However, residue of EO might remain in the products, raising concerns about the potential health risks associated with it. Therefore, it is essential to monitor and analyze ethylene oxide levels in food product to ensure compliance with safety regulations and protect consumer health. In this study, both ethylene oxide (EO) and 2-chloroethanol (2-CE) in rice were analyzed. 2-CE, a potential byproduct of EO fumigation, is also a compound of interest due to its toxicity and potential health implications. By utilising dynamic headspace and GC-MS/MS, precise and sensitive detection of both EO and 2-CE residues in rice samples can be achieved. The limit of quantitation is 5 ng/g with area %RSD (n=5) for EO and 2-CE at 5.6 % and 4.9 %, respectively. The spike recovery study at concentrations of 10 ng/g, 25 ng/g and 100 ng/g yielded recoveries within 70 %-130 %. This comprehensive analytical approach enables accurate quantification and identification of these compounds, significantly contributing to the overall safety and quality assurance of the food supply chain.</p><p><strong>Conclusion</strong></p><p>An internal standard method was successfully developed for the analysis of EO and 2-CE in rice using HS-20 NX Trap with GCMS-TQ8050 NX. The dynamic headspace technique and GC-MS/MS were employed, simplifying the sample preparation process and maximizing the sensitivity. Good linearity (R2 value &gt; 0.997) and recovery within 70 % to 130 % were achieved for all the samples analyzed in this experiment, even without the use of matrix-matched calibration curves.</p><h4><strong>2. Exposomics is the study of the exposome that seeks to understand and quantify the totality of a human's lifetime exposure to various environmental factors, including chemical, physical, and biological agents. In essence, it involves assessing and analyzing the complex interactions between environmental exposures and an individual's genetic makeup, lifestyle, and other factors to understand their impact on human health and disease. By examining the cumulative effects of multiple exposures over time, exposomics provides valuable insights into the underlying mechanisms of disease development and can inform strategies for prevention and intervention.<sup>1</sup></p><p><strong>Summary</strong></p><p>The ability to measure the complexity of the exposome has been limited by the available analytical technology to detect complex exposures patterns at varying concentrations. By using untargeted approaches with high-resolution mass spectrometry, it is possible to detect and identify ongoing exposures that may have not been expected or characterized.</p><ul><li>High-resolution, full scan mass spectrometry using Orbitrap technology provides a solution for:</li><li>Detection and quantification of compounds using an untargeted full scan acquisition that is both highly selective and sensitive.</li><li>Identification and elucidation of the chemical composition of unknown compounds.</li><li>Retrospective analysis of samples long after data acquisition.</li></ul><p>The combined capability of both the Orbitrap Exploris GC 240 mass spectrometer and Compound Discoverer software with spectral libraries is an excellent tool to support exposomics research.</p><h4><strong>3. Agilent Technologies: </strong><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_pesticides_botanicals_captiva_5994_7361en_agilent_53a42de795.pdf"><strong>Comparison of Sample Preparation Methods for Pesticide Analysis in Botanical Dietary Supplement Materials</strong></a></h4><ul><li>Application</li></ul><p><strong>Abstract</strong></p><p>This application note presents a novel streamlined sample preparation method for liquid chromatography/triple quadrupole mass spectrometry (LC/MS/MS) and gas chromatography/triple quadrupole mass spectrometry (GC/MS/MS) multiresidue pesticide (over 440) analysis. This treamlined sample preparation enables analysis in botanical dietary supplement (BDS) materials using the EMR mixed‑mode passthrough cleanup with Agilent Captiva EMR with Carbon S cartridges. Various BDS samples are extracted by traditional QuEChERS extraction, followed by appropriate Captiva EMR cartridge cleanup. The selected Captiva EMR cartridges, based on the BDS sample matrix complexity and pigment intensity, include Captiva EMR–GPD for green tea and peppermint tea, Captiva EMR–LPD for barberry root, and Captiva EMR–GPD + EMR–GPF for curcumin complex. The developed method provides a simple and unified sample preparation workflow for both LC/MS/MS and GC/MS/MS pesticide detection. This method demonstrates acceptable performance, i.e., recovery of 70 to 120% and RSDs &lt; 20%, in representative BDS samples for over 82% of the total 447 pesticides analyzed. Compared to the traditional approach that requires multiple sample preparation methods, the newly developed method demonstrates a more one‑size‑fits‑most approach for efficient cleanup. This approach increases the instrument uptime and reduces both<br>consumables and labor resources. The direct comparison of new method versus old method on actual samples analysis demonstrates acceptable equivalence on reporting results.