Found in products like firefighting foams, non-stick cookware, and water-resistant fabrics, PFAS are now recognized as major environmental and health concerns. These substances accumulate in soil, water, and living organisms, including humans, where they may cause serious health impacts over time.</p><p>Given their potential for long-term contamination, the detection and quantification of PFAS are critical. Advanced analytical methods, particularly LC-MS/MS with Multiple Reaction Monitoring (MRM), offer the specificity and sensitivity needed to accurately measure PFAS concentrations. However, one of the primary challenges in PFAS analysis is minimizing background contamination from PFAS residues within the LC system and during sample collection and preparation. Effective strategies to reduce these sources of contamination are essential for reliable results in PFAS quantification.</p><p>n this application news, we describe a single laboratory validation study with a rapid extraction of trace level concentrations of 32 PFAS (Table 1) in infant formula (baby food) involving the use of the QuEChERS as an extraction technique followed by mass spectrometric analysis using a Shimadzu LCMS-8060NX triple quadrupole mass spectrometer coupled to a Shimadzu Nexera X3 liquid chromatograph (Figure 2).</p><p><strong>Conclusion</strong></p><ul><li>A simple, sensitive, and rapid LC-MS/MS method has been developed to quantify 32 PFAS compounds in infant formula.</li><li>This method utilizes a QuEChERS-based extraction technique for efficient sample preparation.</li><li>With Shimadzu’s LC-MS/MS, the method demonstrated high sensitivity and reproducibility, achieving low relative standard deviations (RSDr and RSDR) in accordance with AOAC SMPR standards at trace levels.</li><li>This approach offers a reliable solution for multi-PFAS analysis in infant formula, making it highly applicable for testing laboratories focused on food safety.</li></ul><h4><strong>3. The choice between direct injection and automated sample preparation depends on the instrument's dynamic range, detection limits, and matrix complexity. Complex matrices like waste/industrial water, soil, food, and biological samples require specific preparation techniques to mitigate matrix effects and ensure accurate quantification. Techniques such as Dispersive Liquid-Liquid Microextraction (DLLME) can significantly improve concentration factors and reduce matrix interference, enhancing overall method performance. Additionally, the choice of the<br>analyzer is a critical consideration.</p><p><strong>Methods: </strong>Direct injection of water sample was performed on Thermo Scientific TSQ Altis Plus triple quadrupole mass spectrometer. For those sample prepared with automated dispersive liquid-liquid microextraction (DLLME), acquisition was performed on a Thermo Scientific Orbitrap Exploris MX high resolution mass spectrometer.</p><p><strong>Results: </strong>The methods presented achieves high-level sensitivity for PFAS analysis in drinking water, detecting compounds in the low ng/L range. Using DLLME, 56 PFAS compounds were quantified to low part per trillion levels from just 15 mL of sample. With a FAPAS®-Drinking Water proficiency test, we validated the<br>three strategies for PFAS analysis: direct injection on TSQ Altis Plus and the DLLME extract injected on both Orbitrap Exploris MX and Thermo Scientific Orbitrap Exploris GC.</p><h5><strong>Conclusions</strong></h5><ul><li>Compared to traditional SPE workflows, direct injection improves sample throughput in laboratories by significantly reducing the required sample preparation.</li><li>The DLLME method is a promising technique for extracting and pre-concentrating PFAS from drinking water and various other matrices. It is widely used for quality control of pharmaceuticals and biologics, research and medical purposes (1). Over the last several decades the gold standard of analysis for these areas of science has been High-Performance Liquid chromatography (HPLC). Typical HPLC systems operate within a 6000 psi pressure limit (2). Consequently, they are commonly paired with columns packed with 3 - 5 μm particles. Although smaller particle-packed columns allow faster separations, their use in HPLC is restricted to shorter lengths due to elevated backpressures (3). Additionally, established methods which are often used in a quality control setting may restrict alterations to particle size or column configuration.</p><p>Improved column efficiency on an HPLC system can be achieved using solid-core particles. Solid-core particles have a non-porous “solid” center and a porous outer layer(4). Columns packed with solid-core particles offer distinct advantages over those packed with fully porous particles, including higher efficiency and reduced operating backpressure. Waters offers a line of solid-core particle columns known as CORTECS Columns.</p><p>By utilizing CORTECS Columns, researchers can increase separation efficiency, achieve faster analysis times, reduced solvent waste and decreased column back<br>pressure. In this poster, we delve into the significance of larger solid-core particles for scalability. We compare columns packed with 5 μm CORTECS Prototype Material, CORTECS UPLC μm) Material and CORTECS HPLC (2.7 μm) C18 Material with columns packed with fully porous particles. Additionally, we explore how CORTECS Columns differ from similar offerings on the market.</p><p><strong>CONCLUSION</strong></p><ul><li>5 μm CORTECS Prototype Columns have 28% lower back pressure in comparison to 3.5 μm XBridge BEH C18 Columns.</li><li>5 μm CORTECS Prototype Columns have 14% higher optimal efficiency in comparison to 3.5 μm XBridge BEH C18 Columns.</li><li>CORTECS Columns are scalable across all 3 particle sizes shown.</li><li>In comparison, Vendor H and A columns are not scalable across all 3 particle sizes due to the significantly lower retention in the 4 and 5 μm particle size columns, shown in Figure 3.</li></ul><h4><strong>5. DCA allows researchers to identify key electrochemical processes, phase transitions, and degradation mechanisms occurring within the battery during charge and discharge cycles. This technique is particularly valuable for characterizing complex systems such as lithium-ion batteries, where multiple electrochemical reactions may overlap.</p><p>This Application Note explores the principles and practical applications of differential capacity analysis, highlighting its role in improving battery performance.<br>This application also reveals how INTELLO, with its dedicated suite of battery-oriented commands and its ability to untether during long measurements, is an excellent choice for battery researchers looking to maximize their time and efficiency when conducting DCA.</p><p><strong>INTRODUCTION</strong></p><p>Within the INTELLO battery cycling environment, it is possible to sample the differential capacity (defined as dQ/dE = |Qn+1-Qn|/(En+1-En), where n is the index of the data point, Q is the value of the charge/discharge capacity, and E is the measured working electrode potential) and plot by default this value versus the measured voltage of the cell. Agilent Technologies: </strong><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/5991_9356_EN_590f86441b.pdf"><strong>Determination of Carbonate Solvents and Additives in Lithium Battery Electrolyte Using the Agilent 5977B GC/MSD</strong></a></h4><ul><li>Application</li></ul><p><strong>Abstract</strong></p><p>This application note describes a method for determining carbonate solvents and additives in lithium battery electrolyte using the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=1&amp;search=5977B"><strong>Agilent 5977B</strong></a> single quadrupole gas chromatography/mass selective detector (GC/MSD). In this method, direct liquid injection is adopted. At a split ratio of 20:1, the target compounds achieved good linearity in a concentration range of 10–500 mg/L. This method has excellent reproducibility, and the instrument detection limits (IDL) for all of the 15 target compounds analyzed were below 1.3 mg/L. During the analysis of the actual electrolyte samples, diluted injection can be used to accurately quantify the target compounds and qualitatively identify unknown additives or impurities.