However, its reactivity can lead to hydrogenation and dechlorination reactions in the mass spectrometer electron ionization (EI) source, complicating its application in many scenarios. To address these issues, Agilent has developed a specially designed extractor source for the Inert Plus GC/MSD and triple quadrupole GC/MS systems, enhancing performance with hydrogen carrier gas. The Agilent HydroInert source maintains mass spectral fidelity and allows users to utilize existing helium-based mass spectral libraries.This study investigates how the 5Q series of columns impact the performance and durability of the HydroInert source tune. The study involved cycling through 10,000 oven temperature ramps without any injections to assess the effects of column bleed from the 5Q phase. Furthermore, the study examined how the combination of sample matrix and column bleed from the 5Q phase affects source longevity.<h4>Experimental</h4><h5>HydroInert Functionality with 5Q Columns (no Injection)&nbsp;</h5>An experiment was designed to investigate the effects of ultra low bleed columns (<a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?instrumentationItems=19&amp;manufacturerItems=1&amp;search=HP-5Q">HP-5Q</a>) on the functionality of the HydroInert source over time. A method simulating a large oven temperature ramp cycle (Table 1) over 12 minutes was created. The syringe was removed from the ALS tower to simulate injections, but to isolate the source of siloxanes to column bleed from the HP-5Q only. 10,000 injections were executed, and a system tune was performed every 100 injection to monitor source parameter shifts.The Agilent <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=1&amp;search=7000E">7000E</a> was run in SCAN mode, scanning from 50-450 m/z during each run. Air leaks were detected and immediately remedied when found to ensure the experiment was free from oxides. Liner and septum were not changed during this experiment.<h4>Results and Discussion</h4><h5>HydroInert Functionality with 5Q Columns (no Injection)&nbsp;</h5>Results of the effects of 10,000 oven temperature cycles on the HydroInert source tune parameters are shown in Figure 2. Repeller and ion body voltages are set automatically by the tune algorithm to maximize the transference of ion on the 7000E. Throughout the 10,000 injections the ion body voltage remained stable. The EM voltage gain across 10,000 injections was as expected. The source was in use for 120,000 minutes during this study and data was being collected throughout the duration of the study in SCAN mode. During this study, three leaks were detected at the MS transfer line and were corrected as soon as they were detected. The leaks are reflected in the spike in O2 percentage. A new ion body will typically have a voltage of 16-22 V on a HydroInert source. When an ion body begins to lose functionality the ion body voltage will begin to rise to compensate for the loss. Eventually the ion body voltage tops out at 29-30 V and the tune begins to show lower abundance of tune ions, until the source fails to tune. In the current study the ion body voltage was set by the tune algorithm between 19-24 V. This is a stable and normal operating range for the ion body. It is important to remember that a single elevated ion body tune is not cause for concern, there needs to be a trend of elevation over time. The overall purpose of this first study was to examine what baseline changes would occur to the HydroInert tune parameters when using a 5Q column in leak free conditions. Water and oxygen are known to speed the liberation of column bleed, which in turn reduces the functionality of the HydroInert source. This study shows that under leak free conditions, with a 5Q column, the HydroInert source can function optimally for an extended period. This study adds to a growing body of evidence that has demonstrated one of the main factors in HydroInert longevity is leak state of the system.<h4>Conclusions</h4>Agilent HydroInert source longevity is dependent on application and method conditions. However, by using the best practices highlighted above, the longevity of HydroInert parts can be prolonged. The HydroInert source cannot be mechanically or chemically cleaned, thus when source parts show reduced functionality, replacement of the recommended parts is required to restore functionality. Replacement intervals will be method and application dependent.<h3>2. Liège University / MDCW: MOSH&amp;MOAH in food ingredients and additives and the advantages of using LC/GC×GC(-FID/TOFMS) for their analysis&nbsp;</h3><ul><li>Presentation</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/FL_02_2025_MDCW_AG_MOSH_MOAH_additives_a05df36b39.pdf">Full PDF for download</a></li></ul>This presentation by Aleksandra Gorska and colleagues (Gembloux Agro-Bio Tech) addresses the analysis of MOSH (mineral oil saturated hydrocarbons) and MOAH (mineral oil aromatic hydrocarbons) in food ingredients and additives, particularly those derived from oils and fats. These contaminants are widespread in the food chain due to packaging, lubricants, or storage conditions. While MOSH can accumulate in human tissues, MOAH—especially those with three or more aromatic rings—pose genotoxic risks. The European Food Safety Authority (EFSA) has recently emphasized a lack of data on such impurities in food additives.To fill this gap, the researchers developed and applied an advanced analytical workflow based on HPLC-GC×GC-FID/TOFMS, replacing the traditional HPLC-GC-FID approach. This multidimensional technique combines high-resolution separation with both flame ionization detection (FID) and time-of-flight mass spectrometry (TOFMS), allowing detailed structural characterization, efficient removal of interfering compounds, and the ability to separate MOAH by the number of aromatic rings.Key benefits of the HPLC-GC×GC-FID/TOFMS system include clearer chromatograms with structured elution patterns, improved purification and separation of complex MOAH profiles, and identification of contamination markers such as DIPN (indicating migration from recycled packaging). Additionally, the system enables simultaneous quantification of MOAH and polycyclic aromatic hydrocarbons (PAHs), which have distinct analytical requirements and toxicological profiles.The study concludes that, despite its complexity, the HPLC-GC×GC-FID/TOFMS technique provides substantial analytical advantages and aligns with EFSA’s recommendations. It offers a powerful tool for ensuring food safety in products containing oil- or fat-derived additives, enabling more accurate risk assessments and source tracking of contamination.<h3>3. Shimadzu / ASMS: Highly sensitive and efficient analysis of aroma components using trapped Headspace GC-MS&nbsp;</h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/ASMS_2025_MP_352_97902375e7.pdf">Full PDF for download</a></li></ul>The aroma composition of foods has been the subject of considerable research in recent years. It is well known that the presence of volatile compounds and their composition determine the specific aroma of foods and the flavor of the resulting products. Static headspace technique enables high sensitivity analysis of volatile compounds, normally extracting 1 mL of headspace. But in situations where it is necessary to evaluate the total chemical profile, it is known that components in small quantities also affect the overall aroma. Thus, the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=3&amp;search=HS-20+NX">HS-20 NX</a> was used running in "Trap mode", where by using trapped headspace sampling, it is possible to extract much more volume and analyze, not only the majority components, but all the compounds with high sensitivity.&nbsp;The system used to achieve the highest sensitivity is the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=3&amp;search=HS-20+NX">HS-20 NX</a> coupled to the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=3&amp;search=GCMS-QP2020+NX">GCMS-QP2020 NX</a> single quadrupole mass spectrometer.<h4>Results and Discussion</h4>After the incubation time, the acquisitions were done by loop mode and trap mode with 3- and 5-times sample concentration. In this way, it is possible to analyze, via trap, the presence of compounds at trace levels, which cannot be observed when analyzed in loop mode. 18 compounds could be identified in the sample when analyzed in loop mode, whereas in trap mode, 35 were identified.Some components such as benzene acetaldehyde, gammacaprolactone, methyl butyrate, o-guaiacol, that have a decisive influence on the overall aroma and flavor were detected only in the trap mode, due to their low concentrations in the sample. Even concentrating the sample 5-times in the trap, it was possible to detect these compounds that require greater sensitivity along with compounds present in high concentrations in the sample, such as butanoic acid, acetic acid, and others, without losing analysis efficiency.