BaySpec: PNNL and BaySpec Launch Compact Mass Spectrometry System for Rapid Narcotics Detection</strong></h3><ul><li>Scientific article</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/PNNL_and_Bay_Spec_Launch_Compact_Mass_Spectrometry_System_for_Rapid_Narcotics_Detection_087ddb9b7e.pdf"><strong>Full PDF for download</strong></a></li></ul><p>The U.S. Department of Energy’s Pacific Northwest National Laboratory’s (PNNL) VaporID, which is a newly developed portable air sampling system incorporating a miniaturized mass spectrometer (MS), can detect trace levels of fentanyl, methamphetamine, cocaine, and even explosives like TNT with great accuracy, the laboratory announced in a press release (1). Because of its success, BaySpec, an optical products manufacturer, is commercializing this instrument, which will be produced later in 2025 (1,2).&nbsp;</p><p>The new system employs a technique known as noncontact detection, which collects airborne molecules and analyzes them in real time using a miniature mass spectrometer (1). The VaporID device was first developed in 2020. The device won GeekWire’s Innovation of the Year award that same year (2). At the 2025 American Society for Mass Spectrometry (ASMS) conference in Baltimore Maryland, Krisztian Torma, a BaySpec scientist and one of the lead developers, presented the latest test results of the device. The VaporID device was field-tested at the U.S.–Mexico border crossing in Nogales, Arizona. Working with the U.S. Customs and Border Protection (CBP) agency, the device was able to successfully identify trace amounts of fentanyl, as well as other narcotics including MDMA (ecstasy), methamphetamine, ketamine, and cocaine (1).</p><p>Fentanyl, explosives, and other drugs have low vapor pressure, which means that they do not willingly evaporate and release very few molecules into the air. Given the fact that there are often other interfering chemicals in the air, this presents a detection challenge. The VaporID system overcomes this hurdle by filtering out irrelevant vapors and enhancing signal clarity (1).&nbsp;</p><p>The success of the VaporID system is the result of a two-year collaboration between PNNL and BaySpec. With plans to commercialize the system by the end of the year, BaySpec aims to bring this advanced narcotics and explosives detection tool into routine use at ports of entry, airports, shipping facilities, and public event venues (1).</p><h3><strong>2. However, some compounds are not registered in the NIST DB. In addition, molecular formula estimation from molecular ions obtained by soft ionization often results in a large number of candidate formulas. To address these issues, we developed two machine learning (ML)–based methods for molecular formula recommendation and structural formula estimation. Our method is considered effective for rapid structural estimation of unknown compounds in GCxGC-MS measurements.</p><h3><strong>3. Therefore, during the sake production process, it is essential to preserve the sake’s aroma profile. One key step in sake production is pasteurization (“hiire”), in which unpasteurized sake is heated to sterilize it, deactivate enzymes, and halt fermentation. This pasteurization step is known to impair the aroma of sake if it is carried out for too long or at too high a temperature. In this analysis, a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=Nexis+GC-2030"><strong>Nexis GC-2030 gas chromatograph</strong></a> was combined with an HS-20 NX headspace sampler (Fig. 1) to quantify individual aroma compounds in sake before and after pasteurization.&nbsp;</p><p>Headspace gas chromatography (HS-GC) is an effective method for analyzing volatile compounds, including aromas. HS-GC requires no pretreatment (e.g., derivatization), and sample preparation is straightforward: simply place the sample in a vial and perform the analysis.&nbsp;</p><p>Hydrogen, which is inexpensive and readily available, was used as the carrier gas. Using H2 gas as the carrier reduces analysis costs while maintaining high separation performance over a wide range of linear velocities. However, because hydrogen is flammable, it must be handled with care. The <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=Nexis+GC-2030"><strong>Nexis GC-2030</strong></a>, <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=Brevis+GC-2050"><strong>Brevis GC-2050</strong></a>, and <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=Nexis+GC-2060"><strong>Nexis GC-2060</strong></a> gas chromatographs can be equipped with an optional hydrogen sensor that measures H2 concentration in the column oven. If the hydrogen sensor detects a leak, the system shiftsto standby mode or automatically powers off, helping prevent accidents before they occur.</p><h4><strong>Conclusion</strong></h4><p>By combining the Nexis GC-2030 gas chromatograph with the HS-20 NX headspace sampler, the aroma compounds can be easily quantified without pretreatment such as derivatization. In addition, using inexpensive, readily available H2 as the carrier gas reduces analysis costs while maintaining high separation performance. Because the Nexis GC-2030 can be equipped with an optional hydrogen sensor, analyses using H2 carrier gas can be performed safely.