</p><p><strong>Conclusion</strong></p><p>A simple, rapid, and reliable method using Agilent Bond Elut QuEChERS extraction followed by EMR mixed‑mode passthrough cleanup using Agilent Captiva EMR with Carbon S cartridges was developed. The results of this method were verified for over 440 pesticides in botanical dietary supplement extracts and products by LC/MS/MS and GC/MS/MS. The EMR mixed‑mode passthrough cleanup method provided a convenient and simplified cleanup method after sample extraction for selective and efficient matrix removal in BDS matrices and acceptable quantitation recovery and reproducibility in large panel pesticide quantitation. The detailed proficiency test confirmed the acceptable method performance for practical<br>sample analysis.</p><p>The ability to streamline sample preparation into a single method allows for a reduction in the total amount of sample preparation consumables and reduces the preparation time. The reduced organic solvent, especially the toxic solvent used per sample, made the entire procedure greener while being more people and environmentally friendly. Also, having one sample to be prepped in a single day allows for much greater traceability regarding shared analytes between LC and GC analyses. This single sample preparation also helps simplify the results and documentation. Metrohm: </strong><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_k_074_download_e3518c084c.pdf"><strong>Water content in crude oil determined with Karl Fischer titration</strong></a></h4><ul><li>Application</li></ul><p><strong>Fully automated determination using the oven method according to ASTM D4928</strong></p><p>Crude oil contains water. Before transport, water is removed to reduce costs. Furthermore, the presence of water in crude can cause corrosion. Therefore, it is essential to determine the water content in crude oil. Previously, ASTM D4928 described a direct coulometric Karl Fischer titration to determine water in crude oil. This quickly contaminated the titration cell, requiring regular cleaning and frequent reagent exchange. ASTM D4928 was then revised to include coulometric Karl Fischer titration in combination with the oven method. In this method, the sample is heated in an oven. The water evaporates and is carried into the titration cell by an inert carrier gas. The water content is determined in the titration cell. The oven method avoids titration cell contamination and significantly reduces reagent consumption. It can be fully automated, minimizing handling errors and workloads, with outstanding reproducibility.</p><p><strong>CONCLUSION</strong></p><p>The oven method is the perfect option to determine water content in crude oils precisely and reliably. Using the 874 Oven Sample Processor allows fully automated determination, freeing up valuable time and thus increasing laboratory productivity. Furthermore, by fully automating the analysis, the reproducibility can be increased and sample analysis failures due to improper handling can be reduced.</p><h4><strong>2. Thermo Fisher Scientific: </strong><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/cs_003393_ov_orbitrap_astral_immunopeptidomics_cs003393_na_en_7b9f76c0f4.pdf"><strong>Immunopeptidomics: Harnessing cutting-edge technology for unprecedented depth and accessibility in analysis to leverage the immune response for precision therapy</strong></a></h4><ul><li>Application</li></ul><p><strong>Introduction</strong></p><p>Immunopeptidomics is the study of the peptides presented by major histocompatibility complex (MHC) molecules on the surface of cells. These MHC peptides have major implications for many areas of research, including immunotherapy and personalized medicine. For example, many studies in this field aim to identify low-level tumor specific antigens (TSAs) with the goal of developing personalized immunotherapies to target cancerous cells with a high degree of specificity. Mass spectrometry (MS) allows for direct immunopeptidomics analysis, enabling simultaneous identification and quantification of thousands of MHC peptides in a single run. The recently developed Thermo Scientific™ Orbitrap™ Astral™ mass spectrometer has enabled new levels of sensitivity and selectivity to provide deeper insights into the immunopeptidome. In this study, we utilized the Orbitrap Astral MS to characterize the immunopeptidome extracted from IM-9 human Multiple Myeloma B-lymphocyte cells to support the<br>detection and annotation of potential neoantigens.