Part 4: Armor CAR T Cells with Enhanced Potency for Cancer Immunotherapy

The success of CAR T cell immunotherapy depends on the differentiation status and metabolic fitness of the CAR T cell product. Our research challenges conventional CAR T cell therapy protocols by demonstrating that functional CAR T cells can be rapidly generated within 24 hours without the need for activation or ex vivo expansion, preserving their maximal fitness. By harnessing the intrinsic stemness of non-activated CAR T cells and co-expressing specific cytokines to enhance anti-tumor efficacy, we aim to elevate metabolic and effector functions, thereby improving long-term persistence and therapeutic impact. These findings offer promising avenues to enhance the efficacy and accessibility of CAR T cell therapy for cancer patients.
