Agilent Technologies: Automated Solid Phase Extraction of PFAS from Aqueous Samples</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_1633_automated_spe_wax_carbon_s_5994_8289en_agilent_1fc425207e.pdf"><strong>Full PDF for download</strong></a></li></ul><p>In this study, three types of dual-phase SPE cartridges were evaluated based on the quality control requirements for aqueous matrices as specified in US EPA Method 1633 using automated SPE. The cartridges contained PFAS WAX and Carbon S for PFAS. In the first configuration, a 200 mg bed of PFAS WAX was layered on top of a 50&nbsp;mg bed of Carbon S with the sorbents separated by a frit. In the second configuration, 200 mg of PFAS WAX was mixed with 50 mg of Carbon S before cartridge packing. In the third configuration, 200 mg of PFAS WAX and 10&nbsp;mg of Carbon S were mixed before cartridge packing. All three configurations were evaluated under identical protocols using automated SPE and were compared to results obtained using the manual addition of carbon and separate WAX extraction. The blended sorbent containing 200 mg of PFAS WAX and 10 mg of Carbon S was selected for validation and further evaluation of several environmental matrices including groundwater from residential private wells, effluent from a wastewater storage lagoon, and a landfill monitoring&nbsp;well.</p><h4><strong>Experimental</strong></h4><h5><strong>Equipment and materials</strong></h5><p>The three Agilent dual-phase cartridges used in the study are listed in Table 1. As a performance benchmark, a single‑phase <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?search=Bond+Elut+PFAS+WAX"><strong>Bond Elut PFAS WAX sorbent</strong></a> cartridge (Agilent, part number 5610‑2151) was used with either Carbon S bulk (Agilent, part number 5610-2095) or Superclean ENVI-Carb (Millipore Sigma, part number 57210-U).</p><p>Samples were extracted using the PromoChrom eight‑channel automated SPE extractors with upside‑down shaker racks (part number SPE-03 with MOD004), anticlogging frits (part&nbsp;number CF-06), high-capacity inline filters (part&nbsp;number&nbsp;F-HC-30), and anticlogging tips (part&nbsp;number F-T-4). Sample analysis was performed using an <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?search=1290+Infinity+II+LC"><strong>Agilent 1290 Infinity II LC</strong></a> system consisting of an Agilent 1290 Infinity&nbsp;II high-speed pump (G7120A), an Agilent 1290 Infinity II multisampler (G7167B), and an Agilent 1290 Infinity II multicolumn thermostat (G7116B). The LC system was modified for PFAS analysis using the Agilent <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?search=InfinityLab+PFC%E2%80%91free+HPLC+conversion+kit"><strong>InfinityLab PFC‑free HPLC conversion kit </strong></a>(part number 5004-0006). The LC system was coupled to an <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?search=6495C"><strong>Agilent 6495C triple quadrupole LC/MS</strong></a> equipped with an Agilent Jet Stream technology ion source. Agilent <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?search=MassHunter+Workstation"><strong>MassHunter Workstation software</strong></a> was used for data acquisition (version 10.1) and <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?search=Quantitative+Analysis+"><strong>Quantitative Analysis </strong></a>for QQQ (version 12.1) was used for data analysis. Instrumental conditions were determined based on detailed multiparametric optimization experiments developed in‑house.&nbsp;</p><h4><strong>Conclusion&nbsp;</strong></h4><p>The adoption of automated solid phase extraction (SPE) for PFAS analysis in environmental matrices significantly enhances laboratory efficiency and reduces the potential for human error. Our evaluation of five extraction protocols revealed that the blended sorbent containing 200 mg of PFAS WAX and 10 mg of Carbon S provided the best performance in terms of recovery, matrix interference reduction, and fewer outliers. This configuration was successfully applied to various environmental samples, demonstrating its versatility and reliability. The findings support the use of automated SPE with optimized sorbent cartridges as a robust method for PFAS analysis, aligning with the quality control metrics of US EPA Method 1633. Future work should focus on further refining automated SPE techniques and exploring their application to other environmental matrices.</p><h3><strong>2. Shimadzu: Automated Preparation of Calibration Curves Using the Autosampler's Dilution Function</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_00914_en_089a8c34fa.pdf"><strong>Full PDF for download</strong></a></li></ul><h5><strong>User Benefits</strong></h5><ul><li>The auto-dilution function equipped in the autosampler eliminates the need for manual dilution during calibration curve creation and sample preparation.</li><li>Samples can be automatically diluted to create a calibration curve by specifying the desired dilution factor in the batch table.