</p><p><strong>Introduction</strong></p><p>Lithium battery electrolyte is the carrier of ion transport in a lithium battery, which is generally composed of lithium salt and organic solvent. In the electrolyte, the commonly used lithium salt is LiPF<sub>6</sub> , and the solvent is a binary, ternary, or multinary system composed of a mixture of cyclic carbonates and chain carbonates. Selecting the right organic electrolyte is the key to achieving a higher energy density, a longer cycle life, and greater battery safety. Lithium battery manufacturers also add specific additives to extend battery life. Therefore, studies on the composition of lithium battery electrolyte play an important role in the development of new lithium batteries.</p><p>GC/MS analysis is commonly used in the studies on lithium battery electrolyte, and can accurately and quantitatively analyze the major components of the organic solvent in the electrolyte. Meanwhile, the strong qualitative capacity of mass spectrometry can also qualitatively identify and analyze unknown additives and impurities.</p><p>This application note describes the method for determining carbonate and carboxylate solvents and additives in lithium battery electrolyte using the 5977B GC/MSD. In regard to the fact that LiPF<sub>6</sub> , the main component of the electrolyte, is unstable and can easily decompose, diluted injection was adopted, which ensured good sensitivity, linearity, and reproducibility.</p><p><strong>Conclusion</strong></p><p>In this application note, a method for analyzing carbonate solvents and additives in lithium battery electrolyte was developed with an Agilent 7890 GC and Agilent 5977B GC/MSD. The method is easy to use, and can achieve great separation, a wide linear range, and excellent reproducibility and sensitivity<br>for various components in the electrolyte, making it very suitable for qualitative and quantitative analysis of organic solvents, additives, and impurities in lithium battery electrolyte.</p><h4><strong>2. To address this problem, hydrogen gas is being used as an alternative carrier gas. The characteristics of hydrogen gas are different from those of helium gas, however, and methods using helium gas must be adjusted to appropriate conditions for hydrogen gas. We have developed a quantitative analysis method for 216 pesticides with GC-MS/MS using hydrogen gas. Using this method, good recovery results were obtained in food samples.</p><p><strong>Conclusions</strong></p><p>Using hydrogen gas as an alternative carrier gas, we simultaneously analyzed 216 food-borne agrochemicals using an ultra-sensitive triple-quadrupole mass spectrometer <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=3&amp;search=GCMS-TQ8050+NX"><strong>GCMS-TQ8050 NX</strong></a> combined with AOC-30i and AOC-20s U as injectors.</p><p>Good recovery rate within 70 ~ 120% were obtained for 70% to 74% of the compounds. This system enables simultaneous analysis of multiple pesticides with high sensitivity and accuracy.</p><h4><strong>3. Metrohm: </strong><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_nir_121_download_0bb1185dc9.pdf"><strong>Water content in propylene glycol monomethyl ether (PGME)</strong></a></h4><ul><li>Application</li></ul><h5>Water determination possible within seconds using NIRS</h5><p>Propylene glycol monomethyl ether (1-methoxy-2-propanol, or PGME) is one of many glycol ether solvents with a wide variety of applications. It is used as an intermediate and in formulations for industrial, professional, or consumer applications, mainly in surface coatings, inks for printing, cleaning solutions, deicing/anti-icing formulations, and agrochemical purposes. It is also used as an extractant and as a coalescing agent and flow improver in water-based paints.</p><p>Water in propylene glycol methyl ether is usually measured by Karl Fischer (KF) titration which requires chemicals and takes about five minutes per determination. This Application Note describes how near-infrared spectroscopy (NIRS) can be used as a faster and more cost-efficient alternative for water determination in PGME.</p><p><strong>CONCLUSION</strong></p><p>This Application Note demonstrates the feasibility to determine a key parameter for the quality control of propylene glycol monomethyl ether (water content) with NIR spectroscopy. The main advantages of NIR spectroscopy over wet chemical methods are that running costs are significantly lower and time-to-result is significantly reduced. Additionally, no chemicals are required, and the technique is non-destructive to samples.</p>

Several scientific studies have provided evidence supporting the importance of element analysis in various aspects of healthcare, whether it can be nutritional assessment, toxic element exposure or medical conditions.</p><p>In recent times, ICP-MS is state of the art choice for routine analysis of elements in blood serum. Different aspects like robustness and throughput form the principles of this method, while not compromising reliability.</p><p>Important aspects to take into consideration are highlighted within this note, especially aiming to reduce sample volume and measurement time, allowing to quantify more than 20 elements in ~2.0 min. while avoiding more complex sample injection techniques.</p><p><strong>Conclusion</strong></p><p>For routine use, the ICPMS-2040 LF is highly suitable for analyzing trace elements in human and veterinary blood serum. The method allows for the measurement of samples from extremely low serum volumes.</p><p>To measure Se more sensitive and without double charged ion interferences of Gd, ICPMS-2050 LF is the instrument of choice.</p><p>More than 20 elements can be measured at a rate of 30 samples per hour. The low consumption of argon and helium, combined with reduced power consumption for operating the Minitorch plasma and short sample cycle time, makes it exceptionally cost-effective without compromising sensitivity.</p><p>Besides the mentioned elements, further ones like nickel (Ni), chromium (Cr) and manganese (Mn) have been successfully evaluated in shorter batches and can be simply added to make the method once more universal.</p><h4><strong>2. Thermo Fisher Scientific: </strong><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_002881_tea_icap_msx_icp_ms_water_analysis_epa6020b_an002881_em_en_f457bce5d2.pdf"><strong>Robust analysis of a variety of water, wastewater, and soil samples according to U.S. EPA Method 6020B (SW-846)</strong></a></h4><ul><li>Application</li></ul><p><strong>Goal</strong></p><p>To evaluate and demonstrate performance of the Thermo Scientific™ iCAP™ MSX ICP-MS for robust analysis of a variety of water, wastewater, and soil samples according to the requirements of U.S. EPA Method 6020B.