Noc vědy 2026 - Komunikace

Třetí den <a target="_blank" rel="noopener noreferrer" href="https://labrulez.com/cs-cz/all/events/930">27. Školy hmotnostní spektrometrie</a>, středa 23. září 2026, se dopoledne nesl ve znamení omických přístupů a metod, které posouvají možnosti běžné LC-MS analýzy. Zdeněk Spáčil otevřel program přehledem akvizičních strategií od full scanu a MS/MS přes DDA, DIA/SWATH a PRM/MRM až po HRMS, na který Jaroslava Friedecká navázala tématem společného vyhodnocování metabolomických, proteomických a lipidomických dat. Sekce „Za hranicemi rutinní MS“ pak ukázala další směry současné hmotnostní spektrometrie: Pavel Hrouzek představil využití stabilních izotopů při studiu metabolických drah, Robert Jirásko se zaměřil na iontovou mobilitu v LC/MS při analýze malých molekul a dopolední odborný blok uzavřela přednáška společnosti <a target="_blank" rel="noopener noreferrer" href="https://lcms.cz/companies/43">Shimadzu</a> věnovaná ionizační technologii <a target="_blank" rel="noopener noreferrer" href="https://labrulez.com/cs-cz/lcms/library?mainauthorItems=53&amp;search=SICRIT">SICRIT®</a>.Po intenzivním odborném programu přišlo na řadu tradiční kulturně-poznávací odpoledne. Účastníci si mohli vybrat mezi návštěvou Vyššího Brodu a tamního kláštera, výletem do Českého Krumlova spojeným s prohlídkou grafitového dolu, pěší túrou na Vítkův kámen nebo individuálním programem, jehož součástí byl také turnaj ve stolním fotbálku. 27. Škola MS 2026 - den 3

Nový grafenový inkoust pro výrobu levných a citlivých tištěných senzorů vyvinuli výzkumníci z CATRIN Univerzity Palackého ve spolupráci s kolegy z VŠB-Technické univerzity Ostrava a Univerzity Karlovy. Při přípravě inkoustu poprvé využili metodu atomárního inženýrství a díky navázaným atomům mědi jsou schopni detekovat velmi citlivě dopamin – neurotransmiter, který umožňuje přenos signálů mezi nervovými buňkami. Přístup <a target="_blank" rel="noopener noreferrer" href="https://advanced.onlinelibrary.wiley.com/doi/10.1002/advs.76796">představený v časopise Advanced Science</a> má velké šance uplatnit se v lékařské diagnostice.<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/CATRIN_Atom_po_atomu_Vedci_ukazali_novy_smer_v_lekarske_diagnostice_8a02ad6a66.webp" alt="CATRIN: Atom po atomu. Jeho širšímu uplatnění v lékařské diagnostice však dosud bránil nedostatek atomárně upravených materiálů i absence na nich založených tisknutelných inkoustů.&nbsp;Nyní se vědcům podařilo vyvinout novou třídu atomárně dopovaných grafenových materiálů a také inkoust pro přípravu citlivých tištěných senzorů. Sázka na měď se vyplatila při detekci dopaminu, jehož sledování je důležité pro biomedicínský výzkum a vývoj pokročilých diagnostických technologií některých onemocnění včetně Alzheimerovy choroby.<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/CATRIN_Atom_po_atomu_Vedci_ukazali_novy_smer_v_lekarske_diagnostice_2_97e419f039.webp" alt="CATRIN: Atom po atomu. Samotný senzor vyžaduje pouze 1,3 mikrogramu aktivního inkoustu, stojí přibližně jednu korunu a dával stabilní odezvu minimálně po dobu tří měsíců. Inkoust je navíc plně kompatibilní s běžně dostupnými inkoustovými materiálovými tiskárnami. To vytváří velký potenciál pro přípravu cenově dostupných, digitálně navržených senzorů, jejichž funkci lze přizpůsobovat atom po atomu,“ uvedl korespondující autor článku Michal Otyepka.Základem pro novou generaci atomárních inkoustů byla dusíkem dopovaná grafenová kyselina (NGA), kterou vědci z CATRIN vyvinuli před několika lety. Nyní ji postupně obohacovali různými jednotlivými atomy kovů včetně mědi, železa nebo manganu.<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/CATRIN_Atom_po_atomu_Vedci_ukazali_novy_smer_v_lekarske_diagnostice_3_c15d831792.webp" alt="CATRIN: Atom po atomu. Zatímco atomy mědi výrazně zesilují signál dopaminu, ostatní kovy jej naopak potlačují. Rozhodující&nbsp;tedy není jen samotná přítomnost izolovaných atomů kovu, ale také to, o jaký konkrétní kov se jedná. To je velmi důležitý poznatek pro další vývoj biosenzorů,“ objasnil první autor Martin-Alex Nalepa.Publikované výsledky představují novou třídu senzorických materiálů s velkým potenciálem v lékařské diagnostice. Vědci nabízí obecně využitelnou platformu pro vývoj grafenových inkoustů s jednotlivými atomy kovů a otevírají cestu ke škálovatelné, reprodukovatelné a materiálově úsporné výrobě pokročilých elektrochemických senzorů. „Jsme na začátku a fascinují nás nové možnosti, které se před námi otevírají. Vidíme, že změnou typu atomů můžeme řídit citlivost a selektivitu senzoru. Kromě dopaminu tak umíme velmi přesně stanovit například imunoglobuliny – protilátky, jejichž množství a typ mohou být důležitým ukazatelem autoimunitních nebo infekčních onemocnění,“ zdůraznil další z autorů článku Radek Zbořil.<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/CATRIN_Atom_po_atomu_Vedci_ukazali_novy_smer_v_lekarske_diagnostice_4_273cd760a5.webp" alt="CATRIN: Atom po atomu. Jejich klíčovou součástí jsou funkcionalizované elektrody, pro jejichž výrobu se ukázaly jako velmi vhodné materiály právě na bázi grafenu. Vznikl integrací vědeckých týmů Centra regionu Haná pro biotechnologický a zemědělský výzkum (CRH), Regionálního centra pokročilých technologií a materiálů (RCPTM) a Ústavu molekulární a translační medicíny (ÚMTM) s cílem úzce propojit vědecké kapacity v oblasti biomedicíny, nanotechnologií, nových materiálů, výzkumu rostlin či biotechnologií a ještě úspěšněji čelit  globálním výzvám současného světa, k nimž patří například dopady klimatických změn, boj s novými typy patogenů, diagnostika a léčba civilizačních onemocnění, odvrácení hrozící potravinové krize, získávání a ukládání „zelené“ energie, zajištění dostatku pitné vody, ochrana životního prostředí a řada dalších.