</li><li>Because the same method file can be used across different dilution factors, only minor adjustments are needed when changing dilution conditions.&nbsp;</li></ul><p>In LC quantification, the dilution ofstandard and sample solutions is typically performed manually using pipettes, which is a laborintensive and time-consuming process. Recent advancements have called for automation to reduce labor and improve efficiency. By automating the dilution process, workflows can be streamlined, leading to increased productivity. The Nexera series autosamplers are equipped with an automatic pretreatment function that enables automated dilution. Using the auto-dilution function, samples can be automatically prepared at any desired dilution factor and directly introduced into the analytical column. This article presents a method to automate the creation of calibration curves and subsequent quantification workflows using the autosampler’s auto-dilution function.</p><h4><strong>Automated Dilution Analysis of Organic Acid Standard Solution</strong></h4><p>Automated dilution analysis was conducted using a 1000 mg/L mixed standard solution of ten organic acids. The analytical conditions are shown in Table 2 (pretreatment program : Table 1). 1.0 mL polypropylene vials were used as mixing vials, and septum-sealed vials were used for the undiluted standard solution. Rinse solution (ultrapure water) was used as the diluent. Fig. 3 shows the chromatogram of the 100-fold diluted organic acid mixed standard solution (concentration after auto-dilution : 10 mg/L for each organic acid). The <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=1&amp;search=Nexera+Organic+Acid+Analysis+System"><strong>Nexera Organic Acid Analysis System</strong></a> (pH-buffered electric conductivity detection) was employed for this analysis.</p><h4><strong>Conclusion</strong></h4><p>By utilizing the auto-dilution function in the autosampler, the standard undiluted solution for calibration curve creation can be automatically diluted at any desired dilution ratio. This eliminates the need for manual sample dilution and streamlines the calibration curve creation workflow. Additionally, since the autodilution function can also be applied to sample solutions, only the preparation of undiluted standard and sample solutions is required, allowing for a fully automated process from calibration curve creation to quantitation. As a result, the reduction in pretreatment work contributes to improved work efficiency and productivity.</p><h3><strong>3. Thermo Fisher Scientific / HPLC Symposium: Wide-ranging polynucleotide separation capabilities using Reversed Phase particles with variable pore geometry</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/po_wide_ranging_polynucleotide_separation_capabilities_po_en_1fdaba9e08.pdf"><strong>Full PDF for download</strong></a></li></ul><p>Polynucleotides such as DNA and RNA serve many uses in the areas of basic research, industrial manufacturing, therapeutics to up/down regulate cellular functions, and as therapeutic and vaccine intermediates (i.e., as an instructional template to produce proteins that 1) supplement/replace ineffective proteins or 2) function as antigens). CRISPR technology using guide RNA (gRNA, ~100mer) to directly modify cellular chromosomes was recently leveraged to create a customized treatment within 6-months for an infant with CPS1 deficiency1. This approach has provided long-term, life-saving enzyme production negating the need for daily treatments. Associated DNA and RNA such as gRNA and mRNA (1000+mer) require detailed characterization to ensure safety and efficacy (e.g., of sufficient quantity and fidelity in delivery vehicles such as lipid nanoparticles). Here, we present a RP particle with complex pore geometry and the effects of mobile phase and temperature for separating DNA/RNA with therapeutic benefits (siRNA-gRNA-mRNA:~ 20-100- 1000+mer). Semi-prep column separations, common for sample recovery for subsequent analyses or sample enrichment for in vitro or in vivo use, is also shown.</p><h4><strong>Materials and methods</strong></h4><h5><strong>Test method(s)&nbsp;</strong></h5><p><a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/library?mainauthorItems=3&amp;search=DNAPac+RP+column"><strong>DNAPac RP columns</strong></a> packed with 4 µm DNAPac RP media include 2.1 × 50 mm (P/N 088924), 2.1 × 50 mm (P/N 088924), and 21.2 × 150 mm (P/N 080922- 1521232). For instruments, mobile phases, gradient conditions, and other chromatographic operating parameters (flow rate, temperature, injection volume, etc.), please reference the figures and associated text. For all analyses, samples were detected by UV absorbance at 260nm. For 21.2x150mm semi-prep separations, a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products?category=40&amp;manufacturerItems=3"><strong>conventional (U)HPLC chromatography system</strong></a> with a binary pump, autosampler and detector without column compartment (ambient temperature separations) was used for all analyses. Ion Pairing Agents (Cation Anion): TEAA – Triethylamine Acetate, HAA – Hexylamine Acetate, HFIP – Hexafluoroisopropanol.