeling, offers a reliable QC method for routine and real-time chocolate analysis.</p><h3><strong>4. On the other hand, because it is a major contributor to environmental issues, such as ocean pollution and global warming, measures to reduce plastic usage are being undertaken in countries around the world. 1)&nbsp;</p><p>In Japan, the Plastic Resource Circulation Promotion Act2) came into effect in April 2022. The act does not regulate plastic but rather promotes the establishment of a sustainable circular economy by encouraging collaboration among businesses, local governments, and consumers throughout the overall process of designing, manufacturing, selling, collecting, and recycling plastic products.&nbsp;</p><p>One of these initiatives is the recycling of PET bottles for beverage use. When recycling beverage bottles, residual odors can be an issue, with substances like acetaldehyde from water bottles and limonene from citrus beverages known to persist in the containers. To address these residual substances, recycling companies are implementing unique strategies, one of which involves using gas chromatography-mass spectrometry (GC-MS) to quantify the residual materials. In this article, we present an example of qualitative and quantitative analysis of aldehydes and limonene in PET bottles using the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMS-QP2020+NX"><strong>GCMS-QP2020 NX</strong></a>/HS-20 NX, with hydrogen (H2) as the carrier gas.</p><h4><strong>Conclusion</strong></h4><p>By using hydrogen (H2) gas as the carrier gas, we were able to avoid the use of the hard-to-obtain helium gas. Additionally, by employing an HS-GC-MS system, we could qualitatively and quantitatively analyze acetaldehyde and limonene from PET without the need for cumbersome pretreatment.</p>

Agilent Technologies: Analysis of Trace Impurities in Monocyclic Aromatic Solvents Following ASTM D7504</strong></h3><p><strong>Dual simultaneous analysis using the Agilent 8860B gas chromatograph</strong></p><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_trace_impurity_aromatic_solvents_analysis_astm_d7504_8860_gc_5994_9316en_agilent_c9fa6868e6.pdf"><strong>Full PDF for download</strong></a></li></ul><section><div><div><div><div><div><div><p>The production of high-purity monocyclic aromatic hydrocarbons, such as benzene, toluene, ethylbenzene, and xylenes (BTEX), is a cornerstone of the modern petrochemical industry. These compounds serve as essential feedstocks for the synthesis of plastics, resins, and textiles. Because even trace impurities can adversely affect downstream chemical reactions and product quality, rigorous analytical testing is required to ensure these intermediates meet strict commercial specifications.&nbsp;</p><p>ASTM D7504 is the industry-standard test method for determining the purity of, and trace impurities in, monocyclic aromatic hydrocarbons using gas chromatography (GC) with flame ionization detector (FID).1 This method is favored for its streamlined workflow; it utilizes effective carbon number (ECN) correction factors for area normalization. This eliminates the need for labor-intensive, multipoint calibrations for every individual impurity, allowing laboratories to calculate purity and quantify trace nonaromatics and aromatics with high precision.&nbsp;</p><p>The application scope of ASTM D7504 covers a wide array of aromatic liquids, including benzene, toluene, mixed xylenes, and styrene. To comply with the method's stringent performance requirements, the GC system must&nbsp;demonstrate:&nbsp;</p><ul><li><strong>High sensitivity: </strong>A limit of quantitation (LOQ) of at least 0.0006% by mass.</li><li><strong>Good resolution: </strong>Maintaining a valley-to-peak ratio of no more than 50% for critical separations (such as p-xylene and m-xylene).&nbsp;</li><li><strong>Strict precision: </strong>Highly reproducible peak areas to ensure accurate purity calculations.</li></ul><p>Implementing ASTM D7504 on a dual-channel GC/FID platform—where two independent analytical channels configured with identical injectors, columns, and detectors operate within a single GC oven—provides significant operational advantages:&nbsp;</p><ol><li>Enhanced laboratory throughput: By performing two injections simultaneously, the test lab can double its sample capacity per instrument, effectively reducing the cost-per-analysis and maximizing hardware ROI.&nbsp;</li><li>Method flexibility and redundancy: A dual-channel set up allows a lab to run different sample types (such as toluene on the front channel and xylene on the back) in a single oven cycle. Furthermore, it provides an immediate "back up" channel to maintain uptime during routine&nbsp;maintenance.&nbsp;</li></ol><p>For the system to be considered reliable, both channels should meet ASTM D7504 sensitivity, resolution, and precision requirements. In addition, both channels should yield consistent quantitative results for the same sample. The reproducibility limit (R) demonstrated in ASTM D7504 interlaboratory testing will be used to evaluate the result consistency between the two channels.