The integration of the LC system into a client-server environment allows the use of additional software, such as Agilent Sample Linking software and Agilent Sample Scheduler for OpenLab, enabling a complete workflow that links samples from any laboratory information management system (LIMS) to the vials and HPLC analysis results. This white paper highlights the benefits of the integrated analytical workflow. In this workflow, barcodes are used to track samples from the sample list to their vial position, and the presence of the correct sample is recognized without manually entering its position. This saves time and improves ease of use, with fewer errors over the complete workflow.</p><p><strong>Conclusion</strong></p><p>This white paper describes a workflow that begins with an analytical sequence initiated by any LIMS, then links the barcodes of sample containers to the barcodes of target pre-barcoded vials. The samples are analyzed to get the final data results, and a report is transmitted back to LIMS. This closed-loop, error-free, and time-saving workflow is enabled by Agilent Sample Linking software together with the Agilent Sample Scheduler for OpenLab. Samples can be tracked through the complete workflow using barcodes to transfer them accurately from their original containers to a final barcoded vial, managed completely by the Sample Linking software. This saves time, enhances ease of use, and results in fewer errors over the complete workflow.</p><h4><strong>3. Waters Corporation: </strong><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/720008468_en_03451ccb29.pdf"><strong>Improving USP Monograph Analysis Time Through Scaling by N on an HPLC System Using CORTECS™ Premier Columns</strong></a></h4><ul><li>Application</li></ul><p><strong>Abstract</strong></p><p>The United States Pharmacopeia General Chapter &lt;621&gt; allows certain changes to monograph methods. Changes in column length and particle size are employed as part of modernization efforts from HPLC to UPLC™ instrumentation. However, some modernization activities can be performed without switching LC systems. For HPLC analyses, reducing particle size from 5 μm to 3.5 μm can improve throughput and run times, however those columns increase backpressure of the system. Additionally, depending on the monograph, modernization may not be possible due to available column configurations, and the guidelines set in General Chapter &lt;621&gt; for scaling by L/dp. Modernizing by plate count (N) is one path forward, especially if the column being modernized to takes advantage of highly efficient particles. This application note scales the USP impurities assay of<br>zidovudine from the original 4.6 x 250 mm column using fully porous 5 μm particles to a 4.6 x 150 mm column using solid-core particles to improve both throughput and solvent usage.</p><p><strong>Benefits</strong></p><ul><li>USP Monograph for zidovudine impurities analyzed on various columns</li><li>Vastly improved separation efficiency using CORTECS Premier 5 μm columns with original USP monograph column configuration</li><li>40% improvement in solvent usage and up to 60% improvement in analysis run time using CORTECS Premier 5 μm Column</li></ul><p><strong>Introduction</strong></p><p>Monograph methods have traditionally employed the use of 5 μm particle size columns due to pressure limitations of older HPLC instrumentation. In combination with the larger particle stationary phases, longer column dimensions were needed in order to achieve appropriate column efficiency. This leads to lengthy analyses with high volumes of solvent being consumed. Monograph methods can be modernized however, and the United States Pharmacopeia (USP) outlines two routes for modernization as outlined in USP General Chapter &lt;621&gt;.1 The first is by changing particle size and column length (L/dp) which can be done but may require the use of more modern LC technology like UPLC. The second route to modernization involves scaling a column based on the plate count (N) for the assay. Changes can be made to either the L/dp or N by -25% and up to 50%. Modernizing by N allows a method to be changed to any configuration as long as the plate count for the new conditions falls within the limits outlined in General Chapter &lt;621&gt;.</p><p>For some assays, modernizing by N can be possible by using higher efficiency stationary phases, like solid-core particles. Solid-core particle columns, like CORTECS columns, have been shown in the past to boost separation efficiency due to particle morphology.2–4 By taking advantage of the solid-core particles, a shorter column may be used while still achieving acceptable plate counts. Thus, improvements to run time and solvent usage could<br>be possible without changing particle size, therefore avoiding the potential increases to system pressure accompanied with smaller particle sizes. In this application note, the assay and impurities monographs for zidovudine were scaled from a 250 mm column down to a 150 mm column using the modernize by N approach. Using the new CORTECS Premier 5 μm columns results in a 40% decrease in solvent usage with up to a 60% reduction in analysis time per sample.