Antibody-based therapeutics are becoming increasingly complex, creating new challenges for obtaining detailed sequence and structural information across critical regions of these molecules.

Join Luca Fornelli, PhD, Associate Professor at the University of Oklahoma, as he explores middle-down and native top-down mass spectrometry approaches for the characterization of antibody-based therapeutics using the Thermo Scientific™ Orbitrap™ Tribrid™ Apex BioPharma Mass Spectrometer.

Luca will show how infrared-assisted electron transfer dissociation (IR-ETD) and electron transfer/higher-energy collision dissociation (IR-EThcD) can be combined with proton transfer charge reduction (PTCR) to generate detailed sequence information from complex biologics. The talk will discuss denaturing and native conditions, automated data analysis, Fab fragments, and intact monoclonal, bispecific, and trispecific antibodies.

Who should attend?

Scientists and researchers involved in biopharmaceutical characterization and development, including monoclonal and multispecific antibodies, middle-down and top-down MS, native MS, biotherapeutic sequence and structural characterization, and advanced fragmentation workflows.

What you can expect to learn
  • How middle-down and native top-down MS provide complementary information
  • How IR-ETD, IR-EThcD, and PTCR can improve fragmentation and simplify spectra
  • Approaches for increasing sequence information from disulfide-protected regions and CDRs
  • How advanced fragmentation can be applied to subunits, Fab fragments, and intact therapeutics
  • How automated data analysis can streamline interpretation