</p><p>ASTM D7504 recommends helium and hydrogen as carrier gas. In China, major Chinese petrochemical companies often align their internal quality control standards directly with ASTM D7504 when testing for export of high-purity chemical intermediates. Because nitrogen is commonly used in China, they commonly adapt the ASTM D7504 method to use nitrogen carrier gas.&nbsp;</p><p>This application note demonstrates how dual-channel analysis on the Agilent 8860B GC, following ASTM D7504, delivers consistent and high-precision data using three types of carrier gas.</p><h4><strong>Experimental</strong></h4><p>An <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8860B"><strong>Agilent 8860B GC</strong></a><strong> </strong>was configured with dual split/splitless inlets and dual flame ionization detectors (FIDs). The sample introduction was made by dual <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=7693A"><strong>Agilent 7693A automatic liquid samplers (ALS)</strong></a> with high density turrets (16-vial). The analytical parameters and consumables are summarized in Tables 1 and 2. Data acquisition and analysis were conducted using <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=OpenLab+CDS"><strong>Agilent OpenLab CDS version 2.8</strong></a></p><h4><strong>Conclusion&nbsp;</strong></h4><p>The Agilent 8860B gas chromatography system configured with two identical analytical channels are applied for impurity analysis in monocyclic aromatic solvents using three carrier gases. Each analytical channel was evaluated according to the ASTM D7504 method in terms of resolution, sensitivity, quantitation precision, and recovery with satisfactory results. The quantitation results between the two channels are consistent with each other.&nbsp;</p><ul><li>Column resolution compounds pair demonstrated good separation with H/V no less than 5:1 for 300 ppm m-xylene and p-xylene solvent.&nbsp;</li><li>The LODs achieved for benzene on each analytical channel are less than 0.0001%, exceeding the 0.0002% LOD required by ASTM D7504.&nbsp;</li><li>The response and mass concentration precisions of most impurities in check standards are below 1%, meeting or exceeding the precision requirements in D7504.&nbsp;</li><li>Excellent recovery results, ranging from 93 to 105%, was achieved for added impurities, indicating good quantification accuracy based on the 8860B GC system.&nbsp;</li><li>The quantitation difference of all probe impurities between two channels is less than the reproducibility thresholds (R), demonstrating excellent across channel consistency</li></ul><p>Besides great instrument performance, the Agilent OpenLab CDS software can realize ECN based normalization for easy and fast quantitation and report generation. The 8860B dual channel GC system coupled with the OpenLab CDS software provides an accurate and reliable analysis of impurities in monocyclic aromatic solvents with satisfactory precision. Lab productivity can be improved significantly compared to the single channel analytical system.</p><h3><strong>2. MDCW / University of Alberta: Leveraging Local Libraries for Positively Parsimonious Peak Tables</strong></h3><ul><li>Presentation</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/O_6_Harynuk_f005bdafaf.pdf"><strong>Full PDF for download</strong></a></li></ul><section><div><div><div><div><div><div><p>James J. Harynuk’s presentation, <strong>“Leveraging Local Libraries for Positively Parsimonious Peak Tables,”</strong> explores how custom mass spectral libraries can simplify and improve compound identification in complex GC×GC-(HR)MS datasets. GC×GC-MS is widely applied in petroleum, environmental, food/flavour, and omics research, where samples can contain hundreds or thousands of peaks. While expanded separation space, cleaner spectra, retention patterns, and high-resolution accurate-mass data provide strong identification capabilities, converting large datasets into coherent peak tables with consistent compound assignments across multiple samples remains challenging.</p><p>The presentation demonstrates the approach using a field pea study investigating how nitrogen availability influences plant metabolism and volatile organic compounds. A conventional workflow based on peak finding and library searching can generate very large peak tables and inconsistent assignments. Incorporating retention indices (RI) and LECO’s Identification Grading System (IGS) substantially reduces the number of questionable peaks. In one example, a pea sample containing <strong>789 detected peaks was reduced to 146 retained peaks</strong> after RI filtering and IGS scoring, producing a much more manageable dataset. However, conventional libraries still struggle with isomers and with compounds that repeatedly occur in a laboratory’s samples but cannot yet be confidently identified—the presentation calls these <strong>“unknown knowns.”</strong></p><p>Harynuk therefore proposes building a <strong>custom local HRMS library</strong> containing not only standards and confidently identified compounds, but also reproducible unknown features. To qualify for inclusion, a peak should have a high-quality spectrum, S/N &gt;100, chromatographic purity, occurrence in multiple technical replicates, an assigned retention index, and a unique identifier. Crucially, the exact chemical identity does not need to be known: an unknown but reproducible feature can receive an internal identifier such as “JJH-001” and be updated later as more information becomes available. This allows laboratories to retain analytically meaningful recurring signals instead of discarding them simply because a definitive library identification is unavailable.