</p><p><strong>Conclusion</strong></p><p>Validated monograph methods often employ HPLC systems with larger particle size columns due to pressure limitations of the system. Modernization of these monograph methods can be performed under USP General Chapter &lt;621&gt; using either the length to particle size ratio (L/dp) or Plate Count (N). In either case, the new column configuration must be within -25% to 50% of the original conditions. Once a new column is selected, changes to gradient profile and flow rate are calculated. This application note focused on the assay and impurity test for zidovudine, which calls for a 250 mm column in the original monograph.</p><p>By using CORTECS Premier 5 μm Column the assay and impurities test for zidovudine was successfully scaled down to a 150 mm column while maintaining the particle size. This greatly improved sample throughput with a 40% reduction in run time and solvent usage. Further improvements to throughput were realized by increasing flow rate from 1.5 mL/min to 2.25 mL/min. The increased flow rate, while not acceptable under USP General Chapter &lt;621&gt;, is still able to be run due to the lower system pressure generated by the 150 mm column. A 60% reduction in run time is realized with the higher flow rate. CORTECS Premier 5 μm Columns offer highly efficient solid-core particles combined with MaxPeak Premier High Performance Surface Technology hardware.</p><h4><strong>4. Perfluorooctane sulfonic acid (PFOS) and perfluorooctanoic acid (PFOA) are representative compounds of PFAS. They are used in a wide range of applications, such as fire retardants, food packaging materials, and non-stick coatings, due to their water-repellent, oil-repellent, heat-resistant, and chemical-resistant properties. Due to their structural stability, PFAS widely remains in the environment. There are concerns about health risks caused by human ingestion of fish that have ingested seawater, river water, or feed contaminated with PFAS. Therefore, quantitative assessment of PFAS levels in fish should be important.</p><p>This application news introduces a quantitative analysis of PFAS in tuna fillets with LC-MS/MS. Thirty principal PFASs targeted by <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/companies/292"><strong>AOAC INTERNATIONAL</strong></a>, a North American organization that standardizes food testing methods and verifies analytical methods, were analyzed using newly created method, which was investigated from the pretreatment procedure then validated by recovery test. By reducing losses during the pretreatment and optimizing the analytical conditions, good recovery rates were obtained for all compounds.</p><p><strong>Conclusion</strong></p><p>This application news described a quantitative analysis of thirty PFAS targeted by AOAC INTERNATIONAL in tuna fillet. The analysis was performed using a quadrupole mass spectrometer LCMS-8060NX equipped with ultra-high performance liquid chromatograph Nexera X3 UHPLC system. Shim-pack Scepter, which provides good separation and peak shape, was used as the column. A spiked recovery test were conducted, and all compounds showed recovery rate within 80-120% and repeatability below 20% at spiked concentrations of 0.1, 1, 5 μg/kg. In particular, recovery rates of PFOS, PFOA, PFNA, and PFHxS were within 95.5-108.4% at all spiked concentrations. Using optimized pretreatment and analytical methods, accurate quantitation is possible from 0.1 μg/kg.</p><h4><strong>5. Thermo Fisher Scientific: </strong><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/AN_003215_Highly_sensitive_method_for_the_determination_of_12_nitrosamine_impurities_in_multiple_AR_Bs_the_class_of_sartan_drug_formulations_c2744b4c40.pdf"><strong>Highly sensitive method for the determination of 12 nitrosamine impurities in multiple ARBs, the class of sartan drug formulations</strong></a></h4><ul><li>Application</li></ul><p><strong>Application benefits</strong></p><ul><li>Single method applicable for highly sensitive quantitation of 12 nitrosamines in three ARB drug substances.</li><li>A robust gradient method with significant chromatographic separation between nitrosamines and ARB drug substances.</li><li>Better retentivity and resolution of nitrosamines and APIs with comparatively shorter runtime, ideal for high throughput screening.</li></ul><p><strong>Goal</strong></p><p>Development of a sensitive and robust LC-MS/MS method to quantitate the following 12 nitrosamine impurities in three ARB drug substances: N-nitrosodimethylamine (NDMA), N-nitroso-diethylamine (NDEA), N-ethyl-N-nitroso-2-propanamine (NEIPA), N-nitroso-diisopropylamine (NDIPA), N-nitroso-di-n-propylamine (NDPA), N-nitroso-methylphenylamine (NMPA), N-nitroso-di-n-butylamine (NDBA), N-nitroso-N-methyl-4-aminobutyric acid (NMBA), N-nitrosomorpholine (NMOR), N-nitrosopyrrolidine (NPYR), N-nitrosomethylethylamine (NMEA), and N-nitrosopiperidine (NPIP).