</p><h3><strong>4. However, there are many cases where a traditional spectrometer’s working distance is not sufficient to achieve this, in which case a focus-independent, long-distance collection proves invaluable. One potential application of this focus-independent “remote” measurement is high-throughput plastics recycling, where many differently sized plastics are passed along at a fixed distance and screened for specific material identification. Similarly, any potential use cases of samples inside a glove box or within a hazardous environment may benefit from long-distance collection, because the laboratory grade spectrometer can be kept clear of the sample enclosure while providing excellent spectral signal. It should be noted that each unique application comes with its own optical beam path and laser safety requirements.</p><p>In addition to this particular modality, the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/2238"><strong>Thermo Scientific™ DXR3 Flex Raman Spectrometer</strong></a> provides excellent experimental versatility with micro scale, bulk sampling, and fiber probe collection modes also available. Shown below is a longdistance analytical setup in which the DXR3 Flex Raman spectrometer passes the unfocused laser beam through a quartz shielding window onto a polystyrene sample (Figure 1). The relationship between pathlength/sample mounting preparation and spectral signal is investigated below, to validate the use of a long-distance collection mode as a viable alternative to more traditional, focus-dependent analysis. It is worth mentioning the practice of standoff Raman spectroscopy, a technique designed to provide Raman data at distances ranging from 1 m to several hundred meters. However, these systems often require significantly different components than a standard Raman spectrometer, such as pulsed lasers, gated detectors, UV excitation wavelengths, and telescopic focusing elements.¹ The work described here performs measurements at a distance with a simple, off-the-shelf, benchtop spectrometer.</p><h4><strong>Practical use case: recycled plastic identification&nbsp;</strong></h4><p>One of the major benefits of this long-distance measurement technique is being able to handle samples of various sizes and thicknesses without needing to adjust the laser focus for specific samples. The following results simulate a potential use case where remote Raman spectroscopy is used to rapidly identify the plastic materials of various sizes. The samples were fixed at a 50 cm pathlength, and collection times were set at 10 s.</p><p>All three samples show clear and identifiable Raman signals. The plastic drum is identified as polyethylene, while the water bottle is a specific, BPA-free polycarbonate alternative. The plastic bag is multilayered, with signals from both polyethylene and polycarbonate, along with a visible fluorescence peak at 2150 cm-1. All of these different polymers are spectroscopically identifiable, and at a range of different sample thicknesses. These results provide an excellent view into the use of this technique on non-reference samples. This is just one example of a potential use for the long-distance collection mode, a mode which enables much greater versatility in experimental design and instrument placement.</p></div></div></div></div></div></div></section>

This method typically involves using a standard 10 mm path length cuvette to measure absorbance of materials, which can then be used in QC labs to quantify or qualify an analyte of interest.&nbsp;</p><p>In UV-Vis measurements, the photometric response of the spectrophotometer follows the Beer‑Lambert law, where a linear relationship exists between the sample absorbance and sample path length. The path length is the distance that the incident light travels through a sample. Increasing the cuvette path length from 10 to 50 mm increases the absorbance of a sample, aiding in more accurate quantification of components with low peak intensity. Alternatively, if the sample concentration is high and exceeds the linear dynamic detection range of the system, a cuvette with a shorter path length can be used instead.<sup>1</sup></p><p>In the pharmaceutical industry, ethanol is widely used in manufacturing as a disinfectant, solvent, and preservative. Due to its amphipathic structure, ethanol can dissolve both hydrophilic and lipophilic substances, serving as a medium to dissolve key components for a broad spectrum of drug formulations. The dual nature of ethanol also allows it to inhibit microbial growth by disrupting cellular membranes and denaturing proteins, processes that are vital for disinfection and the preservation of&nbsp;medications.<sup>2</sup>&nbsp;</p><p>As a result, ethanol used in pharmaceutical manufacturing must comply with pharmacopeia standards to ensure drug safety, efficacy, and quality. The United States Pharmacopeia (USP)<sup>3</sup>, European Pharmacopoeia (EP)<sup>4</sup>, and Japanese Pharmacopoeia (JP)<sup>5</sup> all specify using an ultraviolet absorption test to measure impurities in ethanol at designated wavelengths (Table 1). In this test, the absorption spectrum should show a steadily descending curve with no observable peaks or shoulders, which serve as qualitative indicators of impurities present in the sample. Since the impurities have low absorbance levels, the USP, EP, and JP require the use of a 50 mm cell to improve sensitivity and detection levels.