</p><p><strong>Summary</strong></p><ul><li>The integration of the Vanquish Neo UHPLC system with an Orbitrap Astral mass spectrometer, featuring the FAIMS Pro Duo interface, enhances sensitivity and dynamic detection range in immunopeptide analysis. This configuration enables greater depth of coverage and increased analysis throughput.</li><li>Increased sensitivity allows for compatibility with low levels of material equivalent to samples extracted from tissue biopsy samples.</li></ul><h4><strong>3. Shimadzu: </strong><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_00739_en_712d96e287.pdf"><strong>Quantitative Analysis of Nine Haloacetic Acids in Tap Water Using LCMS™ - 8050RX</strong></a></h4><ul><li>Application</li></ul><p><strong>User Benefits</strong></p><ul><li>Enable users to analyze nine haloacetic acids only 15 minutes per cycle using a simple dechlorination pretreatment.</li><li>Enable users to detect nine haloacetic acids at 2 μg/L in water.</li><li>Enable users to analyze nine haloacetic acids with good repeatability and recovery rates that are compliant with validation guidelines.</li></ul><p><strong>Introduction</strong></p><p>Haloacetic acids in tap water are by-products from disinfection processes during water treatments. In Japan, the Ministerial Ordinance Concerning Water Quality Standards<sup>1)</sup> specifies drinking water quality standards for three haloacetic acids: monochloroacetic acid (MCAA), dichloroacetic acid (DCAA), and trichloroacetic acid (TCAA) (with limit values of 0.02 mg/L for MCAA, 0.03 mg/L for DCAA, and 0.03 mg/L for TCAA, respectively). The ordinance also specifies items for further study for the following six additional acids: bromochloroacetic acid (BCAA), bromodichloroacetic acid (BDCAA), dibromochloroacetic acid (DBCAA), monobromoacetic acid (MBAA), dibromoacetic acid (DBAA), and tribromoacetic acid (TBAA).</p><p>The EU issued a directive, which became effective in 2020, that regulates five haloacetic acids, that is, the three acids governed by Japanese water quality standards as well as MBAA and DBAA.<sup>2)</sup></p><p>Haloacetic acids are analyzed using gas chromatography-mass spectrometry (GC/MS) or liquid chromatography-mass spectrometry (LC/MS). In particular, the LC/MS enables more efficient measurements since dechlorination is the only for required pretreatment.</p><p>This Application News describes an example of analyzing the nine haloacetic acids using the LCMS-8050RX, triple quadrupole mass spectrometer system (Fig. 1).</p><p><strong>Conclusion</strong></p><p>Nine haloacetic acids specified for water quality testing criteria and monitoring were analyzed simultaneously in 15 minutes using the CMS-8050RX system.</p><p>The results indicated that the system offered ample sensitivity for analyzing all haloacetic acids at concentrations of 2 μg/L, and it provided good calibration curve linearity within the 2 to 30 μg/L concentration range. In addition, spike-and-recovery tests of tap water samples (n = 5) provided good results with repeatability (concentration %RSD) of 10 % or less and recovery rates within the 80 to 120 % range. This confirms that analysis<br>with good accuracy and repeatability can be performed with the LCMS-8050RX system.</p><h4><strong>4. Agilent Technologies: </strong><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/to_energy_consumption_1260_prime_hplc_system_5994_7573_EN_agilent_a595b38dc7.pdf"><strong>Energy Consumption of the Agilent 1260 Infinity III Prime LC System</strong></a></h4><ul><li>Technical note</li></ul><p><strong>Abstract</strong></p><p>The Agilent 1260 Infinity III Prime LC System features the Agilent InfinityLab Assist—a module that automates tasks, facilitates maintenance, and assists in troubleshooting, thereby increasing the system's operational efficiency and uptime. To assess the energy consumption of these added benefits, the 1260 Infinity III Prime LC System was compared with the Waters Alliance iS HPLC System. The energy use was measured during different operational states of a typical laboratory day. Results indicate that the Agilent system consumed less energy across all states compared to the Waters system, making it a superior choice for laboratories seeking to work more economically and sustainably.