<h4>Conclusion</h4>The improvement in analysis sensitivity when using trap mode can be observed when comparing the area for some components with a major impact on the overall aroma but present in small quantities. When concentrating the sample 3-times in the trap, the peak intensities increased from 351.17 to 1401.08%. And it was still possible to have a significant increase of another 168%, on average, by concentrating the sample 5-times. The peak intensities increased from 521.48 to 2517.86% compared to loop mode analysis.&nbsp; These analyses demonstrate that trapped headspace can be used to determine an overall aroma profile in a single analysis and with a single trapping material. The trapped headspace sampler enables ultra sensitivity analysis, identifying compounds at low concentrations which were undetectable with conventional headspace samplers.<h3>4. In this technical note, the new SmartStatus instrument tracking capabilities are highlighted with examples of how it can enable analytical laboratories to keep instruments producing consistent results.<h4>SmartStatus software overview&nbsp;</h4>SmartStatus intelligent software, available within Thermo Scientific™ <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/76">Chromeleon™ Chromatography Data System (CDS)</a>, allows monitoring of the entire analytical system including Thermo Scientific™ autosamplers, GC, GC detectors (FID and TCD), and mass spectrometers (MS). Hundreds of key instrument parameters are monitored by SmartStatus software, which provides real-time data and insights informing the user to potential problems before they arise to ensure maximum instrument uptime. This allows users to make informed decisions on which parts of the analytical system require attention and when. By performing preventative maintenance using data-driven guidance, optimal instrument performance is sustained day in and day out. Figure 1 shows the SmartStatus software overview page for a TSQ 9610 triple quadrupole MS coupled to a TRACE 1610 GC and AI/AS 1610 liquid autosampler. This overview page allows a user to quickly determine if the system is ready to run or requires attention.&nbsp;For more information on instrument status, there is a detailed view for each part of the system. This next level view shows how many injections have been performed for the various GC consumables, the lifetime of the MS detector and filament, and when the last tune was performed. Figure 2 shows an example of the GC and MS parameters and how they are monitored. Each parameter is represented as a counter gauge. As the usage increases, the gauge changes color to quickly notify the user of any maintenance that is required or will be needed soon. This allows users to plan maintenance in advance to fit it around their schedule and workload. SmartStatus instrument tracking can be monitored directly in Chromeleon CDS, and consumables usage can also be monitored directly on the TRACE 1610 GC touch screen through the dedicated instrument health icon, as shown in Figure 3.The ISQ 7610 GC-MS and the TSQ 9610 GC-MS/MS detector lifetime is monitored using a predictive algorithm. This assesses the voltages on the detector and predicts the lifetime remaining on the system so that preventative maintenance can be planned. As the ISQ 7610 GC-MS and TSQ 9610 GC-MS/MS are equipped with the XLXR™ electron multiplier detector as standard, the lifetime of the detector is several years.1 By monitoring the detector intelligently, users can avoid unnecessary downtime. Figure 4 shows monitoring of the detector within SmartStatus software.

Agilent Technologies / ASMS: Faster Than the Speed of Life: A Rapid Screening Method for Select PFAS Compounds in Serum by RapidFire-MS</h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/ps_rapidfire_ms_pfas_serum_asms_2025_wp_243_en_agilent_c8b56f4dd5.pdf">Full PDF for download</a></li></ul>Per- and polyfluoroalkyl substances (PFAS) are a broad class of compounds with a high degree of fluorination used in a variety of industries and consumer products over the past seventy years. Because of their widespread use, as well as their persistence and poorly understood effects on the human body, laboratories have started to monitor their presence in biological fluids, including serum. Biological samples are commonly prepared with protein precipitation. Additional cleanup steps such as solid phase extraction (SPE) with either C18 or weak anion exchange (WAX) sorbent can be used for increased sensitivity and cleanup. Alternatively, a pass-through cleanup using Captiva EMR-Lipid removes proteins and phospholipids through filtration, which significantly reduces sample complexity.<h4>Experimental</h4>Purchased serum was aliquoted into two sets of samples, one for protein precipitation (ppt) only and one for a protein precipitation plus Captiva EMR-Lipid based cleanup. For each preparation, 100 µL serum was aliquoted into a tube and spiked with 25 native and extracted isotopically labeled (EIS) compounds (EPA 533 list). The samples were vortexed for 1 minute and then crashed with 1% formic acid in cold ACN. They were vortexed for 3 minutes and then spun for 10 minutes to complete the crash. Samples undergoing the Captiva-EMR Lipid cleanup were then loaded onto a pre-rinsed cartridge and allowed to filter through (Figure 1). All samples were then spiked with the additional non-extracted isotopically labelled (NIS) compounds, vortexed, and transferred to a plate for analysis.The RF/TQ system consisted of a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?manufacturerItems=1&amp;search=RapidFire+400">RapidFire 400</a> front end with a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?manufacturerItems=1&amp;search=6495D+triple+quadrupole">6495D triple quadrupole mass spectrometer</a> for detection. Samples were introduced into the mass spectrometer using a RapidFire cartridge for additional cleanup, and the total cycle time was about 12.5 seconds, with analytes detected in negative polarity. Specific method conditions are detailed in Tables 1 (RF) and 2 (MS).<h4>Results and Discussion</h4><h5>Early Development Work&nbsp;</h5>Feasibility studies for detection of PFAS compounds in serum using the RapidFire started by looking for PFAS background in the instrument and in all steps of the workflow. Early testing demonstrated no detectable background from any stage of the process.RapidFire method development tested a variety of cartridges for optimal binding of the PFAS compounds, as well as determining the best state times and solvent conditions. For the preliminary work, the C18 cartridge appeared to have the best results, while the optimized elution solvent was 5 mM ammonium acetate in methanol.Early experiments were performed to understand the effects of dilution and sample prep on the overall sensitivity of the method. When only a protein precipitation preparation was employed, dilution had a significant positive effect on analyte sensitivity, as shown in Figure 2. However, when the sample prep routine utilized protein precipitation in combination with a Captiva EMR-Lipid cleanup, additional dilution was not necessary and in fact decreased overall sensitivity, as shown in Figure 3. This is attributed to the removal of extraneous matrix using the Captiva EMR-Lipid device, which has a unique selectivity for unbranched alkane chains, capturing phospholipids while allowing PFAS compounds to pass through the sorbent bed unretained.<h5>Custom Cartridge Work&nbsp;</h5>This preliminary work was done using the C18 cartridge, but the cartridge had poor retention of the short-chain PFAS compounds (C5 and below). To address this issue, a cartridge was custom-packed with mixed mode material that included C18 and anion exchange resins. Further testing and method optimization for this specific packing material yielded better results across the board, but especially for the short-chain PFAS compounds. Sensitivity for most analytes was at or below 0.1 ppb, as summarized in Table 3, with a calibration range of 0.01- 10 ppb tested for all analytes.