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>The <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/library?mainauthorItems=10&amp;search=Cary+3500"><strong>Agilent Cary 3500 Flexible UV-Vis spectrophotometer</strong></a> with variable-path-length cell holder demonstrates exceptional performance in the measurement of ethanol impurities according to global pharmacopeia standards. Whey protein is a co-product of cheese production and represents the water-soluble protein remaining in the solution after the milk has been acidified or a coagulation agent has been added. The removal of the non-soluble casein protein portion is referred to as curds. Whey protein is a common ingredient in many dietary supplements due to its availability, solubility, and unique functional characteristics.&nbsp;</p><p>The accurate and precise determination of protein in whey and powdered dairy products not only plays a role in the characterization of nutritional or dietary value in dairy products, but may also be key in determining the quality or category of the dairy product. Protein in whey and other dairy products is most commonly calculated using the measured Nitrogen in the sample and a protein factor multiplier (protein factors vary according to the sample matrix).</p><h4><strong>Instrument Model and Configuration&nbsp;</strong></h4><p>The <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/products/1885"><strong>LECO FP928</strong></a> is a macro combustion Nitrogen/Protein determinator that utilizes a pure Oxygen environment in a high-temperature horizontal ceramic combustion furnace, utilizing ceramic boats designed to handle macro sample masses (~ 1.0 g). A thermoelectric cooler removes moisture from the combustion gases before they are collected in a ballast. The gases equilibrate and mix in the ballast before a representative aliquot (3 cm<sup>3</sup> or 10 cm<sup>3</sup> volume) of the gas is extracted and introduced into a flowing stream of inert carrier gas (Helium or Argon) for analysis. The aliquot of gas is carried through a heated reduction tube, filled with Copper, to convert Nitrogen Oxide combustion gas species (NO<sub>x</sub>) to Nitrogen (N<sub>2</sub>). The aliquot gas is then carried to a thermal conductivity cell (TC) for the detection of Nitrogen (N<sub>2</sub>).&nbsp;</p><p>Thermal conductivity detectors work by detecting changes in the thermal conductivity of the analyte gas compared to a reference/carrier gas. The greater the difference between the thermal conductivity of the carrier gas and the analyte gas, the greater the sensitivity of the detector. The FP928 supports either the use of Helium or Argon as the instrument's carrier gas. When used as a carrier gas, Helium provides the highest sensitivity, and the best performance at the lower limit of the Nitrogen range. The thermal conductivity difference between Argon and Nitrogen is not as great as the thermal conductivity difference between Helium and Nitrogen; therefore, the detector is inherently less sensitive when using Argon as a carrier gas.</p><h4><strong>TYPICAL RESULTS&nbsp;</strong></h4><p>Data was generated utilizing a linear, force through origin calibration using ~0.75 g of LECO 502-896 (Lot 1007) LCRM EDTA (9.59% N). The calibration was verified using ~0.1 g of LECO 502-688 (Lot 1004) LCRM Nicotinic Acid (11.37% N). Samples were weighed and analyzed at ~0.5 grams. A protein † factor of 6.38 was used for all samples to calculate the protein content.</p><h3><strong>3. Shimadzu: High-Temperature Gigacycle Fatigue Test of Inconel 718 and Element Distribution Measurement on a Fracture Surface</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_01082_en_37d9ba3a1a.pdf"><strong>Full PDF for download</strong></a></li></ul><h4><strong>User Benefits</strong></h4><ul><li>Fatigue testing at 20 kHz can be performed using an ultrasonic fatigue testing machine, and 109 cycle fatigue tests can be completed in about 14 hours.</li><li>High-temperature fatigue tests can be performed by using a combination of an ultrasonic fatigue testing system and an induction heating furnace.</li><li>The origins of fatigue fractures and inclusions can be identified by observations and element distribution measurements using an electron probe microanalyzer.</li></ul><p>Inconel 718 has excellent heat and corrosion resistance and high strength. Its mechanical properties can be maintained even under high-temperature environments, which is why it is used in harsh environments, such as in the jet engines and turbine blades in the aerospace industry, and in gas turbines and nuclear power plant parts in the energy industry. In these applications, long-term reliability is crucial, making fatigue testing essential. However, this can be time-consuming; for example, gigacycle fatigue testing exceeding 109 cycles takes three to four months at a frequency of 100 Hz when using a conventional fatigue testing machine. But with the USF-2000A ultrasonic fatigue testing system, tests performed at a frequency of 20 kHz can be completed in approximately 14 hours.&nbsp;</p><p>This article describes using the <strong>USF-2000A</strong> to perform gigacycle fatigue tests on Inconel 718 at 600 °C. In addition, the EPMA8050G, which is an electron probe microanalyzer (EPMA ), was used to observe the fatigue fracture surfaces and measure the element distribution.