This presentation will focus on our use of a RapidFire 400 High Throughput Mass Spectrometry system coupled to a triple quadrupole mass spectrometer, to build and implement a single platform for the rapid biochemical characterization of multiple biological enzyme targets.
Because of the high sample throughput compared to traditional LC-MS/MS, we can comprehensively characterize the activity of multiple enzymes across a variety of substrates including biologically relevant substrates (e.g. oligonucleosomes) and tailor our Medicinal Chemistry small molecule compound design efforts to target the enzyme activity of interest. Employing a single platform with internal redundancy, enables us to both quickly and confidently develop suites of assays within and across multiple enzyme classes, without many of the limitations present with other technologies.

This presentation will focus on our use of a RapidFire 400 High Throughput Mass Spectrometry system coupled to a triple quadrupole mass spectrometer.

Reaction analytes across a diverse group of enzyme targets

Data independent acquisition (DIA) allows for suspect screening in complex matrices with retroactive analysis capabilities important to emerging compounds of interest in food safety. However, with increased analyte scope, the review of questionable identifications can create an analysis burden requiring timely review and reinjections.
Here we present a robust DIA methodology with a novel prototype LC/Q-TOF, leveraging a combination of high-resolution, extended dynamic range, stable accurate mass, and isotopic fidelity with coeluting fragmentation ions for identification, analyzed with MH Quantitative Analysis 12.1 and LC Screener Tool.
Over 200 analytes were identified from a spiked broccoli matrix prepared with QuEChERS, resulting in good calibration curve linearity and low area %RSDs. Fused, is the Intelligent Reflex Workflow used to automatically evaluate and reinject samples in which results were flagged as questionable and/or identified, via targeted MS/MS utilizing the available m/z and RT information. Transferring questionable results into confirmed identifications by acquiring targeted data for present compounds. Leveraging the combination of DIA and automatically enabled targeted MS/MS reinjections via Intelligent Reflex allows for broad screening analysis and confident identification confirmation of analytes.

Robustness testing is a measure of a method’s ability to remain unaffected by the minor changes of chromatographic parameters. The robustness multivariable design of experiments (DoE) can be used to study the effect of individual parameters and their interaction on the chromatographic resolution between peaks.
The Empower Sample Set Generator (SSG) can be used to automatically create a sample set method, which includes instrument methods, and method sets required to perform the entire study in one run. The Effect Plots generated by the Method Validation Manager clearly show which parameters have the greatest effect on method performance and provides knowledge for establishing a set of controls to assure a method meets the expected criteria during routine use.

There are a variety of sample preparation methods used for the analysis of drugs of abuse in physiological sample matrices such as blood, urine, oral fluid.
These include simple ‘dilute and shoot’, protein removal, phospholipid removal, liquid -liquid and solid phase extraction as well as many others.
With each method there is variability in the number of steps involved, specificity, and the suitability to automation.
Choosing the most suitable sample preparation technique can depend on the type of analytical challenge being addressed, for example certain methods can be used to screen for a large number of compounds and others can be selected to enhance sensitivity for a particular compound or class of compounds. 
In this session, a number of sample preparation methods will be discussed in terms of their strengths and limitations and suitability for particular forensic toxicology applications.