</p><p><strong>Introduction</strong></p><p>Modern HPLC systems are more than sheer sample analyzers. Instrument sensors collect nonanalytical data to monitor the wear and tear of consumable parts such as pump seals, needle seats, or detector lamps, and notify users if a change is due. Control software integrates features that facilitate system equilibration, shut down the system after analysis, or automate daily tasks such as priming the pump.</p><p>The InfinityLab Assist elevates this assistance to the next level. This slim module added to the LC stack features a dedicated processor and large touch screen, which makes interacting with the LC system faster, more accessible, and more convenient than interacting through a PC–independently of the chromatography data system (CDS) connected to the LC. Automated tasks to start up and shut down the system, guided maintenance, and assisted troubleshooting are just three of many exciting features of the InfinityLab Assist described in a dedicated white paper.<sup>1</sup></p><p>All the benefits of saved time, increased confidence, and ease of use with the InfinityLab Assist might be contrasted with how much the extra module will add to the energy consumption of the LC. To shed light on this question, this technical overview presents the energy consumption of the 1260 Infinity III Prime LC System under typical use conditions. The same conditions are applied to the Waters Alliance iS HPLC—a competitor system that also features a touch screen for easier operation. As in previous Agilent publications discussing energy consumption of LC systems<sup>2,3</sup>,<br>the analytical method was not optimized to the lowest energy consumption possible but was intended to represent a realistic use case over a typical day.</p><p><strong>Conclusion</strong></p><p>The energy consumption of the Agilent 1260 Infinity III Prime LC System was measured during Idle and Run states, applying typical conditions of an analytical UHPLC method. For comparison, the same method was run on a Waters Alliance iS HPLC System. Energy consumption in different operational states (Idle, Ready, and Run), as well as overall consumption per day and per sample was lower on the Agilent system. Doubling the number of measured samples per day resulted in an energy consumption increase below 10%, which underlines that an LC is most efficiently used if it measures samples all day instead of running idle. The InfinityLab Assist can help increase the uptime of the LC by adding automated tasks, maintenance feedback, and assisted troubleshooting to increase confidence and ease of use.</p><h4><strong>5. Waters Corporation: </strong><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/720008385en_d2611a1349.pdf"><strong>Mapping phosphorylation of peptide biomarkers corresponding with glioblastoma tumours using data-dependent acquisition and multi-reflecting time-of-flight mass spectrometry</strong></a></h4><ul><li>Application</li></ul><p><strong>INTRODUCTION</strong></p><p>Glioblastoma is an aggressive form of brain tumor with little therapeutic options available, and correspondingly the 5-year survival rate remains &lt;10% of those diagnosed, whilst the median life expectancy is 15 months.<sup>1</sup> Phosphorylation is a key post-translational modification (PTM) that is involved in numerous processes, such as activation of key enzymes/kinases for example. Hence, phosphorylation sites are important targets for the regulation and development of glioma and may offer insight for new targeted therapies to perturb specific cellular pathways.<sup>2</sup></p><p>Profiling of phosphorylated residues using mass spectrometry can be problematic due to the labile nature of the phosphoryl group. For example, during collision induced dissociation the group can be lost (H<sub>3</sub>PO<sub>4</sub> or HPO<sub>3</sub> + H<sub>2</sub>O) resulting in a neutral loss of –98Da. When combined with data independent approaches (DIA) approaches and complex spectra, it can make identification of phosphorylated residues difficult. However, utilising data dependent acquisition (DDA) mode allows relation of the precursor m/z and that resulting from a neutral loss to that of the quadrupole set mass, facilitating identification of phosphorylated peptides.<sup>3</sup></p><p>Here, we describe an LC-MS workflow using the Xevo™ MRT Mass Spectrometer; a novel benchtop high resolution, high mass accuracy platform for accurately and characterizing post-translational modifications whilst also providing corresponding quantitative data (Figure 1).</p><p><strong>CONCLUSION</strong></p><p>Phosphopeptide enrichment from human glioblastoma cell lines was successfully implemented using Fe(iii)-NTA spin columns. The ACQUITY Premier System demonstrated excellent non-adsorption of phosphopeptides and in tandem with data dependent acquisition successfully profiled and localised sites of phosphorylation. The Xevo MRT Mass Spectrometer provided mass resolution of &gt;70,000 FWHM at fast scan speeds of 20 Hz and allowed for confident identification of both precursor and product ions.</p>

