Tosoh Tuesday - Continuous Multi-Column Chromatography for Purification of Inter-alpha Inhibitor Proteins from Human Plasma

On September 29, Abbie Breton, Applications Development Manager, will present the next edition of Tosoh Tuesday, a webinar series held by Chromatography Experts. Yow-Pin Lim, Founder and CEO of ProThera Biologics, and Jennifer Knister, Commercial Product Manager - Process Solutions at Tosoh Bioscience, will showcase a robust and scalable purification process of Inter-alpha Inhibitor Proteins (IAIP), plasma proteins that contribute to innate immunity and regulate the host inflammatory response. 

Circulating IAIP levels decline markedly during severe systemic inflammation and sepsis, supporting their potential use as host-response biomarkers. Preclinical studies also demonstrate that administration of exogenous IAIP significantly improves survival in models of neonatal sepsis, resulting in the development of IAIP as a replacement therapy intended to restore immune balance.

A two-step chromatographic process has been developed using an anion-exchange resin for IAIP capture, followed by an affinity-mimetic polishing step.

This webinar will describe the development of the IAIP capture process using TOYOPEARL GigaCap Q-650M. We will then present the transition from batch chromatography to continuous multi-column chromatography (MCC) using the Octave BIO instrument and discuss how this approach can improve process efficiency while reducing buffer consumption, equipment footprint, and hands-on time. 

By using ProThera Biologics’ IAIP purification program as a case study, participants will learn how about the development of scalable purification processes for complex, difficult-to-purify biologics.