</p><p><strong>Introduction</strong></p><p>Angiotensin receptor blockers (also called ARBs or angiotensin II inhibitors) are medicines that dilate blood vessels and are used in the treatment of high blood pressure (hypertension), heart failure, or kidney disease in people with diabetes.<sup>1</sup> These ARBs, also known as sartan medicines, have a specific tetrazole ring-containing structure whose synthesis could potentially lead to the formation of nitrosamine impurities.<sup>2</sup></p><p>If the content of nitrosamine impurities is found to be greater than the acceptable limit, then regulatory authorities need to be informed, and that may ultimately lead to the drug substance product being recalled. There have been numerous drug recalls associated with nitrosamine impurity levels exceeding the allowable limits in drug substances of various pharmacological categories, including ARBs.<sup>3</sup> In 2018 and in 2019, the U.S. Food and Drug Administration (FDA) found traces of NDMA and NDEA impurities in the ARB drug products valsartan, losartan, and irbesartan. Since July 2018, more than two dozen specific ARB products have been recalled owing to the unacceptable presence of potentially carcinogenic nitrosamine impurities.<sup>4</sup></p><p>In 2018, the FDA issued guidance to the industry on how to assess and control impurities. EMA finalized a review under Article 5(3) of Regulation (EC) No 726/2004 in June 2020 to provide guidance to marketing authorization holders on how to avoid the presence of nitrosamine impurities in human medicines.</p><p>The CHMP asked marketing authorization holders to review all chemical and biological human medicines for the possible presence of nitrosamines and test products at risk. Regulators across the globe have been vigilant on nitrosamine testing and subsequent modification of guidelines or testing procedures to ensure the best quality products reach the market. Recently, the FDA<sup>5</sup> and EMEA<sup>6</sup> updated guidelines with a more comprehensive and categorized approach towards nitrosamine testing.</p><p>In this application note, the goal was to develop an LC-MS/MS solution to the increased frequency of demands for higher sensitivity and quantification of an extensive array of nitrosamine impurities in pharmaceutical formulations with a one-method approach for multiple products. From the multiple techniques available today, liquid chromatography-tandem mass spectrometry is one of the best for highly sensitive and reproducible quantification of nitrosamines. It not only benefits sensitivity in sub-nanogram concentrations but also provides a more robust, linear, and reproducible solution for nitrosamine detection. The guidelines followed in this application are from the ICH Q2(R2)<sup>7</sup>, FDA<sup>5</sup>, and EMEA<sup>6</sup>.</p><p>As demonstrated by the excellent results observed in this study, the Thermo Scientific™ TSQ Altis™ Plus Triple Quadrupole mass spectrometer connected to the Thermo Scientific™ Vanquish™ Flex UHPLC system makes a suitable configuration for the highly sensitive analysis of multiple nitrosamines in a single method applicable for 3 ARBs—olmesartan, irbesartan, and valsartan. It is expected to have benefits such as soil improvement, increased agricultural productivity, and carbon sequestration in the soil.</p><p>In recent years, carbon sequestration using biochar has attracted attention as one of the technologies enabling carbon dioxide removal (CDR) from the atmosphere, resulting in active research in this field. In 2019, the IPCC (Intergovernmental Panel on Climate Change) improved the guidelines to include a method for calculating carbon sequestration resulting from the application of biochar to agricultural and grassland soils. Consequently, carbon sequestration with biochar has been globally recognized as a CDR technology.</p><p>Additionally, research and development of high-performance biochar that can achieve both high carbon sequestration efficiency and increased agricultural productivity is also underway to expand the use of biochar.</p><p>Such research requires the evaluation of the total organic carbon (TOC) content in biochar and the TOC solid sample measurement system, which consists of the Shimadzu TOC-L total organic carbon analyzer combined with an SSM-5000A solid sample combustion unit, can be utilized for that purpose.</p><p>This article presents an example of evaluating the TOC content of biochar using the Shimadzu TOC solid sample measurement system.</p><p><strong>Conclusion</strong></p><p>By using the TOC solid sample measurement system, it is possible to measure both TC and IC, and the TOC can be determined from the difference in their concentrations.