<p><strong>27. ročník Školy hmotnostní spektrometrie odstartoval již v neděli 20. září blokem krátkých kurzů zaměřených na praktickou interpretaci EI hmotnostních spekter, troubleshooting LC-MS a publikování vědeckých prací z pohledu editora. V pondělí dopoledne pokračoval odborný program dalšími kurzy věnovanými interpretaci EI spekter, vícerozměrné analýze dat a necílené analýze environmentálních kontaminantů. Odpoledne byl 27. ročník oficiálně zahájen a následovaly zvané přednášky i sekce věnované mladým vědcům a novinkám v hmotnostní spektrometrii. Témata sahala od hledání stopových analytů a necíleného screeningu přes nové přístupy v DIA a lipidomiku karcinomu prostaty až po vývoj hmotnostní spektrometrie a cílené i necílené workflow pro analýzu PFAS. 27. Škola MS 2026 - den 1

<p><strong>Druhý den </strong><a target="_blank" rel="noopener noreferrer" href="https://labrulez.com/cs-cz/all/events/930"><strong>27. Školy hmotnostní spektrometrie</strong></a><strong>, úterý 22. září 2026, se dopoledne zaměřil na klíčové kroky předcházející samotné analýze. Program otevřela sekce věnovaná designu studie a přípravě vzorku, kde </strong><a target="_blank" rel="noopener noreferrer" href="https://labrulez.com/cs-cz/lcms/articles?search=david+friedeck%C3%BD"><strong>David Friedecký</strong></a><strong> rozebral otázku optimálního počtu vzorků a Ivan Petřík praktický vývoj protokolu přípravy biologických vzorků pro LC-MS pomocí statistického plánování experimentů. Následovala sekce o vývoji a validaci metod, která se věnovala stabilitě analytů a zpracování tkání včel, detailům rozhodujícím při vývoji LC-MS metod i rozvoji platformy </strong><a target="_blank" rel="noopener noreferrer" href="https://labrulez.com/cs-cz/all/companies/1"><strong>LabRulez</strong></a><strong> z českého centra směrem ke globální analytické komunitě. Po obědě patřil prostor plakátovým sdělením a poté se program přesunul k praktickým aplikacím hmotnostní spektrometrie.</strong></p><p>Odpolední bloky nabídly velmi široký průřez současnou praxí – od <strong>LC-MS analýzy hyaluronanu</strong>, léčiv, drog a metabolitů v komplexních environmentálních matricích až po screening pomocí <strong>timsTOF</strong> a nové chromatografické kolony. Druhá praktická sekce se věnovala mimo jiné <strong>PFAS v různých environmentálních lokalitách</strong>, identifikaci neznámých píků, kontrole kvality kratomu, <strong>LC-MS/MS charakterizaci RNA</strong> a novým možnostem hmotnostních spektrometrů Thermo Fisher Scientific. <p><strong>Akné patří mezi nejčastější kožní onemocnění. Během dospívání postihuje až 85 % lidí a u mnoha přetrvává i v dospělosti. Kromě fyzických projevů může výrazně ovlivňovat i sebevědomí. Jeho léčba bývá dlouhodobá a některé běžně používané přípravky mohou pokožku vysušovat nebo dráždit. Vědci z CEITEC Vysokého učení technického (VUT) vyvinuli speciálně upravená celulózová mikrovlákna, která v laboratorních testech účinně potlačovala bakterie způsobující akné. Vlákna se dají použít k výrobě pleťových masek. Otevírá se tak cesta k novým možnostem péče o pokožku trpící akné.</strong></p><p>Materiál tvoří <strong>mikrovlákna z acetátu celulózy obohacená o oxid zinečnatý</strong>, která jsou následně upravená kyslíkovým plazmatem. Tento oxid se pak postupně uvolňuje do pokožky, působí na ni antibakteriálně a<strong> omezuje množení bakterií spojených se vznikem akné</strong>. Na rozdíl od běžných přípravků je účinná látka navázána přímo ve vláknech a uvolňuje se postupně, což umožňuje<strong> dlouhodobější účinek</strong>.</p><p><i>„Vlákna si přitom uchovávají vysokou účinnost, a to bez přídavků jakýchkoli aditiv, které jsou běžná v&nbsp;komerčních přípravcích. Materiál obsahuje přesně to, co je potřeba pro účinný boj s akné, a žádné zbytečné materiály navíc. Výzkum prokázal, že antibakteriální účinek mají ještě minimálně rok poté, co projdou úpravou,“</i> říká<strong> Martina Říhová</strong> z týmu <i><strong>Pokročilých nízkodimenzionálních nanomateriálů</strong></i><strong> na CEITEC VUT</strong>, která výzkum vedla. Právě dlouhodobý prokázaný účinek otvírá dveře komerčnímu využití.</p><p>Do budoucna by stejný princip mohl najít využití také při léčbě dalších kožních bakteriálně komplikovaných onemocnění, jako je <strong>atopická dermatitida</strong>. Stejný fyzikální princip se využívá i při výrobě cukrové vaty – místo cukru ale v&nbsp;tomto případě vznikají jemná polymerní vlákna.</p><p>Doslova průlomová pak byla kombinace metody zvané <strong>Vapour Phase Infiltration</strong>, která vlákna obohatila o oxid zinečnatý, s následným ošetřením kyslíkovým plazmatem. To povrch vláken mírně naruší a umožní, aby se při kontaktu s vodou uvolňovaly účinné látky.