Maintaining fertility and quality of soil is an ultimate prerequisite for ensuring human and animal health. Environmental pollution is an important cause for concern in modern society, and applicable regulations and standards focus on regularly testing soils and sediments from different areas. For similar reasons, sludges, which are often used as fertilizers, are also regularly tested to detect harmful elements and compounds that may degrade soil quality and eventually pose a threat to human health and safety. Researchers and applied testing laboratories work to fully understand the root cause and potential impact of soil pollution, as well as its distribution through mobilization or diffusion. Accurate and rapid analysis of the content of metal substances in soil is a key step in accomplishing this important task.</p><p>Inductively coupled plasma mass spectrometry (ICP-MS) is a technique that is widely used for the elemental analysis of complex samples such as soil and sludge digests. In this application note the new Thermo Scientific™ iCAP™ MSX ICP-MS was used for the analysis of soils, sludges, and sediments. Argon Gas Dilution (AGD) reduced the manual work involved in sample dilution, and at the same time, enhanced the overall matrix tolerance of the system to different environmental samples. This study demonstrates how AGD can be used to readily deliver repeatable and reliable results with high accuracy and sensitivity in laboratories tasked to analyze high sample loads.</p><p><strong>Conclusions</strong></p><p>The iCAP MSX ICP-MS is an excellent choice for laboratories performing soil and sediment digests with varying compositions. It provides the robustness and matrix tolerance needed to analyze the often challenging samples, yet at the same time provides the sensitivity required to detect important contaminants at relevant levels. The integrated argon gas dilution for controlled and automatic dilution of the sample aerosol effectively minimizes matrix effects without the need for laborious additional sample preparation. Robustness combined with accuracy and high sensitivity, enabling the analysis of several major as well as trace elements in soil samples with high reliability, makes this method an ideal option for seamless analysis of hundreds of soil samples daily. The most important results of this study are summarized below:</p><ul><li>The use of AGD Level 10 combined with KED mode measurements yielded excellent linearity, wide linear dynamic range, high sensitivity, and interference free data for a range of different elements in soil samples.</li><li>Online sample dilution using integrated AGD offers an automatic solution for diluting soil sample digests to the desired extent and results in accurate and reliable analysis with minimal to no matrix induced signal variation.</li><li>The accuracy of the method is confirmed by the SRM recoveries as well as the stable QC analyte response which were both found to be within 80–120% for the different analytes on repeated analysis over several hours.</li><li>The internal standard recoveries remained stable and extremely consistent within 80–120% throughout the 8-hour robustness test, proving that a reliable uninterrupted and robust high-quality analysis of different types of soil samples can be easily carried out using the iCAP MSX ICP-MS</li></ul><h4><strong>2. Agilent Technologies: </strong><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_cosmetics_analysis_containers_vaya_5994_7595en_agilent_c1f6227a05.pdf"><strong>Cosmetic Raw Material Identification Testing Through Transparent and Opaque Containers</strong></a></h4><ul><li>Application</li></ul><p><strong>Abstract</strong></p><p>With the adoption of stricter requirements for the manufacturing of cosmetic products, the Good Manufacturing Practice landscape has evolved and impacted quality control (QC) processes to meet quality, costs, and efficiency requirements. In particular, raw material identification (ID) testing has seen the adoption of at‑the‑point‑of‑need solutions in the warehouse. The Agilent Vaya Raman is the latest generation Raman handheld spectrometers for raw material ID testing directly through containers. This application note discusses the capability of Vaya for cosmetics raw material ID testing through transparent and opaque containers and describes the impact on raw material receipt processes.