<p><strong>Mikroplasty se staly jedním z nejdiskutovanějších kontaminantů životního prostředí současnosti. Bez ohledu na to, kde žijeme, je každodenní expozice mikroplastům prakticky nevyhnutelná – tyto drobné plastové částice se nacházejí ve vzduchu, který dýcháme, ve vodě, kterou pijeme, i v prostředí, ve kterém žijeme. Vzhledem k tomu, že se vědci snaží lépe porozumět zdrojům mikroplastů, cestám jejich šíření a jejich dopadům, neustále roste poptávka po spolehlivých metodách jejich analýzy.</strong></p><p>Ačkoli se techniky, jako jsou FTIR a Ramanova spektroskopie, staly standardními nástroji pro identifikaci mikroplastů, jejich využití je omezené u menších částic (tj. nanoplastů) a ve složitých matricích. Pokročilejší metody analýzy mikroplastů mohou poskytnout hlubší informace o tom, co tyto částice obsahují a jak se mohou v průběhu času v životním prostředí měnit.</p><h3><strong>Omezení spektroskopických metod analýzy mikroplastů</strong></h3><p><strong>Infračervená spektroskopie s Fourierovou transformací </strong>(<strong>FTIR</strong>) a <strong>Ramanova spektroskopie </strong>patří mezi široce používané metody analýzy mikroplastů, protože umožňují<strong> identifikovat typy polymerů</strong> na základě interakce elektromagnetického záření s materiálem. Tyto techniky jsou užitečné pro určení, zda se jedná například o polyethylen, polypropylen, polystyren nebo jiný běžný plast. Pro mnoho studií jsou tyto informace dostačující k potvrzení přítomnosti mikroplastů, zejména u čistých vzorků s dostatečným množstvím mikroplastových částic.</p><p><a target="_blank" rel="noopener noreferrer" href="https://www.leco.com/application/environmental/"><strong>Vzorky z životního prostředí</strong></a> jsou však málokdy čisté a složením jednoduché. Částice mikroplastů mohou v důsledku působení okolního prostředí postupně stárnout a být překryty či obaleny jinými materiály, což ztěžuje jejich přesnou identifikaci pomocí spektroskopie. <span>Pokud navíc výzkumníci potřebují zjistit, jaká aditiva, degradační produkty nebo znečišťující látky jsou s daným plastem spojeny, samotná identifikace částice nestačí. Metody FTIR a Ramanovy spektroskopie proto mají omezené možnosti při podrobné charakterizaci přítomných chemických látek, zejména v komplexních environmentálních matricích.</span> S rostoucím významem otázek spojených s expozicí mikroplastům proto výzkumníci potřebují analytické techniky, které dokážou podrobněji charakterizovat chemické složení vzorku, a nikoli pouze potvrdit přítomnost samotné plastové částice.</p><h3><strong>Termální desorpce a pyrolýza s GCxGC-TOFMS pro komplexnější charakterizaci mikroplastů</strong></h3><p><span>Pro podrobnější charakterizaci nano- a mikroplastů ve složitých vzorcích lze využít termální <strong>desorpci </strong>(<strong>TD</strong>) a <strong>pyrolýzu </strong>(<strong>Py</strong>) ve spojení s komplexní dvourozměrnou plynovou chromatografií a hmotnostní spektrometrií s analyzátorem doby letu (</span><a target="_blank" rel="noopener noreferrer" href="https://www.leco.com/core-analysis/gc-and-gc-ms/"><span><strong>GCxGC-TOFMS</strong></span></a><span>). Termální desorpce a pyrolýza poskytují vzájemně se doplňující informace o chemickém složení vzorku.</span></p><p><strong>Termální desorpce </strong>umožňuje analyzovat těkavé a adsorbované látky spojené s částicemi mikroplastů. Patří mezi ně například environmentální kontaminanty, degradační produkty nebo těkavé organické látky (VOC). Analýza tak poskytuje informace o chemických látkách, které se nacházejí na povrchu plastu nebo jsou v něm obsaženy.</p><p><strong>Pyrolýza </strong>se naopak zaměřuje na samotný polymer. Působením vysoké teploty jej rozkládá na charakteristické sloučeniny, které lze následně analyzovat pomocí GC. Tyto produkty pyrolýzy slouží jako markery umožňující určit, jaké polymery jsou ve vzorku přítomny. Tento přístup je zvláště užitečný u komplexních nebo výrazně zestárlých mikroplastů, které se pomocí běžných spektroskopických metod identifikují obtížně.</p><p>Nejvýznamnější přínos přináší spojení těchto technik s komplexní<strong> dvourozměrnou plynovou chromatografií</strong>. Environmentální vzorky často obsahují stovky až tisíce sloučenin, které mohou v jednorozměrné GC-MS koeluovat, a jejich jednotlivá identifikace je tak obtížná nebo dokonce nemožná. GCxGC poskytuje výrazně vyšší separační schopnost a pomáhá analytikům odlišit důležité polymerní markery od složek matrice. Lepší separace zároveň zvyšuje schopnost hmotnostního spektrometru identifikovat charakteristické produkty pyrolýzy polymerů, a to i ve vysoce komplexních environmentálních vzorcích. Výzkumníci tak mohou detekovat a identifikovat i obtížně analyzovatelné mikroplasty bez rozsáhlých kroků čištění vzorku, které často vyžadují jiné analytické postupy.</p><p>Další jedinečnou výhodou tohoto přístupu je možnost použít teremální desorpci a pyrolýzu postupně na stejném vzorku. Společně tak poskytují komplexnější chemický profil vzorku.</p><h3><strong>Komplexní analytický postup pro lepší pochopení mikroplastů v životním prostředí</strong></h3><p>FTIR a Ramanova spektroskopie zůstávají cennými nástroji pro rychlý screening a identifikaci vzorků obsahujících mikroplastové částice. <strong>Kombinací teremální desorpce, pyrolýzy a GCxGC-TOFMS</strong> však mohou analytici získat<strong> mnohem komplexnější informace o plastech </strong>přítomných ve vzorcích, jejich chemickém složení a změnách, kterým v životním prostředí podléhají. S postupujícím environmentálním výzkumem a rostoucími obavami z kontaminace mikroplasty budou výzkumníci stále častěji potřebovat informace, které přesahují pouhou odpověď na otázku: <i>„Jaký plast je přítomen?“</i></p><p>Tato hlubší úroveň charakterizace může v konečném důsledku přinést nové poznatky o osudu mikroplastů v životním prostředí a pomoci vědcům lépe porozumět jejich potenciálním dopadům na ekosystémy a expozici člověka.</p>