</p><p><i>„Tento postup jsme u vláken z acetátu celulózy demonstrovali jako<strong> první na světě</strong>,“</i> podotýká vědkyně, která se výzkumu akné a jeho léčby věnuje už sedm let.<strong> </strong><a target="_blank" rel="noopener noreferrer" href="https://www.researchgate.net/publication/403127590_Cellulose_Acetate_Fibers_With_Infiltrated_ZnO_Nanocrystals_Activation_of_Antibacterial_Properties_Against_Acne_vulgaris_by_Oxygen_Plasma_Treatment"><strong>Výsledky</strong></a> proto s kolegy publikovala i ve vědeckém magazínu <i><strong>Small</strong></i>, který patří mezi přední světová periodika zaměřená na nanomateriály a nanotechnologie.</p><h3><strong>Na výzkumu spolupracovali vědci z Brna a Španělska</strong></h3><p>Na vývoji materiálu se podílelo několik výzkumných týmů z různých institucí.&nbsp; <i>„Náš tým pod vedením <strong>Jana Macáka</strong> měl na starosti přípravu vláken. Poté jsme je převezli do španělského San Sebastianu, konkrétně do<strong> Nanogune k&nbsp;Mato Knezovi</strong>, kde se postarali o Vapor Phase Infiltration. Následně je <strong>tým Lenky Zajíčkové na CEITEC VUT</strong> ošetřil kyslíkovým plazmatem. A antibakteriální testování měl na starosti<strong> tým Zbyňka Hegera z Ústavu chemie a biochemie na Mendelově univerzitě</strong>,“</i> vyjmenovává spolupracovníky Martina Říhová.</p><p>Práce na výzkumu trvala přes rok. Antibakteriální účinek, založený na <strong>uvolňování iontů Zn²⁺</strong>, byl aktivován ošetřením kyslíkovým plazmatem. Vlákna CA o průměru přibližně <strong>2 µm</strong> byla připravena metodou <strong>odstředivého zvlákňování</strong> a následně infiltrována ZnO s využitím <strong>infiltrace z plynné fáze (Vapor Phase Infiltration, VPI)</strong> při různém počtu depozičních cyklů; obsah Zn dosahoval až <strong>8 hm. %</strong>. Protože jsou vlákna CA přirozeně hydrofobní, byla ošetřena kyslíkovým plazmatem, aby se stala hydrofilními a zlepšilo se smáčení pokožky. <strong>Kombinace těchto postupů je zde představena poprvé.</strong></p><p>Ošetření plazmatem vedlo ke vzniku <strong>funkčních skupin obsahujících kyslík</strong> na povrchu CA a významně zlepšilo jeho smáčivost. Analýzy upravených vláken pomocí <strong>skenovací elektronové mikroskopie (SEM)</strong> neprokázaly žádné morfologické poškození. Podrobná charakterizace vláken po <strong>32 a 128 cyklech VPI</strong>, provedená pomocí <strong>rentgenové fotoelektronové spektroskopie (XPS), Ramanovy spektroskopie, RTG-fluorescenční spektroskopie (EDXRF), rentgenové difrakce (XRD) a transmisní elektronové mikroskopie (TEM)</strong>, ukázala, že plazma povrch částečně oxidovalo a leptalo, čímž ovlivnilo distribuci ZnO.</p><p>Na rozdíl od neošetřených vláken vykazovala <strong>plazmaticky modifikovaná vlákna antibakteriální aktivitu vůči bakteriím způsobujícím akné</strong>, konkrétně <i>Cutibacterium acnes</i> a <i>Staphylococcus epidermidis</i>, a vytvářela výrazné <strong>inhibiční zóny o velikosti až 5 mm</strong>. Tyto výsledky dokládají jejich potenciál jako slibného terapeutického přístupu. Celkově ošetření plazmatem umožnilo účinnou <strong>funkcionalizaci povrchu</strong> a vedlo k přípravě antibakteriálních vláken se zlepšenými povrchovými vlastnostmi, vhodných pro využití při <strong>léčbě akné</strong>.</p>

<p><strong>Environmental matrices often present challenging interferences that may either be plasma or sample based.</strong></p>
<p>Reduction of these interferences using kinetic energy discrimination and/or collisional dissociation have been proven to be effective using single quad ICP-MS using helium as the cell gas, however, reactive gasses can be very effective in either shifting the interference away from the analyte of interest, or shifting the analyte to a new mass, away from the associated polyatomic interference.</p>
<p>Using a reactive gas on a single quad ICP-MS risks creating undesired polyatomic interferences in the collision/reaction cell on other target analyte masses.  By utilizing a quadrupole with single amu resolution before the collision/reaction cell, only the target analyte and associated interferences of the same mass enter the cell, giving the analyst ultimate control of the reaction products for a given analyte and associated interference.<br/>The optimization of reaction cell technology, combined with single unit mass resolution allows for more precise measurement of analytes in complex matrices, while also enjoying lower detection limits with tighter precision and improved accuracy.  As regulatory detection limits become increasingly lower for environmental matrices, triple quadrupole ICP-MS/MS systems with single unit mass resolution technology has proven to be a superior approach for complex sample matrices.</p>