</p><p><strong>Conclusion</strong></p><p>The Agilent Vaya Raman spectrometer has the capability to distinguish a unique Raman spectrum for each material in this study, enabling their direct ID verification while in quarantine. There is no need for sample or container preparation before scanning a the contents of a container using the ID verification feature of the Vaya. Users are simply asked to place the Vaya directly against the container and initiate the process. The instrument remains stationary during the spectrum acquisition phase. On average, the scanning time for through‑transparent is 10 to 15 seconds, and for opaque containers, 35 to 40 seconds. The Vaya Raman raw material identity verification system, with its rapid analysis times and capability to measure through both transparent and opaque containers, is perfectly adapted for handling large quantities of raw materials used in the formulation of cosmetic products. It enables immediate identification of raw materials in quarantine upon receipt. The Vaya Raman instrument simplifies the process by reducing or eliminating many steps required in FTIR or conventional Raman spectroscopy, such as cleaning instrumentation and the sampling booth or moving containers to and from the quarantine area for sampling and analysis. Using Vaya allows for future preparation of raw material receipts and can accommodate increased production volumes or new sampling requirements such as 100% ID, all with minimal investment.</p><h4><strong>3. For example, the changing of a color from exposure to light is called photochromism, from heat is called thermochromism, and from a change in solvent polarity is called solvatochromism. These chromotropic phenomena have everyday applications, such as checking moisture absorption with silica gel and the printing of receipts. While UV-Vis spectrophotometers are used to measure these phenomena quantitatively, they require additional equipment depending on the type of external stimulus causing the color change. So while, solvatochromism can be measured without additional equipment, measuring thermochromism requires a device to heat the sample. (Application News 01-00016 describes using a UV-Vis spectrophotometer to measure thermochromism in a solid sample.) Measuring photochromism requires irradiating the sample with light, which can be performed using Shimadzu’s dedicated photoreaction evaluation system called Lightway. More detailed information about Lightway can be found in Application News No. A625. This Application News describes an evaluation of the solvatochromism and thermochromism of a metal complex solution. To measure the thermochromism, a thermoelectrically temperature-controlled cell holder (TCC-100) and a six-cell thermoelectrically temperature-controlled cell positioner (CPS-100) were used. <sup>1</sup></p><p><strong>Conclusion</strong></p><p>This Application News evaluated the solvatochromism and thermochromism of solutions of cobalt chloride. The changing of colors due to solvatochromism was evaluated quantitatively using the optional color calculation software for LabSolutions UV-Vis. The TCC-100 enabled the examination of thermochromism over the wide temperature range of 10 to 50 °C. The CPS-100, which can control the temperature of multiple<br>samples concurrently, enabled a more efficient investigation of thermochromism. This shows how selecting the right equipment for the application enables optimal temperature-controlled measurements.</p>

Shimadzu: </strong><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_00_00095_en_c6eed5e008.pdf"><strong>Analysis of Major Psychoactive Compounds in Nutmeg Using GC-MS/MS</strong></a></h4><ul><li>Application</li></ul><p><strong>Abstract</strong></p><p>Nutmeg is a spice used in a variety of recipes that can be easily purchased by anyone. Among younger people, however, excessive consumption of nutmeg powder for recreational purposes is increasing, causing hallucinogenic effects. However, there are few reports on toxic or comatose-fatal blood levels and their changes over time in cases of nutmeg poisoning. Therefore, an analytical method using GC-MS/MS was devised and applied to a case of nutmeg poisoning in order to determine the blood concentrations of the main psychoactive substances (safrole, myristicin, and elemicin) in nutmeg and their change over time. The limit of detection (LOD) and limit of quantification (LOQ) of the constructed method were 0.14 to 0.16 ng/mL and 0.5 ng/mL (the lowest point of the calibration curve), respectively. Calibration curves showed good linearity, with R2 = 0.996 to 0.997 for all substances in the range of 0.5 to 300 ng/mL. The accuracy for QC (low 1 ng/mL, medium 120 ng/mL, and high 240 ng/mL) and LOQ (0.5 ng/mL) was %RSD 2.4 to 11 % for diurnal variation (n = 5) and %RSD 1.5 to 11 % for diurnal variation (6 days). A systematic error of -2.6 to 2.1 % was obtained.