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>Gigacycle fatigue tests were performed on Inconel 718 at 600 °C, using the USF-2000A ultrasonic fatigue testing system. Good results were obtained since the number of cycles to fractures fell in the range 106 to 109 for their respective loading conditions. The EPMA-8050G electron probe microanalyzer was used to perform elemental mapping analysis of fractures originating from inclusions, and the elements of the inclusions were identified.</p><h3><strong>4. Thermo Fisher Scientific: XPS depth profiling of coated nitrided steel using femtosecond laser ablation</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/labrulez-bucket-strapi-h3hsga3/xps_depth_profiling_coated_nitrided_steel_appnote_d19841e6c9.pdf"><strong>Full PDF for download</strong></a></li></ul><p>X-ray photoelectron spectroscopy (XPS) is a powerful surface analysis technique that provides essential information about elemental composition along with chemical and electronic states. Depth profiling, a process in which material is repeatedly and sequentially analyzed with XPS and removed, provides access to buried features, layers, and interfaces. Traditional ion sputtering methods for depth profiling can, however, lead to preferential sputtering and chemical damage, impacting the reliability and accuracy of results. This is particularly problematic for complex, multi-layered structures and coatings, where it is crucial for the stoichiometry and chemical integrity to be maintained throughout the depth profile.&nbsp;</p><p>This application note explores the use of femtosecond laser ablation (fs-LA) as an alternative to conventional ion sputtering for XPS depth profiling. Specifically, this approach is used to analyze a nitrided steel surface coated with a titanium-doped molybdenum disulfide (MoS₂:Ti) layer, which was applied with physical vapor deposition (PVD). Nitriding is typically used in applications where durability is critical, such as in the production of gears, as it increases surface hardness and wear resistance. The addition of a MoS₂:Ti coating offers additional benefits such as enhanced lubrication and wear resistance. Molybdenum disulfide is well-known for its lubricating properties, and doping with titanium improves mechanical strength and adhesion to the substrate. Accurate depth profiling is essential for unraveling how these structures increase performance and for their further optimization and improvement.</p><h4><strong>Experimental</strong></h4><p>A <a target="_blank" rel="noopener noreferrer" href="https://icpms.labrulez.com/products/2206"><strong>Thermo Scientific™ Hypulse™ Surface Analysis System,</strong></a> equipped with both traditional ion sputtering and fs-LA capabilities, was utilized for the analysis. XPS data was collected in SnapShot mode using a 30 µm X-ray spot size. A 500 eV monoatomic Ar⁺ beam from a Thermo Scientific™ MAGCIS™ Dual Beam Ion Source was used for sputter depth profiling. The fs-LA depth profiles were generated using a 1,030-nm laser operating at a 160 fs pulse length. The pulse energy was increased from 83 µJ to 250 µJ when transitioning from the MoS₂:Ti coating into the bulk steel material.</p><h4><strong>Summary&nbsp;</strong></h4><p>XPS depth profiling with femtosecond laser ablation offers a robust alternative to traditional ion sputtering, particularly for materials sensitive to preferential sputtering and chemical damage. The technique provides accurate stoichiometric analysis and preserves chemical state information, making it ideal for complex, multi-layered structures with deeply buried interfaces. The fs-LA method can offer comparable depth resolution to ion sputtering while being able to profile greater depths in a fraction of the time.&nbsp;</p><p>This application note demonstrates the effectiveness of fs-LA in maintaining the chemical integrity of a nitrided steel material with a MoS₂:Ti coating throughout a depth profile, providing accurate and reliable results in a practical timeframe.</p>

<p><strong>This study compares the aroma profiles of beers fermented with </strong><i><strong>Torulaspora delbrueckii</strong></i><strong> and </strong><i><strong>Saccharomyces cerevisiae</strong></i><strong>. Sensory evaluation revealed a more intense fruity character in the </strong><i><strong>T. delbrueckii</strong></i><strong> beer, supported by aroma extract dilution analysis identifying 32 odorants, with most exceeding their odor thresholds.</strong></p><p>Key fruity esters, including ethyl butanoate, ethyl 2-methylpropanoate, and ethyl propanoate, showed higher odor activity values in the <i>T. delbrueckii</i> beer. Although yeast had unintentionally been used to produce beer back then, it was not until the 19th century that yeast was identified as the major active organism accomplishing the fermentation process. <a href="javascript:void(0);">(1)</a> Before the invention of the pure culture process in 1883 by Emil Christian Hansen, beer was the product of a spontaneous mixed fermentation and involved different species of yeast and bacteria. Each of them potentially contributed to the overall aroma of the beer. <a href="javascript:void(0);">(2)</a> Today, most beers worldwide are produced with the top-fermenting yeast <i>Saccharomyces cerevisiae</i> or the bottom-fermenting <i>Saccharomyces pastorianus</i>. <a href="javascript:void(0);">(3)</a> For a long time, extraneous yeasts were only considered spoilage organisms, <a href="javascript:void(0);">(4,5)</a> however, they recently generated interest for beer aroma diversification.