<h4>Conclusions</h4><ul><li>LOQs for most PFAS analytes of interest in serum were 0.1 ppb or below</li><li>Samples were injected every 12 seconds, allowing for high throughput analysis</li><li>Custom mixed mode cartridge had best overall performance&nbsp;</li><li>There was no background PFAS contribution from the RapidFire system</li></ul><h3>2. Shimadzu / ASMS: Benchtop Matrix Assisted Laser Desorption Ionization Mass Spectrometry of PFAS for preliminary screening of environmental samples</h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/ASMS_2025_MP_588_4c415fced0.pdf">Full PDF for download</a></li></ul>Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants which negatively impact human health and the environment.&nbsp; High concentrations of PFAS in test samples can cause contamination of sensitive LC-MS/MS instruments requiring extensive decontamination procedures, resulting in significant downtime and costly delays in sample processing.&nbsp;We present the development of a simple, cost-effective method for the rapid screening of PFAS samples using an entry-level linear benchtop MALDI-TOF mass spectrometer to identify samples containing high levels before they enter the analytical workflow.<h4>Methods</h4>PFOA and PFOS standards were purchased from Merck, UK. Stock solutions (1 ppm prepared in acetonitrile) were diluted in water to the required concentrations.&nbsp;Samples were tested with a variety of MALDI matrices (CHCA, DHB, DHAP, DAN, NRM and TMGN), with norharmane (NRM) and 1,8-bis(tetramethylguanidino)-naphthalene (TMGN) identified for further investigation. Diluted samples were mixed with NRM (1 mg/mL) or TMGN (5 mg/mL) in 70:30 Acetonitrile:Water. Blank samples of UHP water were prepared to ensure no contamination was present. 1 µL of the mixture was spotted onto a FlexiMassSR48 slide and the samples were analysed using a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?manufacturerItems=3&amp;search=MALDI-8030">MALDI-8030 MALDI-TOF mass spectrometer</a> (Shimadzu). Analysis conditions are shown in Fig 1. Peaks at m/z 314.2 and m/z 499.2 were monitored for PFOA and PFOS, respectively. 13C8PFOS (m/z 507) was used as an internal standard for PFOS samples with a final sample concentration of 6.25 ng/mL (see Fig. 2). MALDI-MSI was used to assess spot homogeneity and limits of detection were established.Subsequently, PFHxA and PFBS standards were purchased from Merck, UK and analysed using the same method<h4>Conclusion</h4><ul><li>We have shown, in principle, that it is possible to use MALDITOF mass spectrometry to implement a simple pre-screening method for aqueous samples to prevent widespread contamination in a PFAS monitoring laboratory. Using just a 200 µL sample and minimal preparation, a full slide of samples can be acquired in less than 15 minutes&nbsp;</li><li>Spot imaging during method development highlighted the differences in homogeneity of PFOS and PFOA spots suggesting that multiple internal standards would be necessary to pursue semi-quantitative analysis.&nbsp;</li><li>Whole slide imaging can provide quick results for multiple samples across a range of PFAS concentrations in a single run with easy visual identification of potentially problematic samples.&nbsp;</li><li>As a linear MALDI-TOF method, these results do not confirm the presence of PFAS but may be used as an indicative result to modify further testing protocols. The Thermo Scientific Orbitrap Astral mass spectrometer [1] typically achieves around 60 k resolution at m/z = 138, falling to 50 k under high space charge influence.&nbsp;Here we present an enhanced resolution multi-pass method implemented in the new Thermo Scientific <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1959">Orbitrap Astral Zoom mass spectrometer</a> [2], significantly improving the resolving power at low m/z enabling TMT 32-plex on the Thermo Scientific Astral analyzer. The ions are repeatedly guided through a switchable prism deflector to extend the ion path while the principal aberrations are compensated, thereby more than doubling the resolution.<h4><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1959">The Orbitrap Astral Zoom Mass Spectrometer</a></h4>The Orbitrap Astral Zoom mass spectrometer incorporates a range of novel technologies for higher speed, sensitivity, dynamic range and resolving power. A faster ion filter and quadrupole switching in combination with improved ion transfer times in the ion-routing multipole (IRM) and ion processor allow DIA methods (Thermo Scientific Pierce FlexMix calibration solution, 2 Th isolation window, 350-980 m/z range, Thermo Scientific Orbitrap MS and Astral MS/MS) with scan rates faster than 270 Hz (DDA methods &gt; 180 Hz). A duty cycle of up to 80 % at 270 Hz is made possible by a new accumulation stage in the bent trap (linear to 106 ions) which increases the effective ion injection into the IRM by up to 40 % (additional ~0.75 ms on top of 2 ms IT in a DIA method). Furthermore, an additional mode of the high-dynamic range detector of the Astral analyzer for low-input sample applications like single-cell proteomics boosts the single ion detection efficiency by about 10 % and thereby the sensitivity. In combination with a new peak deconvolution algorithm for enhanced spectral processing, this increases the depth of analysis. In addition, modified IRM hardware allows for faster stepped collision energy experiments for deeper spectral coverage (2 CE: 80 Hz (OT Astral), 180 Hz (Orbitrap Astral Zoom MS), 3 CE: 60 Hz, 150 Hz). A modified AGC filling in the Orbitrap by a new enhanced dynamic range mode (EDR) provides additional spectral complexity and sensitivity. Here, multiplexed ion injections with different accumulation times are used for different mass ranges taking the variation in signal intensity into account. In addition, the Astral analyzer incorporates a new multi-pass mode which enhances the resolving power for small masses by more than a factor of two, enabling TMT 32-plex measurement in the Astral analyzer.<h4>Conclusions&nbsp;</h4>The TMT HR Mode advances the capabilities of the Astral analyzer. Fast electronic switching of the relay prism opens an ion trap and guides the ions after three times their normal flight distance to the detector. This greatly boosts the resolving power, especially for small m/z ratios, thereby enabling baseline-resolved quadruplets of reporter ions in a TMT 32-plex sample. First results show a peak resolution well above 100 k while maintaining high mass accuracy and shot-to-shot stability. The TMT HR Mode is utilized within a novel acquisition scheme delivering greater throughput for protein identification and quantitation for multiplexing up to 32 samples. Achieving a throughput of over 440 samples per day, the proteome coverage included 4,299 quantified protein groups and 22,146 peptides.<h3>4. The need for rapid, accurate, and cost-effective screening is critical in forensic laboratories. The study evaluates the impact of parts-per-billion (ppb) mass accuracy using the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/1148">Waters Xevo MRT Mass Spectrometer</a> compared to previous DIA methods.The method involved analyzing a commercial standard and ten anonymized urine samples, diluted and analyzed via the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/199">ACQUITY UPLC I-Class FTN PLUS System</a> coupled with the Xevo MRT MS. Data acquisition was performed in MSE mode (data-independent acquisition) under positive electrospray ionization (ESI+) using a 15-minute gradient. Data were processed in waters_connect software, leveraging a comprehensive spectral library with over 2000 compounds.Results demonstrated a root mean square (RMS) mass error of just 571 ppb for precursor ions, with 94% of compounds detected within 1 ppm, and 96% of fragment ions within the same tolerance. A key example illustrated how reducing fragment ion tolerance from 2 mDa to 0.2 mDa eliminated a false positive (Anabasine), correctly identifying Nicotine as the true analyte, showing how ppb-level accuracy increases identification confidence and minimizes misidentifications.The study concludes that ppb mass accuracy significantly reduces false positives, lowers review workloads, and facilitates independent analysis by less experienced personnel. Additionally, it supports cost savings by minimizing the need for confirmatory testing, potentially reducing both financial and environmental burdens in forensic laboratories.