</p><p><strong>Introduction</strong></p><p>Nutmeg, the dried seed of Myristica fragrans, has been used as a spice and for medicinal purposes all over the world since ancient times and is widely sold at low prices. Because nutmeg contains safrole, myristicin, and elemicin (Fig. 1) as the major psychoactive compounds, they are sometimes intentionally consumed in large amounts to induce hallucinations or intoxication<sup>8)-10)</sup>. In addition, intoxication by unintentional misuse of nutmeg has recently become a problem. In addition to hallucinogenic effects, too much nutmeg is associated with clinical symptoms such as nausea, vomiting, abdominal pain, agitation, drowsiness, dizziness, tachycardia, blurred vision, dry mouth, and flushing<sup>5), 9), 11)-14)</sup>. However, the majority of nutmeg overdose cases are mild and unlikely to be fatal<sup>14), 15)</sup>. Symptoms of nutmeg poisoning usually begin 2 to 8 hours after ingestion<sup>11), 15), 16)</sup> and subside within a few days<sup>3), 5), 9), 11), 13), 15)-17)</sup>. Only two deaths worldwide have been linked to the consumption of nutmeg<sup>15), 18)</sup>.</p><p>In a recent study, biological samples were collected from volunteers who ingested nutmeg powder to identify metabolites of major psychoactive substances<sup>19)-23)</sup>. However, although many cases of nutmeg poisoning have been reported to date, there have been no reports evaluating the blood levels and half-lives of major psychoactive substances, such as safrole, myristicin, and elemicin, during poisoning. Therefore, we developed an analytical method for three psychoactive substances using GC-MS/MS, measured the blood levels of these main substances in patients with nutmeg poisoning, and evaluated their changes over time.</p><p><strong>Conclusion</strong></p><p>Using a MonoSpin® extraction kit in combination with a GC–MS/MS system, a simple method was devised to detect and quantify safrole, myristicin, and elemicin in human serum. This method was validated and applied successfully to a case of nutmeg poisoning. For this patient, the method quantified the time-course changes and half-life of safrole, myristicin, and elemicin in serum. It is believed that this method will help to provide additional blood-concentration data for nutmeg poisoning cases as well as more accurate toxicokinetic parameters.</p><h4><strong>2. Agilent Technologies: </strong><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_fatty_acid_methyl_ester_analysis_8890_gc_fid_luma_5994_7849en_agilent_677241bc1b.pdf"><strong>Analysis of 37 Fatty Acid Methyl Esters on the Agilent 8890 GC Using FID and LUMA Detectors</strong></a></h4><ul><li>Application</li></ul><p><strong>Introduction</strong></p><p>The analysis of fatty acid methyl esters (FAMEs) is used to characterize lipids in foods, including oils, meats, seeds, and other products. The fatty acid composition of fats is a complex mixture of saturated, monounsaturated, and polyunsaturated compounds with various carbon chain lengths.<sup>1</sup> Because the roles of fatty acids in the body vary depending on their structure, it is necessary to conduct detailed compositional analyses of the fatty acids contained in foods. The analysis of fatty acid composition in food is standard in many governmental, quality control (QC), and contract research laboratories worldwide.</p><p>The nutrition labels on food products include much of this information, detailing food composition to help consumers make informed choices.<sup>3</sup> Gas chromatography-flame ionization detection (GC-FID) is a commonly used method for analyzing the fatty acid composition in foods. In this application brief, an analysis of a neat 37-component FAME mix standard was conducted using an Agilent 8890 GC system equipped with an FID and LUMA vacuum ultraviolet (VUV) detector. For comparison, several branded oils were also purchased and analyzed.</p><p><strong>Conclusion</strong></p><p>These results show the advantages of using the LUMA detector coupled with an Agilent 8890 GC system and FID for FAMEs testing. By combining the robustness of the FID and the UV spectral power of the LUMA, users can be confident in their results.