</p><p><strong>Introduction</strong></p><p>As a result of industrialization and manufacturing, agriculture and farming, and population growth, various types of solid and liquid wastes are introduced or released into the environment that could be hazardous to humans and the ecosystem. Although there are laws, regulations, and measures in place to minimize and control disposal and release into the environment, wastes reduction from anthropogenic sources will continue to be a challenge. The United States Environmental Protection Agency’s (EPA) Office of Land Management and Emergency Response (OLEM) includes the Office of Resource Conservation and Recovery (ORCR) which implements the Resource Conservation and Recovery Act (RCRA). The RCRA gives the EPA the authority to control the generation, transportation, treatment, storage, and disposal of hazardous waste. To do this, the EPA developed regulations, guidelines, and policies for proper and safe management and cleanup of hazardous wastes and programs for pollution prevention and recycling:</p><ul><li>Conserve energy and natural resources by recycling and recovery</li><li>Reduce or eliminate waste</li><li>Clean up waste that may have spilled, leaked, or was disposed of improperly</li></ul><p>The EPA developed various test methods for the analysis of contaminants in environmental samples, which can be found in the publication “Test Methods for Evaluating Solid Waste: &nbsp;Physical/Chemical Methods,” also known as SW-846. This is the EPA’s official compendium of analytical and sampling methods that have been evaluated and approved for use by waste management programs to comply with RCRA regulations. The SW-846 compendium functions primarily as a guidance document setting forth acceptable, although not required, methods for the regulated and regulatory communities to use in response to RCRA-related sampling and analysis requirements. SW-846 is a multi-volume document that changes over time as new information and data are developed. This application note will discuss the workflow developed for the analysis of different types of water, wastewater, and digested soil samples using the iCAP MSX ICP-MS, which offers a comprehensive solution for effective and reliable analysis of high dissolved solid containing samples. Thermo Scientific™ Qtegra™ Intelligent Scientific Data Solution™ (ISDS) Software was used to control the ICP-MS instrument and to generate, process and report analytical data, ensuring that the entire workflow meets the requirements specified in EPA Method 6020B, including quality control samples. To verify the consistent performance of the instrument over time, a sequence of 240 samples was repeated over two consecutive days resulting in a total of 480 samples analyzed.</p><p><strong>Summary</strong></p><p>The iCAP MSX ICP-MS was extensively tested for compliance with EPA Method 6020B. The quality of the analytical data obtained over two consecutive days of measurements demonstrated that the built-in Argon Gas Dilution system for controlled and automatic dilution of the sample aerosol is a powerful solution for laboratories analyzing demanding samples, such as soil digests or wastewater, under high-throughput conditions. The overall performance of the instrument suggests that reliable analysis of these types of samples can be performed without need of any maintenance and with no instrument downtime over three or more days of analytical work. Some of the important outcomes of this study are summarized below:</p><ul><li>All the requirements of EPA Method 6020B were met during the test period of two days, enabling the analysis of a total of 480 samples.</li><li>The instrument detection limits (IDLs) and lower limits of quantification (LLOQs) achieved met and exceeded the requirements given in the method, which suggests that the employed methodology, with its optimized argon gas sample dilution, is suitable for achieving the required robustness and instrument sensitivity for these types of samples.</li><li>Results observed during analysis of ICB and CCB QC standards indicate that the proposed method ensures minimum carryover between samples, enabling trouble-free measurement of high matrix samples across the full calibrated concentration range.</li><li>The accuracy obtained for ICV and CCV standard solutions ensures the reliability and consistency of instrument performance while analyzing challenging high TDS containing samples such as wastewater and solid waste digests.</li><li>The analytical data obtained during analysis of interference check solutions (ICSA and ICSAB) highlight the effectiveness of single KED mode using helium as collision gas in removing potential polyatomic interferences on each analyte, ensuring interference-free analysis every time.</li><li>The data obtained during analysis of matrix spiked and duplicate measurements demonstrate that minimal or no matrix effect was encountered when analyzing complex matrices such as wastewater.</li><li>The observed behavior of the internal standards on two consecutive days of analysis highlights the robustness and consistency of the instrument performance. The consistent internal standard readback within the range of 80–120% suggests that the developed methodology is a reliable solution for effective handling of high TDS containing samples with no impact from the matrix content.</li><li>Qtegra ISDS Software provides all the necessary tools, including different QC functions, automatic calculations and limit and flag functionality, to ensure that the analysis is performed as per the compliance requirements of EPA Method 6020B.</li></ul><h4><strong>4. Metrohm: </strong><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_nir_121_download_0bb1185dc9.pdf"><strong>Water content in propylene glycol monomethyl ether (PGME)</strong></a></h4><ul><li>Application</li></ul><h5>Water determination possible within seconds using NIRS</h5><p>Propylene glycol monomethyl ether (1-methoxy-2-propanol, or PGME) is one of many glycol ether solvents with a wide variety of applications. It is used as an intermediate and in formulations for industrial, professional, or consumer applications, mainly in surface coatings, inks for printing, cleaning solutions, deicing/anti-icing formulations, and agrochemical purposes. It is also used as an extractant and as a coalescing agent and flow improver in water-based paints.</p><p>Water in propylene glycol methyl ether is usually measured by Karl Fischer (KF) titration which requires chemicals and takes about five minutes per determination. This Application Note describes how near-infrared spectroscopy (NIRS) can be used as a faster and more cost-efficient alternative for water determination in PGME.</p><p><strong>CONCLUSION</strong></p><p>This Application Note demonstrates the feasibility to determine a key parameter for the quality control of propylene glycol monomethyl ether (water content) with NIR spectroscopy. The main advantages of NIR spectroscopy over wet chemical methods are that running costs are significantly lower and time-to-result is significantly reduced. Additionally, no chemicals are required, and the technique is non-destructive to samples.</p>

<p><strong>In the study published in the </strong><i><strong>Food Chemistry</strong></i><strong> journal, researchers from Ningbo University, China, &nbsp;the University of Melbourne, Australia, Walailak University, Thailand, and Wilmar (Shanghai) Biotechnology Research and Development Center Co Ltd., China investigated the impact of gestational diabetes mellitus (GDM) on the metabolite profile of mature human milk (MHM) using integrated </strong><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?instrumentationItems=3"><strong>GC–MS</strong></a><strong> and </strong><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?instrumentationItems=31"><strong>LC-MS</strong></a><strong> analysis.</strong></p><p>A total of 268 differentially expressed metabolites were identified, with significant increases in linoleic acid, arachidonic acid, 9R-HODE, and L-glutamic acid, and a decrease in 12,13-DHOME in MHM from women with GDM. These metabolites are linked to key pathways such as linoleic acid metabolism, fatty acid biosynthesis, galactose metabolism, and ABC transporters, which are associated with insulin resistance and impaired glucose metabolism. Integrated Gas Chromatography-Mass Spectrometry (GC–MS) and Liquid Chromatography-Mass Spectrometry (LC-MS) were employed for comprehensive identification and comparison of metabolites in mature human milk (MHM) from women with and without GDM. A total of 268 differentially expressed metabolites (DEMs) were identified. Among these, linoleic acid, arachidonic acid, 9R-HODE and L-glutamic acid were significantly elevated and 12,13-DHOME was significantly decreased in MHM of women with GDM. These metabolites are significantly enriched in linoleic acid metabolism, fatty acid biosynthesis, galactose metabolism and ABC transporters pathways. Disorders in these metabolic pathways are associated with insulin resistance and poor glucose metabolism indicating these conditions may persist postpartum.