Překonání limitů tradičních metod analýzy mikroplastů pomocí termální desorpce a pyrolýzy ve spojení s GCxGC-TOFMS

<p><strong>27. ročník Školy hmotnostní spektrometrie odstartoval již v neděli 20. září blokem krátkých kurzů zaměřených na praktickou interpretaci EI hmotnostních spekter, troubleshooting LC-MS a publikování vědeckých prací z pohledu editora. V pondělí dopoledne pokračoval odborný program dalšími kurzy věnovanými interpretaci EI spekter, vícerozměrné analýze dat a necílené analýze environmentálních kontaminantů. Odpoledne byl 27. ročník oficiálně zahájen a následovaly zvané přednášky i sekce věnované mladým vědcům a novinkám v hmotnostní spektrometrii. Témata sahala od hledání stopových analytů a necíleného screeningu přes nové přístupy v DIA a lipidomiku karcinomu prostaty až po vývoj hmotnostní spektrometrie a cílené i necílené workflow pro analýzu PFAS. <p><strong>The United States Pharmacopeia (USP) is the only major pharmacopeia with a general chapter dedicated to analytical instrument qualification (USP &lt;1058&gt;). This makes &lt;1058&gt; a globally significant regulatory reference. However, despite the significant changes with the 2017 edition of &lt;1058&gt;, 68% of laboratories have not updated their qualification procedures. Additionally, the USP has already started the process to initiate updates to &lt;1058&gt;, when many laboratories may not fully comply with the current edition!</strong></p>
<p>This raises many questions about the role of the current &lt;1058&gt; general chapter and how it aligns with the recently issued 2nd Edition of GAMP® 5.</p>

<p><strong>Raman spectroscopy is used around the globe for identification of unknowns, verification of raw materials, basic R&amp;D, and process monitoring and control. The most commonly deployed technology is based on 785 nm lasers and detectors that leverage innovations from the telecommunications boom in the 1990s. However, this technology has been fundamentally limited by fluorescence interference that overpowers the Raman signal. The most common ways to overcome fluorescence have all had significant drawbacks around cost, safety and sample preparation – until now.</strong></p>
<p>Developed for first responders to help identify unknowns in street narcotics, eXTRaction (XTR) technology uses advanced digital signal processing to separate Raman data from a background. With XTR, 785 nm Raman can be extended to difficult-to-analyze systems.</p>
<p>In this webinar, Dr. Adam J. Hopkins and Elena Hagemann will explain how XTR works and show different use cases for both qualitative and quantitative measurements, including biodiesel and polyurethane production.</p>