&nbsp;</p><h5><strong>Data analysis&nbsp;</strong></h5><p>The <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/76"><strong>Thermo Scientific Chromeleon </strong></a>7.2.10 or 7.3 Chromatography Data Systems (CDS) software was used for data acquisition and analysis.</p><h4><strong>Results&nbsp;</strong></h4><h5><strong>DNAPac RP particle characterization – chemistry and pore structure&nbsp;</strong></h5><p>Figure 1 summarizes the chemical and physical properties of the resin, which is based on polymeric divinylbenzene (top left). Unlike conventional silica particles, the polymeric resin is not susceptible to hydrolytic degradation at neither extreme pH (1-14) nor high temperature (90+oC). The aromatic groups also provide π- π bonding with aromatic nucleotides giving different selectivity from conventional alkyl phases incapable of such interactions. The SEM image visually shows the highly variated porous structure of the particles. Barrett-Joyner-Halenda (BJH) N2 adsorption analysis details the surface area contributions of the different pore sizes. From this analysis it is clear that a continuum of pores from 50Å to 2500Å (suitable for small molecules up to macromolecule adsorption, respectively) contributes to the overall surface area of the particle. Based on the complex geometry and resulting pore structure, we commonly refer to the media as a supermacroporous (SMP) particle.</p><h4><strong>Conclusions&nbsp;</strong></h4><ul><li>High resolution separation of nucleic acids varying in length from 10 to 10,000 nucleotides is achieved using an SMP particle with a variable pore structure.</li><li>Polymeric SMP particles are compatible with different mobile phase IP compositions, high pH and high separation temperature, which are critical parameters for optimizing the separation of polynucleotides&nbsp;</li><li>SMP particles provide high resolution separations in both analytical and semi-prep formats</li></ul><h3><strong>4. Monitoring PFAS in biofluids is possible, but their very low concentrations present analytical challenges.</p><p>Data-Directed Acquisition (DDA) isolates ions that meet defined criteria, fragments them at elevated collision energies, and generates product ion spectra. Because PFAS ions show a characteristic <strong>mass defect</strong> due to fluorine atoms, inclusion lists can enhance their detection. By adding the new Particle Analysis Program to AMsolution software, users can automatically measure the size, mass, and volume of multiple particles and easily visualize the results through graphs and tables.</p><p>The program calculates particle dimensions such as major and minor axes, while also estimating mass and volume using a validated theoretical formula based on recent research from Ehime University. This allows laboratories to determine microplastic mass concentrations with high accuracy, a significant improvement over traditional methods. Statistical data, including component ratios and histograms, can be output directly or exported in CSV format, while customizable analysis conditions allow exclusion of unwanted particle types like fibers.</p><p>To further enhance measurement reliability, Shimadzu offers a <strong>Particle Filter (PF) Holder</strong>. This accessory secures PTFE or stainless steel membrane filters to prevent sagging during drying, keeping the surface level and ensuring more precise focusing and particle detection.</p><p>Together, the AIMsight and AIRsight microscopes, combined with the Particle Analysis Program and PF Holder, provide laboratories with a powerful, automated solution for high-throughput, reproducible microplastics research and contaminant monitoring.</p><h3><strong>3. While some elements are required for biological function, others can pose health risks when present even at trace levels. Because blood is a complex matrix rich in salts and biomolecules, it presents significant challenges for accurate and reproducible analysis. <strong>Triple quadrupole ICP-MS</strong> is particularly well suited for this task, offering high sensitivity, robust interference removal, and wide dynamic range.</p><p>In this study, researchers applied the <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/products/1488"><strong>Thermo Scientific™ iCAP™ MTX ICP-MS</strong></a> equipped with an <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/products/1715"><strong>iSC-65 Autosampler</strong></a> and operated using <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/products/1489"><strong>Qtegra™ ISDS software</strong></a>. Whole blood samples were diluted 50-fold and analyzed for 43 elements ranging from lithium to uranium. By combining <strong>He KED mode</strong> with <strong>TQ-O₂ mass-shift reactions</strong>, the method effectively removed spectral interferences, enabling accurate detection across eight orders of magnitude—from sub-ng/L levels up to 50 mg/L.