</span></p><h3><strong>2. Materials and methods</strong></h3><h4><strong>2.1. Materials</strong></h4><p>Methanol, acetonitrile and formic acid were purchased from Fisher Scientific (Hampton, NH, USA). L-2-chlorophenylalanine was purchased from Shanghai HC Biotech Co., Ltd. (Shanghai, China). Methoxylamine hydrochloride pyridine solution was purchased from Shanghai Macklin Biochemical Co., Ltd. (Shanghai, China). BSTFA was purchased from TCI (Shanghai) Development Co., Ltd. (Shanghai, China). N-hexane was purchased from CNW Technologies GmbH (Dusseldorf, Germany). Methyl octanoate (C8:0) and methyl palmitate (C16:0) were purchased from Dr. Ehrenstorfer GmbH (Augsburg, Germany). Methyl nonanoate (C9:0), methyl decanoate (C10:0), methyl laurate (C12:0), methyl myristate (C14:0), methyl stearate (C18:0), methyl eicosanoate (C20:0), methyl docosanoate (C22:0) and lignoceric acid methyl ester (C24:0) were purchased from Nu-Chek Prep, INC (Minnesota, USA).</p><h4><strong>2.2. Mature human milk collection</strong></h4><p>Mature human milk (3–6&nbsp;months postpartum) was collected from twenty women (GDM mothers: <i>n</i>&nbsp;=&nbsp;10, non-GDM mothers: n&nbsp;=&nbsp;10) living in Ningbo, China. The complete breast expression samples were collected using the electronic breast pump, stored in milk storage bags and immediately frozen at −80&nbsp;°C. All the analysis was conducted within 2&nbsp;months after milk collection. Milk samples from GDM or non-GDM women were randomly allocated into eight groups to minimize the influence of individual and dietary variations (3 samples per group, each sample can only be allocated into three groups maximum). The demographic information of these mothers and their newborns is presented in Table S1. All women were healthy with no pre-existing or ongoing diseases (e.g. mastitis and breast cancer) and did not engage in any lifestyle behaviour that may affect the growth and development of infants during pregnancy and lactation period (e.g., smoking, alcohol and drug abuse). GDM was diagnosed according to the standard of the International Association of Diabetes and Pregnancy Study Groups (IADPSG) guidelines as follows: fasting plasma glucose (FPG)&nbsp;≥&nbsp;5.1&nbsp;mmol/L, FPG 1&nbsp;h post-load glucose ≥10.0&nbsp;mmol/L or FPG 2&nbsp;h post-load glucose ≥8.5&nbsp;mmol/L (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0308814624023136#bb0030"><span>Diabetes, I. A. o. and Panel, P. S. G. C, 2010</span></a>). All participants had signed informed consent and this research was approved by the Medical Ethics Committee of Ningbo University (number: NBU-2021-142; Ningbo, Zhejiang, China).</p><h4><strong>2.3. GC–MS analysis of human milk metabolome</strong></h4><h5><strong>2.3.1. Mature human milk metabolite extraction and derivatization</strong></h5><p>Mature human milk samples were thawed at room temperature. The milk samples (150&nbsp;μL) were mixed with methanol-acetonitrile (450&nbsp;μL, V: V&nbsp;=&nbsp;2:1) solution containing 2&nbsp;μg/mL of L-2-chlorophenylalanine (internal standard) for 1&nbsp;min. Following that, the mixture was ultrasonicated in an ice water bath for 10&nbsp;min, and stored at −40&nbsp;°C for 30&nbsp;min to precipitate proteins thoroughly. The mixture was then centrifuged at 4&nbsp;°C at 12000&nbsp;rpm for 10&nbsp;min. The supernatant (150&nbsp;μL) was collected and dried using a centrifugal freeze dryer (Taicang Huamei Biochemical Instrument Factory, Jiangsu, China). Methoxylamine hydrochloride pyridine solution (80&nbsp;μL, 15&nbsp;mg/mL) was added to the freeze-dried samples to block carbonyl groups (such as α-ketonic acids and sugars) and reduce the generation of derivative byproducts. The oximation process was performed in a shaking incubator (Shanghai Lichen Bangxi Instrument Technology Co., Ltd., Shanghai, China) at 37&nbsp;°C for 60&nbsp;min. Finally, 50&nbsp;μL of BSTFA (with 1% TMCS, introducing silyl groups), 20&nbsp;μL n-hexane and 10&nbsp;μL of internal standard (including C8, C9, C10, C12, C14, C16, C18, C20, C22, C24) were added into the mixture and was derivatized at 70&nbsp;°C for 60&nbsp;min. The samples were left at room temperature for 30&nbsp;min before GC–MS analysis. Equal amounts of the extracted solutions for all samples were mixed as a quality control sample.</p><h5><strong>2.3.2. GC–MS analysis</strong></h5><p>The derivatized milk metabolites were analyzed using an Agilent <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=1&amp;search=8890B"><strong>8890B</strong></a> gas chromatography system coupled to an Agilent <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=1&amp;search=5977B"><strong>5977B </strong></a>MSD system (Agilent Technologies Inc., CA, USA). The derivative milk metabolites were separated using a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=1&amp;search=DB-5MS"><strong>DB-5MS</strong></a> capillary column (30&nbsp;m&nbsp;×&nbsp;0.25&nbsp;mm&nbsp;×&nbsp;0.25&nbsp;μm, Agilent J &amp; W Scientific, Folsom, CA, USA). The helium flow rate (&gt; 99.999%) was 1.0&nbsp;mL/min. The injector temperature and injection volume were set to 260&nbsp;°C and 1&nbsp;μL, respectively. The GC conditions were set as follows: The initial oven temperature was set at 60&nbsp;°C and maintained for 0.5&nbsp;min. Then, the temperature was increased to 125&nbsp;°C at a rate of 8&nbsp;°C/min, to 210&nbsp;°C at a rate of 8&nbsp;°C/min, to 270&nbsp;°C at a rate of 15&nbsp;°C/min, to 305&nbsp;°C at a rate of 20&nbsp;°C/min, and finally held at 305&nbsp;°C for 5&nbsp;min. The MS conditions refer to previous research (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0308814624023136#bb0185"><span>Zhang et al., 2024</span></a>). The GC–MS data were processed according to the methods of <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0308814624023136#bb0135"><span>Wang et al. (2023)</span></a>. The GC–MS data were processed using MS-DIAL software for peak detection, peak recognition, MS2Dec deconvolution, characterization, peak alignment, wave filtering and missing value interpolation. The metabolites were identified using the LUG database.</p><h4><strong>2.4. LC-MS analysis of human milk metabolome</strong></h4><h5><strong>2.4.1. Mature human milk metabolite extraction</strong></h5><p>The milk samples (150&nbsp;μL) were mixed with methanol-acetonitrile (450&nbsp;μL, V: V&nbsp;=&nbsp;2:1) solution containing 2&nbsp;μg/mL of L-2-chlorophenylalanine (internal standard) for 1&nbsp;min. After that, the mixture was placed at −40&nbsp;°C for 2&nbsp;h and centrifuged at 13000&nbsp;rpm and 4&nbsp;°C for 10&nbsp;min. The supernatant (150&nbsp;μL) was filtered through a 0.22&nbsp;μm microfilter and transferred to an LC vial and stored at −80&nbsp;°C until LC-MS analysis. Equal amounts of the extracted solutions for all samples were mixed as a quality control sample.</p><h5><strong>2.4.2. LC-MS analysis</strong></h5><p>A Dionex U3000 UHPLC coupled to <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?manufacturerItems=2&amp;search=Q-Exactive+plus+quadrupole+orbitrap"><strong>Q-Exactive plus quadrupole orbitrap</strong></a> mass spectrometer equipped with a heated electrospray ionization source (Thermo Fisher Scientific, Waltham, MA, USA) was used for metabolic profiling analysis in both ESI positive and ESI negative ion modes (a type of MS scanning mode). <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?manufacturerItems=5&amp;search=ACQUITY+UPLC+HSS+T3"><strong>ACQUITY UPLC HSS T<sub>3</sub></strong></a> column (100&nbsp;mm&nbsp;×&nbsp;2.1&nbsp;mm, 1.8&nbsp;μm, Waters, Milford, Massachusetts, USA) was used in both positive and negative modes. The binary gradient elution system consisted of strongly polar water (solvent A, containing 0.1% formic acid, <i>v</i>/v) and weakly polar acetonitrile (solvent B), which could effectively shorten the analysis cycle, improve separation ability, reduce tailing and increase sensitivity. Gradient separations refer to previous research (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0308814624023136#bb0100"><span>Ren et al., 2023</span></a>). The flow rate and column temperature were 0.35&nbsp;mL/min and 45&nbsp;°C, respectively. All the samples were kept at 4&nbsp;°C during the analysis. The injection volume was 5&nbsp;μL. The LC-MS data were processed according to the method by <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0308814624023136#bb0190"><span>Zhang, Tan, et al. (2023)</span></a>. The raw LC-MS data were processed using MS-DIAL and <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?manufacturerItems=5&amp;search=Progenesis+QI"><strong>Progenesis QI </strong></a>V2.3 for baseline filtering, peak identification, integral, retention time correction, peak alignment and normalization. The metabolites were identified using the Human Metabolome Database (HMDB), Lipidmaps (V2.3), Metlin, EMDB, PMDB and our in-house databases.