</p><p>The results show that combining a custom library with local RI data and IGS filtering improves both the consistency and efficiency of peak-table generation. Compounds with IGS scores ≥3.0 were generally assigned the same identification across samples when the custom library was used, while reliance on NIST and IGS alone produced less consistent results. After correcting only a small number of problematic identifications per peak table, retention-index errors for custom-library entries dropped dramatically—in the evaluated samples, typically to around <strong>0.5–1.9 RI units</strong>. The key recommendation is therefore straightforward: <strong>build a custom MS library, include recurring “unknown knowns,” collect local retention-index data, and apply identification-quality filters</strong> to obtain cleaner and more consistent GC×GC-MS peak tables with substantially less manual effort.</p><h3><strong>3. Shimadzu: Simultaneous Analysis of Pesticide Residues in Food Using Triple Quadrupole GC-MS/MS with SPL Mode of a Multimode injection Unit (MMI)</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_01234_en_d8cff47c74.pdf"><strong>Full PDF for download</strong></a></li></ul><h4><strong>User benefits</strong></h4><ul><li>The MMI’s rapid heating and cooling technology significantly reduces waiting time during inlet maintenance, improving instrument uptime.&nbsp;</li><li>The Smart Pesticides Database enables automatic creation of analytical methods without the need for pesticide standards.&nbsp;</li><li>Peakintelligence for GCMS uses AI-based peak processing to deliver highly accurate analysis with no parameter settings required</li></ul><p>GC-MS/MS is an indispensable tool for the simultaneous analysis of pesticide residues in food. However, improving productivity in high-throughput workflows remains a persistent challenge. With the increasing use of simplified sample preparation methods such as QuEChERS, contamination of the inlet by matrix components is unavoidable, and frequent inlet maintenance, including liner replacement, is often required. Because this maintenance involves both cooling the inlet and waiting for temperature re-stabilization afterward, it can become a major source of instrument downtime.&nbsp;</p><p>The newly developed Multi-Mode Inlet (MMI) incorporates Shimadzu’s proprietary thermal insulation technology, providing excellent temperature stability together with rapid heating and cooling performance. While maintaining high data compatibility with conventional split/splitless inlets, the MMI significantly reduces the waiting time associated with inlet maintenance.</p><p>In this Application News, simultaneous multiresidue analysis in SPL mode is demonstrated for four food matrices with different characteristics - spinach, orange, avocado, and ginger - using the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMS-TQ+8040+RX"><strong>GCMS-TQ 8040 RX triple quadrupole</strong> </a>GC-MS/MS equipped with the MMI.</p><h4><strong>Data Analysis&nbsp;</strong></h4><p>LabSolutions Insight, a quantitative data processing software package designed for high-throughput sample analysis, was used for data analysis, with Peakintelligence for GCMS applied as the peak integration algorithm (Fig. 6). Peakintelligence is an AI-based waveform processing algorithm that uses machine learning to emulate the peak processing performed by experienced analysts. It requires no operator-defined parameter settings and provides peak integration performance comparable to that of skilled users.&nbsp;</p><p>For more information on improving the efficiency of pesticide data analysis using Peakintelligence for GCMS, please refer to Application News 01-00585-EN, “<a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_00585_en_7e311a3c77/an_01-00585-en.pdf"><strong>Time-Saving Data Processing for Pesticide Residues with Peakintelligence for GCMS</strong></a>.”</p><h4><strong>Conclusion&nbsp;</strong></h4><p>In this Application News, an example of simultaneous multiresidue pesticide analysis in food using a triple quadrupole GC-MS/MS equipped with a Multi-Mode Inlet (MMI) was presented. By adopting the MMI, instrument downtime associated with inlet maintenance can be significantly reduced while maintaining high quantitative performance. In addition, by combining automated method creation with the Smart Pesticides Database and efficient data analysis using the AIbased waveform processing algorithm Peakintelligence for GCMS, highly accurate results were obtained even for complex food matrices. This system optimizes the overall analytical workflow, from method development and maintenance to data analysis, and can make a substantial contribution to improving productivity in high-throughput pesticide residue analysis.</p></div></div></div></div></div></div></section></div></div></div></div></div></div></section>