</p><p>In this article, the TOC concentrations of five types of biochar made from different raw materials were measured, which confirmed that the TOC concentrations varied significantly depending on the raw material. The CV value was less than 2 %, indicating good repeatability. This system, which can easily and quickly measure the TC and IC amounts of solid samples, is useful for measuring the TOC content of biochar.</p><h4><strong>3. An automatized and reproducible acid digestion sample preparation combined with a sensitive and robust method based on triple quadrupole ICP-MS allows a wide range of trace elements present in the samples to be quantified without any compromise in respect to applicable regulations.</p><p><strong>Introduction</strong></p><p>Many countries have established their own regulations and guidelines to ensure the safety and quality of baby food products. These regulations are generally based on scientific research and risk assessments conducted by health and food safety authorities. Among a range of other contaminants, toxic heavy metals, such as lead, arsenic, cadmium, and mercury, are subject to screening. Infants and young children up to the age of three represent an especially vulnerable group susceptible to different illnesses and potential lifelong neurological damage through exposure to toxic heavy metals.</p><p>The Baby Food Safety Act of 2021 requires manufacturers and the FDA to take long overdue action by setting maximum levels of inorganic arsenic (10 μg∙kg-1, 15 μg∙kg-1 for cereal), lead (5 μg∙kg-1, 10 μg∙kg-1 for cereal), cadmium (5 μg∙kg-1, 10 μg∙kg-1 for cereal), and mercury (2 μg∙kg-1) allowed in baby food.</p><p>Since January 2022, baby food manufacturers need to meet these maximum levels for toxic elements by testing their final products (because only ingredient testing significantly underestimates toxic heavy metal levels in the final product), and to post the results of their product testing online twice per year. Similarly, as the FDA is working on a “closer to zero plan”, the European Union has established regulations and maximum limits for various contaminants, including toxic elements, in baby food.</p><p>Moreover, essential minerals like iron, zinc, copper, calcium, and magnesium are important for the healthy growth and development of infants. Monitoring their levels ensures that the baby food provides adequate nutrition. Depending on the specific formulation and ingredients of the baby food, additional trace elements such as selenium, manganese, chromium, and molybdenum may also be analyzed to assess their levels and potential impact on infant health.</p><p>ICP-MS (inductively coupled plasma mass spectrometry) is the commonly used analytical technique to measure trace elements in various samples, including baby food. Results: PROTAC drug metabolism was found to be extensive for the investigated compounds. Among the detected metabolites, several isomeric compounds were found where MSn fragmentation data could be used to reduce ambiguity in the transformation site localization, especially for metabolites with higher molecular weights.</p><h5><strong>Introduction</strong></h5><p>The study of a drug’s metabolism is an integral part of drug discovery and development, from improving drug exposure to flagging potential toxicity from its metabolites. The identification of drug metabolites routinely relies on LC/MS experiments, with fragmentation data enabling their structure elucidation. In recent years, hetero-bifunctional small molecule drugs, acting on a given protein of interest (POI) through recruitment of inherent protein degradation pathways rather than inhibition, have gained significant interest as new modalities, with multiple such proteolysis targeting chimera (PROTAC®) drug candidates being investigated in clinical trials.[1] However, due to PROTAC’s unique properties, the identification of their metabolites can be challenging.[2] Here, we describe the utility of multi-stage fragmentation (MSn) for the elucidation of transformation sites on PROTAC drug metabolites, based on analysis of the representative compounds shown in Figure 2.</p><h5><strong>Conclusions</strong></h5><ul><li>The utility of flexible MS data acquisition methods for metabolite identification could be demonstrated with two representative PROTAC compounds, MZ1 and dBET1, which exhibit extensive metabolism in vitro.</li><li>MSn fragmentation data helped to enhance the precision of transformation site localization, particularly for isomeric metabolites and those with high molecular weights.</li></ul><h4><strong>2. After that, the acquired data is analyzed and “preparation” for the subsequent analysis is carried out, followed by starting the next analysis again. The method development progresses by repeating these processes, but in addition to the significant time required to repeatedly create analysis schedules, expertise in chromatography is necessary to explore optimal conditions based on data analysis. In other words, typical method development requires “human intervention”. Therefore, eliminating human involvement and automating such method development processes would be desirable to improve labor efficiency. This article introduces an example of automatic optimization of gradient conditions to separate montelukast (a small molecule drug) and its related impurities using LabSolutions MD (<a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/c190_e309_96044bc345/c190-e309.pdf"><strong>Technical Report C190-E309</strong></a>), a dedicated software for supporting method development.