</p><h4><strong>2.5. Bioinformatic analysis</strong></h4><p>Metaboanalyst 5.0 and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway software database (KEGG PATHWAY Database (genome.jp) were used for metabolic pathway analysis. MetaMapp and Cytoscape 3.9.1 were used for pathway mapping.</p><h4><strong>2.6. Statistical analysis</strong></h4><p>The GC–MS and LC-MS matrix were imported into R, principal component analysis (PCA) and orthogonal partial least-squares-discriminant analysis (OPLS-DA) were performed. <i>t</i>-test and fold-change values were used to screen DEMs between GDM and non-GDM groups. <i>p</i>&nbsp;&lt;&nbsp;0.05 (<i>t</i>-test) and Variable Important in Projection (VIP)&nbsp;&gt;&nbsp;1.0 were considered as significance thresholds. Graphpad prism was used to draw a Venn diagram, volcano plot and heat map. Cytoscape was used to draw an interaction network diagram.</p><h3><strong>4. Conclusion</strong></h3><p>Untargeted metabolomic analysis was performed using integrated GC–MS and LC-MS platforms to unveil the differential expressed metabolites in MHM of GDM and non-GDM women. A total of 268 DEMs were identified with a low abundance of 12,13-DHOME and a high abundance of linoleic acid, arachidonic acid, 9R-HODE and L-glutamic acid in MHM of GDM indicating disorder in linoleic acid metabolism, fatty acid biosynthesis, galactose metabolism and ABC transporters pathways. Abnormal levels of these metabolites showed poor glucose metabolism may persist for a longer period postpartum period. Mitigation strategies need to be developed to improve glucose metabolism, prevent transition to type 2 diabetes in GDM women and ensure healthy and optimal growth in infants. This study was limited in terms of sample sizes. However, findings from this study will pave the way for larger prospective cohort study on GDM physiopathology and the health of GDM mothers and babies.</p></div>

Untargeted metabolomics study of mature human milk from women with and without gestational diabetes mellitus

Within biological systems, endogenous elements are critical for maintaining metal homeostasis in cells, a fundamental aspect in disease progression when disrupted. Additionally, elements from the higher mass range may originate from various sources such as metal-tags for immuno-mass spectrometry, nanoparticles, or metal-based anticancer drugs. This study assesses the efficacy of collision/reacton cell (CCT) mode for simultaneously analysing elements across the complete mass spectrum. Furthermore, we demonstrate the accuracy of the LA-ICP-TOFMS methodology through the analysis of serum reference material.</span></p><h5><span lang="EN"><strong>Results</strong></span></h5><p><span lang="EN">Our findings demonstrate that the CCT mode outperforms the standard/no gas mode for LA-ICP-TOFMS measurements, particularly in quantifying endogenous elements susceptible to interferences. Through the analysis of picolitre-volume micro-droplet standards and serum reference material (deposited as micro-droplets), we observed accurate quantification of elements such as iron and selenium, with isotope ratios closely resembling natural compositions. As key advantage, the utilization of the CCT mode eliminated the need for labor-intensive post-data processing, streamlining analytical procedures. Additionally, the CCT mode also provided enhanced sensitivity (factor of 1.5–2) for elements in the higher mass range compared to the standard mode, without compromising the sensitivity for endogenous elements.</span></p><h5><span lang="EN"><strong>Significance</strong></span></h5><p><span lang="EN">We successfully integrated Seronorm serum reference material-containing micro-droplets into LA-ICPMS bioimaging workflows for quality control, enabling validated measurements of key biological elements. The use of the CCT mode is highly recommended for bioimaging applications that address the analysis of elements across the entire mass range.</span></p><h3><strong>2. Experimental</strong></h3><h4><strong>2.1. Chemicals and reagents</strong></h4><p>Ultrapure water (18.2&nbsp;MΩ&nbsp;cm, ELGA Water purification system, Purelab Ultra MK 2, UK) and nitric acid (&gt;69&nbsp;%, Rotipuran Supra, Carl Roth, Karlsruhe, Germany) were used for dilutions of the standard solutions. A multi-element stock solution and single element standard solutions were purchased from Labkings (Hilversum, The Netherlands). Gelatin was obtained from Sigma-Aldrich (Vienna, Austria). The serum reference material Seronorm (Seronorm™ Trace Elements Serum L-1, Norway) was reconstituted according to the manufacturer's protocol. Solution preparations and LA-ICP-TOFMS measurements were carried out in clean room classes ISO 8 and ISO 7, respectively.</p><h4><strong>2.2. Calibration standards based on multi-element gelatin micro-droplets</strong></h4><p>Gelatin-based micro-droplet standards were prepared according to a previously described procedure [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0003267024011462#bib20"><span>20</span></a>]. For this purpose, liquid multi-element standard solutions were prepared in different concentrations gravimetrically from standard stock solutions and mixed with a fish gelatin solution (10&nbsp;%, w/w) to reach a final concentration of 1&nbsp;% (w/w) gelatin. The resulting solutions were transferred into the sample source of a micro-spotter system. A CellenONE X1 micro-spotter (Cellenion, Lyon, France) was used to generate arrays of the gelatin micro-droplet standards onto glass slides with a droplet volume of around 400&nbsp;pL resulting in droplet sizes of approximately 150–200&nbsp;μm in diameter. The diameter of the droplets was measured by a built-in camera of the micro-spotting system before and after each spot run and used to calculate the exact volumes of the droplets. The picolitre-volumes were used to establish the absolute elemental amounts within the droplet standards. The slides were stored at room temperature in the dark until LA-ICP-TOFMS analysis and proved to be stable for up to 12 months. The entire micro-droplets were selectively and quantitatively ablated [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0003267024011462#bib27"><span>27</span></a><span>] and multi-element analysis was performed by LA-ICP-TOFMS imaging. The multi-element gelatin micro-droplet standards contained 48 elements in different concentrations (summarized in </span><a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0003267024011462#appsec1"><span>Table S1</span></a><span>).</span></p><h4><span><strong>2.3. Dispensing of serum reference material</strong></span></h4><p><span>The serum reference material Seronorm (Seronorm™ Trace Elements Serum L-1, Norway) was reconstituted according to the manufacturer's protocol. The resulting solution was immediately transferred to the sample source of the CellenONE X1 micro-spotter (Cellenion, Lyon, France). Serum reference material was dispensed onto glass slides resulting in micro-droplets with a volume of around 400&nbsp;pL and a droplet size of around 150–200&nbsp;μm in diameter. The laser fluence was adjusted to selectively and quantitatively ablate the entire micro-droplets containing the serum reference material without co-ablation of the glass slide. LA-ICP-TOFMS analysis was performed and the elemental concentrations quantified by using the micro-droplet standards with the same laser ablation parameters. The micro-droplets containing the serum Seronorm reference material proved to be stable over 6 months upon storage at room temperature in the dark (refer to&nbsp;</span><a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0003267024011462#appsec1"><span>Table S2</span></a><span> for stability data).