</p><p>In most industrialized countries, beer sales and consumption have decreased continuously over the past few years. <a href="javascript:void(0);">(6)</a> To act against this negative trend, diversifying beer by developing special beers with aroma characteristics beyond the mainstream is a suitable option. This can be achieved by customization of raw materials and/or processing. The impact of specialty malts on beer aroma has already been studied on the level of the causative substances, and so has the contribution of novel hop varieties with unique aroma profiles in combination with different hopping techniques. <a href="javascript:void(0);">(7−10)</a> In contrast, knowledge about how non-<i>Saccharomyces</i> yeasts influence beer aroma is scarce at the molecular level.</p><p>In recent years, non-<i>Saccharomyces</i> yeasts have demonstrated great potential for producing new beer styles. <a href="javascript:void(0);">(5)</a> However, a majority of yeasts do not grow sufficiently in wort, and some produce unpleasant aromas. Michel et al. <a href="javascript:void(0);">(4)</a> developed a screening method to determine the fermentation ability of non-<i>Saccharomyces</i> yeasts by a combination of tests addressing, e.g., sugar utilization, hop and ethanol resistance, and propagation efficiency. The results particularly showed the outstanding suitability of <i>Torulaspora delbrueckii</i> strains for beer brewing.</p><p><i>T. delbrueckii</i> is well-known in the wine industry, where it is used in single and mixed fermentations to produce wines intense in aroma. <a href="javascript:void(0);">(11)</a> Recent studies have shown that <i>T. delbrueckii,</i> when used in mixed fermentation with <i>S. cerevisiae,</i> can also produce beers with a pronounced fruity aroma corresponding with high amounts of acetyl esters, 3-methylbutan-1-ol, and ethyl butanoate. <a href="javascript:void(0);">(12−18)</a> Further data are available on the volatile compounds formed by <i>T. delbrueckii</i> in single fermentation of synthetic media <a href="javascript:void(0);">(19,20)</a> or wort. <a href="javascript:void(0);">(17,21−24)</a> However, in none of the studies, the aroma contribution of the volatile compounds formed by <i>T. delbrueckii</i> during beer making has been systematically evaluated. To fill this gap, our aim was to apply a molecular sensory science approach to identify the key odorants in a beer brewed with <i>T. delbrueckii</i> in parallel with a beer brewed with <i>S. cerevisiae,</i> serving as a reference. The volatiles were isolated from the beer samples by solvent-assisted flavor evaporation (SAFE) and screened for odor-active compounds by gas chromatography–olfactometry (GC–O) in combination with a comparative aroma extract dilution analysis (AEDA). A dilution analysis of static headspace samples was performed to additionally cover highly volatile odorants. After structure elucidation, major odorants were quantitated. For olfactometric detection in the GC–O/FID and HS–GC–O/MS approaches, a trained and experienced assessor placed the nose directly above the sniffing port and evaluated the effluent. Whenever an odor was detected, the retention time and the odor quality were recorded. All olfactometric analyses were conducted by three assessors with complementary olfactory capabilities, i.e., if one assessor exhibited a specific anosmia, at least one of the other two was normosmic. Awareness of one’s own anosmia was taught in weekly training sessions, which also included familiarization with a standardized flavor language. Retention index (RI) calculation was accomplished after GC analysis with a constant temperature gradient (6 °C/min) by linear interpolation from the retention time of the target odorant and the retention times of adjacent <i>n</i>-alkanes. <a href="javascript:void(0);">(40)</a></p><h4><strong>Results and Discussion</strong></h4><h5><strong>Sensory Characteristics of the Beers</strong></h5><p>The quantitative olfactory profiles of both the reference beer brewed with <i>S. cerevisiae</i> and the special beer brewed with <i>T. delbrueckii</i> showed patterns fairly characteristic of top-fermented beers: high ratings for malty, floral, honey-like, and fruity in combination with moderate ratings for caramel-like, sweaty, and banana-like (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acsfoodscitech.5c01133#fig1">Figure 1</a>). Nevertheless, a closer look also revealed clear quantitative differences. Whereas the reference beer was dominated by the malty aroma note, the special beer was rated higher in the fruity aroma note, the caramel-like aroma note, and the banana-like aroma note. Assessors rated the intensity of each descriptor on a scale from 0 to 3 with 0.1 increments and 0 = not detectable, 1 = weak, 2 = moderate, and 3 = strong." srcset="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/thumbnail_ACS_Food_Sci_Technol_2026_6_1_181_188_Figure_1_Quantitative_olfactory_profiles_of_the_reference_beer_brewed_with_S_cerevisiae_and_the_special_beer_brewed_with_T_delbrueckii_c5e8770229.jpg 165w," sizes="100vw" width="638" height="602"></p><h5><strong>Odor-Active