Dyes and pigments are constantly being developed to improve product stability, boost color saturation, ensure consumer safety, and lower production costs. Quality control tests must be conducted on new raw materials to ensure color consistency throughout product lines. For each of these industrial needs, the analytical methodologies for characterizing these products must be robust. One of the key metrics for coloring agents is their response to UV-visible light. Cosmetics such as makeup can have complex interactions with light, including absorption, specular reflectance, and diffuse transmission and reflectance, which influence characteristics such as color, shine, and UV&nbsp;protection.&nbsp;A powerful technique used to analyze these interactions is UV-Vis spectroscopy with an integrating sphere such as an internal UV-Vis DRA. The Cary 60 UV-Vis DRA is an attachment fitted inside the Agilent <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?manufacturerItems=1&amp;search=Cary+60+UV-Vis">Cary 60 UV-Vis spectrophotometer</a> (Figure 1) with a spherical cavity coated in diffuse reflective polytetrafluoroethylene or PTFE, entrance and exit ports for the UV-Vis beam, and its own detector (Figure 2).If the sample is positioned at the entrance port, light that is diffusely transmitted through the sample can be collected with minimal loss compared to a traditional transmission measurement. Similarly, positioning the sample at the exit port allows the specular and diffusely reflected light to be measured by the integrating sphere. In this application note, diffuse reflectance of several differently colored, powdered concealers is measured to accurately determine their color coordinates using an Agilent Cary 60 UV-Vis spectrophotometer equipped with an internal DRA and the Agilent <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?manufacturerItems=1&amp;search=Cary+WinUV+Color+software">Cary WinUV Color software</a>.<h4>Conclusion&nbsp;</h4>The Agilent Cary 60 UV-Vis spectrophotometer with DRA and the Agilent Cary WinUV Color software is a simple but powerful solution for analyzing the reflectance properties of cosmetics. The DRA is an ideal tool for measuring a range of cosmetic samples in both transmission and reflection modes, and alternative sample holders are available that allow liquids, powders, and gels to be measured. Data collection is fast and reliable, and the software allows the analysis of measurements with multiple illuminants and in many color coordinate systems by simply recalculating the data, making this combination well suited to color-based quality control and color-matching applications.<h3>3. Shimadzu: Mapping Measurement of Paints and Pigments with Thermoelectrically Cooled MCT Detector</h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_00822_en_487ed50ef1.pdf">Full PDF for download</a></li></ul>Automobiles are coated with multiple layers of paint, each with a specific purpose and composition. So paint fragments at the scene of a traffic accident can provide critical information for forensic investigations. Infrared microscopy can analyze fragments that retain this multi-layer structure to identify the component constituents in each paint layer, and this can be used to identify the specific model and year of manufacture of an automobile involvedin an accident.Infrared microscopy is also used in scientific studies of valuable historical works of art to identify the pigment components and their distribution. This can reveal what techniques were used to create them, and it can assist in selecting the techniques and materials used to restore and conserve them.&nbsp;The thermoelectrically cooled MCT (TEC MCT) detector is a new optional detector for infrared microscopes that can measure microscopic targets as small as 25 µm with high resolution and without requiring liquid nitrogen. But while AIMsight infrared microscope and the AIRsight infrared Raman microscope are equipped as standard with a T2SL detector, which can measure microscopic targets down to 10 µm, they require liquid nitrogen to do so. Due to the difficulty of procuring it and the dangers it can pose of frostbite and oxygen displacement, there is an increasing demand for analytical options that avoid using liquid nitrogen.&nbsp;In this article, an infrared microscopy system (<a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?manufacturerItems=3&amp;search=IRTracer-100">IRTracer-100</a>, AIMsight) equipped with a <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?manufacturerItems=3&amp;search=TEC+MCT+detector">TEC MCT detector</a> was used for a mapping measurement of an automobile coating and organic pigments.<h4>Conclusion&nbsp;</h4>An infrared microscopy system equipped with a TEC MCT detector was used to perform mapping measurement of an automobile coating and organic pigments. The TEC MCT detector could perform mapping measurement over a wide target area with good sensitivity and without the need for liquid nitrogen. But for mapping measurement that requires a smaller aperture or if it needs to be quicker and use fewer scans, the T2SL detector, which requiresliquid nitrogen isrecommended.

Disinfection of drinking water is vital for public health but leads to the formation of harmful disinfection by-products (DBPs). To comply with EU Directive 2020/2184, a sensitive and selective HS-HiSorb-TD-GC-MS method was developed and validated for quantifying 12 DBPs in drinking water. The method showed excellent linearity (1–120 ppb), low detection limits (0.33–3.33 ppb), and good precision and recovery.Its performance was verified across concentrations, with intra-day and inter-day variability ranging from 1.3–10% and 3.3–15%, respectively. Recovery ranged from 80–120%. The method was tested for sample stability at multiple storage temperatures and applied to real tap water samples. The World Health Organization (WHO) emphasizes on the critical importance of water safety for public health, including various aspects such as drinking, domestic activities, food production, and recreation [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0001">1</a>]. Various chemical disinfectants have been used to eliminate harmful pathogens (e.g., cholera, dysentery, etc.) that can be present in untreated water, these include the use of chlorine, ozone, chloramine, and chlorine dioxide. However, a number of risks to human health appeared following the use of these means of water disinfection, such as cancer, genetic damage and developmental effects, particularly after long-term exposure [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0002">2</a>]. Therefore, public water supply systems are regularly monitored to ensure that treated water is within the established disinfection by products (DBPs) regulatory limits [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0003">3</a>].Green sample preparation microextraction techniques are considered efficient and cost-effective alternative tools for the extraction of water DBPs, offering improved detection via preconcentration. In particular, solid-phase microextraction (SPME) is a widely used sorbent-based technique, where a coated fiber is used for the adsorption of analytes [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0019">[19]</a>, <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0020">[20]</a>, <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0021">[21]</a>, <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0022">[22]</a>, <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0023">[23]</a>]. An enhanced version of this technique is the SPME arrow, designed with a larger surface area and capacity compared to standard SPME fibers, which increases extraction efficiency. A step forward, is HiSorb™ that offers much higher extraction capacity and is suitable for the analysis of higher molecular weight compounds. HiSorb™ extraction tool consists of a stainless steel rod, part of which is coated with an adsorbent material. It is a passive extraction technique (equilibrium extraction) based on the same principle as the SPME technique. HiSorb features a larger phase volume of 30 µL compared to the 0.6 µL of SPME, allowing for the adsorption of more analytes, thereby enhancing the sensitivity of the method. Additionally, HiSorb™ is more durable than SPME fibers [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0024">24</a>]. These techniques are particularly effective for extracting DBPs at low concentrations, ensuring reliable analytical results [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0025">25</a>,<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0026">26</a>].SPME and SPME arrow analyses require only a standard GC system with a minor modification on the inlet, whereas HiSorb needs an extra thermal desorption (TD) unit directly connected to the GC column hence acting as the inlet for the system. The abundant nature of THMs, in combination with their regulated levels and negative health-effects, render them as potential candidate molecules for water quality studies. Although THMs are primarily associated with water treatment processes, their presence is extended to other water-containing products. Using various microextraction techniques, THMs were detected in a wide array of samples e.g., whey [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0027">27</a>], beer [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0023">23</a>], soft drinks [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0012">12</a>], and swimming pools [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0022">22</a>].