</span></p><h4><span><strong>2.4. LA-ICP-TOFMS measurements</strong></span></h4><p><span>An Iridia 193&nbsp;nm excimer laser ablation system (Teledyne Photon Machines, Bozeman, MT, USA) was coupled to an </span><i><span>icp</span></i><span>TOF 2R (TOFWERK AG, Thun, Switzerland) TOF-based ICP-MS instrument. The LA system was equipped with a low-dispersion ablation cell [</span><a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0003267024011462#bib7"><span>7</span></a><span>,</span><a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0003267024011462#bib28"><span>28</span></a><span>] within the Cobalt ablation chamber and connected to the ICP-TOFMS system via the aerosol rapid introduction system (ARIS). Through the low-dispersion mixing bulb of the ARIS, an Ar make-up gas flow (</span><span style="font-family:&quot;Cambria Math&quot;,serif;"><span>∼</span></span><span>0.90–1.10&nbsp;L&nbsp;min<sup>−1</sup>) was introduced into the He carrier gas flow (0.60&nbsp;L&nbsp;min<sup>−1</sup>) before entering the plasma. The LA and ICP-TOFMS settings were optimized on a daily basis while ablating NIST SRM612 glass certified reference material (National Institute for Standards and Technology, Gaithersburg, MD, USA). Optimization was based on high intensities for <sup>26</sup>Mg<sup>+</sup>, <sup>59</sup>Co<sup>+</sup>, <sup>115</sup>In<sup>+</sup> and <sup>238</sup>U<sup>+</sup>, low oxide formation based on the <sup>238</sup>U<sup>16</sup>O<sup>+</sup>/<sup>238</sup>U<sup>+</sup> ratio (&lt;2&nbsp;%) and low elemental fractionation based on the <sup>238</sup>U<sup>+</sup>/<sup>232</sup>Th&nbsp;<sup>+</sup>&nbsp;ratio (</span><span style="font-family:&quot;Cambria Math&quot;,serif;"><span>∼</span></span><span>1.1).</span></p><p><span>Laser ablation sampling was performed in fixed dosage mode 2, at a repetition rate of 250&nbsp;Hz and using different spot sizes: 20, 10, 5 and 2&nbsp;μm, resulting in pixel sizes of 10, 5, 2.5 and 1&nbsp;μm. Selective ablation of the standards and the sample micro-droplets was achieved by selecting an energy density below the ablation threshold of glass and above the ablation threshold of gelatin and the samples [</span><a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0003267024011462#bib27"><span>27</span></a><span>]. Gelatin micro-droplet standards and Seronorm containing micro-droplets were ablated quantitatively using the same LA parameters and a fluence of 0.40&nbsp;J&nbsp;cm<sup>−2</sup>.</span></p><p><span>The </span><i><span>icp</span></i><span>TOF 2R ICP-TOFMS instrument has a specified mass resolution (R&nbsp;=&nbsp;m/Δm) of 6000 (full width half-maximum definition). The standard operation mode was used, which balances mass resolving power, sensitivity and ion transmission across the entire measured mass range and which allows the analysis of ions from </span><i><span>m/z</span></i><span>&nbsp;=&nbsp;14–256. The integration and read-out rate match the LA repetition rate. The instrument was equipped with a torch injector of 2.5&nbsp;mm inner diameter and nickel sample and skimmer cones with a skimmer cone insert of 2.8&nbsp;mm in diameter. A radio frequency power of 1440&nbsp;W, an auxiliary Ar gas flow rate of 0.80&nbsp;L&nbsp;min<sup>−1</sup> and a plasma Ar gas flow rate of 14&nbsp;L&nbsp;min<sup>−1</sup> was used. In case the CCT mode was used, the collision/reaction cell was pressurized with a mixture of H<sub>2</sub>/He gas with an optimized gas flow rate of 4.20–5.0&nbsp;mL&nbsp;min<sup>−1</sup>. Instrumental parameters for LA-ICP-TOFMS measurements are summarized in&nbsp;</span><a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0003267024011462#appsec1"><span>Table S3</span></a>.</p><h4><strong>2.5. Data acquisition and processing</strong></h4><p>Data was recorded using TofPilot 2.10.3.0 (TOFWERK AG, Thun, Switzerland). The LA-ICP-TOFMS data were saved in the open-source hierarchical data format (HDF5, <a target="_blank" rel="noopener noreferrer" href="http://www.hdfgroup.org">www.hdfgroup.org</a>). Post-acquisition data processing of data acquired in standard/no gas mode was performed with Tofware v3.2.2.1, which is a TOFWERK data analysis package and used as an add-on on IgorPro (Wavemetrics Inc., Oregon, USA). The data processing comprised the following steps: (1) drift correction of the mass peak position in the spectra over time via time-dependent mass calibration (2) determining the peak shape and (3) fitting and subtracting the mass spectral base-line.</p><p>Data from TofPilot (in case the CCT mode was used) and from Tofware (in case the standard mode was used) were further processed with HDIP version 1.8.5. (Teledyne Photon Machines, Bozeman, MT, USA). An integrated script was used to automatically process the files generated by TofPilot or Tofware and to generate 2D elemental distribution maps. For calibration, signal responses for each mass channel monitored during ablation of a single spiked droplet were integrated using HDIP. The sum of the elemental signal intensities and the absolute masses of the respective elements within the droplet standards were used to set up calibration curves. Signal responses for each mass channel monitored during ablation were also integrated for the droplets containing serum reference material, using HDIP.</p><h3><strong>4. Conclusions</strong></h3><p>The use of the CCT mode (cell pressurized with a He/H<sub>2</sub> gas mixture) proved to be superior to the standard/no gas mode for LA-ICP-TOFMS bioimaging applications aimed at analyzing elements across the entire mass range. Accurate quantification of endogenous elements that are subjected to interferences was achieved from picolitre-volume micro-droplet standards, as demonstrated by the analysis of micro-droplets containing serum reference material. For iron and selenium, isotope ratios in the CCT mode closely matched the natural ones. In addition, the sensitivity of elements from the higher mass range was better by a factor of 1.5 to 2.0 in the CCT mode compared to the standard mode. In case the CCT mode was used, no laborious post-data strategies were required to obtain accurate results, which is another advantage towards the use of the standard mode.</p><p>We have demonstrated the successful integration of Seronorm™ serum reference material into quantitative LA-ICPMS bioimaging workflows as an accuracy control. This validation step of quantitative measurement results can be especially attractive for industry, where quality control is a main requirement. The analysis of n&nbsp;=&nbsp;3 quality control samples adds around 5&nbsp;min of analysis time to a calibration sequence providing validated measurements of endogenous elements with biological key functions. Future directions towards one point-calibration approaches can further reduce the time required for calibration and quality control of LA-ICPMS results.</p>