</p><p><strong>Conclusion</strong></p><p>Automatic optimization of gradient conditions using AI algorithm of LabSolutions MD was applied to montelukast and its related impurity. As a result, gradient conditions that met the criteria (“resolution for montelukast and related impurity &gt; 3.0” and “elution time of the last peak &lt; 15 minutes”) were successfully explored. This result indicates that significant labor saving in method development can be expected by LabSolutions MD. This article introduces an automatic optimization of gradient conditions in method development while LabSolutions MD also supports a series of workflow of method development, including the screening phase and robustness evaluation phase. This setup makes it harder for users to control the LC and monitor what is happening with the instrument at the same time. Adding a tablet-style user interface to each LC solves this issue by bringing important controls and diagnostics to the user’s fingertips right at the instrument. The Agilent InfinityLab Assist not only fulfills these functions, but also incorporates automated, guided, and scheduled features that enable the user to perform daily tasks easier, faster, and with increased confidence.</p><p><strong>Introduction</strong></p><p>Laboratories with many different analytical instruments often have their instruments connected to a central data system that allows sample submission and data analysis from a remote PC. Using an Agilent OpenLab client/server system, multiple instruments can be controlled from a single client or Agilent Instrument Controller, which is not necessarily located at the same bench, or even in the same room as the instrument. This setup might be advantageous with respect to safety and optimal use of lab space. Tasks that require interaction with both the instrument and the controlling PC, however, become more tedious and error-prone if the user needs to swap positions constantly. A local user interface resolves this issue and adds confidence and convenience to instrument control. As mass spectrometry (MS) technology rapidly advances and with data acquisition methods such as data independent analysis (DIA) progressing, the ability to delve deeper into the metabolome and lipidome is significantly improved, whilst at the same time generating high-dimensional datasets. In this study, we describe a data processing pipeline which converts MS data (DIA and DDA based) that were collected using a novel, multi-reflecting ToF mass spectrometry into a generic file format, that can be readily processed by a variety of third-party informatic tools. Data were primarily processed via Skyline1 and MS-Dial2 for lipidomic datasets, whilst metabolomic datasets were processed using XCMS3.</p><p><strong>CONCLUSION</strong></p><ul><li>Data Convert provides a flexible and easy approach to generating mzML files from DDA and DIA acquisitions.</li><li>Integration of Data Convert into waters_connect allows a seamless workflow of automatically generating mzML during data acquisition, in addition to having flexibility for users to export mzML from projects at any point in time.</li><li>mzML files can be tailored for the intended third party informatics package (i.e., centroided, compressed etc).</li><li>Data from the benchtop Xevo MRT has demonstrated the workflow from data acquisition through to data processing with third party tools, such as Skyline, MS-Dial and XCMS using the waters_connect/Data Convert pipeline.</li><li>Metabolomic and lipidomic datasets highlight the high quality data that can be generated from the Xevo MRT mass spectrometer:<ul><li>Superior mass accuracy (sub 1 ppm)</li><li>High mass resolution which can be maintained with fast, high throughput LC gradients</li><li>High levels of sensitivity/dynamic range</li></ul></li></ul>

These chemicals are colorless, odorless, and water-soluble. There have been multiple incidents involving the deaths of children due to medicinal cough syrups contaminated with these impurities. These impurities often originated from the raw materials (such as glycerin, propylene glycol, and sorbitol solutions) or excipients used in their formulation (1-2). Substandard and falsified raw materials frequently enter the excipient supply chain, causing significant challenges for drug manufacturers and egulatory bodies. The complexity of the global supply chain, which often involves importing raw materials from various countries, worsened this issue. Therefore, it is crucial to frequently test these raw materials upon receipt to prevent the reoccurrence of such incidents and protect public health.