<div>The aim of the present work was to develop a headspace (HS)-HiSorb-TD-GC-MS analytical method for the simultaneous analysis of DBPs (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#tbl0001">Table 1</a>) in line with the EU 2020/2184 directive. A limited number of green and sustainable analytical methods exist for monitoring tap water DBPs [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0028">28</a>,<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0029">29</a>]. Given the urgent need to transition to more sustainable and environmentally friendly analytical chemistry techniques, this method incorporates these essential characteristics for effectively monitoring DBPs in tap water.<h4>2. Materials and methods</h4><h5>2.4. HiSorb-TD-GC-MS</h5>TD was performed using an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=7&amp;search=Unity-xr+thermal+desorber">Unity-xr thermal desorber </a>(Markes International Ltd.) connected to an Agilent <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?search=7890B">GC-7890B gas chromatograph</a> and an Agilent <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?search=5977B+GC%2FMSD">MSD-5977B</a> mass spectrometer (TD-GC-MS). The analytical details of the TD-GC-MS method are presented in the supplementary material (Table SM1). Helium (99.999 %, Linde) was used as the inert carrier gas at a flow rate of 3.45 mL/min. Chromatographic separation was achieved using a DB-5MS capillary column that was heated as follows: initial temperature 35°C, maintained for 2 min, reached 250°C at a rate of 15°C min−1, and maintained for 0.5 min. The total analysis time was 16.83 min that included a 1 min solvent delay to protect the electron ionization (EI) filament. EI (70 eV) mass spectra were collected in full scan mode in the range 35–350 m/z. The quadrupole, interface and ion source temperatures were set at 150°C, 250°C, and 230°C, respectively. Chromatograms were processed using <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=1&amp;search=ChemStation">MSD ChemStation</a> (Agilent), and the NIST20 library (National Institute of Standards and Technology) was used to identify the detected analytes. Three replicates were performed per sample, and the results were expressed as the area ratios of the analyte to IS2 (1,2-Dibromoethane) area.<h4>3. Results and discussion</h4><h5>3.1. Optimization of HiSorb extraction</h5><h6>3.1.8. Selection of chromatographic column</h6>After investigating the optimal sampling conditions using SPB-624 column, two chromatographic columns were examined for the analysis of DBPs. Initially, a SPB-624 column was tested but although it separated the four THMs, it resulted in co-elution of the two HKs. To address this issue, an alternative stationary phase was investigated. According to the literature, a potentially suitable column is the DB-5MS column [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0044">44</a>]. Upon changing the column, the co-elution issue was resolved, and the total analysis time significantly improved providing several advantages, both for the analyst in terms of time, and for the environment, as less energy was required for the analysis. Additionally, as shown in the respective chromatograms (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#fig0002">Figs. 2</a>a and b), the peak intensity for all THMs at 50 ppb was significantly higher using the DB-5MS column. Finally, due to the higher temperature limits of the DB-5MS compared to the SPB-624 column (350 vs. 250°C), it could be possible to also analyze DBPs with higher boiling points using a DB-5MS column. It was anticipated that the DB-5MS column would deliver better results and require shorter analysis times compared to the SPB-624 column due to its shorter length and thinner film thickness. It is important to note that both columns displayed additional peaks unrelated to the samples, such as siloxanes. After the DB-5MS column was selected as the most suitable chromatographic column for all the compounds analyzed, the necessary checks were performed to ensure that the results from the optimization experiments were validated. Therefore, method validation and all subsequent water sample analyses were performed using the optimized HS-HiSorb-TD-GC-MS method with the DB-5MS column.<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/ADVANC_3_66593bcddf.jpg" alt="Advances in Sample Preparation, Volume 14, 2025, 100168: Fig. 2. Monitoring DBPs in tap water samples</h5>The suitability of the developed method was tested by analyzing tap water samples from Cyprus. Note that, in Cyprus, water disinfection is carried out using NaOCl. As anticipated, the four THMs (TCM, BDCM, DBCM, and TBM), along with CH and DBAN, were detected and quantified, with measured concentrations of 2.22, 18.31, 29.97, 20.27, 9.49, and 10.75 ppb, respectively. The total THMs concentration was 70.78 ppb, within the regulatory guidelines. An exemplary HS-HiSorb-TD-GC-MS chromatogram of a tap water sample is shown in <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#fig0004">Fig. 4</a>, presenting the six detected DBPs. Additional compounds were identified in the water samples that are not of significance to this study but could be used for other tap water quality studies. These include various organic compounds, such as bromo‑chloro‑methane, dibromo-methane, 1‑bromo-2‑chloro-ethane, nonanal, 2,4-di‑tert-butylphenol, o-hydroxybiphenyl, etc. Some of these compounds are not necessarily associated with the water disinfection process but have exogenous origins. For example, o-hydroxybiphenyl is a biocide commonly used as a preservative and a fungicide, particularly for waxing citrus fruits. Additionally, the tallest peaks in the chromatogram primarily correspond to siloxanes likely originating from the thermal degradation of the PDMS adsorbent material used in the analysis. This highlights an advantage of full-scan analysis over SIM mode analysis, as it enables the detection of ionizable compounds within the sample, providing a comprehensive overview of its composition [<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S277258202500021X#bib0052">52</a>].<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/Advances_in_Sample_Preparation_Volume_14_2025_100168_Fig_4_Typical_HS_Hi_Sorb_TD_GC_MS_chromatogram_of_a_tap_water_sample_6c2151937f.jpg" alt="Advances in Sample Preparation, Volume 14, 2025, 100168: Fig. 4. Conclusions</h4>A green sample preparation microextraction method was developed and optimized for the simultaneous monitoring of 12 low molecular weight DBPs in water samples using HiSorb combined with TD-GC-MS. The optimized conditions - headspace PDMS sampling, 2 g NaCl addition, a 24-hour equilibrium time, 90 min HiSorb sampling, simultaneous heating in a water bath at 60°C, and stirring at 1000 rpm - maximized extraction efficiency, particularly for THMs.The method demonstrated excellent linearity (R² = 0.984–0.999), low detection limits (0.33–3.33 ppb), and good precision (1.3–10 % intra-day, 3.3–15 % inter-day), with recoveries ranging from 80 to 120 %. Finally, the stability of THMs in the water samples was examined, revealing that they remained stable for up to 3 days when stored at 4°C.However, the high initial cost of thermal desorption systems may limit accessibility, and extended equilibrium times may be a drawback for time-sensitive analyses. Despite these limitations, the method represents a step toward more sustainable and efficient DBP monitoring in water samples.</div>