</p><p>Performance testing demonstrated excellent accuracy, with results closely matching certified reference materials for whole blood. The setup also showed outstanding robustness, sustaining more than 15 hours of continuous acquisition for 287 samples with minimal drift. The Step Ahead feature of the autosampler reduced analysis time by 9%, while the humidifier and interface maintenance hardware improved stability and minimized downtime. Notably, after over 1,000 sample injections, the sampler and skimmer cones remained clean, underlining the durability of the system.</p><p>The study also compared human, horse, and pig blood, revealing significant differences in elemental composition between species. These findings highlight the potential of triple quadrupole ICP-MS to accelerate large-scale studies on biological variation and exposure assessment. Together, the <strong>iCAP MTX ICP-MS</strong> and <strong>iSC-65 Autosampler</strong> provide a powerful, high-throughput solution for multi-element analysis in one of the most challenging sample types: whole blood.</p>

Formats and hyperlinks were adapted from the original.</strong></p><p>Spectroelectrochemistry (SEC) provides a great amount of information in a single experiment, since it permits the following of a system from two different points of view: electrochemical and spectroscopic. <a href="javascript:void(0);">(1)</a> To enhance the capabilities of electrochemical analysis, we propose to implement new multiresponse techniques that enable the simultaneous recording of multiple signals. <a href="javascript:void(0);">(2,3)</a> Several SEC devices combining two spectroscopic techniques have been developed by our group in the past, promoting the development of new SEC techniques such as bidimensional UV/vis absorption SEC, <a href="javascript:void(0);">(3,4)</a> the combination of fluorescence and UV/vis absorption SEC, <a href="javascript:void(0);">(5)</a> or the combination of Raman scattering and UV/vis absorption SEC. <a href="javascript:void(0);">(6)</a> Nevertheless, to the best of our knowledge, this is the first time that electrochemistry is coupled to three different spectroscopic techniques simultaneously.</p><p>In this technical note, we propose a novel design that allows the straightforward and reproducible coupling of <i>operando</i> and simultaneous SEC measurements of UV/vis absorption spectroscopy, Raman scattering, and photoluminescence (PL). This new setup focuses on the wealth of information that all these techniques can provide in the study of a chemical system. UV/vis SEC provides molecular information about the compounds present or electrogenerated at the electrode/solution interface and in the diffusion layer. PL SEC also allows the acquisition of data regarding the location and characteristics of molecules at the electrode/solution interface and within the diffusion layer, achieving a higher degree of specificity and sensitivity than UV/vis absorption SEC, as these molecules are fluorescent. Finally, Raman SEC provides vibrational characterization of the compounds, being surface-sensitive due to the <i>Surface Enhanced Raman Scattering</i> (SERS) phenomenon.</p><p>To characterize complex systems, such as electrode processes, <a href="javascript:void(0);">(7,8)</a> SEC experiments can be performed separately, changing the optical technique used in each case. Nevertheless, our proposal is that the simultaneous combination of different analytical techniques represents a better strategy, as it facilitates the study of the same chemical system under the same experimental conditions, thereby avoiding any influence of irreproducibility among consecutive experiments. In some cases, the studied system can exhibit inherent reproducibility issues, making it difficult to establish reliable correlations between experimental results.</p><h4><strong>Experimental Section</strong></h4><h5><strong>Design and Fabrication of the Device for UV–vis absorption/PL/Raman SEC Measurements</strong></h5><p><a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acselectrochem.5c00038#fig1">Figure 1</a> shows the multi-SEC cell for the simultaneous recording of UV/vis absorption, PL, Raman, and electrochemical signals. This cell consisted of three main parts: I) support of the screen-printed electrode (SPE), II) holder of the optical fibers for UV/vis absorption measurements in a parallel configuration, and III) holder of the optical fibers for PL measurements. (I) Lower piece, (II) Middle piece and (III) Upper piece." srcset="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/thumbnail_ACS_Electrochem_2025_1_6_997_1002_Figure_1_a_Schematic_of_the_assembled_UV_vis_absorption_PL_Raman_SEC_cell_a8ecc7b83f.jpg 140w," sizes="100vw" width="1000" height="1117"></p><h4><strong>Results and Discussion</strong></h4><h5><strong>Application of the Cell to Study a Bactericidal Molecule</strong></h5><p>Experiments with Ru(bpy)<sub>3</sub><sup>2+</sup> allowed us to