</p><p>This application news demonstrates the use of GC-FID for analyzing EG and DEG contaminants in glycerin, propylene glycol (PG), and sorbitol solutions (70%) (both crystallizing and non-crystallizing) in accordance with the Indian Pharmacopoeia (IP) monographs. Shimadzu’s Brevis GC, featuring a compact design with a width that is approximately 35% smaller than existing GC models, is used in this experiment (Fig. 1). This design allows the Brevis GC to occupy less space in the laboratory and to be eco-friendly with reduced power consumption, while maintaining analytical performance and enhancing productivity.</p><p><strong>Conclusion</strong></p><p>In this study, the analysis of EG and DEG in three types of raw materials was successfully performed using the compact Brevis GC coupled with AOC-30i. Despite its small size, the Brevis GC demonstrated excellent capability in analyzing EG and DEG impurities in the raw materials according to the IP monographs, making it an ideal and more eco-friendly solution for this application.</p><h4><strong>2. In many cases, due to previously existing methodologies, samples need to undergo several analyses on different instrument configurations, with either specific consumables, including the analytical column or a dedicated detector. This leads to a variety of protocols and potential instrument setups required in a laboratory, some of which must be run in a dual-column configuration or a confirmation on a separate instrument using a second injection is required.</p><p>While mass spectrometric detection provides a much higher selectivity than analog detectors, only triple quadrupole systems provide the selectivity needed to effectively leverage selected rection monitoring (SRM) as a tool for method standardization and consolidation. The power of triple quadrupole mass spectrometry can be easily used to modernize existing workflows. Laboratories can reduce their need for instrument-specific consumables and components on-hand to reduce costs and complexity with a single instrument configuration. This application demonstrates the use of a single hardware configuration for the analysis of different environmental contaminants.</p><p><strong>Conclusion</strong></p><p>The utilization of a single configuration GC-MS/MS for multiple environmental methods has been demonstrated in this application note. As well as being able to meet the regulatory requirements for different compound classes, there are advantages to using a single configuration for multiple analysis:</p><ul><li>Reduction in operation cost, including reduced training needs and centralization of consumables</li><li>Ability to react and be ready to run the required method without lengthy set up or changing instrument analytical column and hardware</li><li>Increase in instrument capacity as all systems can run all methods</li></ul><p>The Thermo Scientific GC-MS/MS system also increases productivity:</p><ul><li>The TRACE 1610 GC has a unique module design allowing injectors and detectors to be exchanged by the user and maintenance to be performed rapidly offline. The TSI injector allows a wider range of compound classes with different volatilities to be covered in a single injection. The easy-to-use touch screen provides real-time monitoring of the system and video tutorials for quick adoption.</li><li>The TSQ 9610 GC-MS/MS is equipped with Thermo Scientific™ NeverVent™ technology, which allows the GC column to be exchanged, the ionization source to be removed, and filaments to be removed without the need for breaking vacuum. This allows routine maintenance to be performed in minutes and increases system uptime drastically.</li></ul><p>In summary, this single GC-MS/MS configuration can greatly simplify the workflows of environmental laboratories and increase efficiency and productivity, reduce costs, and maximize revenue.</p><h4><strong>3. Shimadzu: </strong><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/ap_aplnote96_en_5f0a892b7f.pdf"><strong>Study on the Mechanism of Early Delivery by Multi-Omics Analysis of Metabolites, Elements and Bacterial Flora in Amniotic Fluid</strong></a></h4><ul><li>Application</li></ul><p><strong>Abstract</strong></p><p>A gas chromatograph mass spectrometer (GCMS-TQ™8040 NX) and an energy-dispersive X-ray fluorescence spectrometer (EDX-7200) were used to analyze metabolites (488 components) and elements (78 elements) contained in the amniotic fluid of pregnant women. These data were combined with amniotic fluid microbiota (approximately 250 species of microorganisms) measured using a miniature next-generation sequencer<br>(MinION Mk1C System, Oxford Nanopore Technologies, Inc.) and 16 items of labor data (e.g., neonatal weight) to analyze differences between preterm and normal births. Volcano plot and metabolic pathway analyses were performed using Multi-omics Analysis Package based on the 740 items detected. In this application, we introduce an example of multi-omics analysis conducted by gender and time series.