</p><h4><strong>3. Thermo Fisher Scientific: </strong><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_003254_gc_helium_hydrocarbon_oil_index_water_soil_an003254_na_en_4b0ceb0e48.pdf"><strong>Dual injection for Hydrocarbon Oil Index (HOI) determination in water and soil with helium carrier gas conservation</strong></a></h4><ul><li>Application</li></ul><p><strong>Goal</strong></p><p>The aim of this application note is to demonstrate the performance of a high-throughput and cost-effective method for the determination of the Hydrocarbon Oil Index (HOI) also known as Total Petroleum Hydrocarbon (TPH), in water and soil samples by using a dual injection configuration and conserving helium gas.</p><p><strong>Introduction</strong></p><p>Petroleum products are complex mixtures of hundreds of hydrocarbon compounds, ranging from light, volatile, short-chained organic compounds to heavy, long-chained, branched compounds. Petroleum hydrocarbons can enter the environment through industrial accidents, spills, or leaks, and as by-products from commercial or private uses. Contamination of water and soil is a growing concern as lighter TPH fractions float in water and form thin surface films, whereas the heavier fractions accumulate in the sediment at the bottom of the water, affecting bottom-feeding fish and organisms. TPH released in soil can evaporate into the air, dissolve into the groundwater, and move away from the spillage area, contaminating the drinking water supplies, or remain bound to the soil particles for a long period, thus reducing the usability of land for development.</p><p>To protect public health from toxic effects due to TPH exposure, many European countries and the United States have made the determination of the content of mineral oils and petroleum products in water and soils a compulsory requirement for quality certifications. Because there are so many petroleum products, it is not practical to measure each one individually, therefore the total amount of all hydrocarbons found in a sample of soil, water, or air is usually monitored.<sup>1</sup></p><p>The United States Environmental Protection Agency (U.S. EPA), the International Organization for Standardization (ISO), as well as the Italian Institute of Environmental Protection and Research (ISPRA) have published several methods (U.S. EPA 8015 D,<sup>2</sup> ISO 16703:2011,<sup>3</sup> ISO 14039:2004,<sup>4</sup> ISO 9377:2000<sup>5</sup>, ISPRA Doc. N. 46/14,<sup>6</sup> ISPRA Doc. n. 04/11<sup>7</sup>) that can be considered when analyzing TPH in water and soil using gas chromatography<br>coupled to flame ionization detection.</p><p>The Hydrocarbon Oil Index (HOI) represents the total amount of compounds that can be extracted from the sample (potable water, surface water, and wastewater) with a non-polar solvent. The extracted compounds must not absorb on Florisil™ and must elute between n-decane (C10) and n-tetracontane (C40) when analyzed by GC on a non-polar column. Such fraction includes heating oils, diesel fuels, kerosene, lubricants, and transmission fluids.</p><p>In this work, the Thermo Scientific™ AI/AS 1610 autosampler in the Gemini configuration for simultaneous dual injections combined with the Thermo Scientific™ HeSaver-H<sub>2</sub> Safer™ carrier gas saving technology was employed to increase the sample throughput and double the productivity while reducing helium consumption and cost per analysis. Method compliance in terms of sensitivity, recovery (C40/C20 ratio), linearity and precision were evaluated for quantitative assessment of the HOI in water and soil samples.</p><p><strong>Conclusions</strong></p><ul><li>The Hydrocarbon Oil Index analysis in water and soil is a routine test for environmental laboratories. Robust solutions offering time- and cost-saving opportunities are key to maintain laboratory efficiency and face increasing workloads.</li><li>The TRACE 1610 GC with a dual channel SSL-FID and the AS 1610 autosampler in the Gemini high-throughput configuration can double the number of samples through simultaneous injections, while Chromeleon CDS allows easy management of instrument control, data processing, and reporting for both channels.</li><li>The system performance exceeds the regulatory requirements in terms of recovery, repeatability, linearity, and limit of quantitation, well below the minimum acceptable hydrocarbon contamination.</li><li>The system robustness helps minimize maintenance for extended instrument uptime with consistent results over longer sequences.</li><li>The innovative HeSaver-H<sup>2</sup> Safer technology used on both channels provides a significant reduction of the helium gas consumption, also during sample analysis, extending 4.5-fold the cylinder lifetime and providing a significant cost savings to the laboratory.</li></ul>