Monitoring of 12 DBPs in drinking water using a microextraction TD-GC-MS method

This study presents novel LC-TQ-MS/MS and cost-effective UPLC methods for quantifying N-nitroso drug substance-related impurity (N-MFA) in mefenamic acid tablets and pediatric suspensions. Both methods were successfully validated according to ICH guidelines, demonstrating excellent sensitivity, linearity, and accuracy. LC-MS/MS achieved quantification at 0.01 ng/mL (1 mg/mL MFA), while UPLC quantified down to 0.036 μg/mL (10 mg/mL MFA). Linearity was confirmed across 0.01–125 ppm for LC-MS/MS and 3.6–125 ppm for UPLC with R² = 0.999.Accuracy recoveries ranged from 90.43% to 101.34% for LC-MS/MS and 92.12% to 101.21% for UPLC. Both methods were applied to commercial formulations and demonstrated suitability for routine use in R&amp;D and quality control labs. These impurities, which include nitrosamines derived from active pharmaceutical ingredients, are referred to as Nitrosamine Drug Substance Related Impurities (NDSRIs). Nitrosamines, or N-nitrosamines, are molecules characterized by their N-nitroso functional group. They are classified as a group of concern due to their status as human carcinogens, with evidence showing that long-term intake above acceptable levels can increase cancer risks.These NDSRI substances are categorized into one of three “Cohort-of-Concern” (CoC) groups: aflatoxin-like, alkyl-azoxy, and N-nitroso. “CoC” denotes a collection of highly potent mutagenic compounds requiring special attention due to potential health risks. The focus of pharmaceutical regulatory authorities has shifted from common nitrosamines to NDSRIs because of their potential carcinogenicity and their presence in drug products at unacceptable levels.1–6 As a result, regulatory authorities recalled various drug products worldwide. Furthermore, the detection and quantification of NDSRIs are of significant concern to analytical researchers.7–11Nonsteroidal anti-inflammatory drugs (NSAIDs) are a class of medications that relieve pain and inflammation, including in renal transplants and chronic non-cancer pain in children.12,13 About 30 million people worldwide use NSAIDs every day, according to a report by Medical News Today. The worldwide market for NSAIDs was valued at USD 15.58 billion in 2019 and is envisioned to reach USD 24.35 billion by 2027.Mefenamic acid (MFA) is chemically known as 2-[(2,3-dimethylphenyl)amino]benzoic acid, with the chemical formula C15H15NO2 and a molecular weight of 241.28 g mol−1. MFA is classified as an anthranilic acid (phenolate) NSAID that exhibits both antipyretic and analgesic properties. This medication treats moderate pain and inflammation associated with osteoarthritis, post-operative pain, menstrual pain, rheumatoid arthritis, toothaches, and severe back and muscle pain. Additionally, it is also used to manage menorrhagia.14,15To our knowledge, this is the first UPLC-MS/MS and environmentally friendly UPLC method for quantifying the target compound N-MFA. As a result, there are no existing methods available to compare with the proposed UPLC-TQ-MS/MS and the cost-effective green UPLC method. Various regulatory agencies, including the US FDA and Health Canada, have proposed that the acceptable intake (AI) value for N-MFA is 78 000 ng per day.16,17 The interim control limit for N-MFA is 62.4 ppm, which can be derived from the AI value of the impurity and the maximum daily dose of the MFA. Hence, simple and highly sensitive UPLC-MS/MS and UPLC methods have been developed for the accurate quantification of N-MFA in MFA drug formulations, including tablets and pediatric suspensions.<div>The current study aimed to assess genotoxicity using (Q)-SAR (Quantitative Structure–Activity Relationship) models and to develop new LC-TQ-MS/MS and UPLC methods for quantifying N-MFA in both the MFA drug substance and its formulation dosage forms. Recent regulatory guidelines demonstrate that setting the quantitation limit (QL) at 30% is considered reasonably good and acceptable. If the values consistently fall below the QL across all process validation batches and corresponding stability samples (both accelerated and long-term), it is acceptable to proceed with skip lot testing. UPLC can detect the N-MFA impurity below the 30% control limit (∼19 ppm). Therefore, this cost-effective UPLC method is proposed alongside the LC-TQD-MS/MS for use in quality control laboratories. Both methods have been successfully validated regarding the QL, linearity, precision, accuracy, and robustness. The sustainability and eco-friendliness of the UPLC method were evaluated using greenness and whiteness assessment metrics, including GAPI, the analytical eco-scale, AGREE, AGREEprep, BAGI, and RGB12 tools.18–20 The results indicated that the proposed method exhibits a high level of greenness and whiteness. <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/org/science/article/pii/S2046206925001597#fig1">Fig. 1</a> illustrates the chemical structures of MFA, N-DPA (N-nitroso diphenylamine), and N-MFA. The applicability of these methods was successfully verified through the analysis of commercial samples of APIs and dosage forms of MFA.<h4>Materials and method</h4><h5>Instrument and software</h5>The chromatographic investigation was accomplished on the <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/198">Waters H class UPLC system</a>, which includes a PDA-photo diode array detector model (L20UPD127A) modules, an Auto Sampler Manager with a Flow-Through Needle (FTN-L20FTP352G), Quaternary Solvent Manager (QSM-L20QSP358A) (Waters Corporation, USA). For the LC-TQ-MS/MS method (MRM analysis) a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/195">Waters Xevo TQ-XS MS</a> system equipped with an ESI ion source was utilized. A Mettler-Toledo (Model: XPE205, Columbus, Ohio, USA) analytical balance was employed to weigh the samples and the impurity standard. The analysis utilized high-purity water obtained from the Millipore Milli-Q purification system (Bedford, MA, USA). Centrifuge (Model: 5810R, Hemburg, Germany) from Eppendorf. ChemDraw Professional 15.0 sketched MFA, N-DPA, and N-MFA molecular structures. The two primary (Q)-SAR methodologies employed were the Derek Nexus (version 6.2.0, featuring the Derek KB 2022 1.0 knowledge base) and the Sarah Nexus (version 3.2.0, which includes the Nexus version 1.9 and the Sarah Model 2022.1). <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=5&amp;search=TargetLynx">Target Lynx software</a> v 4.2 was used to compute all LC-TQ-MS/MS acquisition and process parameters.<h4>Results and discussion</h4><h5>Development and optimization of mass spectrometry conditions</h5>To develop an accurate quantification of N-MFA in MFA tablets and pediatric suspensions, optimizing mass parameters is a decisive step. We intended to attain a sensitive, precise, and accurate method. For N-MFA quantification, the MRM technique was utilized. For initial mass conditions optimization by injecting the 1000 ng mL−1 concentration of N-MFA into mass spectrometry using infusion mode to identify the prominent product ion in electron spray ionization in positive ion mode (m/z 271.13 → 241.14). Due to the pump flow rate being set at 0.7 mL min−1 and the recommended desolvation gas temperature and gas flow being 450 °C and 900 L per Hr, hence performed the analysis with the values mentioned earlier. Employing the Intellistart software, which is built into Waters mass lynx software, to fine-tune MS/MS parameters, including cone voltage, capillary voltage, and collision energy values with different mass transitions. Intellistart identified three major MRM transitions, including m/z 271.13 → 241.14, m/z 271.13 → 223.12, and m/z 271.13 → 208.10 (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/org/science/article/pii/S2046206925001597#fig2">Fig. 2</a>). After careful optimization of cone voltage and collision energies finalized, the quantifier ion transition as m/z 271.13 → 241.14 and the qualifier ion transition as m/z 271.13 → 223.12 (Fig. S4<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/org/science/article/pii/S2046206925001597#fn1">†</a>). A mass spectrometer was installed, a 50 μL loop, which improves the