Compounds in the Beers</strong></h5><p>To approach the compounds that were responsible for the aroma difference between the reference beer brewed with <i>S. cerevisiae</i> and the special beer brewed with <i>T. delbrueckii</i>, we first screened the volatiles isolated from the beers by solvent extraction and SAFE using GC–O in combination with a comparative AEDA. As a result, we obtained 32 odor-active compounds, among which 31 were detected in both beers, and FD factors up to 16384 in the reference beer and up to 32768 in the special beer (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acsfoodscitech.5c01133#tbl1">Table 1</a>). Structure assignments were achieved with the following approach: RIs on two columns with different polarity (DB-FFAP and DB-5) and odor descriptions were compared to published data, including those compiled in the Leibniz-LSB@TUM Odorant Database. <a href="javascript:void(0);">(44)</a> The resulting structure proposals were confirmed by parallel GC–O and GC–MS analyses of the beer volatile isolates and authentic reference compound solutions.</p><p><img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/ACS_Food_Sci_Technol_2026_6_1_181_188_Table_1_Odorants_in_the_Volatile_Isolates_Obtained_from_the_Reference_Beer_Brewed_with_S_cerevisiae_and_the_Special_Beer_Brewed_with_T_delbrueckii_1_d6e079edf1.jpg" alt="ACS Food Sci. The compound with the highest overall FD factor of 32768 was caramel-like smelling 4-hydroxy-2,5-dimethylfuran-3(2<i>H</i>)-one (HDMF; <strong>23</strong>) in the special beer. With 8192 it also showed a high FD factor in the reference beer. HDMF is a well-known beer odorant thermally generated during malt production. <a href="javascript:void(0);">(7)</a> With an FD factor of 16384, the most potent odorant in the reference beer was floral, honey-like smelling 2-phenylethan-1-ol (<strong>21</strong>), followed by malty smelling 2- and 3-methylbutan-1-ol (<strong>8</strong>) with a combined FD factor of 8192. 2- and 3-methylbutan-1-ol were not separated on the GC column used for AEDA, but the mass spectra in EI mode indicated a mixture of both compounds. 2-Phenylethan-1-ol, 2-methylbutan-1-ol, and 3-methylbutan-1-ol also showed high FD factors in the special beer, namely 16384 and 512. These important beer odorants, <a href="javascript:void(0);">(45,46)</a> are formed as byproducts of yeast metabolism during fermentation. <a href="javascript:void(0);">(47)</a> Further five odorants with FD factors as high as 1024 to 4096 in at least one of the two beer samples were 2-methoxy-4-vinylphenol (<strong>27</strong>; smoky, clove-like; FD factors 1024 and 4096), phenylacetic acid (<strong>31</strong>; honey-like, beeswax-like; FD factors 2048 and 2048), sotolon (<strong>26</strong>; fenugreek-like; FD factors 1024 and 512), (<i>E</i>)-β-damascenone (<strong>18</strong>; cooked apple-like; FD factors 1024 and 256), and 2,3-dihydromaltol (<strong>20</strong>; caramel-like; FD factors 256 and 1024). All these compounds have been reported as beer odorants before. <a href="javascript:void(0);">(10,46,48)</a> The entire results of the odorant identification are summarized in <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acsfoodscitech.5c01133#tbl1">Table 1</a>. While the volatiles in beers brewed with <i>T. delbrueckii</i> have previously been analyzed by GC–FID <a href="javascript:void(0);">(5,13−15,18,23)</a> and GC–MS, <a href="javascript:void(0);">(19,20,24)</a> to our knowledge, this is the first study to apply activity-guided odorant screening using GC–O approaches.</p><p>Approximately half of the compounds compiled in <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acsfoodscitech.5c01133#tbl1">Table 1</a> showed no substantial FD factor differences between the reference beer brewed with <i>S. cerevisiae</i> and the special beer brewed with <i>T. delbrueckii</i>. FD factors were either identical, such as for 2-phenylethan-1-ol (<strong>21</strong>; both 16384) and phenylacetic acid (<strong>31</strong>; both 2048), or differed only by a factor of 2, which cannot be considered an indication of a quantitative difference, e.g., for sotolon (<strong>26</strong>; 1024 vs 512) and ethyl 2-methylpropanoate (<strong>2</strong>; 256 vs 128). Among the compounds that showed substantial FD factor differences between the reference beer and the special beer were the important malty smelling beer odorants 2- and 3-methylbutan-1-ol with common FD factors of 8192 in the reference and 512 in the special beer. This difference corresponded well to a higher rating of the malty aroma note in the olfactory profile of the reference beer (cf. <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acsfoodscitech.5c01133#fig1">Figure 1</a>). On the other hand, the FD factors of fruity smelling compounds were not able to explain the stronger fruitiness of the special beer brewed with <i>T. delbrueckii</i>. In fact, the fruity smelling esters ethyl propanoate (<strong>1</strong>), ethyl butanoate (<strong>3</strong>), and ethyl 3-methylbutanoate (<strong>5</strong>) showed higher FD factors in the special beer (16 vs 8, 128 vs 64, and 8 vs 4); however, other fruity smelling esters showed higher FD factors in the reference beer, namely ethyl 2-methylpropanoate (<strong>2</strong>; 256 vs 128), ethyl 2-methylbutanoate (<strong>4</strong>; 8 vs 2), and ethyl hexanoate (<strong>10</strong>; 32 vs 16). To clarify the compounds being causative for the more intense fruity aroma of the special beer, sensory experiments based on exact quantitative data would, therefore, be essential. Before starting these experiments, however, the screening for odor-active compounds in the beers was extended to the fraction of highly volatile compounds.