</p><p><strong>Introduction</strong></p><p>Preterm delivery refers to delivery at a gestational age of 22 weeks or less than 37 weeks, which can have a significant impact on the health and development of the newborn and the life of the family. This problem occurs worldwide: in 2020, 1/10 of all births were preterm<sup>1)</sup> (Fig. 1). Prematurity is the leading cause of early childhood mortality, so there is an urgent need to promote research on preterm birth.</p><p>However, the global rate of premature birth reduction from 2010 to 2020 was only 0.14 %, indicating that the birth cycle has not improved. It is known that preterm birth is closely related to amniotic fluid, and it is used as a research object in preterm birth<sup>2)</sup>. Amniotic fluid is approximately 500 mL of water, contains fetal urine, sloughed cells, inflammatory substances, etc., and is a valuable source of information for understanding<br>fetal growth and health<sup>3)</sup>. Measuring metabolites in amniotic fluid helps assess the fetus's metabolic activity and nutritional status. Elemental measurements can also determine the extent of fetal development and environmental exposure. It is also essential to measure the bacterial flora in the amniotic fluid. It has been found that about 80 % of preterm births occur in pregnant women who have a chain of bacterial infections from the vagina to the uterus and from the uterus to the amniotic fluid <sup>4)</sup>.</p><p>In this study, we used a gas chromatograph mass spectrometer GCMS-TQ8040 NX, an energy-dispersive X-ray fluorescence spectrometer EDX-7200, and a miniature next-generation sequencer MinION Mk1C to measure primary metabolites, elements, and flora in the amniotic fluid of preterm and term pregnant women (Fig. 2). These data were combined with 16 birth delivery data items (e.g., female age, newborn weight, etc.),<br>and the differences in amniotic fluid content between preterm and term births were visualized using Multi-omics Analysis Package.</p><p><strong>Conclusion</strong></p><p>Amniotic fluid from 6 pregnant women (3 preterm births and 3 term births) was measured using a next-generation sequencer (MinION Systems, Oxford Nanopore Technologies, Inc.), a gas chromatography-mass spectrometer (GCMS-TQ8040 NX), and an energy-dispersive X-ray fluorescence spectrometer (EDX-7200) to evaluate the intestinal microbiota (approximately 250 species of microorganisms), metabolites (approximately 500 components), and elements (approximately 80 elements). Metabolic pathway analysis was performed using Multi-omics Analysis Package based on approximately 700 items detected.</p><p>In metabolic pathway analysis, not only the accumulated amount of metabolites, but also the ratio of product/substrate or its reciprocal substrate/product was automatically created in Multi-omics Analysis Package and used as a new variable to be read into class cluster analysis, etc., thereby enabling statistical analysis from a perspective different from the accumulated amount.</p><h4><strong>4. Cosmetic products are complex mixtures, which can be challenging to analyze due to the presence of many compounds that could cause chromatographic and spectral interferences.</p><p>In this application note, an Agilent 8890 GC with a dual-column setup and an Agilent 7000E GC/TQ provide high selectivity for the analysis of 57 allergen fragrance substances and their relevant isomers in cosmetic products. This GC/TQ method is quickly and easily transferred from an existing gas chromatograph/mass selective detector (GC/MSD) method using Agilent MassHunter Optimizer software. The high selectivity and reliable quantitation of the GC/TQ method increase confidence in data by reducing uncertainty caused by coelution and therefore the risk of false positives and negatives.</p><p><strong>Introduction</strong></p><p>Allergens include diverse compounds, both synthetic and natural, that cause allergic reactions in sensitive people. Allergens can be found in many cosmetic products, including shower gels, soaps, shampoos, creams, makeup, etc. These compounds are often not intentionally added to cosmetics<br>during manufacturing, but are substances contained in the base ingredients that make up the final product. This work focuses on a method to determine allergens commonly found in fragrance substances.</p><p>Regulating agencies around the world have—and are frequently updating—regulations around allergens in consumer products, like cosmetics and fragrances. In 2012, the European Scientific Committee on Consumer Safety (SCCS) identified 87 fragrance substances that can cause allergies in humans. So, in 2023, the European Commission published Regulation 2023/1545, amending Regulation (EC) Number 1223/2009 of the European Parliament and of the Council, concerning the labeling of allergenic substances in cosmetic products.<sup>1</sup> The new regulation expanded the historical list of 26 allergens to 87 compounds.