peak shape when injecting the samples containing a high percentage of organic solvents. The highest intensity of quantifier and qualifier ions are detected by using the below MS/MS optimized conditions: cone voltage is 18 V, capillary voltage is 2.48 kV, collision energy is 8 V and 18 V for qualifier and quantifier respectively, desolvation gas temperature is 450 °C, and gas flow 900 L h−1. In the MS/MS method, the events program flow state was directed from 0 to 5.4 min set to mass to detect the N-MFA and the flow state from 5.5 to 10 minutes to avoid the high concentration of MFA contamination triggered by formulation tablet and pediatric suspension samples 1 mg mL−1 concentration. (A) Mass spectrum of NMFA, and (B) MS/MS fragmentation pattern of N-MFA." srcset="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/thumbnail_RSC_Advances_Volume_15_Issue_3_2025_Pages_1957_1969_Fig_2_A_Mass_spectrum_of_NMFA_and_B_MSMS_fragmentation_pattern_of_N_MFA_09e3975748.jpg 182w," sizes="100vw" width="1500" height="1286"><h5>Method validation</h5>The present proposed LC-TQ-MS/MS and UPLC methods have been fully validated by ICH Q2R2, USP 〈1225〉 guidelines,<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/org/science/article/pii/S2046206925001597#cit23">23</a> and published journals,24–26 following the method validation parameters, such as specificity, limit of detection, limit of quantitation, linearity, precision, accuracy, robustness, and solution stability. <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/org/science/article/pii/S2046206925001597#tab3">Table 3</a> presents the results of the validation study, demonstrating that the method is specific, precise, linear, and accurate.<h5>Specificity</h5>The current analytical method's specificity is demonstrated by the liquid chromatography system's capability to differentiate between the diluent, placebo, MFA, N-MFA, and N-MFA spiked at the specification level. Injected each 10 μL for LC-MS/MS and 50 μL for UPLC of diluent, placebo, N-MFA standard solution, and sample solution of MFA. The analysis showed that no co-eluting peaks were apparent at the retention time (tR) of N-MFA, allowing for the specific and accurate estimation of N-MFA in MFA tablet and pediatric suspension formulations. Diphenylamine is a precursor that can lead to the formation of N-MFA. The acceptable intake (AI) of N-MFA was derived from N-DPA, and the determined AI for N-MFA is 78 000 ng; hence the specification limit is 62.4 ppm. Proposed novel LC-TQ-MS/MS and cost-effective UPLC methods are proposed to accurately quantify N-nitroso mefenamic acid (N-MFA) in mefenamic acid (MFA) tablets and pediatric suspension dosage forms. The proposed method has been successfully validated in compliance with ICH guidelines. The developed methods were further applied to commercially available mefenamic acid formulations to determine accurate and precise quantification of N-MFA. For routine quality control laboratories, this cost-effective UPLC method can be used to detect 30% of the interim control limit (∼19 ppm). Furthermore, sustainability and eco-friendliness were evaluated using greenness and whiteness assessment metrics, including the analytical eco-scale, GAPI, AGREE, AGREEprep, BAGI, and RGB12 tools. The results indicated that the method proposed exhibits a high level of greenness and whiteness. Both methods can be used to determine N-MFA content in mefenamic acid formulation dosage based on the availability of instruments LC-TQ-MS/MS and UPLC in R&amp;D and quality control laboratories.</div>

A novel LC-TQ-MS/MS method for quantifying mefenamic acid-NDSRI (N-nitroso drug substance-related impurity) in mefenamic acid tablet and pediatric suspension dosage forms: a comparative study with a cost-effective white, green, and blue UPLC method

This study analyzed the elemental composition of 102 non-PDO (non–Protected Designation of Origin) Greek cheeses from both mainland and island regions, made from various milk types and production methods. Using ICP-MS, concentrations of essential elements (Cr, Cu, Fe, Mn, Mo, Se, Zn) and heavy metals were determined. Nutritional values were calculated based on these elements.Results showed low levels of heavy metals, confirming the cheeses are safe for consumption, and highlighted their contribution to daily intake of essential nutrients like Cr and Zn. Discriminant analysis demonstrated differentiation based on milk and cheese type. Twenty-two of these cheeses, such as Feta, Formaella Arachovas Parnassou, and Ladotyri Mytilinis, hold a Protected Designation of Origin (PDO) status, ensuring their authenticity and regional specificity. In 2021, production included 23,473 tons of hard/semi-hard cheeses, 23,891 tons of whey cheeses, and 41,769 tons of soft cheeses that do not have PDO/PGI designation (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib30">Pappa &amp; Kondyli, 2023</a>). These cheeses contribute substantially to local economies, reflecting regional traditions and manufacturing techniques. Their production is inherited from one generation to another, while some of them are consumed seasonally and are associated with specific festivities. Variability in production methods and local traditions endow these cheeses with distinct identities. Many of these cheeses despite being recognized within their production areas, they remain unfamiliar to Greek, let alone European consumers. Nowadays, consumers are interested in local food products and are willing to pay higher prices taking into account the culture of these products as well as their characteristics and manufacturing conditions (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib22">Licitra, 2010</a>; <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib4">Braghieri et al., 2014</a>; <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib20">Kamber, 2015</a>).Cheese contains important nutrients such as protein, fat, vitamins and minerals and has a crucial nutritional role (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib34">Walther et al., 2008</a>). There are twenty minerals essential for humans which are divided into macro-elements, sodium (Na), potassium (K), chloride (Cl), calcium (Ca), magnesium (Mg), phosphorus (P) and trace elements iron (Fe), copper (Cu), zinc (Zn), manganese (Mn), selenium (Se), iodine (I), chromium (Cr), cobalt (Co), molybdenum (Mo), fluoride (F), arsenic (As), nickel (Ni), silicon (Si), boron (B). Also, several other elements such as lithium (Li) and aluminum (Al) occur but are not nutritionally essential (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib6">Cashman, 2002</a>). Iron, although found in low concentrations, is an essential trace element which is involved as a catalyst in certain metabolic reactions and plays an important role in the transport, storage and utilization of oxygen. Copper is an essential element that enables the proper function of many enzymes involved in the antioxidant defense of the body. Zinc is a trace element essential for growth, development, wound healing, and normal functioning of the immune system. Manganese is involved in the metabolism of biogenic amines and participates in the regulation of carbohydrate metabolism. Selenium is a component of selenoprotein glutathione peroxide which helps to protect cell components from oxidative damage. Chromium is regarded as an essential nutrient for humans and the earliest detectable effect of deficiency is an impairment of glucose tolerance. Therefore, determination of elemental profile is a very important analysis (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib18">Georgiou &amp; Danezis, 2015</a>).Generally, the elements’ levels of a cheese depend on their levels in raw milk, reflecting the composition of soil used for pasture and the environmental contamination. The type of animal, i.e., sheep, goat, cow, their genetical characteristics, the type of pasture used, the stage of lactation, the seasonal and regional variations can influence the elemental concentration of a cheese (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib25">Miller et al., 2007</a>; <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib33">Sola-Larranaga &amp; Navarro-Blasco, 2009</a>). During cheese manufacture, different treatments such as heating of milk, curdling and salting can modify the element composition (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib21">Khozam et al., 