</p><p>In summary, the use of <i>T. delbrueckii</i> for beer brewing resulted in a beer with an intense fruity aroma associated with higher concentrations of ethyl butanoate, ethyl 2-methylpropanoate, and ethyl propanoate than in the corresponding <i>S. cerevisiae</i> beer. Our study thus confirmed that the usage of non-<i>Saccharomyces</i> yeasts for beer brewing can be an appropriate tool for diversifying beer aroma and developing unique beers with aroma characteristics beyond the mainstream. However, further research is needed to substantiate our results, assess their reproducibility by analyzing biological replicates, and study the effects of different yeast strains.</p></div>

Characterization of the Major Odorants in Beer Brewed with Torulaspora delbrueckii─Differences to Beer Brewed with Saccharomyces cerevisiae

<p><strong>This study investigates the impact of emitter positioning on signal intensity in nanoflow electrospray ionization mass spectrometry. By systematically varying emitter position in three dimensions, the authors demonstrate that signal robustness improves at greater distances along the z-axis, with consistent performance maintained within 1–2 mm of the optimal position.</strong></p><p>Signal behavior was consistent across the m/z range, indicating that precise repositioning for different analytes is unnecessary. Formats and hyperlinks were adapted from the original.</strong></p><div><p>Owing to its high sensitivity, nanoflow electrospray liquid chromatography coupled with tandem mass spectrometry (nESI-LC-MS/MS) is widely used for protein sequence analysis. <a href="javascript:void(0);">(1,2)</a> Whereas high-flow electrospray sources typically have relatively fixed emitter alignments and are more robust to changes in emitter positioning, nESI sources frequently require manual alignment in three dimensions. <a href="javascript:void(0);">(3,4)</a> Since the number of ions reaching the MS system can vary depending on the location of the nESI emitter, positioning is a critical aspect of achieving robust and reproducible results.</p><p>Multiple reports illustrate the relationship between emitter positioning and electrospray signal intensity; <a href="javascript:void(0);">(2,4−10)</a> however, these studies typically examine a single dimension or evaluate emitters and flow regimes not typical of shotgun proteomic workflows. To understand these relationships, we investigate here the effect of emitter positioning on signal intensity when using fused-silica capillaries with integrated emitters at a flow rate of 300 nL/min, a common choice across the field. <a href="javascript:void(0);">(11−17)</a> Experiments were conducted using an Orbitrap Ascend mass spectrometer, a notable example of instruments commonly used in proteome analysis, <a href="javascript:void(0);">(18)</a> which features a nanoflow source and asymmetric inlet capillary (high-capacity transfer tube, HCTT). This work represents, to our knowledge, the first report in the literature characterizing how emitter positioning impacts signal intensity for nanoflow electrospray ionization into an inlet capillary without radial symmetry.</p><p>Since most nESI emitters are manually positioned, understanding the precision in positioning that is required could assist in reducing batch-to-batch variation that is common in larger proteomic experiments. Further, recent efforts to improve the throughput of proteome analysis feature the use of multiple columns with multiple emitters aligned with the source at the same time. <a href="javascript:void(0);">(19−23)</a> By performing measurements in all three dimensions, this report provides insight for implementing such a multiemitter setup.</p><h4><strong>Experimental Section</strong></h4><p>Emitter positioning measurements were performed by infusing BSA peptides at 300 nL/min into an <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/748"><strong>Orbitrap Ascend mass spectrometer</strong></a> (Thermo Scientific) using a fused silica capillary (360 μm O.D, 75 μm I.D.) with integrated emitter (∼10 μm opening). <a href="javascript:void(0);">(24)</a> The emitter was aligned to the inlet using the source camera and the <i>x</i>/<i>y</i>/<i>z</i> micrometer on a NanoSpray Flex source (Thermo Scientific). The origin of the coordinate system used here was defined as the center of the inlet capillary opening. In particular, the <i>z</i> = 0 plane is orthogonal to the opening of the inlet capillary, and the <i>x</i> = 0 and <i>y</i> = 0 planes are parallel to the inlet capillary axis. Additional experimental and data analysis details are described in the <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/jasms.5c00441/suppl_file/js5c00441_si_001.pdf">Supporting Information</a>.