demonstrate that simultaneous information from four different points of view can be obtained with the cell proposed in this work. As proof of concept, the oxidation of a fluorescent organic molecule, ofloxacin (OFL), a commonly used antibiotic, is proposed. This system was selected because its spectroscopic behavior is similar to Ru(bpy)<sub>3</sub><sup>2+</sup>, but its Raman response is quite different. In addition, the study of this molecule is of great interest because of its widespread use as a bactericidal agent. OFL exhibits characteristic UV/vis absorption, fluorescence emission, and Raman spectra that change when subjected to electrochemical oxidation processes. The performance of time-resolved experiments in which the evolution of the electrochemical signal, molecular absorption, fluorescence emission and Raman spectra are recorded simultaneously will be of great help in determining its reaction mechanism, which may help to understand its mode of action as a bactericidal antibiotic. <a href="javascript:void(0);">(17,18)</a></p><p>For this purpose, a 5 × 10<sup>–4</sup> M OFL solution in Britton-Robinson buffer (BR, pH = 4.25) was prepared to perform SEC experiments while maintaining the above experimental conditions. Similarly, CV was selected as the electrochemical technique. In this case, changes were recorded between the vertex potentials +0.00 V and +1.10 V, starting at +0.60 V in the cathodic direction. Two scans were registered at 0.02 V·s<sup>–1</sup>, and the initial spectrum at OCP was selected as reference for UV/vis absorption measurements. All spectra obtained using the three techniques were recorded at an integration time of 1 s. However, a different integration time could be used for each individual technique, depending on the signal to noise ratio.</p><p><a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acselectrochem.5c00038/suppl_file/ec5c00038_si_001.pdf">Figure S5</a> shows the contour plots for each spectroscopic technique registered during the SEC experiment for the oxidation of OFL. In this case, the evolution of UV/vis absorption spectra (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acselectrochem.5c00038/suppl_file/ec5c00038_si_001.pdf">Figure S5a</a>) is characterized by a strong negative absorption band at 303 nm which corresponds to a π–π* electronic transition of the chromophore of OFL that involves a carboxyl group and the nitrogen atom at position 1 in the quinolone group, and a broad negative shoulder between 330 and 370 nm, linked to an n−π* electronic transition of the chromophore of OFL from the nitrogen of the piperazinyl group to the carbon at position 7 in the quinolone group attached to the carbonyl group; both bands are related to the consumption of OFL on the WE surface due to its oxidation. <a href="javascript:void(0);">(2)</a> The evolution of PL spectra (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acselectrochem.5c00038/suppl_file/ec5c00038_si_001.pdf">Figure S5b</a>) shows a band at 521 nm that decreases during the oxidation of OFL that it is accompanied by a small shoulder between 390 and 460 nm that increased during this oxidation process. <a href="javascript:void(0);">(19)</a> The emission band peaking at 521 nm is related to the protonated form of OFL, indicating again the consumption of this molecule by the oxidation process. The increase in the shoulder at shorter wavelengths must be linked with the oxidation product electrogenerated. <a href="javascript:void(0);">(20)</a> To facilitate a more accurate visual representation of the photoluminescence signal’s evolution, <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acselectrochem.5c00038/suppl_file/ec5c00038_si_001.pdf">Figure S5b</a> illustrates its variation throughout the experiment, taking the initial fluorescence spectrum of the solution as a reference spectrum (ΔI<sub>PL</sub> = I<sub>PL,t</sub> – I<sub>0</sub>, where I<sub>PL,t</sub> is the photoluminescence intensity at different times/potentials applied during the experiment, and I<sub>0</sub> corresponds to the photoluminescence intensity of the initial spectrum of the solution). Finally, the evolution of the Raman spectra (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acselectrochem.5c00038/suppl_file/ec5c00038_si_001.pdf">Figure S5c</a>) is dominated by two peaks at 1397 and 1619 cm<sup>–1</sup> that evolve during the second potential cycle. The first one is due to the symmetric stretching mode of the carboxylate group (ν<sub>ring</sub>, COO<sup>–</sup>), and the second characteristic band is attributed to the C═C stretching vibration of the quinolone aromatic ring (ν<sub>s</sub>, C═C). <a href="javascript:void(0);">(21,22)</a></p><p><a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acselectrochem.5c00038#fig3">Figure 3</a>a shows the corresponding CV. No reduction peak was observed during the first cathodic scan. When the potential is reversed in the anodic direction, a first oxidation peak (O<sub>1</sub>) at +0.95 V is observed due to the oxidation of the AuNPs and OFL. OFL is irreversibly oxidized, as was demonstrated in previous works. <a href="javascript:void(0);">(2)</a> Regarding the second cycle, a cathodic peak (R<sub>2</sub>) is observed at +0.28 V, corresponding to the reduction of AuNPs, oxidized at potentials higher than +0.75 V, which agrees with the experiments with Ru(bpy)<sub>3</sub><sup>2+</sup>.