2012</a>; <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib32">Schone et al., 2003</a>). Moreover, the concentration of some elements can be increased by the transfer of metals from containers and tools, with which milk or curd come into contact, especially when metal containers are used for cheese storage such as brined cheeses (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib3">Blunden &amp; Wallace, 2003</a>; <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib8">Coni et al., 1995</a>).The elemental profile of PDO and PGI cheeses was previously determined (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib10">Danezis et al., 2020</a>). The aim of the present work was to provide a compositional elemental description of cheeses from the Greek market which do not bear a certification of geographical indication. Their physicochemical content was studied previously by <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib31">Pappa et al. (2023)</a>. Their elemental profile may add value and greater significance contributing to better consumer recognition and to increased producers’ income. Therefore, in this work twenty elements, namely Al, vanadium (V), Cr, Mn, Fe, Co, Cu, Zn, As, Se, rubidium (Rb), strontium (Sr), niobium (Nb), Mo, tin (Sn), cesium (Cs), barium (Ba), gold (AU), lead (Pb), uranium (U) were determined and a database was created. Also, the nutritional value, in terms of dietary recommended intakes, for some elements (Cr, Cu, Fe, Mn, Mo, Se, Zn), was calculated.<h4>2. Material and methods</h4><h5>2.4. ICP-MS analysis</h5>Elemental content was determined with <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?mainauthorItems=3&amp;search=iCAP+RQ+ICP-MS">iCAP RQ ICP-MS</a> (Thermo Scientific, MA, USA) taking advantage of its rapid ultra-trace level multi-element determinations capability (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib18">Georgiou &amp; Danezis, 2015</a>). The ICP-MS parameters were as follows: nebulizer gas flow of 1.04&nbsp;L&nbsp;min−1; cooling gas flow of 14.00&nbsp;L&nbsp;min−1, auxiliary gas flow of 0.8&nbsp;L&nbsp;min−1; spray chamber temperature of 270&nbsp;°C; ICP RF power of 1550&nbsp;W; extraction lens of −200.0&nbsp;V; CCT focus lens of 1.80&nbsp;V; detector voltages of −1800/2000 V; Q-cell active in KED mode to solve potential polyatomic interferences and matrix effect. Collision gas He-flow of 8.5&nbsp;mL&nbsp;min−1; pole bias of −15.0&nbsp;V and CCT bias of −8.0&nbsp;V. Twenty elements were determined, namely 27Al, 51V, 52Cr, 55Mn, 57Fe, 59Co, 63Cu, 66Zn, 75As, 77Se, 85Rb, 88Sr, 93Nb, 95Mo, 118Sn, 133Cs, 137Ba, 197Au, 207Pb and, 238U. Each of these elements was selected based on its relevance to food analysis, its nutritional value, its potential to be a geographical indicator and/or an indicator of authenticity, as well as for the detection of eventual exposure to environmental contaminants. By analyzing a wide range of heavy metals, essential elements and toxic elements, a detailed elemental fingerprint for different cheese varieties can be developed, which can be used for quality control, as well as ensuring compliance of cheeses with food safety standards.<h4>3. Results and discussion</h4><h5>3.1. Elemental profile of local non-PDO cheeses</h5>Traditional Greek hard cheeses exhibited statistically significant differences (P&nbsp;&lt;&nbsp;0.05) for Nb, Mo, Cs, Ba, and Au, while no significant differences (P&nbsp;&gt;&nbsp;0.05) were observed for other elements, as presented in <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#tbl1">Table 1</a>. These elements could serve as markers for differentiating cheeses based on their origin or production methods, while the absence of significant differences for other elements implies consistency across samples, indicating a stable baseline for those elements in traditional Greek cheeses. In detail, Saganaki exhibited the highest concentrations of Nb (10&nbsp;μg/kg) and Au (46&nbsp;μg/kg), Pecorino had the lowest concentrations of Au (1&nbsp;μg/kg), Mo (9&nbsp;μg/kg), and Cs (&lt;LoD) -as was also evident for Tyri Mpournias of Andros-, but the highest concentration of Se (25&nbsp;μg/kg). Melichloro of Limnos showed the highest concentration of Cs (12&nbsp;μg/kg), and Ba (2200&nbsp;μg/kg). Tyri of Mpounias from Andros had the highest concentrations of Mo (180&nbsp;μg/kg), the lowest concentration of Ba (300&nbsp;μg/kg), and Cs below the LoD. Although statistically not significant, Fe concentration showed the highest concentration in Tyri of Mpournias from Andros (13000&nbsp;μg/kg), while the lowest was found in Malathouni of Tinos (2800&nbsp;μg/kg). Variations of certain elements in animal products, like meat or cheese, could be attributed to variations or differences in soil composition, water sources, or feed (<a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib11">Danezis et al., 2017</a>).<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/International_Dairy_Journal_Volume_169_2025_106315_Table_1_Concentration_of_elements_mg_kg_1_in_traditional_Greek_hard_cheeses_050ed9802b.jpg" alt="International Dairy Journal, Volume 169, 2025, 106315: Table 1. Discrimination among cheese types</h5>Discriminant analysis of the elemental fingerprint according to the type of cheese is presented in <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#fig1">Fig. 1</a>. Three discriminant functions were significant (P&nbsp;&lt;&nbsp;0.01) for the classification of samples among different cheese type groups. <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#fig1">Fig. 1</a> depicts the first two discriminant functions. Two clusters were formed, one consisting of hard and semihard cheese samples and one with soft, spread and whey cheese samples.<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/International_Dairy_Journal_Volume_169_2025_106315_Figure_1_68e58863fe.jpg" alt="International Dairy Journal, Volume 169, 2025, 106315: Fig. 1. Discriminant analysis for different cheese types using two discriminant functions of elemental profile of cheese samples (+ indicates group centroid)." srcset="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/thumbnail_International_Dairy_Journal_Volume_169_2025_106315_Figure_1_68e58863fe.jpg 245w," sizes="100vw" width="1509" height="900">Additionally, in <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#tbl6">Table 6</a>, the different cheese types were classified based on their total elemental profile through discriminant analysis. 70.4&nbsp;% of the samples were correctly classified into the appropriate cheese type group. The highest rate of correct classification was observed for hard cheese samples (90&nbsp;%) whereas the rest of the samples were correctly classified at rates lower than 72.2&nbsp;%. The elements Cr, Co, Cu, Zn, Sr, Au and U were found responsible for the discrimination of samples into cheese type groups according to the stepwise discriminant analysis applied. Under this context, previous studies applied elemental profile as a mean to discriminate among different cheese types. <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib1">Aceto et al. (2017)</a> reported that the distribution of lanthanides could be considered as a fingerprint tool to relate the soil of the pastureland on which cows graze and to the bottled milk. <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib23">Magdas et al. (2019)</a> reported that Mn content together with rare earth element concentrations were good predictors of two traditional cheese specialties of Romania. <a target="_blank" rel="noopener noreferrer" href="https://www.sciencedirect.com/science/article/pii/S0958694625001347#bib24">Mara et al. Conclusions</h4>The present study provides important input on the levels of selected elements of commercially available, non-designated, Greek cheeses. The findings of the study indicate that these cheeses can be regarded as safe, given their low levels in heavy metals, and on the other hand, they cover a considerable percentage of the dietary recommended intake for elements like Cr and Zn. Additionally, the results of this work indicated that discriminant analysis may facilitate cheese differentiation based on cheese- and milk type, as an initial step enhancing their unique properties regarding milk type and cheese making. The cheese elemental profile assessed in the present study may provide added value and better consumer recognition towards an increased producers’ income.