</p><h4><strong>Results and Discussion</strong></h4><p>We characterized signal intensity dependence on emitter position for nanoflow electrospray by infusing a mixture of peptides generated via trypsin digestion of purified BSA in an Orbitrap-quadrupole linear ion trap hybrid mass spectrometer (Orbitrap Ascend Tribrid). To ensure LC–MS applicability, we used an emitter size (∼10 μm orifice) and flow rate (300 nL/min) identical to those typical of capillary LC–MS. <a href="javascript:void(0);">(11−17)</a> The resulting MS1 analysis produced numerous ions across the <i>m</i>/<i>z</i> range (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/jasms.5c00441/suppl_file/js5c00441_si_001.pdf">Figure S3</a>). We selected two of these ions, a doubly protonated species (LGEYGFQNALIVR at <i>m</i>/<i>z</i> 740.4) and a triply protonated species (RHPEYAVSVLLR at <i>m</i>/<i>z</i> 480.6), for their strong signal intensity and stability over the time range of the experiments here (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/jasms.5c00441/suppl_file/js5c00441_si_001.pdf">Figures S4 and S5</a>). With the above setup, we tracked those <i>m</i>/<i>z</i> peaks as a function of emitter position in three dimensions and recorded intensity distributions (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/jasms.5c00441#fig1">Figure 1</a>). <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/jasms.5c00441#fig1">Figure 1</a>C presents the ion signal across x-positions, where the <i>x</i> = 0 plane is aligned to the central axis of the inlet capillary and the dashed lines depict the MS inlet opening width in this dimension (0.6 mm, <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/jasms.5c00441/suppl_file/js5c00441_si_001.pdf">Figure S6</a>). <a href="javascript:void(0);">(25,26)</a> Strikingly, we observe that the signal is relatively stable and high across two millimeters of the <i>x</i>-dimension. Further, greater than 50% of the ion signal is retained at distances up to ∼five times the width of the inlet (i.e., full width half max (fwhm)). <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/jasms.5c00441#fig1">Figure 1</a>D presents the same concept but in the y-dimension, where good signal stability is also observed across a similar length of two millimeters; however, the larger inlet opening in the y-dimension (1.6 mm) does not appear to proportionally impact the width of the intensity distribution. The ion plume is roughly symmetrical in both <i>x</i>- and <i>y</i>-dimensions; <a href="javascript:void(0);">(8)</a> therefore, it is expected that the smaller inlet width in the <i>x</i>-dimension would have a higher probability of being in the high density region of the plume than in the <i>y</i>-dimension. Proportional to inlet opening, the <i>x</i> position of the emitter would consequently have less of an effect on the intensity than the <i>y</i> position. We note the intensity distributions appear to be slightly off-center; however, we find the centroids of the distributions (−0.1 and 0.3 mm, <i>x</i>- and <i>y</i>-respectively, <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/jasms.5c00441/suppl_file/js5c00441_si_001.pdf">Figure S7</a>) were within the tolerance of the micrometer. <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/jasms.5c00441#fig1">Figure 1</a>E presents the ion intensities as a function of emitter position in the <i>z</i>-dimension. Impressively, signal continues to be observed up to ∼7 mm removed from the inlet at levels close to 50% of the highest. Not surprisingly, the highest signal is observed at the closest position, but, as noted above, that signal gradually declines with increasing distance. The relative intensities of these two peptide ions remain fairly constant at all observed z positions (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/jasms.5c00441/suppl_file/js5c00441_si_001.pdf">Figure S8</a>). Overall, identical trends were observed for both peptides over all dimensions. As these peptides are fairly high signal intensity, we wanted to confirm that the trends reported here were not biased by signal saturation, so we extracted signal intensities for low intensity features and compared the <i>x</i>, <i>y</i>, and <i>z</i> intensity distributions (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/jasms.5c00441/suppl_file/js5c00441_si_001.pdf">Figure S9</a>). Specifically, within 1–2 mm in any dimension, one can achieve reasonably consistent and robust signal. Distinct intensity profiles we observed here for the <i>x</i>- and <i>y</i>-dimensions likely arise from the asymmetric shape of the ion capillary. We demonstrate improved robustness of signal intensity to <i>x</i>/<i>y</i> variation at increasing <i>z</i> distances, an observation that will be helpful for positioning multiple emitters, for example. <a href="javascript:void(0);">(19−23)</a></p><p>We provide evidence that the effect of the emitter position on signal intensity does not strongly depend on the analyte for bottom-up proteomics analysis. This report provides insight into the role of emitter positioning on signal intensity for bottom-up proteomics and represents the first characterization of the effect of emitter position on electrospray signal intensity on an instrument with an inlet capillary lacking radial symmetry. Future areas of interest would be an examination of how trends vary across different flow rates or emitter types, as well as assessing how the exact intensity profiles vary across instruments with differences in their atmospheric interface.</p></div>