</p><p>These regulations are not limited to Europe. The Modernization of Cosmetics Regulation Act of 2022 (MoCRA) is the most significant expansion of the U.S. Food and Drug Administration’s authority to regulate cosmetics since the Federal Food, Drug, and Cosmetic (FD&amp;C) Act was passed in 1938.<sup>2</sup> These updated laws and regulations help to ensure the safety of cosmetic products. To determine the presence of allergens in cosmetics, a sensitive and selective analytical technique is required, such as analysis by GC/TQ.</p><p><strong>Conclusion</strong></p><p>Migrating an established GC/MSD-based allergen analysis to GC/TQ is quick and easy, and it improves method selectivity. The same chromatographic and MS source method parameters, columns, and consumables can be used for the new GC/TQ method. For the new MS acquisition method, Agilent MassHunter Optimizer software can be automated to determine optimal MRM transitions for the GC/TQ method using the SIM ions from the original GC/MSD method. The resulting improved GC/TQ method selectivity increases confidence in data, reducing the risk of false positives and negatives for coeluting target allergens. The new GC/TQ method in this work provides reliable quantitation for the analysis of 57 allergen fragrance substances and their relevant isomers in cosmetic products using an Agilent 8890 GC with an Agilent 7000E GC/TQ.</p>

Data was acquired using Thermo Scientific™ Qtegra™ Intelligent Scientific Data Solution™ (ISDS) Software and processed using Iolite™ Laser Ablation Data Reduction Software.</p><p><strong>Results:</strong> A series of tooth samples containing a range of elements in localized phases was analyzed to demonstrate the ease of coupling to a commercially available high-speed mapping laser ablation system, ease of dwell-time optimization for best signal-to-background ratio and image contrast across all scanned elements, and finally fast analysis times from sampling to image.</p><h5><strong>CONCLUSIONS</strong></h5><p>Through careful optimization of laser ablation and ICP-MS parameters, it is possible to generate high quality multi-elemental images at a fast rate. The use of new low-dispersion laser ablation cells and aerosol transfer technology is not limited to simultaneous mass spectrometers and is also applicable to sequential scanning mass spectrometers such as Quadrupole ICP-MS.</p><p>The major limitation of coupling low-dispersion Laser Ablation systems to sequential mass spectrometers is the limited number of elements that can be scanned; however, this is offset by the sensitivity and the specificity of, especially, triple quadrupole ICP-MS systems for the reduction of interferences on select analytes.</p><p>Most imaging experiments can be reduced to three or four analytes of interest, and in these cases LA-ICP-MS coupled to Triple Quadrupole ICP-MS using a low-dispersion, fast imaging setup can provide the analytical data quality required to answer challenging research questions.</p><h4><strong>2. For samples analyzed at large angles of incidence (AOI), such as bandpass and edge filters (e.g., beam splitters), maintaining a high degree of beam collimation is necessary to achieve optimal results. This control is achieved by adjusting the incident beam using apertures within the spectrophotometer that can be customized to meet the specific requirements of the application.</p><p>In this work, an Agilent Cary 7000 UV-Vis-NIR spectrophotometer that includes the Agilent Cary Universal Measurement Accessory (UMA) was used. The degree of beam collimation was investigated using different apertures within the UMA to enhance data quality for the determination of the edge steepness of an optical filter.</p><p><strong>Conclusion</strong></p><p>Characterizing the edge steepness of optical filters is an important measurement for manufacturers and users of these types of devices. The steeper the edge, the better the quality of the filter, so careful determination of edge steepness is required to differentiate between high and low quality filters. Accurate measurements depend on the half cone angle of the apertures used in spectrophotometers to control the degree of collimation.</p><p>This study has shown that the degree of collimation on the Agilent Cary 7000 Universal Measurement Spectrophotometer (UMS) that includes the Agilent Cary Universal Measurement Accessory (UMA) can be controlled precisely using different horizontal apertures ranging from 0.25 to 3°.</p><p>Exploring the use of higher f-number optics in the incident beam of the UMA can lead to improved sensitivity and accuracy in various spectroscopic applications. This performance was demonstrated by the investigation of the edge steepness of a beam splitter. The best data were acquired by the Cary UMA with collimation control using a 0.25° (f/35) horizontal aperture.</p><p>The UMA with optimized and controlled beam collimation enables the complete characterization of optical samples, ensuring optimal performance of final components.</p><h4><strong>4.