</p><p><img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/ACS_Electrochem_2025_1_6_997_1002_Figure_3_SEC_experiment_390710c95f.jpg" alt="ACS Electrochem. 2025, 1, 6, 997–1002: Figure 3. (a) CV and (b) CVA at 303 nm, (c) CVF at 521 and 437 nm and (c) CVR at 1397 and 1619 cm–1." srcset="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/thumbnail_ACS_Electrochem_2025_1_6_997_1002_Figure_3_SEC_experiment_390710c95f.jpg 94w," sizes="100vw" width="625" height="1040"></p><p>The CVA at 303 nm (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acselectrochem.5c00038#fig3">Figure 3</a>b) and the CVFs at 521 and 437 nm (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acselectrochem.5c00038#fig3">Figure 3</a>c) show the same behavior. As it is expected, if in the first potential cycle the oxidation potential exceeds +0.75 V, the absorbance at 303 nm and the fluorescence at 521 nm decrease due to the consumption and irreversible oxidation of OFL, while the fluorescence at 437 nm increases due to the generation of the OFL oxidation product. During the backward scan, when the potential of +0.75 V is reached, the CVA at 303 nm and CVF at 521 nm signals increase due to the diffusion of OFL from the bulk solution to the diffusion layer interrogated by the optical probes.</p><p>Furthermore, the CVF signal at 437 nm is diminished in intensity when the potential is lower than +0.75 V, owing to the diffusion of the oxidation product into the solution.</p><p>The behavior observed in the second potential scan is analogous to that observed during the initial scan. Lastly, the CVRs at the two characteristic bands of OFL (1397 and 1619 cm<sup>–1</sup>, <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acselectrochem.5c00038#fig3">Figure 3</a>d) show that in the first potential scan OFL is not detected by SERS in the Raman spectra. The Raman intensity does not increase until the AuNPs are reduced in the second cathodic scan, as is observed in <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acselectrochem.5c00038/suppl_file/ec5c00038_si_001.pdf">Figure S4b</a> (orange lines). The Raman signal at 570 cm<sup>–1</sup> demonstrates the oxidation/reduction of AuNPs, <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acselectrochem.5c00038/suppl_file/ec5c00038_si_001.pdf">Figure S4b</a>. The Raman signal at 1397 cm<sup>–1</sup> demonstrates that Raman amplification did not occur when the experiment started toward reduction. The AuNPs must first be oxidized and then reduced to observe the corresponding Raman spectrum, suggesting that the reconstruction of the NPs surface plays a key role in the activation of the SERS phenomenon.</p><p>From the results, it can be concluded that the new device allowed us to obtain four independent signals in a single experiment, helping us to understand the chemical processes taking place during an electrochemical process, with all the signals being correlated with each other, providing a full picture of the electrode process. While the UV/vis absorption and PL optical signals show changes throughout the redox process of OFL, the Raman signal demonstrates that an activation of the SERS substrate is needed without prior oxidation and subsequent reduction of the AuNPs, no substrate capable of amplifying the Raman signal is created, and therefore no OFL spectrum is observed up to that point. The oxidation/reduction of AuNPs in this case illustrates that SERS signals are influenced not only by the size and shape of the particles but also by their surface condition, which can facilitate the chemical mechanism of enhancing the Raman signal.</p><h4><strong>Conclusions</strong></h4><p>The coupling of electrochemistry with three spectroscopic techniques (UV/vis absorption, PL, and Raman) provides a comprehensive understanding of all the chemical processes involved in the electrode process. This is a major asset, offering a starting point for the analytical procedures used to identify and quantify the molecules of interest in multiple domains.</p><p>Raman analysis of OFL has demonstrated that when the experiment begins with a reduction step, no amplification of the Raman signal occurs. In other words, the initial AuNPs do not act as a SERS substrate, because the OFL Raman spectrum is not defined. However, when AuNPs are first oxidized and then reduced, the corresponding Raman spectrum emerges, demonstrating that the reconstruction of the AuNP surface plays a key role in the activation of the SERS phenomenon. This reconstruction suggests a new experimental pathway for the detection of this family of antibiotics using SERS.</p>