They are caused by pathogenic variants in genes that control the production of hemoglobin subunits. Hemoglobin (Hb, approximately 62 kDa) is a tetramer, consisting of two alpha chains and two beta or beta-like chains. It is responsible for physiological oxygen transport in all vertebrates except for the <i>Channichthyidae</i>. Adult Hb consists of 96–98% Hb A (α2β2), 2.0–3.3% Hb A2 (α2δ2), and less than 1.0% Hb F (α2γ2).<sup>1</sup> Hemoglobinopathy is a genetic defect characterized by the abnormal structure of one of the Hb subunits, usually resulting from single amino acid substitution. Thalassemia is characterized by the absence or decreased expression of one of the Hb subunits. The most common hemoglobinopathy is sickle cell disease (SCD, Hb S (β<sup>6</sup> Glu→Val)). Around 5.2% of the global population carries significant hemoglobinopathy-causing genes with more than 330,000 affected births occurring annually.<sup>2</sup></p><p>Current techniques used in Hb analysis are electrophoretic and chromatographic assays. However, due to the lack of resolution, they are limited in differentiating isomers (i.e., between Hb C (β<sup>6</sup> Glu→Lys) and Hb E (β<sup>26</sup> Glu→Lys)), very similar masses (i.e., &lt;1 Da difference between normal Hb, Hb C, Hb E, and Hb D-Punjab (β<sup>121</sup> Glu→Gln)), or new variants. Mass spectrometry has gained popularity in clinical research because of its robustness and versatility in analyzing a wide range of samples and analytes from small molecules to proteins. Targeted protein and peptide quantitation joined this trend by applying various proteomics approaches such as bottom-up, middle-down, and top-down methods. Recently, the top-down approach targeting intact protein biomarkers was enabled by high-resolution accurate-mass (HRAM) mass spectrometers such as Thermo Scientific™ Orbitrap™-based instruments. Here, we present the top-down analysis for enhanced detection of various hemoglobin variants using the Orbitrap Exploris 240 mass spectrometer for clinical research.</p><p><strong>Conclusion</strong></p><p>This study successfully demonstrates the application of an LC-MS/MS top-down approach for detecting hemoglobin variants using dried blood spots and the Orbitrap Exploris 240 mass spectrometer. The method enables direct protein extraction with minimal sample preparation, allowing high-throughput analysis. The high resolution of the Orbitrap Exploris 240 MS effectively isolates and identifies hemoglobin variants, even with minimal mass differences, and detects low-abundance variants such as the Hb delta chain. The ProSightPD software enhances data analysis by generating sequence maps for variant identification. This approach provides a robust and efficient workflow for clinical research, offering significant improvements over traditional methods. This technical note demonstrated the feasibility of hemoglobin variant detection workflow, highlighting its potential for broader clinical research applications.</p><h4><strong>2. It is also attracting attention as a biomarker for various endocrine disorders<sup>1)</sup>.</p><p>Gas chromatography (GC) or gas chromatography-mass spectrometry (GC/MS) generally requires pretreatment such as methyl esterification for long-chain fatty acids and polyunsaturated fatty acids analysis. Multiple reaction monitoring (MRM) using liquid chromatography-mass spectrometry (LC/MS) enables highly sensitive quantitative analysis of fatty acids without derivatization. However, fatty acids widely found in the environment, such as palmitic acid and stearic acid, are introduced as impurities in LC system. It has been reported that there is difficulty in accurate quantification of these fatty acids in real samples analysis<sup>2)</sup>.</p><p>This report presents an example of quantitative analysis of free fatty acids in human plasma and serum using the highly stable LCMS-8060RX system coupled with a delay column.</p><p><strong>Conclusion</strong></p><p>Simultaneous analysis of 19 free fatty acids in human plasma and serum was performed using a high-performance liquid chromatograph mass spectrometer LCMS-8060RX. Good linearity was obtained in the range of 1-1000 for all compounds, and we also confirmed that the repeatability at lowest calibration point was less than 10%.</p><p>The use of a delay column allowed removing the matrix peaks derived from the mobile phase. This analytical method provides more stable and accurate quantitative analysis with simple pretreatment.</p><h4><strong>3. Industry processes reduce bitter taste in citrus flour by removal or modification of limonin and other limonoids. The method presented here is useful in the quantitation of limonin in foods, food additives, juices, and plants. This method yields good accuracy, precision, linearity, and range in matrix. The use of an Agilent 6230B Time of Flight LC/MS allows limonin to be distinguished and quantitated. Congeners can also be identified and quantitated.</p><p><strong>Introduction</strong></p><p>The recent rise in popularity of meat and dairy plant-based substitutes have necessitated the use of citrus fiber or flour to replace starches, gums, chemical emulsifiers, and stabilizers. Citrus flour holds water, emulsifies, binds oil, and is vegan. This natural citrus flour improves texture and stability over time and is particularly useful in baked goods, beverages, dairy products, dressings, meats, sauces, frozen foods, pet foods, dairy alternatives, and plant-based meats.</p><p>The global citrus flour market size was valued at USD 427.3 million in 2022 and is projected to reach USD 726.9 million by 2032, growing at a compound annual growth rate (CAGR) of 5.7% from 2023 to 2032.<sup>1</sup> The citrus flour market is being driven &nbsp;in part by the increased focus on health and well-being throughout the world. Being a high-quality dietary fiber source, citrus flour supports satiety and digestive health. Citrus fiber is becoming a sought-after component in formulas with a health focus as consumers become more conscious of the connection between nutrition and overall health and seek goods that have beneficial nutritional qualities and taste good.</p><p>Limonin is enriched in citrus fruits and is often found in higher concentrations in seeds, pulp and peel of oranges, grapefruit, lemons, limes, pumellos, bergamots, and mandarins.<sup>2</sup> Citrus flour is made from leftover dried fibrous material after the fruit is juiced and can have high concentrations of limonin and other limonoids. Limonin and other limonoid compounds contribute to the delayed bitter taste of some citrus food products and is therefore undesirable.</p><p>Limonin is a limonoid: a bitter, white, crystalline substance found in citrus and other plants. It is also known as limonoate, D-ring-lactone, and limonoic acid di-delta-lactone.<sup>3</sup> Chemically, it is a member of the class of compounds known as furanolactones. Limonin has an exact mass of 470.19406791 g/mol.<sup>4</sup> The method presented in this application note is useful in the quantitation of limonin in foods, food additives, juices, and plants. The use of a Time of Flight LC/MS allows excellent quantitation of limonin as well as the characterization of other congeners.</p><p><strong>Conclusion</strong></p><p>The method described in this application note for the determination and quantitation of limonin in citrus flour using an Agilent Time of Flight LC/MS has been shown to be linear, accurate, and precise over typical industrial concentrations. The use of Agilent Time of Flight LC/MS technology also allows the characterization and quantization of congeners and other matrix components. Post-run screening for other compounds is easily accomplished.</p><h4><strong>4. The metabolome is very complex, containing thousands of analytes with a bias towards smaller molecular weight compounds. Once potential biomarkers have been highlighted using statistical analysis of the data, the challenge can be to confidently identify the compounds of interest.</p><p>Higher mass resolution leads to narrower spectral peaks leading to more accurate mass measurement, allowing for tighter mass tolerances for elemental composition and/or database identification searches, improving the confidence in putative identifications. Many small molecules are also observed share similar molecular weights (isoforms). By increasing the mass resolution of the mass spectrometer data clarity is improved, with a higher mass resolution it is possible to separate two compounds with similar molecular weights. With lower mass resolution these isoforms may have merged to form a single spectral peak, leading to erroneous mass determination and incorrect compound identification.</p><p>Using a commercially available urine metabolomics study, we demonstrate the benefits of the Xevo MRT QTof mass spectrometer: delivering routine sub-ppm mass accuracy and high mass resolution. This workflow utilizes the software package MARS (MassAnalytica, Barcelona, Spain) for data processing, statistical analysis and putative identification. A more detailed overview of this software and its functionality can be found in the Waters™ application note: Screening Metabolomics of Human urine using a Waters High Resolution QTof Mass Spectrometer and MARS Data Processing Software.<sup>1</sup> The resulting data showed significant changes in exogenous or environmental markers for each of the different volunteer lifestyles.</p><p><strong>Conclusion</strong></p><p>We demonstrate that the Xevo MRT Mass Spectrometer coupled with an ACQUITY Premier LC System delivers rapid, robust data acquisition for metabolomic studies. When data is imported into MARS software for data processing this platform provides a powerful tool for acquisition, statistical analysis and putative identification of compounds of interest for metabolomic studies.</p><p>This workflow provides the acquisition and data processing needed to confidently identify and pathway profile a typical metabolomic study. The resulting data shows a clear unsupervised PCA separation between the three study groups, putatively identifying several exogenous or environmental markers with a sub 1 ppm mass accuracy and fragmentation patterns, which match closely to theoretical. When these analytes are visualized using a trend plot, significant fold changes indicate differentiation between the three smoker levels. The Xevo MRT MS, delivering a high mass resolution of up to 100,000 FWHM and routine sub-ppm mass accuracy, gives the analyst increased confidence in putative feature identification.</p>

Specific mechanical or chemical properties are often dependent on the presence and level of certain elements in the material. As a large proportion of construction materials are based on metallurgical products, it is critical to understand the concentrations of the trace elements in pure metals or alloys. Modern metal-based &nbsp;materials can be classified into non-ferrous and ferrous metals. Ferrous materials are based on iron as the main component, and therefore include different types of steel (i.e., cast iron or stainless steel). This leads to properties such as high temperature resistance as well as the<br>potential for being recycled easily. On the contrary, non-ferrous materials do not contain iron and refer to metals such as aluminium, copper, or zinc, and alloys such as brass.</p><p>Both ferrous and non-ferrous materials play crucial roles in various industries, such as modern architecture and construction, automotive, aerospace, electronics, and (clean) energy generation. Understanding the characteristics, properties, and potential impact of processing techniques is essential for metallurgical engineers to design and produce high-quality products for different applications. The exact composition, including major components, additives, and trace impurities, needs to be thoroughly determined to ensure the final product meets the performance requirements in terms of corrosion and heat resistance, conductivity, or formability.</p><p>Inductively coupled plasma mass spectrometry (ICP-MS) is often the technique of choice for the determination of impurities at trace and ultra-trace levels due to its outstanding sensitivity. As a full multi-elemental analysis can be accomplished in only a few minutes, it also allows high sample throughput. However, ICP-MS is also well known for its historically limited ability to analyze samples with elevated matrix load. The sample matrix could significantly affect the sensitivity of the instrument, cause intensity fluctuation of the internal standard (suppression and drift), and lead to increased system maintenance with unwanted downtime due to obstruction of the interface cones, torch and injector,<br>or the nebulizer. To overcome this limitation, samples must be diluted, either using liquid diluents or using argon gas provided directly from the instrument itself.</p><p>This application note focuses on the development of a fast, robust, and accurate method for the analysis of bulk and trace elements in ferrous and non-ferrous materials using the Thermo Scientific™ iCAP™ MSX ICP-MS with Argon Gas Dilution (AGD).</p><p><strong>Conclusions</strong></p><p>The iCAP MSX ICP-MS was used to analyze 20 elements in digested nickel superalloys certified reference materials, as well as selected single element standard solutions containing 1,000 mg·L-1 of elements typically used for the preparation of non-ferrous materials and alloys. Among the analytes, several critical interferences can cause unexpected bias, however He KED mode allowed consistent removal in most cases. This analytical method was rigorously tested, and the results obtained clearly demonstrated the following analytical advantages:</p><ul><li>The use of a single measurement mode in combination with other features to increase productivity of the instrument allowed the total analysis time to be reduced to &lt;3 min/sample (including uptake and wash time).</li><li>The analysis results for the 20 elements under investigation (comprising both major components as well as trace and ultra-trace level impurities) showed excellent agreement with certified values.</li><li>The developed method provides the required detection limits and a linear response for all analytes and allows coverage of a dynamic range between 25 to 1,000,000 μg·L-1.&nbsp;</li></ul><p>The use of Argon Gas Dilution (AGD Level 5) was proven to be an effective way to overcome the challenges associated with high and moreover variable matrix content. Dilution is accomplished automatically inside the instrument with no additional sample handling required. AGD is fully integrated and supported in the<br>Qtegra ISDS Software, so that it is easy to set up and operate.</p><h4><strong>2. Shimadzu: </strong><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_00803_en_c98879da3a.pdf"><strong>Screening Analysis of Trace Heavy Metals in Powdered Milk</strong></a></h4><ul><li>Application</li></ul><p><strong>User Benefits</strong></p><ul><li>ALTRACE realizes the same analysis sensitivity as the conventional instrument (EDX-7000) in 1/10 the measurement time.</li><li>Improved workability can be expected, as continuous analysis of up to 48 specimens is possible.</li><li>Simple screening analysis of heavy metals without complex sample preparation is possible because it is only necessary to set the sample in the sample holder.</li></ul><p><strong>Introduction</strong></p><p>One test method for heavy metals in food products is the color reaction (staining) method, but because that method cannot identify metals (elements) or is influenced by the sample components, AA (atomic absorption spectrophotometry), ICP-AES (inductively coupled plasma atomic emission spectrometry) or ICP-MS (inductively coupled plasma mass spectrometry) is used for elemental analysis. However, sample preparation for these test and analysis methods requires a lot of work, such as component extraction with reagents and acidic cleavage, and in the case of the staining method, individual differences in judgment have become an issue in production process and quality control. For this reason, we propose a method using energy dispersive X-ray fluorescence spectrometry (EDXRF), which enables simple analysis in the entire process from sample preparation to measurement and judgment.</p><p>In general, quantitation by EDXRF at levels below 1 ppm is difficult, as shown in Table 1, because these values are around or below the limit of quantitation. However, in the case of powdered milk, the standard values for the powder form before dissolution in hot water are about 7.7 times higher than the values shown in Table 1. This means that a screening analysis to determine whether the concentration is below the standard value or not is possible with EDXRF, which can analyze powders as-is.</p><p>Since the sensitivity of the ALTRACE for heavy metal elements was substantially improved in comparison with the conventional EDXRF (EDX-7000) by installing a high output X-ray tube, it is now possible to analyze samples with same sensitivity as the conventional instrument in only 1/10 the measurement time.</p><p><strong>Conclusion</strong></p><p>Excellent results were obtained for both the calibration curves and the analysis results of powdered milk.
